Rapid functional impairment of natural killer cells following tumor entry limits anti-tumor immunity
Immune cell dysfunction within the tumor microenvironment (TME) undermines the control of cancer progression. Established tumors contain phenotypically distinct, tumor-specific natural killer (NK) cells; however, the temporal dynamics, mechanistic underpinning and functional significance of the NK cell compartment remains incompletely understood. Here, we use photo-labeling, combined with longitudinal transcriptomic and cellular analyses, to interrogate the fate of intratumoral NK cells. We reveal that NK cells rapidly lose effector functions and adopt a distinct phenotypic state with features associated with tissue residency. NK cell depletion from established tumors did not alter tumor growth, indicating that intratumoral NK cells cease to actively contribute to anti-tumor responses. IL-15 administration prevented loss of function and improved tumor control, generating intratumoral NK cells with both tissue-residency characteristics and enhanced effector function. Collectively, our data reveals the fate of NK cells after recruitment into tumors and provides insight into how their function may be revived
Thoracic electrical impedance tomography identifies heterogeneity in lungs associated with respiratory disease in cattle. A pilot study
Respiratory disease in cattle is a significant global concern, yet current diagnostic methods are limited, and there is a lack of crush-side tests for detecting active disease. To address this gap, we propose utilizing electrical impedance tomography (EIT), a non-invasive imaging technique that provides real-time visualization of lung ventilation dynamics. The study included adult cattle from farms in Western Australia. The cattle were restrained in a crush. A standardized respiratory scoring system, which combined visual, auscultation, and clinical scores, was conducted by two non-conferring clinicians for each animal. The scores were blinded and averaged. During assessment, an EIT electrode belt was placed around the thorax. EIT recordings of ten suitable breaths were taken for analysis before the cattle were released back to the herd. Based on the combined examination scoring, the cattle were categorized as having healthy or diseased lungs. To allow visual interpretation of each breath and enable the creation of the quartile ventilation ratio (VQR), Flow/Tidal Impedance Variation curves (F/TIV) were generated for each breath. The analysis focused on two EIT variables: The novel VQR over time during inhalation and exhalation and global expiratory impedance (TIVEXP) adjusted by breath length. A mixed effects model was used to compare these variables between healthy and diseased cattle. Ten adult cattle of mixed ages were used in the current analysis. Five cattle were scored as healthy and five as diseased. There was a significant difference in the examination scores between the healthy and diseased group (P = 0.03). A significant difference in VQR during inhalation (P = 0.03) was observed between the healthy and diseased groups. No difference was seen in VQR over time during exhalation (P = 0.3). The TIVEXP was not different between groups (P = 0.36). In this study, EIT was able to detect differences in inhalation mechanics when comparing healthy and diseased cattle as defined via clinical examination, highlighting the clinical utility of EIT
Expression of the cobalamin transporters cubam and MRP1 in the canine ileum-Upregulation in chronic inflammatory enteropathy
Chronic inflammatory enteropathy (CIE) in dogs, a spontaneous model of human inflammatory bowel disease (IBD), is associated with a high rate of cobalamin deficiency. The etiology of hypocobalaminemia in human IBD and canine CIE remains unknown, and compromised intestinal uptake of cobalamin resulting from ileal cobalamin receptor deficiency has been proposed as a possible cause. Here, we evaluated the intestinal expression of the cobalamin receptor subunits, amnionless (AMN) and cubilin (CUBN), and the basolateral efflux transporter multi-drug resistance protein 1 (MRP1) in 22 dogs with CIE in comparison to healthy dogs. Epithelial CUBN and AMN levels were quantified by confocal laser scanning microscopy using immunohistochemistry in endoscopic ileal biopsies from dogs with (i) CIE and normocobalaminemia, (ii) CIE and suboptimal serum cobalamin status, (iii) CIE and severe hypocobalaminemia, and (iv) healthy controls. CUBN and MRP1 expression was quantified by RT-qPCR. Receptor expression was evaluated for correlation with clinical patient data. Ileal mucosal protein levels of AMN and CUBN as well as mRNA levels of CUBN and MRP1 were significantly increased in dogs with CIE compared to healthy controls. Ileal cobalamin receptor expression was positively correlated with age, clinical disease activity index (CCECAI) score, and lacteal dilation in the ileum, inversely correlated with serum folate concentrations, but was not associated with serum cobalamin concentrations. Cobalamin receptor downregulation does not appear to be the primary cause of hypocobalaminemia in canine CIE. In dogs of older age with severe clinical signs and/or microscopic intestinal lesions, intestinal cobalamin receptor upregulation is proposed as a mechanism to compensate for CIE-associated hypocobalaminemia. These results support oral supplementation strategies in hypocobalaminemic CIE patients
