    Agency und Partizipation in interaktiven Spielstrukturen : Analyse von Interaktionsebenen und Grenzverschiebungen im digitalen Gamedesign

    Digitale Spiele sind keine abgeschlossenen Medienprodukte mehr, sondern offene, dynamische Systeme, in denen Gestaltung, Rezeption und Weiterentwicklung eng miteinander verflochten sind. Diese Arbeit untersucht, wie Agency als Handlungsmacht im Spiel entsteht und wie daraus Formen von Partizipation hervorgehen, die über das Medium hinausreichen. Zentral ist dabei die Unterscheidung zwischen Interaktivität innerhalb des Spiels und Partizipation im kulturellen Umgang mit dem Medium. Die Analyse gliedert sich in drei Ebenen: Die Produktionsebene betrachtet Entscheidungsräume und Spielstrukturen, die Agency ermöglichen. Die Konsumebene analysiert Jenkins’ Konzept der Participatory Culture anhand von Praktiken wie Modding, Beta-Testing und Community-Diskursen. Die dritte Ebene beschreibt Rollenverschiebungen, bei denen Konsument:innen zu Prosument:innen oder Co-Produzent:innen werden. An Beispielen wie The Sims, Minecraft, Episode oder Mass Effect wird deutlich, dass partizipative Spielkulturen vielfältige Ausdrucksformen annehmen. Die Analyse zeigt, dass diese Prozesse nicht beliebig verlaufen, sondern von Spielstruktur, Zugänglichkeit und ökonomischen Interessen abhängig sind. Digitale Spiele sind unter diesem Verständnis nicht mehr als abgeschlossene Produkte zu begreifen, sondern als Medien, die sich im kontinuierlichen Dialog mit ihren Nutzer:innen entwickeln und verändern und deshalb niemals vollständig abgeschlossen sind.Digital games can no longer be understood as finished media products but as open, dynamic systems in which design, reception, and ongoing development are closely interconnected. This thesis examines how agency as a form of player empowerment emerges within games and how it gives rise to forms of participation that extend beyond the medium itself. Central to this is the distinction between interactivity within the game and participation as a cultural engagement with the medium. The analysis is structured across three levels: The production level explores decision-making spaces and game structures that enable agency. The consumption level discusses Jenkins’ concept of participatory culture through practices such as modding, beta testing, and community discourse. The third level addresses shifts in roles, where consumers become prosumers or co-producers. Case studies such as The Sims, Minecraft, Episode, and Mass Effect illustrate the diverse expressions of participatory game cultures. The analysis shows that these processes are not arbitrary but are shaped by game structure, accessibility, and economic interests. Under this understanding, digital games can no longer be regarded as closed or linear products, but rather as media that evolve and transform through ongoing dialogue with their users and therefore can never be considered truly complete.Masterarbeit Universität Innsbruck 202

    Neoliberale ideologische Überzeugungen und Subjekterleben bei Jugendlichen kurz vor dem Berufseinstieg : Eine Kritische Diskursanalyse

    Der Neoliberalismus gilt als primäres Wirtschafts- und Gesellschaftssystem der westlichen Welt und geht mit ideologischen Überzeugungen in Bezug auf Arbeit und Leistung einher. Negative Auswirkungen des neoliberalen Systems werden über neoliberale ideologische Überzeugungen (NLI) gerechtfertigt. Bereits im Jugendalter, das als besonders stressexponierte Lebensphase gilt, lassen sich internalisierte NLI verzeichnen. Die vorliegende Studie untersucht NLI im Jugendalter, um einen Beitrag zur Erforschung ihrer Internalisierung zu leisten, mit einem zusätzlichen Fokus auf das Subjekterleben im Angesicht des anstehenden Berufseinstiegs. Mit 10 Jugendlichen erfolgte eine umfassende Erhebung von NLI unter Verwendung eines Interviews, eines Fragebogens und eines selbstkonstruierten projektiven Testverfahrens. Die Ergebnisse wiesen auf internalisierte NLI bei den Jugendlichen hin. Insbesondere Individualismus war über alle Testverfahren hinweg ausgeprägt. Es ließen sich auch Hinweise auf ein neoliberales Subjekterleben feststellen. Die