A novel function of STAT3? in suppressing interferon response improves outcome in acute myeloid leukemia
Signal transducer and activator of transcription 3 (STAT3) is frequently overexpressed in patients with acute myeloid leukemia (AML). STAT3 exists in two distinct alternatively spliced isoforms, the full-length isoform STAT3α and the C-terminally truncated isoform STAT3β. While STAT3α is predominantly described as an oncogenic driver, STAT3β has been suggested to act as a tumor suppressor. To elucidate the role of STAT3β in AML, we established a mouse model of STAT3β-deficient, MLL-AF9-driven AML. STAT3β deficiency significantly shortened survival of leukemic mice confirming its role as a tumor suppressor. Furthermore, RNA sequencing revealed enhanced STAT1 expression and interferon (IFN) signaling upon loss of STAT3β. Accordingly, STAT3β-deficient leukemia cells displayed enhanced sensitivity to blockade of IFN signaling through both an IFNAR1 blocking antibody and the JAK1/2 inhibitor Ruxolitinib. Analysis of human AML patient samples confirmed that elevated expression of IFN-inducible genes correlated with poor overall survival and low STAT3β expression. Together, our data corroborate the tumor suppressive role of STAT3β in a mouse model in vivo. Moreover, they provide evidence that its tumor suppressive function is linked to repression of the STAT1-mediated IFN response. These findings suggest that the STAT3β/α mRNA ratio is a significant prognostic marker in AML and holds crucial information for targeted treatment approaches. Patients displaying a low STAT3β/α mRNA ratio and unfavorable prognosis could benefit from therapeutic interventions directed at STAT1/IFN signaling
Mixed ensiling of drought-impaired grass with agro-industrial by-products and silage additives improves the nutritive value and shapes the microbial community of silages
Droughts lead to reduced biomass production and unfavourable nutrient composition in grassland. As an alleviation, yet unexploited strategy, mixed ensiling of grass with agro-industrial by-products may improve the ensilability and nutritive value of drought-impaired grassland. This study investigated first whether mixed ensiling of drought-impaired grass with either sugar beet pulp (SBP), wheat gluten feed (WGF) or brewers\u27 grains (BG) has a beneficial impact on chemical composition, fermentation characteristics, in vitro gas production (GP) and physically effective neutral detergent fibre (peNDF) of silages. Secondly, it was tested whether the application of anaerobic fungi culture supernatant (AF), mixed ruminal fluid (RF) or lactic acid bacteria (LAB) provides further advantages. Additionally, the microbial community composition was evaluated in selected silages. All silages showed satisfying conservation characteristics with high lactic acid levels and low dry matter losses, and peNDF values typically found for conserved forages. Mixed ensiling with BG substantially increased the crude protein concentration, whereas SBP increased the total degradability and WGF enhanced both. The further addition of fresh AF resulted in the overall highest lactic acid levels, especially in SBP-based silages, but without changes in in vitro GP. The in vitro GP was higher with RF, particularly in mixed silages, suggesting an improved degradability. The LAB-treated silages showed lower pH compared to controls, but had no impact on in vitro GP kinetics. Concluding, mixed ensiling holds potential to produce high-quality silages from drought-impaired grassland. The further addition of silage additives can be useful for certain substrates, but appeared not mandatory
The evolution of transglutaminases underlies the origin and loss of cornified skin appendages in vertebrates
