Das polytraumatisierte Kniegelenk bei der Katze - eine Literaturübersicht und Fallevaluierung an der Chirurgischen Kleintierklinik
Diploma thesis - University of Veterinary Medicine Vienna - 2024This thesis addresses polytraumatic knee joint injuries in cats, comprising a literature review and a retrospective case study conducted at the University of Veterinary Medicine, Vienna. Polytraumatic injuries of the knee joint frequently result from substantial external forces, such as caraccidents or falls from highs, and are defined by the involvement of multiple structures, including cruciate and collateral ligaments, as well as the menisci. Our hypothesis—that surgical treatment of multiligament knee injuries in cats, based on literature findings and retrospective case analysis from the University of Veterinary Medicine, Vienna, would result in pain-free and functionally stable limb function in the majority of documented treatments— was only partially confirmed due to the limited significance of our findings, which were constrained by insufficient data. The diagnosis of polytraumatic knee joint injuries is typically made through clinical orthopedic examination and imaging methods, such as X-ray and CT scans. Due to joint instability, surgical treatment is necessary, as conservative methods have limited success. The retrospective case study at the University of Veterinary Medicine involved a cohort of 21 cats treated between 2001 and 2023. The surgical methods included a variety of stabilization techniques, such as extracapsular suture stabilization and transarticular pinning. Postoperative complications were observed in some cases, including infections and implant loosening, which occasionally necessitated further surgical intervention. A comparative analysis of cases treated at the University of Veterinary Medicine with those described in the literature revealed a high degree of similarity in terms of treatment choice, therapeutic success, and complications findings. A total of nine major and one minor complication were recorded at the University, including infections, swelling, and implant loosening. A review of the literature reveals comparable findings. Eight major and three minor complications were observed in 22 cats. No correlation was identified between the selected treatment option and the occurrence of major or minor complications. In general, no surgical method was found to have a higher or lower incidence of complications. Consequently, no method was identified as being particularly recommended or advised against. The postoperative care regimen at the University of Veterinary Medicine is characterized by a higher frequency of follow-ups compared to the postoperative regimens described in the literature. This approach may facilitate the earlier identification and treatment of potential complications, which could ultimately lead to improved healing outcomes. The follow-up appointments at the University were scheduled at an earlier point in time and with more 44 frequent intervals than is typically described in the literature. However, the intervals for imaging follow-ups (usually after four to six weeks) were consistent across both sources. Additionally, a questionnaire was employed to evaluate the owner satisfaction with the treatment provided at the University of Veterinary Medicine and to assess the quality of life of the cats. The questionnaire was based on existing assessment systems, and a point-based rating system was employed to assign responses. In this study, a score below 20 was considered to indicate the absence of pain. The analysis of the questionnaires revealed a notable reduction in pain among the animals. The responses from the owners indicated that they perceived their cats to be free from pain, suggesting a favorable treatment outcome with restored stability and functionality of the limb. The results of this study indicate that careful diagnostics and individually selected surgical interventions for polytraumatic knee injuries in cats can result in positive long-term treatment outcomes. Nevertheless, further studies with larger sample sizes and a prospective design would be beneficial to more comprehensively evaluate long-term outcomes and potential relationships between treatment options and complications.Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Diese Diplomarbeit befasst sich mit polytraumatisierten Kniegelenksverletzungen bei Katzen und umfasst eine Literaturübersicht sowie eine retrospektive Fallstudie an der Veterinärmedizinischen Universität Wien. Polytraumatische Verletzungen des Kniegelenks treten häufig in Verbindung mit starken äußeren Einwirkungen wie Unfällen oder Stürzen auf und sind dadurch gekennzeichnet, dass oft mehrere Strukturen wie Kreuz- und Kollateralbänder sowie Menisken betroffen sind. Unsere Hypothese, dass die chirurgische Versorgung multiligamentöser Kniegelenksverletzungen bei Katzen – basierend auf Literaturergebnissen und retrospektiv analysierten Fällen an der Veterinärmedizinischen Universität Wien – in der Mehrheit der dokumentierten Behandlungen zu einer schmerzfreien und funktionell stabilen Gliedmaßenfunktion führt, konnten wir nur teilweise bestätigen, da die Aussagekraft unserer gewonnen Studienergebnisse aufgrund der unzureichenden Datenmenge eingeschränkt ist. Die Diagnosestellung polytraumatischer Kniegelenksverletzungen erfolgt typischerweise durch klinisch-orthopädische Untersuchung und bildgebende Verfahren. Aufgrund der Instabilität des Gelenks ist eine chirurgische Behandlung notwendig, da konservative Methoden wenig Erfolg versprechen. Die retrospektive Fallstudie an der VMU umfasst 21 Katzen, die zwischen 2001 und 2023 behandelt wurden. Die operativen Behandlungsmethoden umfassten verschiedene Stabilisierungstechniken wie unter anderem extrakapsuläre Fadenzügel und transartikuläres Pinning. Postoperative Komplikationen traten in einigen Fällen auf, darunter Infektionen und Implantatlockerungen, die teilweise eine erneute Operation erforderten. Eine Gegenüberstellung der behandelten Fälle an der VMU und der in der Literatur beschriebenen Fälle zeigte eine große Übereinstimmung hinsichtlich der Wahl der Behandlungsoption, des therapeutischen Erfolgs und der aufgetretenen Komplikationen. An der VMU wurden insgesamt neun schwere (major) und eine leichte (minor) Komplikationen verzeichnet, darunter Infektionen, Schwellungen und Implantatlockerungen. Die Literatur berichtet ähnliche Zahlen: bei 22 Katzen traten 8 major und 3 minor Komplikationen. Ein Zusammenhang zwischen gewählter Therapieoption und Auftreten von major/minor Komplikationen konnte nicht festgestellt werden. Insgesamt ergab sich keine Operationsmethode, die mit mehr oder weniger Komplikationen einherging und daher empfohlen/ vermieden werden sollte. 42 Bei der postoperativen Versorgung zeigt sich an der VMU eine Tendenz zu engmaschigeren Kontrollen gegenüber dem postoperativen Regime in der Literatur, was zu einer frühzeitlichen Behandlung möglicher Komplikationen und damit besseren Heilungschancen beitragen könnte. So wurden Kontrolltermine an der VMU früher und häufiger angesetzt, als in der Literatur beschrieben. Übereinstimmungen gab es in den Zeitabständen der bildgebenden Kontrolluntersuchungen (i.d.R nach 4 - 6 Wochen). Ergänzend wurde mittels Fragebögen die BesitzerInnenzufriedenheit an der VMU mit der Behandlung sowie die Lebensqualität der Katzen ermittelt. Dafür wurde ein Fragebogen in Anlehnung an bereits existierende Bewertungssysteme erstellt. Die Antworten wurden einem Punktebewertungssystem zugeteilt. Eine Punktzahl von unter 20 wurde definiert als “schmerzfrei”. Bei der Auswertung der Fragebögen konnte eine signifikante Schmerzlinderung bei den Tieren festgestellt werden: alle BesitzerInnenbefragungen ergaben, dass die HalterInnen ihre Katzen als schmerzfrei beschrieben. Dies weist auf einen erfolgreichen Behandlungsverlauf und eine wiederhergestellte Stabilität und Funktionalität der Gliedmaße hin. Unsere Ergebnisse belegen, dass eine sorgfältige Diagnostik sowie individuell ausgewählte chirurgische Eingriffe bei polytraumatisierten Kniegelenken von Katzen langfristig zu positiven Behandlungsergebnissen führen können. Um die Langzeitergebnisse und mögliche Zusammenhänge zwischen gewählter Therapieoption und auftretenden Komplikationen umfassender bewerten zu können, wären jedoch weitere Studien mit größeren Fallzahlen und prospektivem Charakter wünschenswert
Giardiose bei Hund und Katze: Risikofaktoren und therapeutische Optionen – eine Literaturübersicht

