    No full text
Diploma thesis - University of Veterinary Medicine Vienna - 2024Introduction: Carboxyhemoglobin forms when carbon monoxide binds to hemoglobin. Carboxyhemoglobin can be a reliable indicator of blood carbon monoxide levels. Critically ill patients often tend to form carboxyhemoglobin. Immune-mediated hemolytic anemia is a severe disease and the most common form of hemolytic anemia in dogs. Hypothesis: This thesis aims to assess carboxyhemoglobin levels in dogs with immune-mediated hemolytic anemia in contrast to those with non-hemolytic anemia and dogs suffering from acute hemorrhagic diarrhea syndrome. The hypothesis is that measurement of carboxyhemoglobin is higher in dogs with immune-mediated hemolytic anemia than in animals with non-hemolytic anemia and dogs with acute hemorrhagic diarrhea syndrome. Material and Methods: Retrospective study based on medical records at the clinic for small animals of the University of Veterinary Medicine in Vienna, Austria. The data from patients hospitalized for immune-mediated hemolytic anemia (n=28), non-hemolytic anemia (n=29), or for acute hemorrhagic diarrhea syndrome (Control, n=44) were analyzed retrospectively. Results and Conclusion: Carboxyhemoglobin significantly differed between dogs with immune-mediated hemolytic anemia and dogs with non-hemolytic anemia. Also, there was a significant difference in dogs with immune-mediated hemolytic anemia in comparison to those with acute hemorrhagic diarrhea syndrome. There was no significant difference in carboxyhemoglobin levels between the patients with non-hemolytic anemia and the ones with acute hemorrhagic diarrhea syndrome.Diploma thesis - University of Veterinary Medicine Vienna - 2024Introduction: Carboxyhemoglobin forms when carbon monoxide binds to hemoglobin.

Diploma thesis - University of Veterinary Medicine Vienna - 2024This thesis aimed to determine whether there is a demand for a simulator in the ultrasound diagnostic training within equine medicine and what the requirements for such a simulator are. Additionally, it sought to examine the fundamental competencies of students and explore whether there are skills that need to be expanded upon or introduced and how a simulator can be useful in this aspect. Given the increasing digitalization and the rising expectations for students to master fundamental competencies by the end of their studies, the advancement of ultrasound education in veterinary medicine is gaining importance. Using semi-structured interviews with experts in equine medicine and subsequent analysis, an attempt was made to answer these research questions. The results indicate that the interviewees believed that an ultrasound simulator in equine medicine would have a positive impact on education and training. The key issues identified by the interviewees in teaching include the lack of practice animals, teaching staff, and sufficient practice time, or the ability for students to practice freely outside of teaching hours. Additionally, teaching content cannot adequately be presented to students due to the lack of opportunities to demonstrate pathologies on animals. According to experts, the current skills of students are primarily in reproduction and andrology, colic diagnostics, and orthopaedics with a focus on the distal limb. According to the interviewees, some skills cannot be deepened or taught at all due to a lack of cases in the clinic. There is also a lack of opportunities for students to consolidate and expand their Day One Competencies. With the numerous potential applications of a simulator in ultrasound education, it could address the problems identified by the interviewees. Furthermore, some of respondents also believed that a simulator would eliminate the need for practice animals in teaching. These results suggest that an ultrasound simulator could be a promising solution for some of the challenges identified in this study.

