Structural modification of a natural angiotensin converting enzyme inhibitor and the effect on activity.
Includes bibliographical references (leaves 50-54).54 leaves : illustrationsHypertension is a leading risk factor for mortality that affects nearly a third of adult US residents, and it is primarily combatted through the use of angiotensin converting enzyme inhibitors (ACEIs). In addition to ACEIs currently approved as anti- hypertension drugs, there are a number of natural ACEIs that have not been studied, some of them more effective than others. This study sought to alter the structure of alanine- valine-phenylalanine (AVF), a natural, relatively ineffective ACEI, to create modified peptides that would hopefully act as more effective inhibitors. After a literature analysis, phenylalanine-valine-alanine (FVA) was determined to be more likely to act as an effective inhibitor, so four novel peptides were synthesized. One novel peptide was designed to contain unmodified phenylalanine while the other three contained one of the following para-substituted, N-protected, L-phenylalanine amino acids: L-Phenylalanine, N-benzoyl-4-methyl, L-Phenylalanine, N-benzoyl-4-methoxy-, L-Phenylalanine, N- benzoyl-4-fluoro-. In implementing these peptide modifications, first the respective modified phenylalanine amino acids were synthesized, then the FVA peptides were synthesized, with naturally occurring phenylalanine as well as the three modified phenylalanine amino acids. Using eclipses to probe physical conditions along the jet in SS433.
Includes bibliographical references (leaves 94-99).vi, 99 leaves : illustrationsThe Galactic X-ray binary SS 433 is the only known astrophysical object to exhibit strong relativistically red- and blue-shifted lines from elements such as S, Si, Fe, Ni. The X-ray emission lines originate in a jet outflow that is launched somewhere very close to the compact accretion (a black hole or a neutron star). During 2018 August 10-14, SS433 was observed using the High Energy Transmission Grating Spectrometer system on the Chandra X-ray observatory. A total of 116 ksec of Chandra HETGS observation was made during 2018 August 10-14, which was split in one 20 ksec and one 96 ksec observation. The 20 ksec observation started at an orbital phase of 0.802, three days before an eclipse and the 96 ksec observation started at an orbital phase of 0.109 in the middle of the eclipse. The observations were designed to take advantage of the eclipse and carry out time-resolved spectroscopy and timing studies to infer spatial variation of physical properties such as composition, temperature, and density at different distances among the jet. In addition to phenomenological fits to determine properties of the observed emission lines, we will present results from fitting collisionally ionized plasma models. The observation indicates that the redshift of the western jet was 0.034 while the predicted redshift value is -0.0097. This suggests the independent motion of the two jets. Chronic exposure to methylmercury adversely affects response to oxidative stress.
Includes bibliographical references (leaves 22-28).ii, 34 leaves : illustrationsMethylmercury (MeHg) is an environmental neurotoxicant that has been shown to lead to physiological inefficiencies. Environmental contamination is a major concern, due to the high amounts of mercury pollution that affects a variety of food webs. Chronic exposure to MeHg has been shown to lead to the generation of reactive oxygen species (ROS) and inhibition of cell defense systems. The specific activities of three antioxidant enzymes, thioredoxin reductase (TrxR), glutathione peroxidase (GPx), and glutathione reductase (GR) are altered after chronic exposure to MeHg. Prior studies have shown generally, after MeHg exposure, the activity of TrxR and GPx decrease, while GR activity increases. These data imply that the glutathione system upregulates in the presence of MeHg, compensating for the inhibited activity of TrxR. This current study used kinetic spectroscopic methods to measure the activity and concentration of TrxR, GPx, and GR in Fundulus Heteroclitus exposed to MeHg in food at a rate of 35% of body weight per day for 3.5 weeks. We hypothesized that chronic exposure to MeHg lowers the activities of TrxR and GPx, and increases the activity of GR in brain and liver tissue. Results demonstrate that TrxR activity is unaffected after MeHg exposure in the liver of F. heteroclitus, whereas GPx and GR activity significantly decreased. Both TrxR and GPx activity decreased in the brain following MeHg treatment, while GR activity decreased only in males and remained unaffected in females. Analysis of ciliary gene expression in sea urchin development.
Includes bibliographical references (leaves 61-63).72 leaves : illustrationsCilia are important cellular organelles tied to many disease states due to their pivotal roll in important cellular processes, including early organism development. In this Honors Thesis, cilia were studied in developing Strongylocentrotus purpuratus embryos during the blastula stage of development. The blastula stage is when the sea urchin embryos develop motile cilia. The upstream DNA regions of 37 ciliary genes expressed during the blastula stage of development were analyzed using a motif discovery algorithm MEME and further analyzed with a motif comparison algorithm TOMTOM, in search of shared regulatory elements related to transcription factor binding proteins. This experiment resulted in three promising motifs aligned with known transcription factors Klf6, Klf11, Klf2, and Glis-3. Our data supports a model were Klf6, Klf11, and Klf2 may be transcriptional enhancers of the blastula cluster genes while Glis-3 may be a transcriptional repressor.