TeilnehmerInnen zeigten sowohl autonomes als auch ohnmächtiges Subjekterleben im Angesicht des Berufseinstiegs. Die Ergebnisse wurden in Bezug auf die bisherige Studienlage diskutiert und insbesondere palliative und widerspruchsauflösende Funktionen von Ideologie in Erklärungsansätzen erläutert. Eine weitere Erforschung von NLI und Subjekterleben im Jugendalter wird empfohlen. Die Verwendung des projektiven Testverfahrens soll Anstoß geben, Instrumente zur Erhebung von NLI methodisch zu erweitern und auch unbewusste Komponenten des Konstrukts zu erfassen.Neoliberalism is considered the primary economic and social system of the Western world and is accompanied by ideological beliefs regarding work and merit. Negative effects of the neolib-eral system are justified by neoliberal ideological beliefs (NIB). Already in adolescence, which is considered a particularly challenging phase of life, internalised NIB can be observed. This study examines NIB in adolescence in order to contribute to research on its internalisation, with an additional focus on subject experience in the face of the upcoming entry into working life. A comprehensive exploration of NIB was conducted with 10 adolescents using an inter-view, a questionnaire and a self-constructed projective test. The results indicated internalised NIB among the adolescents. Individualism in particular was highly pronounced across all test procedures. There were also indications of a neoliberal subject experience. The participants showed both autonomous and powerless subject experiences in the face of entering the job market. The results were discussed in relation to previous studies and the palliative functions of ideology. Further research into NIB and subject experience in adolescence is recommend-ed. The use of projective testing is intended to promote the methodological expanding of in-struments for assessing NIB and also capturing unconscious components of the construct.Masterarbeit Universität Innsbruck 202

    Genetic perturbation of stem-cell derived human in vitro brain models using CRISPRi

    Die Unzugänglichkeit des menschlichen Gehirns hat den Fortschritt der Neurowissenschaften im Verständnis der menschlichen Neuroentwicklung behindert, die im komplexesten und am stärksten vernetzten Gehirn gipfelt, das in der Natur beobachtet werden kann. Tierstudien haben zwar wichtige genetische, biologische und umweltbedingte Mechanismen identifiziert, diese sind jedoch oft nicht direkt auf den Menschen übertragbar. Um dieses Problem zu lösen, setzten wir modernste Technologien ein, darunter aus Stammzellen gewonnene menschliche Modelle und genetische Perturbation. Eine CRISPRi-Strategie wurde etabliert und in menschlichen In-vitro-Modellen validiert, die frühe neurologische Entwicklungsereignisse wie die Bildung neuraler Stamm-/Progenitorzellen (NSPC) und die spätere Neurogenese unter Verwendung zerebraler Organoide (COs) abdeckt. Induzierte pluripotente Stammzellen (iPSCs), die die KRAB-dCas9-Maschinerie enthielten, wurden einer Qualitätskontrolle durch Analyse wichtiger Oberflächen- und Kernmarker unterzogen. Die Induzierbarkeit von Doxycyclin (DOX) wurde mittels Durchflusszytometrie für mCherry-Fluoreszenz und qPCR für KRAB-dCas9-mRNA-Spiegel verifiziert. Die abgeleiteten Zelltypen wurden getestet, gefolgt von einer Analyse der Oberflächenmarker und der KRAB-dCas9-Expression nach der Ableitung. Nach der Etablierung wurde CRISPRi eingesetzt, um die Rolle von FOXO3, einem Transkriptionsfaktor (TF), der an mehreren zellulären Prozessen beteiligt ist, die die menschliche Langlebigkeit beeinflussen, und PRTG, einem transmembranproteinkodierenden Gen, zu untersuchen. Beide sind in menschlichen neuronalen Modi wenig erforscht. Der FOXO3-Knockdown (KD) in neuralen Vorläuferzellen (NPCs) führte zu einem signifikanten Rückgang der mitophagiebezogenen Gene LC3B, PINK1 und des neurogenesebezogenen Gens ASCL1. Mitochondriale Untersuchungen zeigten Beeinträchtigungen der Atmung und Unterschiede in der mitochondrialen Morphologie in FOXO3-defizienten Zellen. Bezüglich PRTG zeigten 2D- und 3D-Modelle der neurologischen Entwicklung eine Abweichung in den