Innovative problem solving by wild falcons
Innovation (i.e., a new solution to a familiar problem, or applying an existing behavior to a novel problem) plays a fundamental role in species’ ecology and evolution. It can be a useful measure for cross-group comparisons of behavioral and cognitive flexibility and a proxy for general intelligence. Among birds, experimental studies of innovation (and cognition more generally) are largely from captive corvids and parrots, though we lack serious models for avian technical intelligence outside these taxa. Striated caracaras (Phalcoboenus australis) are Falconiformes, sister clade to parrots and passerines, and those endemic to the Falkland Islands (Malvinas) show curiosity and neophilia similar to notoriously neophilic kea parrots and face similar socio-ecological pressures to corvids and parrots. We tested wild striated caracaras as a new avian model for technical cognition and innovation using a field-applicable 8-task comparative paradigm (adapted from Rössler et al.and Auersperg et al.). The setup allowed us to assess behavior, rate, and flexibility of problem solving over repeated exposure in a natural setting. Like other generalist species with low neophobia, we predicted caracaras to demonstrate a haptic approach to solving tasks, flexibly switching to new, unsolved problems and improving their performance over time. Striated caracaras performed comparably to tool-using parrots, nearly reaching ceiling levels of innovation in few trials, repeatedly and flexibly solving tasks, and rapidly learning. We attribute our findings to the birds’ ecology, including geographic restriction, resource unpredictability, and opportunistic generalism, and encourage future work investigating their cognitive abilities in the wild
Dairy cows housed both full- and part-time with their calves form strong maternal bonds
Dairy cow and calf are typically separated shortly after calving preventing the formation of a maternal -filial bond. To allow some cow -calf contact, part-time contact during the first weeks is thought to be a feasible solution, but it is unknown if it weakens maternal bond, i.e., if maternal motivation is lower. This study aimed to investigate how different amounts of calf contact (full-time, part-time, and no contact) affect cows\u27 maternal motivation. Using pneumatic push gates, we assessed cows\u27 motivation to access their own calf using the maximum price paid (MPP) method. To mitigate frustration at high prices, cows could also access an unfamiliar calf at a constant low price. We expected that cows would access the unfamiliar calf when reaching the maximum price that they were motivated to pay to access their own calf. Following 48 h in a calving pen, cow -calf pairs were allocated to three different treatments: full-time (23 h contact/d, 28 pairs), part-time (10 h contact/d, 27 pairs), and no contact (0 h contact/d, 26 pairs). Approximately 40 d after calving, cows were trained to pass through each of two push gates: one leading to their own and one leading to an unfamiliar calf. The weight on the gate leading to the cows\u27 own calf increased following each passing, while the gate leading to the unfamiliar calf remained light. Cows were tested once daily, until they failed to pass through the gate leading to their own calf on two consecutive days. MPP was analysed using a Cox\u27s proportional hazards mixed effects model. Fewer nocontact cows than full- and part-time cows fulfilled the learning criteria. Furthermore, no -contact cows paid a lower maximum price compared to the two contact treatments, while the MPP of full- and part-time cows did not differ. Most cows remained in the start box if they did not pass the gate to their own calf, indicating that an unfamiliar calf could not substitute for their own calf at high prices. We conclude that cows with part-time calf contact form a maternal bond of similar strength to cows with full-time calf contact. Additionally, cows separated from their calf at 48 h after calving have a weaker maternal motivation at 40 days postpartum
Rethinking connectivity modeling for high-mobility ungulates: insights from a globally endangered equid