Transglutaminases (TGMs) cross-link proteins by introducing covalent bonds between glutamine and lysine residues. These cross-links are essential for epithelial cornification which enables tetrapods to live on land. Here, we investigated which evolutionary adaptations of vertebrates were associated with specific changes in the family of TGM genes. We determined the catalog of TGMs in the main clades of vertebrates, performed a comprehensive phylogenetic analysis of TGMs, and localized the distribution of selected TGMs in tissues. Our data suggest that TGM1 is the phylogenetically oldest epithelial TGM, with orthologs being expressed in the cornified teeth of the lamprey, a basal vertebrate. Gene duplications led to the origin of TGM10 in stem vertebrates, the origin of TGM2 in jawed vertebrates, and an increasing number of epithelium-associated TGM genes in the lineage leading to terrestrial vertebrates. TGM9 is expressed in the epithelial egg tooth, and its evolutionary origin in stem amniotes coincided with the evolution of embryonic development in eggs that are surrounded by a protective shell. Conversely, viviparous mammals have lost both the epithelial egg tooth and TGM9. TGM3 and TGM6 evolved as regulators of cornification in hair follicles and underwent pseudogenization upon the evolutionary loss of hair in cetaceans. Taken together, this study reveals the gain and loss of vertebrate TGM genes in association with the evolution of cornified skin appendages and suggests an important role of TGM9 in the evolution of amniotes
Bewertung von mikrobiellen Metaboliten und Wirtsmetaboliten als Biomarker für die Darmgesundheit von Schweinen
Diploma thesis - University of Veterinary Medicine Vienna - 2024Weaning is a difficult time in the life of a piglet. It is associated with numerous stressors that have a negative impact on the health and well-being of the piglets. Among other factors, the change in feed is one of the most important stressors in this phase of life, which entails a change in the microbiome and often leads to dysbiosis in the digestive tract. As a result, diarrhoeaoften occurs during this sensitive phase of life, which is not only negative for the piglets, but also means economic losses. The aim of this trial was to generate different conditions in the digestive tract with the help of different feed additives. Thiswas achieved with the two feed additives carboxymethylcellulose (CMC) and benzoic acid. The effects of the supplements in the rations were assessed based on growth, diarrhoeamonitoring, gut integrity and serum parameters. Based on the above data, an attempt was made to identify biomarkers for the intestinal health of pigs. In order to influence the intestinal health of the piglets, the dams were divided into two feeding groups in late pregnancy. Half of the sows received 5 mg/kg benzoic acid added to the feed in late pregnancy and during lactation. During the suckling phase,the piglets received 5 mg/kg benzoic acid or the control complementary feed, depending on their mother sow. The piglets remained in the two feeding-groups even after weaning (28th day of life) until the 32nd day of life. The main trial of the experiment began on the piglets\u27 33rd day of life. The piglets were divided into four different feeding groups: Acid group (5 mg/kg benzoic acid), fibregroup (3% carboxymethylcellulose), acid andfibregroup (5 mg/kg benzoic acidand 3% carboxymethylcellulose) and control group. Regular weight measurementsand diarrhoeamonitoring were carried out during the experiment. Invasive sampling was carried outin selected pigletsfrom 39-42 days of the piglets\u27 life, whereby blood samples were taken, the gastrointestinal tract was measured, the intestinal mucosa was checked for signs of inflammation and tissue samples were taken from the jejunum for intestinal electrophysiological examinations using the Ussing-chamber. The analysis of the parameters considered showed that the intestinal mucosa of the acid group showed significantly lower intestinal scoring values in the stomach and mid-colon areas. The intestinal scoring reflects the degree of inflammation of the intestinal mucosa, which means that the addition of benzoic acid to the ration has a positive effect on intestinal health. The serum parameters analysedshowed no significant differences between the feeding groups. The short-circuit current flow in the acid groupwas significantly lower than in the other feeding groups. This could indicate that less fluid is secreted in the intestinal lumenof the acid feeding-group than in the other groups. In summary, it was possible to identify the extent of inflammationof the intestinal mucosa and the electrical short-circuit current flow of the intestine as biomarkers for intestinal health. In addition, the positive influence of benzoic acid as a feed additive on the intestinal health of pigs was confirmed.Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Das Absetzen ist ein schwieriger Zeitpunkt im Leben eines Ferkels. Es ist mit zahlreichen Stressoren verbunden, die sich negativ auf den Gesundheitszustand und das Wohlbefinden der Ferkel auswirken. Unter anderem zählt die Futterumstellung zu den wichtigsten Stressoren in dieser Lebensphase, welche eine Änderung des Mikrobioms mit sich zieht und nicht selten zu Dysbiosen im Verdauungstrakt führt. Deshalb kommt es nicht selten in dieser sensiblen Lebensphase zu Durchfallerkrankungen, welche nicht nur für die Ferkel negativ ist, sondern auch wirtschaftliche Verluste bedeutet. Ziel dieses Versuchs war es mithilfe von verschiedenen Futtermittelzusätzen unterschiedliche Bedingungen im Verdauungstrakt zu generieren. Dies wurde durch die beiden Futtermittelzusätze Carboxymethylcellulose (CMC) und Benzoesäure umgesetzt. Die Auswirkungen von den Ergänzungen in den Rationen wurde anhand von Wachstum, Durchfall-Monitoring, Darmintegrität und Serumparameter beurteilt. Anhand dieser Daten wurde versucht Biomarker für die Darmgesundheit von Schweinen zu identifizieren. Um die Darmgesundheit der Ferkel zu beeinflussen, wurden bereits die Muttertiere in der späten Trächtigkeit in zwei Fütterungsgruppen eingeteilt. Die Hälfte der Sauen erhielten in der späten Trächtigkeit, sowie während der Laktation 5 mg/kg Benzoesäure, die dem Futter beigemengt wurde. Während der Säugephase erhielten die Ferkel entsprechend ihrer Muttersau Beifutter mit 5 mg/kg Benzoesäure oder das Kontroll-Beifutter. Die Ferkel verblieben nach dem Absetzen (28. Lebenstag) bis zum 32. Lebenstag in den beiden Fütterungsgruppen. Der Hauptversuch des Experiments begann am 33. Lebenstag der Ferkel. Die Ferkel wurden in vier verschiedene Fütterungsgruppen aufgeteilt: Säure-Gruppe (5 mg/kg Benzoesäure), Fasergruppe (3% Carboxymethylcellulose), Säure und Faser-Gruppe (5 mg/kg Benzoesäure und 3% Carboxymethylcellulose) und Kontrollgruppe. Während des Versuchs gab es regelmäßige Gewichtsmessungen, sowie Durchfallmonitoring. Eine invasive Beprobung wurde vom 39.-42. Lebenstag bei ausgewähltem Ferkel durchgeführt, wobei Blutproben entnommen, der Gastrointestinaltrakt vermessen, die Darmschleimhaut auf Entzündungsanzeichen kontrolliert und Gewebestücke vom Jejunum für Darmelektrophysiologische Untersuchungen mittels der Ussing-Kammer entnommen wurden. Die Analyse der betrachteten Parameter zeigte, dass die intestinale Schleimhaut der Säure-Gruppe signifikant geringere Werte im Darm-Scoring zeigte, in den Bereichen Magen und Mitte Kolon. Das Darm-Scoring spiegelt den Grad der Inflammation der Darmmukosa wider, was bedeutet das die Ergänzung von Benzoesäure in der Ration positive Auswirkungen auf die Darmgesundheit hat. Die betrachteten Serumparameter zeigten keine signifikanten Unterschiede zwischen den Fütterungsgruppen. Der Kurzschlussstromfluss bei der Säuregruppe war signifikant geringer als bei den anderen Fütterungsgruppen. Dies könnte darauf hindeuten, dass weniger Flüssigkeit ins Darmlumen in der Säure-Gruppe sekretiert wurde als in den anderen Fütterungsgruppen. Zusammenfassend lässt sich sagen, dass es möglich war, den Inflammationsgrad der Darmschleimhaut, sowie den elektrischen Kurzschluss-Stromfluss des Darms als Biomarker für die Darmgesundheit zu identifizieren. Außerdem wurde der positive Einfluss von Benzoesäure als Futtermittelzusatzstoff auf die Darmgesundheit von Schweinen bestätigt
Nachweis von Parasitenstadien an den Pfoten von Hunden und Schuhen ihrer Halter:innen