rosettenartigen Strukturen in PRTG-defizienten Zellen. Diese Daten enthüllten einen bisher unbekannten Einfluss von PRTG auf die menschliche Neuroentwicklung, wie beispielsweise die Verdickung des Neuralrohrs. Zusammenfassend ermöglicht unser System die kontrollierbare und zeitgesteuerte Aufklärung intrinsischer Faktoren, die die NSPC-Proliferation und -Differenzierung steuern. Dies könnte mit Hochdurchsatz-Screening-Technologien kombiniert werden. Dieser Ansatz hat das Potenzial, die Krankheitsmodellierung, die regenerative Medizin und die Alterungsforschung maßgeblich zu beeinflussen.The inaccessibility of the human brain has hindered the advancement of neuroscience in understanding human neurodevelopment, which culminates in the most complex and highly connected brain observed in nature. While animal studies have identified key genetic, biological, and environmental mechanisms, they often lack direct applicability to humans. To address this, we employed cutting-edge technologies, including stem cell-derived human models and genetic perturbation. A CRISPRi strategy was established and validated in human in vitro models, covering early neurodevelopmental events like neural stem/progenitor cell (NSPC) generation and later neurogenesis using cerebral organoids (COs). Induced pluripotent stem cells (iPSCs) containing the KRAB-dCas9 machinery underwent quality control by analyzing key surface and nuclear markers. Doxycycline (DOX) inducibility was verified through flow cytometry for mCherry fluorescence and qPCR for KRAB-dCas9 mRNA levels. Derived cell types were tested, with a post-derivation analysis of surface markers and KRAB-dCas9 expression. Once established, CRISPRi was used to investigate the roles of FOXO3, a transcription factor (TF) involved in multiple cellular processes impacting human longevity, and PRTG, a transmembrane protein coding gene, both understudied in human neural modes. FOXO3 knock-down (KD) in neural progenitor cells (NPCs) led to a significant decrease in mitophagy-related genes LC3B, PINK1, and neurogenesis-related gene ASCL1. Mitochondrial-related assays revealed impairments in respiration and mitochondrial morphology differences in FOXO3-deficient cells. Concerning PRTG, 2D and 3D neurodevelopmental models showed an aberration in rosette-like structures in PRTG-deficient cells. This data revealed previously unknown PRTG influence in human neurodevelopment such as thickening of the neural tube. In summary, our system allows the elucidation of intrinsic factors governing NSPC proliferation and differentiation in a controllable and timed manner, which could be paired with high-throughput screening technologies.     Low temperature reaction kinetics of small negative ions with atomic and molecular hydrogen

    This thesis deals with the interaction between small atomic and molecular anions and the most abundant species in our universe, molecular and atomic hydrogen. All presented results provide temperature dependent reaction rate coefficients measured in a 16-pole radio frequency wire ion trap. The first major part of the thesis details the three-body reaction mechanisms between H2 and the two small negative ions Cl– and C2 – . Through the measurements of the chlorine-H2 system we were able to provide a proof-of-principle measurement for our newly installed 16-pole ion trap. We confirmed the efficient thermalization of the ions translational temperatures at buffer gas temperatures above 16 K. The determined reaction rate coefficient has a temperature dependence of ??−0.98(9). Additionally, we observe an, as of now, unexplained backwards reaction depending on the cube of the hydrogen density, which leads to the dissociation of the formed Cl– (H2) complexes. The second system details the reaction between H2 and C2 – , which leads to the formation of a new chemical bond through the product C2H– . From our experimental results we were able to extract a three-body reaction rate coefficient. A comparison with ab initio calculations confirmed that only a three-body mechanism reproduces the experimental results with very good agreement. The second major part of this thesis entails the first