ContextMaintaining connectivity is crucial for wildlife conservation in human-occupied landscapes. Structural connectivity modeling (SCM) attempts to quantify the degree to which physical features facilitate or impede movement of individuals and has been widely used to identify corridors, but its accuracy is rarely validated against empirical data.ObjectivesWe evaluated SCM\u27s ability to identify suitable habitat and corridors for onagers (Equus hemionus onager) through a comparison with functional connectivity (i.e., actual movement of individuals) using satellite tracking data.MethodsWe used MaxEnt to predict suitable habitat and evaluated the ability of three SCM approaches: circuit theory, factorial least cost path, and landscape corridors approaches to identify corridors. The performance of the three SCM approaches was validated against independently collected GPS telemetry data.ResultsOnagers selected water sources and dense vegetation while avoiding areas grazed intensely by livestock. The three approaches to SCMs identified similar movement corridors, which were interrupted by roads, affecting major high-flow movement corridors. The SCMs overlapped with functional connectivity by about 21%.ConclusionMovement corridors derived from SCMs did not align with the locations or intensity of corridors identified using the functional connectivity model. This finding suggests that SCMs might have a tendency to overestimate landscape resistance in areas with low habitat suitability. Therefore, SCM may not adequately capture individual decisions about habitat selection and movement. To protect corridors linking suitable habitat, data on functional connectivity (i.e., telemetry data) can be coupled with SCM to better understand habitat selection and movements of populations as a consequence of landscape features
Targeting NRAS via miR-1304-5p or farnesyltransferase inhibition confers sensitivity to ALK inhibitors in ALK-mutant neuroblastoma
Targeting Anaplastic lymphoma kinase (ALK) is a promising therapeutic strategy for aberrant ALK-expressing malignancies including neuroblastoma, but resistance to ALK tyrosine kinase inhibitors (ALK TKI) is a distinct possibility necessitating drug combination therapeutic approaches. Using high-throughput, genome-wide CRISPR-Cas9 knockout screens, we identify miR-1304-5p loss as a desensitizer to ALK TKIs in aberrant ALK-expressing neuroblastoma; inhibition of miR-1304-5p decreases, while mimics of this miRNA increase the sensitivity of neuroblastoma cells to ALK TKIs. We show that miR-1304-5p targets NRAS, decreasing cell viability via induction of apoptosis. It follows that the farnesyltransferase inhibitor (FTI) lonafarnib in addition to ALK TKIs act synergistically in neuroblastoma, inducing apoptosis in vitro. In particular, on combined treatment of neuroblastoma patient derived xenografts with an FTI and an ALK TKI complete regression of tumour growth is observed although tumours rapidly regrow on cessation of therapy. Overall, our data suggests that combined use of ALK TKIs and FTIs, constitutes a therapeutic approach to treat high risk neuroblastoma although prolonged therapy is likely required to prevent relapse
Parasitologie in der Zootiermedizin: Literaturrecherche und Analyse parasitologischer Beiträge in den Verhandlungsberichten des Internationalen Symposiums über die Erkrankungen der Zoo- und Wildtiere und deren Nachfolgeveranstaltungen 2001 bis 2023
Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Zootiere können aufgrund des Zusammenlebens verschiedener Spezies in näherer Umgebung und veränderten klimatischen Verhältnissen im Vergleich zu ihrem natürlichen Lebensraum mit einem breiten Spektrum an Parasiten befallen sein. Infektionen mit Parasiten auf ein möglichst geringes Ausmaß zu begrenzen, sollte somit das Ziel jeder zoologischen Einrichtung darstellen. Aus diesem Grund wird der parasitologischen Diagnostik eine große Bedeutung im Bereich der Zootiermedizin beigemessen. Insbesondere eine regelmäßig durchgeführte koproskopische Untersuchung des Tierbestands spielt eine wichtige Rolle bei dem Erhalt der Tiergesundheit in Zoos. Um die Bedeutung der Parasitologie in der Zootiermedizin genauer zu betrachten, wurden im Rahmen der vorliegenden Diplomarbeit die Konferenzberichte der European Association of Zoo and Wildlife Veterinarians (EAZWV) und deren Vorgängerorganisationen