Diploma thesis - University of Veterinary Medicine Vienna - 2024This diploma thesis focuses on the detection of parasite stages on the paws of dogs and the soles of their owners\u27 shoes in Vienna and the surrounding area. Between August 2023 and January 2024, the paws of 125 dogs and the soles of 120 owners\u27 shoes were washed. Additionally, fecal samples from the participating dogs were examined. Among the three sample types, paw wash water, sole wash water, and feces, only one sample from each type tested positive for parasites. In the sole wash water unidentified coccidian oocysts were found. In the paw wash water of a Samoyed, an egg of Trichuris vulpis was detected. Additionally, soil mites, insect and plant fragments and fungal spores were found. In the positive fecal sample Cystoisospora ohioensis and T. vulpis were detected. The results refute the null hypothesis that no parasite stages can be detected on paws and shoes. However, the overall detection rate was low. In a study from Vienna, T. vulpis was identified as the most common endoparasite, which is also reflected in the results of this study. Parasites with high zoonotic potential, such as Toxocara canis or Toxoplasma gondii, were not found. Nevertheless, the findings of this thesis show that parasite stages could enter living spaces via paws and shoe soles. There is also the risk that dogs could become infected with various parasites if they lick their contaminated paws after a walk. The results of this diploma thesis highlight the importance of proper disposal of dog feces to prevent the spread of parasites. However, compared to another study from Moscow, fewer parasites were detected on the paws and shoe soles in Vienna.Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Diese Diplomarbeit beschäftigt sich mit dem Nachweis von Parasitenstadien auf den Pfoten von Hunden und den Schuhsohlen ihrer Halter:innen in Wien und der näheren Umgebung. Insgesamt wurden im Zeitraum von August 2023 bis Jänner 2024 125 Hunden und 120 Halter:innen die Pfoten beziehungsweise die Schuhsohlen gewaschen. Zudem wurden Kotproben der teilnehmenden Hunde untersucht. Von den drei Probentypen Pfotenwaschwasser, Sohlenwaschwasser und Kot, war jeweils nur eine Probe positiv. Im Sohlenwaschwasser eines Halters wurden nicht weiter identifizierte Kokzidien-Oozysten gefunden. Im Pfotenwaschwasser eines Samojeden wurde ein Ei von Trichuris vulpis nachgewiesen. Zusätzlich wurden in den Waschproben lebende Erdmilben, Insekten- und Pflanzenteile sowie Pilzsporen gefunden. In der positiven Kotprobe wurde Cystoisospora ohioensis und ebenfalls T. vulpis nachgewiesen. Die Ergebnisse widerlegen die Nullhypothese, dass auf den Pfoten der Hunde und den Schuhsohlen der Halter:innen keine Parasitenstadien nachweisbar sind. Insgesamt war die Nachweisrate aber gering. In einer Studie aus Wien konnte T. vulpis als häufigster Endoparasit festgestellt werden, was sich auch in den Ergebnissen dieser Studie widerspiegelt. Parasiten mit hohen Zoonosepotential, wie Toxocara canis oder Toxoplasma gondii wurden nicht gefunden. Trotzdem zeigen die Ergebnisse dieser Diplomarbeit, dass über die Pfoten und Schuhsohlen Parasitenstadien in Wohnräume gelangen könnten. Es besteht auch die Gefahr, dass sich Hunde über diese Art mit verschiedenen Parasiten infizieren, wenn sie sich nach einem Spaziergang die kontaminierten Pfoten abschlecken. Die Ergebnisse dieser Diplomarbeit zeigen die Wichtigkeit einer korrekten Entsorgung von Hundekot, um die Verbreitung von Parasitenstadien zu verhindern. Im Vergleich zu einer anderen Studie aus Moskau konnten in Wien allerdings weniger Parasiten an den Pfoten und Schuhsohlen nachgewiesen werden
Sequence diversity in mammalian Y chromosomes