temperature dependent measurements of the associative detachment reaction between atomic hydrogen and astrochemically relevant cyanopolyyne and carbon chain anions. These reactions constitute a major destruction channel for negative ions in parts of the interstellar medium. The measurements with our 16-pole ion trap were undertaken in the temperature regime between 7 K and 296 K. Our results for cyanopolyynes with H indicate a negative temperature dependence for the reaction rate coefficients. Additionally, the CN– plus H coefficients show an unexpected modulation near 150 K. We propose our new temperature dependent results should be incorporated into astrochemical databases for more accurate results of chemical ISM modeling. The results of the reactions between H atoms and small bare carbon chain anions and hydrocarbon anions do not show significant changes with temperature. Except for C2H– + H, where below 50 K, we see an interesting change in the rate coefficient of about a factor of 2.     Hintergrund Der Myokardinfarkt (MI) stellt eine große Belastung für die Gesundheit dar, da er zu Herzinsuffizienz mit erhöhter Mortalität und Morbidität führt. Das Herz von erwachsenen Säugetieren kann sich nach einer hypoxischen Schädigung nicht ausreichend regenerieren. In einem neonatalen Maus-MI-Modell, das durch die Ligatur der Vorderwandarterie (LAD) ausgelöst wird, konnte jedoch eine vollständige Regeneration des Herzens nachgewiesen werden. Ähnliche Beobachtungen wurden auch bei anderen Säugetieren, sowie bei menschlichen Neugeborenen gemacht. In dieser Thesis wird die entscheidende Rolle von Igf1r für die Regenerationsfähigkeit des neonatalen Mausherzens dargestellt. Methoden Igf1r wurde spezifisch in Kardiomyozyten von C57BL6/J Mäusen am Tag der Geburt (P0) mit Hilfe von shRNAmirs, transportiert durch rekombinante adeno-assoziierten Viren (rAAV), reduziert (KD) und mit Renilla KD Kontrollen verglichen. Am ersten Tag nach der Geburt (P1) wurden die Mäuse entweder einer SHAM- oder einer LAD-Ligationsoperation unterzogen. Die Herzfunktion und die Regeneration wurden einen Tag nach Induktion des Herzinfarkts (dpi) und 21 dpi echokardiografisch untersucht und die Herzen anschließend weiter mittels Histologie und Immunoblotting analysiert. Ergebnisse Die LAD-Ligatur führte zu einem signifikanten MI, wie die deutlich reduzierte linksventrikuläre Auswurffraktion (EF) 1 dpi zeigte. Beide SHAM-Gruppen wiesen 21 dpi vergleichbare funktionelle Herzparameter auf. Somit weist der alleinige KD von Igf1r ohne Ligatur der LAD keine Auswirkungen auf das Wachstum und die Entwicklung des Herzens auf. Im Gegensatz dazu präsentierten Igf1r KD-Mäuse nach LAD-Ligatur eine anhaltend signifikant reduzierte EF. Die histologische Analyse demonstrierte eine deutliche Fibrosierung sowie eine reduzierte Karyokinese in den Igf1r KD LAD-Herzen. Immunoblotting zeigte signifikante Unterschiede in der Phosphorylierung von AKT. Schlussfolgerung Während Igf1r KD oder Kontroll-KD durch shRNAmirs transportiert durch rAAVs die physiologische Herzentwicklung nicht verändert, beeinträchtigt der Igf1r KD die neonatale Herzregeneration nach ischämischer Schädigung durch eine LAD-Ligation deutlich.Background Myocardial infarction (MI) represents a major health burden by leading to heart failure with increased mortality and morbidity. The heart of adult mammals cannot regenerate sufficiently after hypoxic damage. However, complete cardiac regeneration was demonstrated in a neonatal mouse model of MI induced by ligation of the left anterior descending artery (LAD). Similar observations in other mammals, as well as in newborn human babies were made. This thesis highlights the crucial role of Igf1r on the regenerative capacity of neonatal mouse hearts. Methods Igf1r was specifically knocked down (KD) in cardiomyocytes of C57BL6/J postnatal day one (P0) mice using recombinant adeno-associated virus (rAAV) delivered shRNAmirs and compared to Renilla KD controls. On the first day after birth (P1) mice underwent either SHAM or LAD ligation surgery. Cardiac function and regeneration were subsequently assessed by echocardiography one day post injury (dpi) and 