auf das Vorkommen parasitologischer Beiträge analysiert. Der Schwerpunkt wurde dabei auf das Protozoon Toxoplasma gondii gelegt, welches einen weitverbreiteten Parasiten in zoologischen Einrichtungen darstellt. Insgesamt konnten in den Tagungsbänden der EAZWV von 2001 bis 2023 444 Beiträge zu Parasiten gefunden werden, wobei 531 verschiedene Parasitenspezies bei 472 unterschiedlichen Tierarten behandelt wurden. Toxoplasma gondii (T. gondii) stellte dabei mit 48 Artikeln den am häufigsten erwähnten parasitären Erreger in den Konferenzberichten dar. Die mit Abstand meisten Beiträge über diesen Parasiten konnten zu Katzen (Felidae) gefunden werden, was darauf zurückzuführen ist, dass Arten dieser Familie die einzigen Endwirte für T. gondii darstellen. Der Manul (Otocolobus manul) war die am häufigsten untersuchte Spezies in Zusammenhang mit Toxoplasmose und gilt genauso wie Neuweltaffen, Lemuren und australische Beuteltiere als besonders empfänglich für die Erkrankung, die bei diesen Tierarten tödlich enden kann. Daher sollte in zukünftigen Studien der Erforschung von Therapiemöglichkeiten bei Zootieren, vor allem bei hoch empfänglichen Arten, mehr Bedeutung beigemessen werden.Diploma thesis - University of Veterinary Medicine Vienna - 2024Parasitology in zoological medicine: literature research and analysis of parasitological contributions in the proceedings of the International Conference on Diseases of Zoo and Wild Animals and its follow-up events 2001 to 2023 Zoo animals can be infected with a wide range of parasites due to the cohabitation of different species in close proximity and different climatic conditions compared to their natural habitat. Limiting infections with parasites to as little as possible should therefore be the goal of every zoological institution. For this reason, parasitological diagnostics are of great importance in the field of zoological medicine. In particular, regular coproscopical examinations of the animal population play an important role in the maintenance of animal health in zoos. In order to examine the importance of parasitology in zoological medicine, the conference proceedings of the European Association of Zoo and Wildlife Veterinarians (EAZWV) and its predecessor organisations were analysed for the occurrence of parasitological articles as part of this diploma thesis. The focus was placed on the protozoon Toxoplasma gondii, which is a widespread parasite in zoological institutions. Altogether 444 articles on parasites could be found in the EAZWV conference proceedings from 2001 to 2023 which dealt with 531 different parasite species in 472 different animal species. Toxoplasma gondii (T. gondii) was the most frequently mentioned parasitic pathogen in the conference proceedings with 48 contributions. By far the most articles about this parasite were found in relation to cats (Felidae), which is due to the fact that members of this family are the only definitive hosts for T. gondii. The manul (Otocolobus manul) was the most studied species regarding toxoplasmosis and, like New World monkeys, lemurs and Australian marsupials, is considered especially susceptible to this disease, which can be fatal in these species. Therefore, in future studies researching treatment options for zoo animals, in particular for highly susceptible species, should be prioritized

These three pictures illustrate the ‘Choosing-wisely recommendations: Patients with asymptomatic bacteriuria should not be treated with antibiotics.’ They were designed for the creative competition for the “German Society for Infectious Diseases” in 2023 and were awarded second place.
The core message of the recommendations was presented in various ways with these images. In Fig. 1 an owl is used as a symbol of wisdom. An angel and a devil as an expression of the “good” versus “bad” are used in the 2nd picture. The 3rd figure illustrates a sporting scene where the goal can be reached when making the correct decision
Medizinisch-biochemische Verlaufsuntersuchungen bei Katzen mit gastrointestinalen Lymphomen