Vor allem junge Hunde und Katzen sind häufiger infiziert, wobei breiiger bis wässriger Durchfall besonders häufig zu beobachten ist. Die Diagnostik erweist sich in vielen Fällen oft als schwierig, da die Zysten häufig intermittierend ausgeschieden werden. Für die Behandlung einer Giardiose haben sich Fenbendazol und Metronidazol als wirksam erwiesen. Allerdings ist ein andauernder Therapieerfolg nicht immer gewährleistet. Durch entsprechende Hygienemaßnahmen kann das Risiko einer Reinfektion reduziert und der Erfolg der Therapie maßgeblich gesteigert werden. In den vergangenen Jahren wurde bei der Behandlung von Giardiose zunehmend auch das intestinale Mikrobiom mit einbezogen, denn die Darmflora trägt einen entscheidenden Faktor zur Abwehr von Parasiten und zur klinischen Heilung bei. Im Vergleich zu Metronidazol zeigt Fenbendazol dabei eine geringere Auswirkung auf das Darmikrobiom. Eine Infektion mit Giardien führt unabhängig von der gewählten Behandlung selbst zu einer Veränderung der Bakterienzusammensetzung der Darmflora. Probiotika fördern die Diversität des Mikrobioms und indirekt auch das Immunsystem, was sich positiv auf die Parasitenabwehr und damit auf den Heilungsprozess auswirken kann. Zudem spielt die Ernährung eine entscheidende Rolle bei der Bekämpfung intestinaler Krankheitserreger. Eine ballaststoffreiche, fettarme, sowie laktose- und zuckerfreie Ernährung ist bei Giardiosepatienten zu bevorzugen. Es existiert eine Vielzahl von Medikamenten, die in experimentellen Studien eine Wirkung gegen Giardien zeigen, jedoch besitzen sie ähnliche Vor- und Nachteile wie Metronidazol und Fenbendazol. Als alternative Ansätze sind pflanzliche Wirkstoffe sowie eine Impfung zu erwähnen. Hinsichtlich der Entwicklung neuer Impfstoffe lassen sich unterschiedliche Ansätze beobachten. Especially young dogs and cats are frequently affected, with semi-liquid to liquid diarrhea being particularly common. The diagnosis of giardiosis can be challenging due to frequently intermittent cyst excretion. For the treatment of giardiosis, fenbendazole and metronidazole have been shown to be effective. However, sustainable treatment success cannot be achieved in all cases. Appropriate hygiene measures can reduce the risk of reinfection and significantly increase the success of therapy. In recent years, the intestinal microbiome was included in the treatment of giardiosis, because the intestinal flora plays a crucial role in parasite defense and clinical resolution. In comparison to metronidazole, fenbendazole has a less pronounced effect on the gut microbiome. Infection with Giardia itself results in modification of the bacterial composition of its gastrointestinal microbiome, irrespective of the chosen treatment. Probiotics promote microbiome diversity and indirectly also the immune system which can have a positive effect on parasite control and therefore on the healing process. Diet also plays a crucial role in the control of intestinal pathogens. A high-fiber, low-fat, lactose- and sugar-free diet is preferred for patients with giardiosis. Numerous compounds have been shown to be effective against Giardia, but they have similar advantages and disadvantages to metronidazole and fenbendazole. Alternative approaches include plant-based active ingredients and vaccination. Different approaches regarding the development of new vaccines can be observed. Further research is needed, especially on the interactions between Giardia, antiprotozoal compounds and the intestinal microbiome as well as on vaccination to optimize the control of this parasitosis in domestic animals
Comparative conflict resolution: cooperative cichlids outperform less social species
Evolution towards complex forms of sociality is dependent on overcoming conflicts of interest between prospective social partners. Competition for limited resources is a key aspect of such conflicts. Behaviour that results in conflict resolution without termination of the social relationship is thus predicted to be more prevalent in more social species. We here tested this hypothesis by staging contests between conspecifics over a crucial resource, using three lamprologine cichlid species that differ in their sociality. The cooperatively breeding species (Neolamprologus pulcher) indeed demonstrated improved conflict resolution, showing more egalitarian sharing of the resource and establishing mutually tolerant relationships more often compared to the pair-bonded species (Variabilichormis moorii) and the mostly solitary species (Lepidiolamprologus elongatus). We suggest that this is the result of behavioural differences between these cichlids: N. pulcher already showed more de-escalating behaviour during the earliest stages of the contests and increased rates of de-escalating behaviours more in response to heightened levels of aggression. Our results thus provide comparative support for the notion that socially competent behaviour, resolving conflicts while maintaining social relations, is a key behavioural aspect of evolutionary transitions to complex societies