Diploma thesis - University of Veterinary Medicine Vienna - 2024Abomasal ulcers in growing calves are a well-known and under-recognized problem worldwide. In this study, 53 samples of perforating abomasal ulcers were collected from suckler calves in Canada to analyze and compare microbial composition. The samples were shipped refrigerated and stored at -80 degrees Celsius at the Sherbrooke Research and Development Center. DNA extraction of the samples was performed by Dr. Renée M. Petri in Quebec according to the manufacturer\u27s protocol of. PowerSoil® DNA Isolation Kit (QIAGEN®, Venlo, The Netherlands). After DNA extraction, the DNA concentration was determined and 16S rRNA sequencing was performed using the Illumina MiSeq and the V2 500 cycle kit. Sequence processing and analysis was performed in the program R-Studio version 4.1.2 (RStudio Team (2020). RStudio: Integrated Development for R. RStudio, PBC, Boston, MA). The quality control turned out that the samples were of good quality to be further analyzed and taxonomically assigned. From the taxonomic determination, graphs could be generated to pro-vide a visual insight into the microbial composition of the abomasum samples. The microbiome of the animals with perforating abomasal ulcers differed from the animals without. The difference in the number and distribution of bacterial families and genera also appears to be greater in animals younger than 30 days. The microbiome shows a major development during this period. In older animals, the microbiome also changes due to the presence of an ulcer in the abomasum, although animals from the younger age group appear to react much more sensitively to this. The use of antibiotics also changes the microbial composition in the abomasum, as expected. The question of a potentially pathogenic microorganism in connection with abomasal ulcers appears to be the bacterial family Clostridiceae_1. Whether with or without antibiotics and regardless of age, this family of bacteria always seems to be present in ulcerations. As this study relates to the visual differences in the graphs, only tendencies and assumptions can be made, which must be substantiated with the aid of statistical data analysis.

Lethal Borna disease virus 1 infections of humans and animals - in-depth molecular epidemiology and phylogeography

    No full text
Borna disease virus 1 (BoDV-1) is the causative agent of Borna disease, a fatal neurologic disorder of domestic mammals and humans, resulting from spill-over infection from its natural reservoir host, the bicolored white-toothed shrew (Crocidura leucodon). The known BoDV-1-endemic area is remarkably restricted to parts of Germany, Austria, Switzerland and Liechtenstein. To gain comprehensive data on its occurrence, we analysed diagnostic material from suspected BoDV-1-induced encephalitis cases based on clinical and/or histopathological diagnosis. BoDV-1 infection was confirmed by RT-qPCR in 207 domestic mammals, 28 humans and seven wild shrews. Thereby, this study markedly raises the number of published laboratory-confirmed human BoDV-1 infections and provides a first comprehensive summary. Generation of 136 new BoDV-1 genome sequences from animals and humans facilitated an in-depth phylogeographic analysis, allowing for the definition of risk areas for zoonotic BoDV-1 transmission and facilitating the assessment of geographical infection sources. Consistent with the low mobility of its reservoir host, BoDV-1 sequences showed a remarkable geographic association, with individual phylogenetic clades occupying distinct areas. The closest genetic relatives of most human-derived BoDV-1 sequences were located at distances of less than 40 km, indicating that spill-over transmission from the natural reservoir usually occurs in the patient´s home region

Four new genome sequences of the Pallas\u27s cat (Otocolobus manul): an insight into the patterns of within-species variability

    No full text
Manul (Otocolobus manul) is the only representative of the genus Otocolobus, which makes up the Leopard Cat lineage along with the genus Prionailurus. Their habitat is characterized by harsh environmental conditions. Although their populations are probably more stable than previously thought, it is still the case that their population size is declining. Conservation programs exist to protect manuls, but those based on captive breeding are often unsuccessful due to their increased susceptibility to diseases. The manul is therefore a suitable model species for evolutionary and diversity studies as well as for studying mechanisms of adaptation to harsh environment and mechanisms of susceptibility to diseases. Recently, the genome of the O. manul based on nanopore long-range sequencing has been published. Aiming to better understand inter- and intraspecific variation of the species, we obtained information on genome sequences of four other manuls, based on whole genome resequencing via the Illumina platform. On average, we detected a total of 3,636,571 polymorphic variants. Information on different types of structural variants and on the extent of SNP homozygosity, not available from the reference genome, was retrieved. The average whole-genome heterozygosity was almost identical to that found in the O. manul reference genome. In this context, we performed a more detailed analysis of the candidate gene EPAS1 potentially related to adaptation to the hypoxic environment. This analysis revealed both inter- and intraspecific variation, confirmed the presence of a previously described non-synonymous substitution in exon 15 unique to manuls and identified three additional unique non-synonymous substitutions located in so far not analyzed EPAS1 exonic sequences. The analysis of lncRNA located in the intron 7 of EPAS1 revealed interspecific variability and monomorphic nature of the sequence among analyzed manuls. The data obtained will allow more detailed analyses of the manul genome, focusing on genes and pathways involved in their adaptation to the environment and in susceptibility to diseases. This information can be helpful for optimizing conservation programs for this understudied species

