    Functional analysis of the p66Shc in BRAFV600E mutated melanoma cells

    The adaptor protein p66Shc1, a splice variant of the SHC1 gene, has been implicated in cellular responses and tumour suppression, particularly in the context of BRAFV600E-mutated melanoma. This study aimed to investigate the specific function of p66Shc1 in human melanoma cells using CRISPR-Cas9 genome editing to disrupt its expression selectively in the A375 melanoma cell line. We targeted the start codon of p66Shc1 to abolish expression of the isoform without affecting the shorter isoforms (p46Shc1 and p52Shc1), preserving their function. Despite initial challenges in establishing a conditional knockout model via FRT recombination, successful genome editing was confirmed through sequencing and TIDE analysis, revealing efficient induction of indels at the targeted locus. Functional analyses revealed altered expression of key signalling proteins in p66Shc1-deficient cells, indicating a regulatory role of p66Shc1 in intracellular signalling pathways and supporting its potential tumour suppressive function in BRAFV600E-transformed melanoma. Furthermore, the difficulty in isolating complete knockout clones and the persistence of residual p66Shc1 expression in edited populations suggest a potential essentiality of p66Shc1 for A375 cell viability or homeostasis.     Das Ziel dieser Arbeit ist die Entwicklung eines Software Tools, welches den Nachweis des Sprödbruchs für den Anwender automatisiert und anwendungsfreundlich durchführt. Dabei liegt der Fokus auf einer Alternative für den meist konservativen Nachweis aus EN 1993-1-10 Tabelle 2.1. Die Arbeit umfasst neben der Dokumentation und Verifikation des Softwaretools, auch eine detaillierte Erklärung des Sprödbruchnachweis. Genauer eingegangen wird auf den theoretischen Hintergrund des vereinfachten Nachweises aus EN 1993-1-10 und der umfangreicheren bruchmechanischen Methode. Das Softwaretool wird so entwickelt, dass Erweiterungen und Änderungen einfach durchgeführt werden können.The aim of this work is to develop a software tool that performs the verification of brittle fracture in an automated and user-friendly manner. The focus is on an alternative for the mostly conservative verification from EN 1993-1-10 Table 2.1. In addition to the documentation and verification of the software tool, the work also includes a detailed explanation of the brittle fracture check. The theoretical background of the simplified verification from EN 1993-1-10 and the more comprehensive fracture mechanics method is explained in more detail.     On the security of deep learning applications in supply chain management : a game-theoretical approach

    Driven by the promise of increased operational efficiency, artificial intelligence and deep learning are increasingly deployed in supply chain management. However, these systems are vulnerable to attacks, such as adversarial examples, backdoor, and jailbreak attacks, which pose significant threats to supply chain management operations. The interconnectedness of modern supply chains makes artificial intelligence systems particularly susceptible due to the potential for propagation effects. Despite extensive research on the security of machine learning models in general, supply chain-specific applications remain underexplored. Moreover, existing threat evaluation methods apply a purely technical perspective and fail to account for contextual factors that influence security. This research complements the technical perspective with economic and strategic considerations. It develops methods to generate adversarial examples for prediction with time-series data, a method commonly applied in supply chain management, and evaluates their impact through empirical experiments. A game-theoretic model is then formulated to examine the strategic interactions between adversaries and defenders, with empirical results informing the model's instantiation and its practical relevance. The findings are integrated into a framework that systematically addresses technical, economic, and contextual factors to qualitatively assess and mitigate security implications for the deployment of deep learning. This thesis demonstrates that deep learning models commonly used in supply chain management are highly vulnerable to adversarial attacks, particularly for the forecasting and image-based quality control task. Including non-technical factors in the evaluation challenges the common notions of attacks and defenses in the literature. This approach reveals the importance of economic considerations in evaluating adversarial machine learning threats. The framework provides a comprehensive approach to assess the susceptibility of deep learning systems. Its application in two supply chain management use cases further demonstrates that technologically similar use cases can exhibit vastly different security implications. The results underscore the urgent need for securing deep learning applications in supply chain management and provide a systematic approach to addressing security implications across domains.     