Bitte einloggen!Ein tiefgreifendes Verständnis der Demografie und der Evolutionsgeschichte der Arten ist für eine harmonische Zukunft von entscheidender Bedeutung. Zahlreiche Forschungsbereiche beschäftigen sich derzeit mit dieser Thematik. Mit den Fortschritten in der Genomforschung und der Verbesserung von Sequenzierungstechnologien werden täglich genomische Daten von immer mehr Tierarten zur Untersuchung der obengenannten Fragestellungen verfügbar. Das Y Chromosom der Säugetiere, welches, mit Ausnahme der pseudoautosomalen Region, ausschließlich in der männlichen Abstammungslinie vererbt wird hat eine eigene Evolutionsgeschichte. Die Analyse der nicht rekombinierenden männlich-spezifischen Region des Y Chromosoms (MSY) über Zeit und Raum ermöglicht es, die Geschichte und die männlichvermittelte Demografie von Populationen offenzulegen. Trotz des Potenzials wird die Analyse des Y Chromosoms in genomischen Studien oft vernachlässigt. In dieser Arbeit werden Analysestrategien und die Aussagekraft von Y-chromosomalen Untersuchungen bei Säugetieren auf Sequenzebene, mit dem Ziel die Geschichte von Populationen und die von männlichen Individuen vermittelte Demografie aufzuklären, erörtert. Im Rahmen von drei Projekten, an denen zwei Arten - Pferde (Equus caballus) und Rentiere (Rangifer tarandus) - im Fokus standen, habe ich versucht, robuste Verfahren für die Analyse von Short-Read- Sequenzierungsdaten auf dem MSY zu entwickeln und anzuwenden. Die Ergebnisse boten neue Einblicke in die historische Entwicklung der jeweiligen Arten und trugen auch zu einem umfassenderen Verständnis der Analyse der Y-Diversität bei Säugetieren bei. Das erste Projekt bei Pferden befasste sich mit der Unzulänglichkeit bestehender Y-Phylogenien, welche nicht das gesamte Spektrum der Y-Diversität erfassen und keine genaue Datierung zulassen. Zu diesem Zweck wurden Y-Chromosomen-Sequenzierungsdaten von lokalen Pferdepopulationen gemeinsam mit öffentlich verfügbaren Daten von modernen Pferderassen sowie von historischen Pferdeproben analysiert. Die Ergebnisse lieferten die bisher unbestimmte Haplotypendiversität in isolierten Pferdepopulationen. Die Implementierung alter Proben in die moderne Y-Phylogenie zeigte, dass die in domestizierten Pferden beobachteten Y-Haplogruppen erstmals vor etwa 4,000 Jahren auftraten, was mit den autosomalen Ergebnissen übereinstimmt. Das zweite Projekt konzentrierte sich auf das Beschreiben von Y-Haplotypen, die mit der jüngsten selektiven Zucht beim Arabischen Pferd in Zusammenhang stehen. Es wurde eine beträchtliche Anzahl an Araberpferden auf Y-Marker genotypisiert, die im Vorfeld auf Basis von, auf YChromosom angereicherte, Sequenzierungsdaten ermittelt wurden. Es wurden drei verschiedene männliche Linien innerhalb der Araberrasse unterschieden, und die Ergebnisse zeigten die weltweite Verteilung der Araberlinien. Im dritten Projekt wurden Y-chromosomale Sequenzen aus Short-Read-Sequenzierungsdaten assembliert und die erste Y-Haplotyp-Phylogenie für Rentiere erstellt. Diese Y-Phylogenie, die Rentiere und Karibus aus verschiedenen Populationen repräsentiert, spiegelte wichtige demografische Ereignisse, wie etwa die Populationsausweitungen nach der letzten Eiszeit, wieder. Rentier Y und mtDNA wiesen ähnliche Topologien auf, was im Gegensatz zu den Beobachtungen in den meisten anderen Säugetieren steht. Bei den eurasischen Rentieren wurden fünf verschiedene domestizierte Y-Linien identifiziert, was darauf hindeutet, dass mehrere männliche Linien domestiziert wurden. Neben der Beantwortung der spezifischen Fragestellungen trugen die oben genannten Studien auch zum Verständnis der Y-Chromosomen-Diversität in einem breiteren Kontext bei. Die verfeinerte Y-Topologie des Pferdes brachte die Verfolgung der väterlichen Abstammungslinien des Pferdes auf ein Niveau wie es beim Menschen angewendet wird. Die Analyse der arabischen Hengstlinien ist ein Beispiel das Potential der Untersuchung der Y-Haplotypen auf Rasseebene, indem kürzliche populationsdynamische Prozesse offengelegt werden. Beim Rentier, eine im Vergleich zu den meisten Haustieren erst kürzlich domestizierte Art, lieferte die beobachtete hohe Y-chromosomale Diversität neue Erkenntnisse über die Entwicklung der Y-Diversität bei domestizierten Populationen. Zusammenfassend lässt sich sagen, dass die Studie verbesserte Methoden zur YAnalyse vorstellt und das Potential der Y-Analysen zum besseren Verständnis der Demografie und Evolution von Säugetieren aufzeigt.PhD thesis - University of Veterinary Medicine Vienna - 2024