Diploma thesis - University of Veterinary Medicine Vienna - 2024The aim of the current study was to investigate the effects of natural betaine and synthetic betaine hydrochloride (HCl) on gut health, gene expression, and growth performance in broiler chickens. One hundred and five 1-day-old broiler chicks were divided into three groups: control, natural betaine (NB), and synthetic betaine HCl (SB). Ussing chamber technique was utilized to assess intestinal permeability, real-time quantitative PCR was applied for the analysis of genes expression, and body performance metrics were measured. The findings demonstrated that chickens allocated to group NB exhibited enhanced growth performance, characterized by elevated body weight and improved feed efficiency in comparison to their counterparts in group SB. Ussing chamber analysis revealed a reduction in intestinal permeability in group NB, suggesting a maintenance of gut barrier integrity. Gene expression analysis highlighted that natural betaine supplementation led to more balanced expression of tight junction proteins, thereby contributing to stable gut integrity. In contrast, the synthetic betaine HCl fed group exhibited inconsistent barrier functionality, resulting in increased paracellular permeability. Furthermore, pro-inflammatory cytokines were more consistently upregulated in group SB, indicating ongoing immune activation, while group NB had no effect on cytokine levels. The results suggest that natural betaine has the potential to promote gut health and growth performance compared to synthetic betaine HCl. Further research is recommended to explore these effects in more detail, particularly under pathogen-challenged conditions

GWAS and comparative genomics reveal candidate antibiotic resistance genes in the avian pathogen Gallibacterium anatis for six widespread antibiotics

    No full text
Gallibacterium anatis is a Gram-negative bacterium found in the respiratory and genital tracts of various animals, primarily poultry. Its association with septicemia and high mortality in poultry, along with the rise in multidrug-resistant strains, has amplified concerns. Recent research uncovered significant variability in antibiotic resistance profiles among G. anatis isolates from different Austrian flocks, and even between different organs within the same bird. In response, in the present study 60 of these isolates were sequenced and a combination of comparative genomics and genome-wide association study (GWAS) analysis was applied to understand the genetic variability of G. anatis across flocks and organs and to identify genes related to antibiotic resistance. The results showed that each flock harbored one or two strains of G. anatis with only a few strains shared between flocks, demonstrating a great variability among flocks. We identified genes associated with resistance to nalidixic acid, trimethoprim, cefoxitin, tetracycline, ampicillin and sulfamethoxazole. Our findings revealed that G. anatis may develop antibiotic resistance through two mechanisms: single-nucleotide mutations and the presence of specific genes that confer resistance. Unexpectedly, some tetracycline-resistant isolates lacked all known tetracycline-associated genes, suggesting the involvement of novel mechanisms of tetracycline resistance that require additional exploration. Furthermore, our functional annotation of resistance genes highlighted the citric acid cycle pathway as a potential key modulator of antibiotic resistance in G. anatis. In summary, this study describes the first application of GWAS analysis to G. anatis and provides new insights into the acquisition of multidrug resistance in this important avian pathogen