Bei der Untersuchung des literarischen Stilmittels des musikalischen Lyrismus, das trotz seiner Allgegenwärtigkeit in der literarischen Analyse oft untererforscht bleibt, konzentriert sich diese Arbeit darauf, Aspekte innerhalb der Texte der renommierten Songwriter Lily Allen und Taylor Swift aufzudecken, die entweder als feministisch oder nicht-feministisch angesehen werden können. Diese einflussreichen Künstlerinnen haben in den letzten zwei Jahrzehnten bis heute maßgeblich die gesellschaftlichen Wahrnehmungen geprägt, insbesondere unter jungen Frauen. Diese Arbeit untersucht daher die feministischen Dimensionen im Songwriting der Studioalben von Lily Allen und Taylor Swift und übernimmt dabei eine kritische Perspektive, um deren Darstellungen feministischer Perspektiven zu erkunden und ihr Schreiben in einen sozialen Kontext zu setzen. Trotz ihres weitreichenden Einflusses haben Allen und Swift durch ihr Engagement für den Feminismus Debatten ausgelöst, insbesondere hinsichtlich Vorwürfen von "Weißen Feminismus" oder "Trendfeminismus". Diese Masterarbeit zielt darauf ab, diese Komplexitäten zu hinterfragen, indem sie die Feinheiten feministischer Themen in ihren Texten genau untersucht. Die Analyse erforscht eingehend die Diskografien von Allen und Swift, kategorisiert Themen und verwendet Techniken von close reading. Durch die Untersuchung der von beiden Künstlern verwendeten erzählerischen Mittel versucht diese Studie, das komplexe Zusammenspiel zwischen feministischen und nicht-feministischen Themen in ihrer Musik zu enträtseln. Darüber hinaus erkennt die Arbeit die Skepsis gegenüber der feministischen Glaubwürdigkeit von Allen und Swift an. Durch diese kritische Untersuchung zielt die Arbeit darauf ab, zu den laufenden Diskussionen über feministische Diskurse in der populären Musik und zur kulturellen Bedeutung des Lyrismus von Allen und Swift beizutragen.In examining the literary device of musical lyricism, often underexplored in literary analysis despite its omnipresence, this thesis focuses on uncovering aspects within the lyrics of renowned songwriters Lily Allen and Taylor Swift, which can be either seen as feminist or non-feminist. These influential female artists have shaped societal perceptions, particularly among young women, over the past two decades up until now. Therefore, this thesis delves into the feminist dimensions within the songwriting of Lily Allen's and Taylor Swift's studio albums, adopting a critical lens to explore their portrayals of feminist perspectives and putting their writing into a social context. Despite their widespread influence, Allen and Swift's engagement with feminism has sparked debates, particularly regarding accusations of "White Feminism". This study aims to interrogate these complexities by closely examining the nuances of feminist themes within their lyrics. The analysis thoroughly explores Allen and Swift's discographies, categorising themes and employing close reading and narrative analysis techniques. By examining the narrative devices utilised by both artists, this study seeks to unravel the intricate interplay between feminist and non-feminist themes in their music. Furthermore, the thesis acknowledges the scepticism surrounding Allen and Swift's feminist credentials, acknowledging the prevalent discourse accusing the songwriters as "White Feminists".     This theoretical and practical Master‘s thesis examines Sou Fujimoto and his architectural methodology through a comprehensive study of his works and a theoretical exploration of his philosophy. Fujimoto‘s philosophy emphasizes the pursuit of a dynamic balance between nature and architecture, integrating ambiguity and diversity into spatial concepts. Central to his philosophy is the concept of the „In-Between“, seen as a space of potentials and fluidity. The formal analysis concludes that Fujimoto often begins with simple primitives and evolves them into projects through scaling, stacking, or nesting. This insight inspired the development of a new method of form-finding in this study. Inspired by the concept of the „In-Between“, the study chooses to identify a simple initial form for Fujimoto‘s projects and achieve the final project through an animated transformation of this initial form. This new method allows for detailed examination of the intermediate steps of transformation and their combination in case studies to explore the spectrum of potentials in architectural form-finding.     