21 dpi followed by further analysis by histology and immunoblotting. Results LAD ligation resulted in significant MI as shown by markedly reduced left ventricular ejection fraction (EF) 1 dpi. 21 dpi both SHAM groups, independent of the Igf1r KD status, displayed comparable functional cardiac parameters, proving no effect of Igf1r KD on neonatal cardiac growth and development without ligation of the LAD. In contrast, Igf1r KD after LAD ligation presented sustained significantly reduced LVEF compared to the controls. Histological analysis revealed marked fibrosis as well as reduced cardiomyocyte karyokinesis in the Igf1r LAD group. Furthermore, immunoblotting revealed significant differences in AKT phosphorylation. Conclusion Whereas Igf1r KD, or control KD, by shRNAmirs delivered by rAAVs does not alter physiologic cardiac development, Igf1r KD markedly impairs neonatal cardiac regeneration following ischemic damage induced by LAD ligation surgery.eingereicht von Dr. med. univ.     The integrity of the EU’s strategic communication : how trustworthy is the EU’s strategic communication?

    This thesis critically examines the trustworthiness and legitimacy of the European Union’s (EU) strategic communication, particularly in the context of the European Green Deal and related cases: the inclusion of nuclear energy and natural gas in the EU Taxonomy, the Regulation on Sustainable Use of Pesticides (SUR), and the Carbon Border Adjustment Mechanism (CBAM). Drawing on the theories of neorealism and ethical neorealism, the analysis explores whether EU communication serves as a genuine vehicle for ethical values or functions primarily as an interest-driven, potentially propagandistic tool. Employing both qualitative and quantitative methods—including discourse analysis and a categorization framework distinguishing ethical, selective, and deceptive communication—the study characterizes the EU’s messaging strategies, stakeholder engagement, and alignment between communicated values and actual decision-making. The findings reveal a complex picture: while the SUR illustrates value-driven, participatory communication, both the EU Taxonomy and CBAM cases demonstrate selective communication, framing, and evidence use that prioritize institutional interests over ethical norms. Notably, the analysis finds no evidence of outright deception but highlights a recurring gap between rhetoric and practice in two of the three cases, raising concerns about the EU’s legitimacy and the risks of uncritical public trust. This thesis concludes that EU strategic communication alternates between trustworthy and interest-driven, with transparency and accountability remaining uneven.     Analyse der Leistungsfähigkeit verschiedener Mauermörtel auf Grundlage von Natürlich Hydraulischem Kalk

    Diese Masterarbeit befasst sich mit der Untersuchung und dem Vergleich verschiedener Mauermörtel auf Basis von Natürlich Hydraulischem Kalk (NHL). Ziel der Arbeit ist es, die Leistungsfähigkeit unterschiedlicher Mörtelrezepturen systematisch zu analysieren und zu bewerten. Untersucht wurden zum einen zwei kommerzielle NHL 5-Produkte der Hersteller Röfix und Tigre, zum anderen modifizierte Varianten dieser Grundmörtel mit puzzolanischen Zusatzstoffen wie Metakaolin und Trass sowie mit Luftporenbildnern. Im Rahmen umfassender Laboruntersuchungen wurden zentrale Eigenschaften der Mörtel bestimmt. Dazu zählen die Biegezug- und Druckfestigkeit, die Karbonatisierungstiefe, die offene Porosität, das Wassertransportverhalten sowie die kapillare Wasseraufnahme. Ergänzend wurden die puzzolanische Aktivität der Zusatzstoffe mit dem modifizierten Chapelle-Test sowie die mineralogische Zusammensetzung mithilfe der Röntgendiffraktometrie untersucht. Die Ergebnisse zeigen deutliche Unterschiede zwischen den beiden NHL 5-Produkten hinsichtlich ihrer mineralogischen Zusammensetzung und Festigkeitseigenschaften. Die Zugabe von Trass und insbesondere von Metakaolin führen zu einer spürbaren Erhöhung der mechanischen Festigkeiten und zu einer verbesserten Wasserbeständigkeit der Mörtel. Metakaolin wießt eine deutlich höhere puzzolanische Reaktivität auf als Trass. Der