Please log in!Understanding the demography and the evolutionary history of species is crucial for a harmonious future on Earth, and numerous research fields focus on these phenomena. As the genomics studies advance and sequencing technologies improve, genomic data from more animals and different loci are studied daily. The mammalian Y chromosome, inherited solely through male lineages without recombination, except in the pseudoautosomal region, possesses a distinct evolutionary history and sequence content. Studying the sequence variation within the non-recombining male-specific region of the Y chromosome (MSY) over time and space unveils the population history and malemediated demography. Despite its potential, the Y chromosome analysis is often overlooked in genomic studies. In this thesis, I discuss power of mammalian Y-chromosome variation data and the analytical strategies used for Y chromosomal data analysis. Conducting three projects involving two species, horses (Equus caballus) and reindeer (Rangifer tarandus), I endeavored to establish and apply best practices for analyzing short-read sequencing data on the MSY. The outcomes offered novel insights into the respective species and also contributed to a more comprehensive understanding of Y diversity analysis in mammals. In the first project, the inadequacy of existing horse Y phylogenies, which failed to capture the full spectrum of Y diversity and lacked accurate dating was addressed. For this, Y chromosomal sequencing data from isolated horse populations were jointly analyzed with publicly available data from modern and ancient horses. The results revealed previously undetermined haplotypes in isolated horse populations. Placing ancient samples on the modern topology suggested the initial appearance of domestic Y haplotypes around 4,000 years ago, consistent with autosomal findings. The second project focused on resolving Y haplotypes associated with recent selective breeding in the Arabian horses. A large set of Arabian horses were genotyped for Y markers ascertained from target enriched Y chromosome sequencing data. The genotyping results revealed the worldwide distribution of Arabian breeds and distinguished three distinct male lineages in this breed. In the third project, I constructed Y chromosomal contigs from short-read sequencing data and established the first Y haplotype topology for reindeer. The calibrated Y topology, representing reindeer and caribou from diverse populations, revealed major demographic events, including population expansions after the last glacial maximum. Unlike in most mammals, reindeer Y and mtDNA revealed similar topologies. Five different domesticated Y-lineages were identified in Eurasian reindeer suggesting several male lineages underwent domestication in different regions. Besides their specific questions, each study contributed to the understanding of Y chromosomal diversity studies in a broader context. The refined horse Y topology pushed horse Y lineage tracking towards the human-level. The analysis of Arabian stallion lines showcased an exemplary breed-level examination of Y haplotypes, applicable to any other domesticated mammal. In reindeer, which is a recently domesticated species compared to others, the observed high diversity provided new insights into the evolution of Y diversity since the onset of domestication. In summary, the thesis introduces effective data analysis methods and demonstrates the contribution of Y chromosomal diversity analysis to comprehending mammalian demography and evolution
Knochensequester bei Rindern und Neuweltkamelen (2007 - 2024) – Retrospektive Auswertung aus dem Patientengut der Klinik (Tierspitalinformationssystem) - Behandlungsergebnisse und postoperative Überlebenszeit
Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Zwischen 2007 und 2024 wurden 24 Tiere (21 Rinder, 2 Alpakas, 1 Lama) mit Sequestern an einer oder mehreren Gliedmaßenknochen an der Universitätsklinik für Wiederkäuer der Veterinärmedizinischen Universität Wien vorgestellt. Bei diesen Patienten wurden klinische Befunde, Größe der Knochensequester, Behandlungsregime, postoperative Komplikationen, Behandlungserfolg und die postoperative Überlebenszeit retrospektiv analysiert. Zum Zeitpunkt der Diagnosestellung wiesen die Tiere ein mittleres Alter von 24,3 Monaten (±24,1) auf. Von den 30 diagnostizierten Sequestern befanden sich 21 (66,5%) an den Metakarpal- und Metatarsalknochen, der Rest verteilte sich auf andere Gliedmaßenknochen. Bei der Erstuntersuchung wiesen die Patienten einen mittleren Locomotion-Score von 3,3 (von 5; ±1,3) auf. Die Knochensequester waren 1,5 bis 10,0 cm lang, 0,5 bis 4 cm breit und 0,2 bis 1,0 cm tief. Für die statistische Auswertung wurde ihre Größe in drei Größengruppen eingeteilt. Drei von insgesamt 24 Patienten waren aufgrund von ernsthaften Begleiterkrankungen, und damit einer schlechten Prognose, nach der Diagnosestellung euthanasiert worden. Neunzehn Tiere wurden operativ mittels Sequestrektomie behandelt und bei zwei Tieren wurden radiologisch kleine und dünne Sequester diagnostiziert, so dass hierbei deren Resorption abgewartet wurde. Fünf der 21 behandelten Patienten wurden aufgrund postoperativer Komplikationen euthanasiert, und 16 Patienten (76,2%) wurden erfolgreich behandelt. Die mediane kumulative postoperative Überlebenszeit dieser 16 Tiere betrug 42,1 Monate (± 39,5). Weder zwischen dem Alter der Patienten zum Zeitpunkt der Diagnosestellung des Sequesters und der postoperativen Überlebenszeit (p = 0,854) noch zwischen Sequestergröße und Behandlungserfolg (p = 0,77) konnte ein statistisch signifikanter Zusammenhang nachgewiesen werden. Hingegen ließ sich ein signifikanter Zusammenhang zwischen Sequestergröße und postoperativer Überlebenszeit (p = 0,011) feststellen; Tiere der Gruppe 3, also die Patienten mit den größten Sequestern, hatten eine signifikant längere postoperative Überlebenszeit als die der Gruppen 1 und 2 (p = 0,017). Aus den Ergebnissen dieser und anderer Studien lässt sich schlussfolgern, dass die Heilungschancen von Rindern und Neuweltkamelen mit Knochensequestern bei adäquater Wahl des Zeitpunktes für die Sequestrektomie - der Sequester muss im Röntgenbild sehr deutlich von allen umgebenden vitalen Knochenanteilen abgrenzbar sein - als gut einzustufen sind, sofern nicht schwerwiegende Begleiterkrankungen zusätzlich vorliegen.Diploma thesis - University of Veterinary Medicine Vienna - 2024From 2007 to 2024, twenty-four animals (21 cattle, 2 alpacas, 1 llama) with bone sequestration on one or more extremities were presented at the University Clinic for Ruminants at the University of Veterinary Medicine Vienna. In these cases, clinical findings, size of the bone sequestra, treatment, postoperative complications, outcome and postoperative survival time were analyzed retrospectively. At the time of diagnosis, the animals had a mean age of 24.3 months (± 24.1). Of the 30 sequestra diagnosed, 21 (66.5%) were located in the metatarsal and metacarpal bones, with the remaining distributed among other limb bones. The animals had a mean locomotion score of 3.3 (out of 5; ± 1.3) at initial examination. The bone sequestra ranged in size from 1.5 to 10.0 cm in length, 0.5 to 4.0 cm in width and 0.2 to 1.0 cm in depth. Their size was categorized into three groups for statistical evaluation. Three out of the total of 24 patients were euthanized directly after diagnosis due to comorbidities and a poor prognosis. Nineteen of the remaining 21 animals received surgical treatment with sequestrectomy. In two out of the 21 patients small and thin sequestra were diagnosed by radiography, and these resolved on their own. Five out of the 21 treated patients were euthanized because of postoperative complications, while 16 patients (76.2%) had a successful recovery. The median cumulative postoperative survival time for these 16 animals was 42.1 months (± 39.5). No statistically significant correlation was found between the age of the patients at the time of sequestrum diagnosis and postoperative survival time (p = 0.854), nor between the size of sequestra and treatment success (p = 0.77). However, a significant correlation was observed between the size of sequestra and postoperative survival time (p = 0.011). Animals in group 3, the patients with the largest sequestra, had a significantly longer postoperative survival time than those in groups 1 and 2 (p = 0.017). Based on the results of the current study and previous research, it can be concluded that ruminants suffering from bone sequestra have a good chance of recovery if the sequestrectomy is performed at the appropriate time, and assuming there are no other serious concomitant diseases present simultaneously. It is crucial that the bone sequestrum is clearly demarcated from surrounding vital bone parts on the X-ray image