Exploratory study on the body distribution of skin color, pigmentation and, degree of tan in Central European Caucasian Women
It is well known that skin color varies by body site and with season. However, little quantitative data on the topography of skin color and pigmentation are available. Therefore, exploratory cutaneous colorimetric measurements in 20 in central European Caucasian women aged 20 to 60 years have been made at 18 body sites. Tri-stimulus L*a*b*-values, hue, and chroma are considered to describe skin color. Based on the “Individual Typology Angle”, the “Degree of Tan” was introduced to quantify the difference between constitutive and facultative pigmentation. Measurements were done in late winter and early summer to estimate potential changes by solar ultraviolet radiation. These measurements made evident that skin color obviously differs across the body in late winter. Even nearby body sites can be recognized as differently colored. A remaining degree of tan was found at permanent and intermittent exposed body sites. The remaining tan was not most pronounced at the permanently exposed sites but on the intermediate ones like the shoulder. In early summer, the degree of tan has most developed at the hands, arms, and instep, followed by the face. This study showed that besides basic differences between body sites in winter, accumulation, and degradation of tan also vary between body sites
Seeing the benefits, but not taking advantage of them: Dog and cat owners\u27 beliefs about veterinary telemedicine
Telemedicine offers benefits to clients and their animals, but potential disadvantages are also being debated. Using a questionnaire, we investigated dog and cat owners’ (N = 2117) use of and beliefs about telemedicine and whether beliefs impact past and expected future use. Although the majority of owners are aware that telemedicine can lead to the risk of something being missed, they see great potential in remote consultation in terms of usefulness for follow-up appointments or improving access to a specialist. However, only 12% of dog owners and 6% of cat owners have used telemedicine, and around 25% of owners who have never used it would be willing to use it in the future. Owners with a larger number of recent veterinary visits were more likely to have used telemedicine.
Although a definition of ‘telemedicine’ was provided, respondents may have had different perceptions of what this meant. Owners of dogs and cats recognise the potential benefits of telemedicine, but there is a mismatch with the actual uptake. This not only raises questions about the current availability of telemedicine but also should increase veterinary professionals’ understanding of its potential benefits in veterinary practice
[11C]metoclopramide is a sensitive radiotracer to measure moderate decreases in P-glycoprotein function at the blood-brain barrier
The efflux transporter P-glycoprotein (P-gp) at the blood-brain barrier limits the cerebral uptake of various xenobiotics. To assess the sensitivity of [11C]metoclopramide to measure decreased cerebral P-gp function, we performed [11C]metoclopramide PET scans without (baseline) and with partial P-gp inhibition by tariquidar in wild-type, heterozygous Abcb1a/b(+/−) and homozygous Abcb1a/b(−/−) mice as models with controlled levels of cerebral P-gp expression. Brains were collected to quantify P-gp expression with immunohistochemistry. Brain uptake of [11C]metoclopramide was expressed as the area under the brain time-activity curve (AUCbrain) and compared with data previously obtained with (R)-[11C]verapamil and [11C]N-desmethyl-loperamide. Abcb1a/b(+/−) mice had intermediate P-gp expression compared to wild-type and Abcb1a/b(−/−) mice. In baseline scans, all three radiotracers were able to discriminate Abcb1a/b(−/−) from wild-type mice (2.5- to 4.6-fold increased AUCbrain, p ≤ 0.0001). However, only [11C]metoclopramide could discriminate Abcb1a/b(+/−) from wild-type mice (1.46-fold increased AUCbrain, p ≤ 0.001). After partial P-gp inhibition, differences in [11C]metoclopramide AUCbrain between Abcb1a/b(+/−) and wild-type mice (1.39-fold, p ≤ 0.001) remained comparable to baseline. There was a negative correlation between baseline [11C]metoclopramide AUCbrain and ex-vivo-measured P-gp immunofluorescence (r = −0.9875, p ≤ 0.0001). Our data suggest that [11C]metoclopramide is a sensitive radiotracer to measure moderate, but (patho-)physiologically relevant decreases in cerebral P-gp function without the need to co-administer a P-gp inhibitor