Diploma thesis - University of Veterinary Medicine Vienna - 2024In this diploma thesis, biochemical laboratory findings of cats with gastrointestinal lymphoma were retrospectively evaluated regarding diagnostic and prognostic significance throughout the disease and were compared using three categories: immunophenotype, WHO-entity and survival time. The laboratory values stem from 33 cats diagnosed with gastrointestinal lymphoma, which were treated in a standardized manner. The classification of patients according to immunophenotype revealed 17 animals with B-cell lymphoma and 15 animals with T-cell lymphoma. One patient showed a mixed immunophenotype. In the classification according to WHO-entity, 17 diffuse large B-cell lymphomas, 3 peripheral T-cell lymphomas, 11 enteropathy-associated T-cell lymphomas of type I and 2 enteropathy-associated T-cell lymphomas of type II were compared. 5 cats showed an ultrashort, 8 a short, 8 an intermediate, 3 a long and 9 an ultralong survival time. The following values were assessed: Glucose, urea, creatinine, uric acid, total protein, albumin, AP, AST, ALT, GLDH, GGT, bilirubin, bile acid, triglycerides, cholesterol, lipase, LDH, Na, K, Cl, Ca, P, Mg, Fe, Vit B12, USG, UPC. The values of creatinine, total protein, albumin, LDH and calcium showed statistically significant differences with probable prognostic relevance. Hypoproteinemia, hypalbuminemia, and increased LDH-activities proved to be a negative prognostic factor. Significantly lower calcium levels were associated with ultrashort survival. B-cell lymphomas showed significantly higher creatinine and calcium levels and a significantly longer survival time than T-cell lymphomas. Animals with EATL I showed the shortest survival time. Some of these changes have already been described in the literature. They can be explained by chronic inflammation and gastrointestinal changes. For conclusions regarding prognosis, further studies would be desirable to further substantiate the data.Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Im Rahmen dieser Diplomarbeit wurden biochemische Labortestergebnisse hinsichtlich prognostisch-diagnostischer Bedeutung im Krankheitsverlauf des felinen gastrointestinalen Lymphoms retrospektiv ausgewertet und anhand dreier Kategorien (Immunphänotyp, WHO- Entität und Überlebenszeit) miteinander verglichen. Die Laborwerte stammen von 33 Katzen, die mit gastrointestinalem Lymphom diagnostiziert und standardisiert behandelt wurden. Die Einteilung der Patienten nach Immunphänotyp ergab 17 Tiere mit B-Zell, 15 Tiere mit T-Zell Lymphom. Ein Patient zeigt einen gemischten Immunophänotyp. In der Klassifizierung nach WHO-Entität wurden 17 diffuse großzellige B-Zell Lymphome, 3 periphere T-Zell Lymphome, 11 Enteropathie-assoziierte T-Zell Lymphome vom Typ I und 2 Enteropathie-assoziierte T-Zell Lymphome vom Typ II verglichen. 5 Katzen zeigen eine ultrakurze, 8 eine kurze, 8 eine mittellange, drei eine lange und 9 eine ultralange Überlebenszeit. Folgende Werte wurden für die jeweiligen Gruppen beurteilt: Glucose, Harnstoff, Kreatinin, Harnsäure, Totalprotein, Albumin, AP, AST, ALT, GLDH, GGT, Bilirubin, Gallensäure, Triglyzeride, Cholesterol, Lipase, LDH, Na, K, Cl, Ca, P, Mg, Fe, Vit B12, USG, UPC. Statistisch signifikante Unterschiede mit vermutlich prognostischer Relevanz zeigten Creatinin-, Totalprotein-, Albumin-, LDH- und Calcium-Werte. Hypoproteinämie, Hypalbuminämie und erhöhte LDH-Aktivitäten erwiesen sich als negativ prognostischer Faktor. Signifikant niedrigere Calcium-Werte waren mit ultrakurzer Überlebenszeit assoziiert. B-Zell Lymphome zeigten signifikant höhere Creatinin- und Kalzium-Werte und eine deutlich längere Überlebenszeit als T-Zell Lymphome. Tiere mit EATL I zeigten die kürzeste Überlebenszeit. Diese Veränderungen sind zu einem Teil bereits in der Literatur beschrieben worden. Sie sind durch das chronische Entzündungsgeschehen und gastrointestinale Veränderungen erklärbar. Für Rückschlüsse in punkto Prognose wären weitere Untersuchungen wünschenswert, um die Daten noch weiter zu untermauern
Vorkommen von Treponema spp. in Dermatitis digitalis– assoziierten Klauenhornläsionen versus eitrigen, tiefen Klaueninfektionen beim Rind