The assessment of paraspinal muscle epimuscular fat in participants with and without low back pain: A case-control study
It remains unclear whether paraspinal muscle fatty infiltration in low back pain (LBP) is i) solely intramuscular, ii) is lying outside the epimysium between the muscle and fascial plane (epimuscular) or iii) or combination of both, as imaging studies often use different segmentation protocols that are not thoroughly described. Epimuscular fat possibly disturbs force generation of paraspinal muscles, but is seldomly explored. This project aimed to 1) compare epimuscular fat in participants with and without chronic LBP, and 2) determine whether epimuscular fat is different across lumbar spinal levels and associated with BMI, age, sex and LBP status, duration or intensity. Fat and water lumbosacral MRIs of 50 chronic LBP participants and 41 healthy controls were used. The presence and extent of epimuscular fat for the paraspinal muscle group (erector spinae and multifidus) was assessed using a qualitative score (0–5 scale; 0 = no epimuscular fat and 5 = epimuscular fat present along the entire muscle) and quantitative manual segmentation method. Chi-squared tests evaluated associations between qualitative epimuscular fat ratings and LBP status at each lumbar level. Bivariate and partial spearman’s rho correlation assessed relationships between quantitative and qualitative epimuscular fat with participants’ characteristics. Epimuscular fat was more frequent at the L4-L5 (X2 = 13.781, p = 0.017) and L5-S1 level (X2 = 27.825, p < 0.001) in participants with LBP compared to controls, which was not found for the higher lumbar levels. The total qualitative score (combined from all levels) showed a significant positive correlation with BMI, age, sex (female) and LBP status (r = 0.23–0.55; p < 0.05). Similarly, the total area of epimuscular fat (quantitative measure) was significantly correlated with BMI, age and LBP status (r = 0.26–0.57; p < 0.05). No correlations were found between epimuscular fat and LBP duration or intensity. Paraspinal muscle epimuscular fat is more common in chronic LBP patients. The functional implications of epimuscular fat should be further explored
Widespread horse-based mobility arose around 2200 Bce in Eurasia
Horses revolutionized human history with fast mobility. However, the timeline between their domestication and their widespread integration as a means of transport remains contentious. Here we assemble a collection of 475 ancient horse genomes to assess the period when these animals were first reshaped by human agency in Eurasia. We find that reproductive control of the modern domestic lineage emerged around 2200 bce, through close-kin mating and shortened generation times. Reproductive control emerged following a severe domestication bottleneck starting no earlier than approximately 2700 bce, and coincided with a sudden expansion across Eurasia that ultimately resulted in the replacement of nearly every local horse lineage. This expansion marked the rise of widespread horse-based mobility in human history, which refutes the commonly held narrative of large horse herds accompanying the massive migration of steppe peoples across Europe around 3000 bce and earlier. Finally, we detect significantly shortened generation times at Botai around 3500 bce, a settlement from central Asia associated with corrals and a subsistence economy centred on horses. This supports local horse husbandry before the rise of modern domestic bloodlines