Einsatz von Luftporenbildnern beeinflusst vor allem die Frischmörteleigenschaften und die Porenstruktur. In einigen Fällen wird eine leichte Reduktion der Druckfestigkeit beobachtet, gleichzeitig kann durch die gezielte Steuerung der Luftporen der kapillare Wassertransport verringert werden, was sich positiv auf die Dauerhaftigkeit auswirkt. Die Arbeit zeigt, dass durch die Kombination von NHL mit reaktiven Zusatzstoffen und Luftporenbildnern leistungsfähige Mörtel mit gezielt einstellbaren Eigenschaften entwickelt werden können. Diese lassen sich entsprechend den Anforderungen an Verarbeitung, mechanische Eigenschaften und Witterungsbeständigkeit anpassen. Die Ergebnisse sind besonders für den Einsatz im Bereich der Restaurierung und Denkmalpflege relevant, da sie die Entwicklung nachhaltiger und materialgerechter Mörtelrezepturen unterstützen. Damit leistet die Arbeit einen wichtigen Beitrag zur Weiterentwicklung moderner Bau- und Instandhaltungsmaterialien auf Basis von Natürlich Hydraulischem Kalk.This master’s thesis investigates and compares various masonry mortars based on Natural Hydraulic Lime (NHL). The aim of the study is to systematically analyze the performance of different mortar formulations and assess their suitability for use in historic and sustainable masonry construction. Two commercial NHL 5 products, manufactured by Röfix and Tigre, were evaluated alongside modified versions of these base mortars with additions of pozzolanic materials such as metakaolin and trass, as well as air-entraining agents. Comprehensive laboratory tests were conducted to determine key properties of the mortars, including flexural and compressive strength, carbonation depth, open porosity, water transport behavior, and capillary water absorption. In addition, the pozzolanic activity of metakaolin and trass was assessed using the modified Chapelle test, and the mineralogical composition was analyzed by X-ray diffraction. The results indicate significant differences between the two NHL 5 products in terms of mineralogical composition and mechanical properties. The incorporation of metakaolin and trass notably improved the mechanical strength and water resistance of the mortars, with metakaolin showing higher pozzolanic reactivity. Air-entraining agents influenced the workability and pore structure of the fresh mortar. Although they slightly reduced compressive strength in some cases, they also helped to reduce capillary water transport, contributing positively to the durability of the mortars. Overall, the study demonstrates that combining NHL with reactive pozzolanic additives and air-entraining agents can produce high-performance mortars with tailored properties.     Melting realities : stakeholders perception and strategic adaptation to the challenges of climate change

    Climate change is regarded as one of the biggest challenges for the tourism industry. Due to its heavy dependence on snow, favorable weather conditions, and temperatures, winter tourism is perceived to be the most directly and immediately affected sector. While existing literature either focuses on projected consequences or technical adaptation measures, such as artificial snowmaking, limited attention has been given to the perception and strategic adaptation at the destination level. This thesis contributes to fill this gap by analyzing how tourism stakeholders in Tyrol and Salzburg, Austria, perceive and strategically adapt to the projected challenges. The analysis was framed by the Triple Bottom Line Theory (TBL) and Trawöger’s adaptation typology. Based on eleven semi-structured interviews, results show that awareness is widespread, however differences in the perception of urgency occur, often closely linked to varying altitudes. Adaptations remain predominantly reactive or concurrent, however, regions have also started to invest in anticipatory strategies, redirecting the focus from traditional winter offerings to a more diversified product palette. Results suggest that many destinations are in a strategic transition phase – from solely short-term, often profit oriented adjustments towards more sustainable, long-term strategies.