Diploma thesis - University of Veterinary Medicine Vienna - 2024Cattle can be affected by both non-infectious and infectious claw diseases. The latter includes bovine digital dermatitis (BDD) and interdigital phlegmon. In BDD, anaerobic spirochetes of the Treponema species have been identified as the main cause of the disease. Starting from sole ulcers and white-line abscesses, deep digital infections involving tendons, bones and joints often develop in cattle. Fusobacterium necrophorum, Dichelobacter nodosus and other pyogenic bacteria play an active role in these deep digital infections. It is still unclear to what extent treponemes are involved in the pathogenesis of deep digital sepsis. Conversely, there is currently no reasonable explanation as to why BDD-associated claw horn lesions often persist for many months or even a year without infection of deep supporting structures of the claw, such as the pedal bone, the deep flexor tendon, or the distal interphalangeal joint, by pyogenic bacteria. The aim of this study was to examine samples from purulent, deep digital infections and BDD- associated claw horn lesions for the presence of treponemal DNA. The goal was to determine if infections caused by BDD-treponemes, and pyogenic bacteria can coexist or if they exclude each other. Nineteen samples of deep, purulent digital infections were taken from eleven cattle, while 18 samples of BDD-associated claw horn lesions were collected from 15 cattle. DNA was extracted from a total of 37 samples, quantified and tested for PCR-compatible quality using beta-actin PCR. The DNA extracts were then tested for the presence of treponemal DNA using a "universal spirochete PCR". Amplification products of the correct size were purified and sequenced. The sequences obtained were compared with sequences from the gene database using a BLAST program. Treponemal sequences were clearly detected in 8 of 15 cattle with BDD-associated claw horn lesions, with Treponema pedis being the most common. Additionally, a feline oral bacterium was identified in six cases, which is often found in association with BDD. In contrast, no treponemal DNA was detectable in deep, purulent digital infections. However, a pathogen that presumably corresponds to oral Parvimonas micra was frequently detected in samples from deep, purulent digital infections. The results indicate that BDD treponemes play only a very minor role, if any, in deep, purulent digital infections. This circumstance can possibly be explained by the phenomenon of "bacterial interference", whereby it is assumed that one group of bacteria displaces another group through various mechanisms. A possible causal contribution of Parvimonas micra to the development and progression of purulent claw defects should be investigated more closely.Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Rinder können von nicht-infektiösen und von infektiösen Klauenerkrankungen betroffen sein, zu letzteren zählt Dermatitis digitalis (DD) und die Zwischenklauenphlegmone. Bei DD wurden anaerobe Spirochäten der Gattung Treponema als wesentliche Verursacher der Erkrankung identifiziert. Ausgehend von Sohlengeschwüren und Weiße-Linie Abszessen, entwickeln sich beim Rind häufig tiefe Infektionen mit Beteiligung von Sehnen, Knochen und Gelenken. Bei diesen tiefen Klaueninfektionen spielen Fusobacterium necrophorum, Dichelobacter nodosus und weitere Eitererreger eine aktive Rolle. Bislang ist unklar, inwieweit auch Treponemen bei der Pathogenese von eitrigen Klauenerkrankungen involviert sind. Umgekehrt gibt es dato keine begründete Erklärung dafür, weshalb DD-assoziierte Klauenhornläsionen häufig über viele Monate bis hin zu einem Jahr bestehen, ohne dass es zu einer Infektion tiefer Stützstrukturen der Klaue, wie dem Klauenbein, der tiefen Beugesehne oder dem Klauengelenk durch Eiterbakterien kommt. Ziel dieser Studie war es daher, Proben von eitrigen, tiefen Klaueninfektionen versus an DD- assoziierten Klauenhornläsionen auf die Anwesenheit von Treponemen-DNS zu untersuchen, um Hinweise zu erhalten, ob Infektionen durch Treponemen und Eitererreger nebeneinander persistieren können, oder einander eher ausschließen. Von 11 Rindern wurden 18 Proben von tiefen, eitrigen Klaueninfektionen entnommen. Von 15 Rindern wurden 18 Proben von DD-assoziierten Klauenhornläsionen gesammelt. Aus den insgesamt 36 Proben wurde DNS extrahiert, quantifiziert und mittels Beta-Aktin-PCR auf PCR- kompatible Qualität geprüft. Im Anschluss wurden die DNS-Extrakte mit einer „Universal- Spirochäten-PCR“ auf Anwesenheit von Treponemen-DNS getestet. Amplifikationsprodukte