UV-Schutz bei Nutztieren: Beschattungsnetze in der Freilandhaltung von Schweinen
Nowadays, pig breeds do not have fur. Their skin sensitivity to ultraviolet radiation (UVR) is similar to that of light-skinned humans. To protect animals from the sun, livestock shade cloths are in use. The shading ability of the shade cloth is reported by the „Shade Rate“, which is the percentage of visible light passing through the shade cloth. However, no information is available on the potential protection from UVR. The aim of this study was to evaluate the effectiveness of shading cloths against erythemally weighted solar UV radiation. To estimate the UVR protection, more than 20 electronic UVR-meters (type Sunsaver) were mounted on a true-to-life pig model, positioned on a rotating platform. Measurements were taken in the blazing sun and under different shading cloths. The Sun Protection Factor was calculated from UV exposure measurements without net for all body sites and measurements with shade net. From our measurements we can see that shade cloths can protect pigs effectively from solar UVR. Protection varies depending on the Shade Rate of the shade cloth, the body site and with solar elevation. The Exposure Ratio to Ambient (ERTA) was calculated from UV exposure measurements all over the body sites and nearby measurements from a Biometer Model 501 (Solar Light) and expressed in percentage. Free-range pigs can be effectively protected from solar UV radiation by shade cloths. However, shade structures can substitute neither the primordial fur, nor the possibility to wallow in mud and should be considered as additional measure.Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Die Hautempfindlichkeit von in der konventionellen Landwirtschaft genutzten Schweinen gegenüber ultravioletter Strahlung (UV-Strahlung) ähnelt der von hellhäutigen Menschen. Um Tiere vor der Sonne zu schützen, können Beschattungsnetze verwendet werden. Der Schattierwert der Schutznetze wird als „Shade Rate“ vom Hersteller angegeben und beschreibt den Prozentsatz des sichtbaren Lichts, der durch das Schutznetz hindurchtritt. Es gibt jedoch keine Informationen darüber, wie gut es vor UV-Strahlung schützt. Das Ziel dieser Studie war es, die Effektivität von Schutznetzen gegen Erythem-gewichtete solare UVStrahlung am Schweinemodell zu bewerten. Zur Abschätzung des Schutzes vor UV-Strahlung wurde mit mehr als 20 elektronischen UVStrahlungsmessgeräten, welche an einem lebensechten Schweinemodell angebracht waren, bei strahlendem Sonnenschein und unter verschiedenen Schutznetzen gemessen. Der Sonnenschutzfaktor wurde aus Messungen der UV-Bestrahlungsstärke ohne Netz für alle Körperstellen berechnet und als Verhältnis ausgedrückt. Unsere Messungen zeigen, dass Schutznetze Schweine effektiv vor solarer UV-Strahlung schützen können. Der Schutz variiert je nach Schattierwert des Schutznetzes, dem Sonnenstand und hängt von der Körperstelle ab. Weiters wurde das Verhältnis der Exposition im Vergleich zur ambienten Strahlung berechnet und in Prozent ausgedrückt. Freilaufende Schweine können effektiv vor solarer UV-Strahlung durch Schutznetze geschützt werden. Allerdings können Schattenstrukturen weder das ursprüngliche Fell noch die Möglichkeit, sich im Schlamm zu suhlen, ersetzen, und sollten daher als ergänzende Maßnahme betrachtet werden
Vorstudie zur Etablierung der Methoden für Sonographie, Gelenkinjektionen mit Kunststoff und Kontrastmittel, sowie Präparation der distalen Manica flexoria an 4 Pferde - Kadaverbeinen
Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Die vorliegende Studie dient als Vorstudie für die Untersuchung tiefgefrorener und wieder aufgetauter Eselgliedmaßen, mit dem Ziel, die anatomischen Grundlagen der distalen Gliedmaße des Esels zu erforschen. Die angewandten Methoden umfassen Ultraschall, Kontrastmittel-Röntgen, Kunststoffausgüsse und abschließende Sektion. Besonderes Augenmerk gilt der digitalen Manica flexoria, die bisher bei Eseln unerforscht blieb. Aufgrund begrenzter Eselproben wurde die Vorstudie an Pferdegliedmaßen durchgeführt. Die optimale Vorbereitung der Präparate für den Ultraschall wurde untersucht. Weiters wurden die geeigneten Punktionsstellen für synoviale Strukturen geübt sowie passende Kontrastmittelund Kunststoffmischungen bestimmt, um die Ausbreitung und Kommunikation dieser Strukturen zu visualisieren. Abschließend