Hintergrund: Long COVID ist durch anhaltende Symptome nach einer akuten SARS-CoV-2- Infektion gekennzeichnet und stellt eine wachsende Herausforderung für Gesundheitssysteme weltweit dar. Betroffene leiden häufig unter Fatigue, kognitiven Beeinträchtigungen und körperlichen Einschränkungen, die über Monate oder sogar Jahre bestehen bleiben. Aktuelle Forschungsergebnisse deuten darauf hin, dass eine veränderte mitochondriale Funktion eine zentrale Rolle in der Pathophysiologie von Long COVID spielen könnte. Zielsetzung: Diese Studie untersuchte Unterschiede in der mitochondrialen Funktion bei Personen mit und ohne Long COVID. Zudem wurden mögliche Zusammenhänge zwischen Veränderungen der mitochondrialen Funktion und Fatigue, psychische Gesundheit sowie Körpergewicht bei Long COVID analysiert. Methoden: Die mitochondriale Atmungsfunktion wurde mittels O2K-Hochauflösungs- Respirometrie in peripheren mononukleären Blutzellen (PBMCs) gemessen. Dabei wurden sowohl primäre als auch sekundäre respiratorische Parameter analysiert. Fatigue wurde mit der Chalder Fatigue Scale erfasst, während die psychische Gesundheit in der Kontrollgruppe mit dem Beck Depression Inventory-II und in der Long COVID-Gruppe mit der Hospital Anxiety and Depression Scale untersucht wurde. Körpergewicht und -größe dienten zur Berechnung des Body Mass Index (BMI) und des Body Mass Excess (BME). Die Daten von Long COVID- Patient*innen (n=50) wurden mit einer psychisch und physisch gesunden Kontrollgruppe (n=118) verglichen. Ergebnisse: Die mitochondriale Funktion war in der Long COVID-Gruppe, im Vergleich zur Kontrollgruppe, signifikant eingeschränkt. Es konnten jedoch keine Zusammenhänge zwischen mitochondrialen Parametern und Fatigue-Scores oder Körpergewicht innerhalb der Long COVID-Gruppe festgestellt werden. Eine Analyse der psychischen Gesundheit war aufgrund fehlender Daten in der Long COVID-Gruppe nicht möglich. Diese Ergebnisse deuten darauf hin, dass komplexe, nicht-lineare und multifaktorielle Mechanismen zur klinischen Ausprägung von Long COVID beitragen. Schlussfolgerung: Die Ergebnisse dieser Studie liefern empirische Hinweise darauf, dass eine mitochondriale Dysfunktion in Immunzellen eine pathophysiologische Rolle bei Long COVID spielt. Es konnte keine messbare Dosis-Wirkungs-Beziehung zwischen der Schwere der Fatigue und der mitochondrialen Funktion festgestellt werden. Dies legt nahe, dass Long COVID die bioenergetische Kapazität von Immunzellen unabhängig von der individuellen Fatigue stark einschränken könnte. Künftige Studien sollten diese Erkenntnisse weiter untersuchen, vorzugsweise in einem longitudinalen Studiendesign, um kausale Zusammenhänge besser zu verstehen.Background: Long COVID, characterized by persistent symptoms following acute SARS- CoV-2 infection, represents a growing challenge for healthcare systems worldwide. Affected individuals report clinically relevant symptoms including fatigue, cognitive impairments, and physical limitations that persist for months or even years after the initial infection. Emerging evidence suggests that mitochondrial impairment may play a central role in the pathophysiology of Long COVID. Objective: This study investigated possible differences in mitochondrial function between Long COVID patients and Long COVID-free controls. In addition, associations between changes in mitochondrial function and scores of fatigue, psychological health, and body weight in Long COVID were tested. Methods: Mitochondrial respiratory function was evaluated using O2K high-resolution respirometry in peripheral blood mononuclear cells (PBMCs). Key parameters, including primary and secondary respiration metrics, were analyzed. Fatigue burden was assessed using the Chalder Fatigue Scale, while psychological health was measured using the Beck Depression Inventory-II in the control group and the Hospital Anxiety and Depression Scale in the Long COVID group. These measures were based on self-reports. Body weight and height did define measures for Body Mass Index (BMI) and Body Mass Excess (BME). Data from patients with Long COVID (n=50) were compared with a physically and psychologically stable control group (n=118). Results: Significant differences in mitochondrial respiration metrics were observed between patients with Long COVID and the control group, demonstrating a relevant decline in mitochondrial function in the Long COVID group. However, no significant associations were found between mitochondrial parameters and fatigue scores or body weight in the Long COVID group. Psychological health could not be analyzed due to missing data in the Long COVID group. These findings highlight the need for further exploration into non-linear and multifactorial mechanisms influencing the clinical phenotype of Long COVID. Conclusion: This study provides empirical evidence that mitochondrial dysfunction in immune-cell bioenergetics plays a pathophysiological role in Long COVID. There appears to be no measurable dose-response relationship between fatigue severity and mitochondrial function, suggesting that Long COVID may strongly limit immune-cell bioenergetics, independent of fatigue burden.