Presence of Duncker\u27s muscle fluke (Alaria alata mesocercariae) in wild boars from east Austria, 2015-2024
Introduction Alaria alata is a trematode that dwells in its adult form in the intestinal tract of carnivores. The life cycle of the parasite is complex and comprises several developmental stages with two obligatory intermediate hosts (snails and amphibians). Wild boars can act as paratenic hosts by ingesting second intermediate hosts (amphibians) that carry Alaria alata mesocercariae. Larval alariosis is a potential health risk to consumers of wild boar meat, although the few cases of human infestations by Alaria mesocercariae were probably attributable to Alaria americana and not to Alaria alata . Materials and methods Between August 2022 and January 2024, we took tissue samples (comprising glandular, fat and muscle tissue; approximately 50-80 g total weight) from the throat area and, if present, from the diaphragm of 429 hunted wild boars. We took samples at wild game processing facilities and collection centers. The day and region (postal code) of killing and the weight of the carcasses were provided. We examined the carcasses within 24 hours of sampling using the Alaria alata mesocercariae migration technique. Data for the period 2015-2018 were provided by the Institute of Food Hygiene and Technology, University of Veterinary Medicine Vienna. Sampling and examination methods were identical to those in the 2022-2024 study. We prepared descriptive statistics for season and origin and for days from killing to testing. For a well described hot-spot in the March-Thaya region (Hohenau-RabensburgBernhardsthal-Drosing-Bockflie ss Neusiedl/Zaya), we compared the frequencies of mesocercariae-positive carcasses with those from previous studies. Results We detected no motile Alaria alata mesocercariae in the samples collected from 2022 to 2024 (0/429), although six of the 482 samples taken between 2015 and 2018 had tested positive for mesocercariae. All six positive samples in 2015-2018 originated from the 58 specimens from the area HohenauRabensburg-Bernhardsthal-Dr & ouml;sing-Bockflie ss-Neusiedl/ Zaya but we detected no mesocercariae in the 26 samples from the same region examined in 2022-2024. The finding was significantly lower than the frequencies reported for previous study periods (p<0.001). Conclusion From a food hygiene perspective, it appears that motile Alaria alata mesocercariae occur less frequently in Austrian wild boar meat than in 2015-2018. Various geographical, climatic and seasonal effects may be partly responsible.Wir untersuchten in den Jahren 2022–2024 Gewebsproben (Drüsen-, Muskel- und Fettgewebe der Hals- Wangen-Region und, wenn verfüg- bar, Zwerchfell) von 429 jagdlich erlegten und zur Verarbeitung in zu- gelassenen Wildbearbeitungsbetrieben vorgesehenen Wildschweinen auf das Vorkommen motiler Alaria alata Mesozerkarien. Es handelte sich um Proben von 275 Wildschweinen aus 16 Bezirken in Niederösterreich, 143 Proben aus 4 Bezirken im Burgenland sowie 11 Proben aus der Steiermark, Oberösterreich und Wien. Mit dem Aus- wanderverfahren („Alaria alata mesocercariae migration technique“) konnten keine motilen Mesozerkarien nachgewiesen werden. Während in den Gemeindegebieten Hohenau- Rabensburg-Bernhardsthal-Drösing- Bockfließ-Neusiedl/Zaya im Zeitraum 2015–2018 noch Alaria alata Mesozerkarien (6/58) nachgewiesen werden konnten, war das 2022–2024 nicht der Fall (0/26), trotz vergleich- baren Studiendesigns und identer Untersuchungsverfahren. Für die Interpretation der Daten wären – derzeit fehlende – rezente Daten zum Vorkommen von Alarien Stadien in Endwirten oder Zwischenwirten hilfreich. Die Ergebnisse können aus lebensmittel¬hygienischer Sicht so interpretiert werden, dass motile Mesozerkarien in Tierkörpern von österreichischem Schwarzwild heute beim Großhandel seltener vor¬kommen als dies noch 2015–2018 der Fall war. Dafür können geographische, klimatische und saisonale Effekte mitverantwortlich sein
Heads up-Four Giraffa species have distinct cranial morphology