wurde ein Sektionsprotokoll für die distale Gliedmaße entwickelt. Die Ergebnisse zeigen eine signifikante Verbesserung der Ultraschalldarstellung der tiefgefrorenen und wieder aufgetauten Präparate bei Einwirkung von Ultraschallgel über 24 Stunden, umhüllt von feuchten Tüchern. Die gemeinsame Injektion von Bariumsulfat und Kunststoff erwies sich als ungeeignet für die Darstellung der synovialen Strukturen im Röntgen, als auch bei der Sektion. Hingegen führte die separate Injektion von Bariumsulfat oder Kunststoff zu guten Ergebnissen, wobei Mischverhältnisse von 1:1,5 (Bariumsulfatpulver/Wasser) bzw. 1:1 oder 1:2 (Technovitflüssigkeit/Technovitpulver) erfolgreich waren. Die Punktion der synovialen Struktur an tiefgefrorenen und wieder aufgetauten Gliedmaßen wurde erfolgreich nach den Vorgaben von Moyer et al., 2011 durchgeführt (9). Die Sektion der Manica flexoria digitalis konnte mit dem angepassten Sektionsprotokoll von Garcia et al.,2015 durchgeführt werden (32). Employed methods encompass ultrasound, contrast radiography, anatomic modeling, and concluding dissection, with focus on the previously unexplored digital Manica flexoria. Due to limited donkey samples, the preliminary study was conducted on horse limbs. The optimal preparation of the frozen and thawed specimens for ultrasound was investigated. Suitable puncture sites for synovial structures were practiced and appropriate contrast agent and plastic mixture to visualize the spread and communication of these structures. Subsequently, a dissection protocol for the distal limb was developed. Results indicate a significant improvement in the ultrasound depiction of frozen and thawed specimens when ultrasound gel was applied over 24 hours and the specimens were enveloped in moist towels. The combined injection of barium sulfate and metylmethacrylat (Technovit) proved inappropriate for both X-ray and dissection imaging. Conversely, separate injections of either barium sulfate or metylmethacrylat yielded successful results, with mixing ratios of 1:1.5 (barium sulfate powder/water) and 1:1 or 1:2 (Technovit liquid/Technovit powder) being effective. Puncture of synovial structures in frozen and thawed limbs was successfully performed following the guidelines of Moyer et al., 2011(9). The dissection of the digital manica flexoria was accomplished using the modified dissection protocol by Garcia et al., 2015 (32). The primary outcome is the successful development of a protocol for the comprehensive examination of frozen and thawed donkey limbs
Textureigenschaften von konventionellem Käse und analogen veganen Erzeugnissen
Diplomarbeit - Veterinärmedizinische Universität Wien - 2024In dieser Arbeit wurden die Textureigenschaften und die Zusammensetzung von konventionellem Käse und veganen Analogprodukten bestimmt und verglichen. Die Textureigenschaften wurden mittels Scherkraftmessung (Warner-Bratzler-Methode) und Texturprofilanalyse (TPA) bestimmt. Im Vorfeld wurden Angaben zu Typ und Zusammensetzung veganer Käse-Analoga erhoben. Es zeigte sich, dass die mengenmäßig bedeutendsten Inhaltsstoffe solcher Produkte Wasser, Stärke und Pflanzenfette (Kokosfett oder Rapsöl) waren. Umgekehrt waren die Proteingehalte niedriger als in vergleichbaren Käsen. Für die Texturmessungen wurden Cheddar, Reibkäse, Gouda, Bergkäse, Feta, Mozzarella in Blöcken und vergleichbare vegane Produkte herangezogen. Da Messungen mit schnittfesten Käsen zeigten, dass die Probentemperatur einen deutlichen Einfluss auf die Messwerte hatte, wurden die Proben vor der Untersuchung auf Raumtemperatur gebracht. Für Reibkäse und Feta wurden immer signifikant höhere Werte für die Scherkraft, Härte, Gummiartigkeit, Adhäsion, Kohäsion und Elastizität gemessen als für die vergleichbaren veganen Erzeugnisse. Bei 6 und 14 Monate gereiftem Cheddar war die Scherkraft höher als beim veganen Erzeugnis; die Härte, Gummiartigkeit, Kohäsion, und Elastizität waren aber niedriger. Mozzarella wies höhere Scherkraftwerte auf als das vegane vergleichbare Produkt. Für die Produkte Gouda, Bergkäse und ein äußerlich vergleichbares veganes Erzeugnis wurde nur die Scherkraft bestimmt, wobei das vegane Erzeugnis zwischen den beiden Käsen lag. Textural properties were analyzed by shear force measurement (Warner-Bratzler method) and Texture Profile Analysis (TPA). Prior to analysis, information was collected on type and composition of vegan cheese analogs. The major ingredients (on a quantitative basis) were water, starch and vegetable fat/oil (coconut fat, rapeseed oil). The protein content was lower than that of conventional cheese. Texture studies were done for Cheddar, grated cheese, Gouda, „Bergkäse“, Feta, Mozzarella and plant-based analogous products. Since measurements in hard cheeses demonstrated a major effect of temperature on the results, samples were brought to ambient temperature before analysis. Grated cheese and Feta demonstrated higher values for shear force, hardness, gumminess, adhesiveness, cohesiveness and springiness than the plant-based analogous products. Cheddar (aged for 6 and 14 months) had higher shear force values than the vegan product, whereas hardness, gumminess, adhesiveness, cohesiveness and springiness were lower. Mozzarella had higher shear strength values than the vegan analogon. For Gouda, „Bergkäse“ and an apparently analogous vegan product, only shear force could be determined, with the vegan product showing an intermediate value. Based on the results, it can be expected that consumers will experience a different texture when biting and chewing the investigated cheeses and their vegan counterparts