Streptococcus uberis is a gram-positive bacterium within the Streptococcaceae group and one of the most important mastitis pathogens in dairy cows. Due to the increase in antimicrobial resistance, it is crucial to analyze and monitor both the resistance patterns of the pathogen strains and the sensitivity of the antimicrobial agents in the one health context. The aim of this diploma thesis, in collaboration with the Istituto Zooprofilattico Sperimentale delle Venezie, was to shed light on antimicrobial resistance of Streptococcus uberis isolates and their development over a period of five years (2019 - 2023). Through bacteriological examinations, Streptococcus uberis was identified in 859 quarter milk samples during the observation period. Antimicrobial resistance was determined using the microbouillon dilution method. The highest values for antimicrobial resistance of Streptococcus uberis were found for trimetoprim sulfonamide with a mean of 35.43 %, lincomycin 32.84 %, pirlimycin 32.66 %, tylosin 32.47 %, oxacillin 24.51 % and erythromycin 20.22 %, respectively. The proportion of multiresistant Streptococcus uberis strains (mean 22.79 %) first increased and then decreased again significantly during the study period. Overall, no increase in antimicrobial resistance was recorded in the defined period. While some of the antibiotics tested (amoxicillin clavulanic acid, oxacillin, trimetoprim sulfonamide) were subject to significant fluctuations in the respective years, others (kanamycin, penicillin, spiramycin) showed a more stable trend. The reasons for this could be the varying use of the antimicrobial agents and the mandatory documentation (electronic prescription) of the antibiotics used in South Tyrol, which is in place since 2019. Reasons for the large differences in antimicrobial resistance between the antimicrobial agents are various and may be due to the different physiology of the antibiotics or the different resistance mechanisms of Streptococcus uberis. Ultimately, reducing the use of antibiotics by improving animal husbandry systems, management practices and competent veterinary care are the prerequisites for preventing antimicrobial resistance in veterinary medicine.

The aim of the present work is to test different indices for the differentiation between normal and brachycephalic cat skulls and to evaluate their effectiveness. In addition, the reliability of the individual indices will be compared using different screening methods. The study included eleven normal and four brachycephalic macerated skulls of adult cats. The epiphyseal fossae were closed. A total of four measurements were determined: the total length of the skull, the facial skull length, the brain skull length and the zygomatic bone width. These measurements were recorded using calipers on the one hand, and digitally using X-ray and computed tomography images on the other hand. Three indices were then calculated. There were two length-to-lenth-ratios and one length-to-width ratio. All values were recorded in an Excel spreadsheet and analyzed using descriptive and inductive statistics. A mixed model analysis revealed a significant difference between normal and brachycephalic cats. The reliability of the indices and the correlation between the methods were tested using Cronbach\u27s alpha and Pearson\u27s correlation coefficient (p ≤ 0.01). Computed tomography provided values with the least significant difference to the sliding gauge, whereby the LW-Index showed the highest reliability and therefore proved to be the best screening method. It is recommended that in future studies the measurements are carried out on larger populations and on live animals to possibly determine a cut-off value. A comparison of different brachycephalic breeds would also be of interest.

Range of Ixodes laguri, a nidicolous tick that parasitizes critically endangered rodents, with details on its western distribution limit in Austria

    No full text
The nidicolous tick Ixodes laguri is a nest-dwelling parasite of small mammals that mainly infest rodents of the families Cricetidae, Gliridae, Muridae and Sciuridae. There is no proven vectorial role for I. laguri, although it is suggested that it is a vector of Francisella tularensis. In this study, a first map depicting the entire geographical distribution of I. laguri based on georeferenced locations is presented. For this purpose, a digital data set of 142 georeferenced locations from 16 countries was compiled. Particular attention is paid to the description of the westernmost record of I. laguri in the city of Vienna, Austria. There, I. laguri is specifically associated with its main hosts, the critically endangered European hamster (Cricetus cricetus) and the European ground squirrel (Spermophilus citellus). These two host species have also been mapped in the present paper to estimate the potential distribution of I. laguri in the Vienna metropolitan region. The range of I. laguri extends between 16-108∘ E and 38-54∘ N, i.e. from Vienna in the east of Austria to Ulaanbaatar, the capital of Mongolia. In contrast to tick species that are expanding their range and are also becoming more abundant as a result of global warming, I. laguri has become increasingly rare throughout its range. However, I. laguri is not threatened by climate change, but by anthropogenic influences on its hosts and their habitats, which are typically open grasslands and steppes. Rural habitats are threatened by the intensification of agriculture and semi-urban habitats are increasingly being destroyed by urban development