Giraffe (Giraffa spp.) are among the most unique extant mammals in terms of anatomy, phylogeny, and ecology. However, aspects of their evolution, ontogeny, and taxonomy are unresolved, retaining lingering questions that are pivotal for their conservation. We assembled the largest known dataset of Giraffa skulls (n = 515) to investigate patterns of cranial variability using 3D geometric morphometrics. The results show distinct sexual dimorphism and divergent ontogenetic trajectories of skull shape for the north clade (G. camelopardalis antiquorum, G. c. camelopardalis, G. c. peralta, and G. reticulata) and the south clade (G. giraffa angolensis, G. g. giraffa, G. tippelskirchi tippelskirchi, and G. t. thornicrofti) which was further supported statistically. Discriminant functions found statistically significant cranial shape differences between all four Giraffa species, and in some cases also between subspecies of the same species. Our 3D morphometric analysis shows that the four genetically distinct Giraffa spp. also have distinct cranial morphologies, largely addressable to features of display (ossicones). Our results highlight the importance of focusing future giraffe conservation efforts on each taxon to maintain their unique characteristics and biodiversity in the wild
Unraveling the distinctive gut microbiome of khulans (Equus hemionus hemionus) in comparison to their drinking water and closely related equids
The microbial composition of host-associated microbiomes is influenced by co-evolutionary interactions, host genetics, domestication, and the environment. This study investigates the contribution of environmental microbiota from freshwater bodies to the gastrointestinal microbiomes of wild khulans (Equus hemionus hemionus, n?=?21) and compares them with those of captive khulans (n?=?12) and other equids-Przewalski\u27s horse (n?=?82) and domestic horse (n?=?26). Using PacBio technology and the LotuS pipeline for 16S rRNA gene sequencing, we analyze microbial diversity and conduct differential abundance, alpha, and beta diversity analyses. Results indicate limited microbial sharing between wild khulans and their waterhole environments, suggesting minimal environmental influence on their gut microbiomes and low levels of water contamination by khulans. Wild khulans exhibit greater microbial diversity and richness compared to captive ones, likely due to adaptations to the harsh nutritional conditions of the Gobi desert. Conversely, captive khulans show reduced microbial diversity, potentially affected by dietary changes during captivity. These findings highlight the significant impact of environment and lifestyle on the gut microbiomes of equids
In situ isotropic 3D imaging of vasculature perfusion specimens using x-ray microscopic dual-energy CT
Ex vivo x-ray angiography provides high-resolution, three-dimensional information on vascular phenotypes down to the level of capillaries. Sample preparation for ex vivo angiography starts with the removal of blood from the vascular system, followed by perfusion with an x-ray dense contrast agent mixed with a carrier such as gelatine or a polymer. Subsequently, the vascular micro-architecture of harvested organs is imaged in the intact fixed organ. In the present study, we present novel microscopic dual-energy CT (microDECT) imaging protocols that allow to visualise and analyse microvasculature in situ with reference to the morphology of hard and soft tissue. We show that the spectral contrast of µAngiofil and Micropaque barium sulphate in perfused specimens allows for the effective separation of vasculature from mineralised skeletal tissues. Furthermore, we demonstrate the counterstaining of perfused specimens using established x-ray dense contrast agents to depict blood vessels together with the morphology of soft tissue. Phosphotungstic acid (PTA) is used as a counterstain that shows excellent spectral contrast in both µAngiofil and Micropaque barium sulphate-perfused specimens. A novel Sorensen-buffered PTA protocol is introduced as a counterstain for µAngiofil specimens, as the polyurethane polymer is susceptible to artefacts when using conventional staining solutions. Finally, we demonstrate that counterstained samples can be automatically processed into three separate image channels (skeletal tissue, vasculature and stained soft tissue), which offers multiple new options for data analysis. The presented microDECT workflows are suited as tools to screen and quantify microvasculature and can be implemented in various correlative imaging pipelines to target regions of interest for downstream light microscopic investigation