Measuring kea parrot (Nestor notabilis) stress and reproductive hormone metabolites by non-invasive faecal sampling
Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Ziel dieser Studie war es, endokrinologische Gegebenheiten von Kea-Papageien (Nestor notabilis) mithilfe einer nicht-invasiven Methode der Kotprobenahme zu entschlüsseln. Der erste Schritt war die Validierung von Enzymimmunoassays (EIAs) für Metaboliten von Reproduktionshormonen - Androgene, Östrogene und Progesteron. Außerdem wurden Referenzwerte der validierten Hormonassays sowie der in einer unveröffentlichten Studie validierten Glukokortikoid-Metaboliten ermittelt. Dies sowohl während als auch außerhalb der Brutsaison. An der Studie nahmen zwei Gruppen von in Gefangenschaft lebenden Kea-Papageien teil, bestehend aus Erwachsenen und Jungtieren, was für die Validierung des EIA erforderlich war, da der Test, der den größten Unterschied zwischen Erwachsenen und Jungtieren aufwies, für die Art am besten geeignet war. Obwohl die Studie nicht in der Lage war, einen EIA für einen der getesteten Metaboliten der Fortpflanzungshormone zu validieren, zeigten die Glukokortikoid-Metaboliten in den Sommermonaten höhere Werte, was auf Hitzestress hindeutet. Außerdem wiesen die beiden Gruppen, die von zwei verschiedenen Standorten stammten, unterschiedliche Glukokortikoid-Metabolitenwerte auf, wobei sie im Tiergarten Wels signifikant höher waren als in der Forschungsstation Haidlhof. Monate nach der Probenahme wurde bei den Vögeln im Tiergarten Wels eine Aspergillose diagnostiziert, was zu der Annahme führte, dass die Glukokortikoidspiegel vor einer Erkrankung und vor dem Auftreten von Symptomen ansteigen. Die Ergebnisse zeigten, dass es herausfordernd sein kann einen EIA für eine Tierart zu validieren. Darüber hinaus zeigte die Studie, dass Glukokortikoid-Metaboliten ein geeigneter Marker für Umweltstressoren und Gesundheitsprobleme bei Kea sein könnten.Diploma thesis - University of Veterinary Medicine Vienna - 2024This study aimed to unravel endocrinological dynamics of kea parrots (Nestor notabilis) using a non-invasive method of faecal sampling. The first step was to validate enzyme immunoassays (EIAs) for measuring metabolites of reproductive hormones - androgens, oestrogens, and progesterone. Additionally, baseline levels of the assays, along with glucocorticoid metabolites validated in an unpublished study, were established within and outside of the breeding season. Two groups of captive kea parrots participated in the study, consisting of adults and juveniles, necessary for EIA validation, as the test showing the largest difference between adults and juveniles was rated the most suitable for the species. Although the study was not able to validate an EIA for any of the tested reproductive hormone metabolites, glucocorticoid metabolite baselines showed higher levels during the summer months, indicating heat-stress. Furthermore, the two groups, originating from two different locations, showed different glucocorticoid metabolite levels being significantly higher at Tiergarten Wels compared to Haidlhof Research Station. Months after sample collection, the birds at Tiergarten Wels were diagnosed with Aspergillosis, which led to the theory that glucocorticoid levels rise prior to an illness and to the display of symptoms. The results showed that it can be challenging to validate EIAs for a species. Furthermore, the study provided evidence that glucocorticoid metabolites might provide a suitable marker of environmental stressors and health issues in kea
Identification of cost-effective biosecurity measures to reduce Salmonella along the pork production chain
The continued occurrence of salmonellosis cases in Europe attributed to the consumption of pork products highlights the importance of identifying cost-effective interventions. Certain biosecurity measures (BSMs) may be effective in reducing the prevalence of specific pathogens along the pork production chain and their presence in food products. The objective of this study was to identify pathogen-specific, cost-effective BSMs to reduce Salmonella at different stages of the pork production chain in two European countries - Austria (AT) and the United Kingdom (UK). For this purpose, a cost-benefit analysis was conducted based on the epidemiological output of an established quantitative microbiological risk assessment that simulated the implementation effect of the BSMs based on their risk ratios. For each of the BSMs, the associated costs and benefits were assessed individually and country-specifically. For both AT and UK, nine different BSMs were evaluated assuming a countrywide implementation rate of 100%. The results showed that four BSMs were cost-effective (benefit-cost ratio > 1) for AT and five for the UK. The uncertainty regarding the cost-effectiveness of the BSMs resulted from the variability of individual risk ratios, and the variability of benefits associated with the implementation of the BSMs. The low number of cost-effective BSMs highlights the need for holistic risk-based models and economic assessments. To increase the willingness to implement BSMs and maximize the benefits for stakeholders, who carry the majority of the implementation costs, epidemiological assessments of BSM effectiveness should consider the impact on several relevant pathogens simultaneously
Duration of maternally derived antibodies of porcine parvovirus in growing pigs and presence of antibodies in gilts and sows vaccinated with three different parvovirus vaccines
While gilts and sows are regularly vaccinated against the porcine parvovirus (PPV), little is known on the presence of antibodies in vaccinated sows nor the decline of maternally derived antibodies (MDA) in their offspring. On twelve farms serum samples were taken from 180 gilts and sows vaccinated at least twice with one of three different commercial PPV vaccines. On nine farms, additional 270 serum samples were collected from growing pigs of three different age categories. All 450 samples were examined for PPV antibodies (Abs) by ELISA and haemagglutination inhibition (HI) assay. In total, 65% of all gilts vaccinated twice with either vaccine 1 or vaccine 3 were seronegative by HI assay. In each farm, there were at least three animals with high Ab titres (≥ 1:1280) indicating the presence of PPV in all twelve study farms. However, PPV DNA could not be detected in collected faecal samples. While low to moderately high Ab titres (1:10-1:640) were measured in 98% of twelve-weeks-old pigs, ELISA was only positive in 30% of the same pigs. Though, the statement on the duration of MDA may depend on the applied test, we could confirm an exponential decay of MDA. In addition, we could demonstrate that applied serological tools are insufficient for the confirmation of successful vaccination
A systematic review and meta-analysis on the efficacy of vaccination against colibacillosis in broiler production
Colibacillosis, a disease caused by Escherichia coli in broiler chickens has serious implications on food safety, security, and economic sustainability. Antibiotics are required for treating the disease, while vaccination and biosecurity are used for its prevention. This systematic review and meta-analysis, conducted under the COST Action CA18217-European Network for Optimization of Veterinary Antimicrobial Treatment (ENOVAT), aimed to assess the efficacy of E. coli vaccination in broiler production and provide evidence-based recommendations. A comprehensive search of bibliographic databases, including, PubMed, CAB Abstracts, Web of Science and Agricola, yielded 2,722 articles. Following a defined protocol, 39 studies were selected for data extraction. Most of the studies were experimental infection trials, with only three field studies identified, underscoring the need for more field-based research. The selected studies reported various types of vaccines, including killed (n = 5), subunit (n = 8), outer membrane vesicles/protein-based (n = 4), live/live-attenuated (n = 16), and CpG oligodeoxynucleotides (ODN) (n = 6) vaccines. The risk of bias assessment revealed that a significant proportion of studies reporting mortality (92.3%) or feed conversion ratio (94.8%) as outcomes, had "unclear" regarding bias. The meta-analysis, focused on live-attenuated and CpG ODN vaccines, demonstrated a significant trend favoring both vaccination types in reducing mortality. However, the review also highlighted the challenges in reproducing colibacillosis in experimental setups, due to considerable variation in challenge models involving different routes of infection, predisposing factors, and challenge doses. This highlights the need for standardizing the challenge model to facilitate comparisons between studies and ensure consistent evaluation of vaccine candidates. While progress has been made in the development of E. coli vaccines for broilers, further research is needed to address concerns such as limited heterologous protection, practicability for application, evaluation of efficacy in field conditions and adoption of novel approaches