korrekter Größe wurden gereinigt und sequenziert. Erhaltene Sequenzen wurden mittels eines BLAST-Programmes mit Sequenzen aus der Gen-Datenbank verglichen. Bei 8 von 15 Rindern mit DD-assoziierten Klauenhornläsionen wurden eindeutig Treponemen- Sequenzen nachgewiesen, wobei Treponema pedis am häufigsten vertreten war. Daneben wurde in sechs Fällen ein felines orales Bakterium identifiziert, welches häufig in Assoziation mit DD gefunden wird. Hingegen war bei tiefen, eitrigen Klaueninfektionen keine Treponemen- DNS nachweisbar. In den Proben von tiefen, eitrigen Klaueninfektionen wurde jedoch häufig ein Pathogen nachgewiesen, welches vermutlich dem oralen Bakterium Parvimonas micra entspricht. Die Ergebnisse deuten darauf hin, dass Treponemen bei tiefen, eitrigen Klaueninfektionen, wenn überhaupt, nur eine sehr untergeordnete Rolle spielen. Möglicherweise kann dieser Umstand mit dem Phänomen der „bakteriellen Interferenz“ erklärt werden, wobei man davon ausgeht, dass eine Gruppe von Bakterien eine andere Gruppe durch verschiedene Mechanismen verdrängt. Ein möglicher kausaler Beitrag von Parvimonas micra an der Entstehung und dem Fortschreiten eitriger Klauendefekten sollte wissenschaftlich näher untersucht werden
Analyse der Treffsicherheit von Annotierungen zwei verschiedener Video-tracking Programme vorbestimmter Körperteile bei hospitalisierten Pferden
Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Ziel dieser Arbeit ist es, die zwei Videotracking Programme „Coco Annotator“ und „Loopy“ auf ihre Treffgenauigkeit der Setzung vordefinierter Körperpunkte zu vergleichen. Der Einsatz an künstlicher Intelligenz wird in vielen Teilen der Veterinärmedizin bereits regelmäßig angewendet, Beispiele dafür sind die automatisierte Erkennung von Körperpositionen oder im Bereich des „Pain Scorings“. Für diese Diplomarbeit sind Videos von sechs hospitalisierten Pferden auf der Veterinärmedizinischen Universität Wien verwendet worden, die mithilfe eines Modells des jeweiligen Programms automatisch annotiert wurden. Nach Setzung der Körperpunkte durch KI-Modelle in den Videos, sind jeweils 2000 Bilder pro Pferd manuell überprüft worden und die Körperpunkte „Nase“, „Widerrist“, „Schweif“, „linke“ und „rechte Vorderextremität“ hinsichtlich mehrerer Kriterien überprüft worden. Die Punkte konnten dabei als „true positive“, „true negative“, „false positive“ und „false negative“ gelabelt werden. Die Auswertung der richtigen Bilder ergab eine höhere Trefferquote des Programms „Coco Annotator“ als bei „Loopy“, die Anzahl der falsch annotierten Bilder ist bei „Loopy“ höher gewesen. Die Bland-Altman Analyse ergab, dass es einen signifikanten Unterschied zwischen beiden Annotierungsprogrammen gibt. Die Beurteilung des Datenmaterials zeigte mehrere Limitationen hinsichtlich der Bildqualität und der Umgebung, manche Pferde erschwerten eine korrekte Annotation durch ihre Fellfarbe oder Abzeichen. Körperpunkte wie die „Nase“ oder der „Schweif“ wurden bei beiden Programmen besser annotiert als der Widerrist. „Coco Annotator“ zeigte über alle Körperpunkte hinweg eine höhere Trefferquote an richtig annotierten Bildern als „Loopy“ und hat bei dieser Studie im Rahmen der Diplomarbeit besser abgeschnitten.Diploma thesis - University of Veterinary Medicine Vienna - 2024The aim of this thesis is to compare the accuracy of two video tracking programs, "Coco Annotator" and "Loopy," in placing predefined body points. Artificial intelligence is already regularly applied in various areas of veterinary medicine, such as automated recognition of body positions or pain scoring. For this thesis, videos of six hospitalized horses at the University of Veterinary Medicine Vienna were used. These videos were automatically annotated using a model from each program. After the body points were set by the AI models in the videos, 2,000 images per horse were manually reviewed, and the body points nose, withers, tail, left and right forelimbs were evaluated based on several criteria. The points could be labelled as "true positive", "true negative", "false positive" and “false negative". The evaluation of the correct images showed a higher hit rate for the "Coco Annotator" program compared to "Loopy", with the number of incorrectly annotated images being higher in "Loopy". The Bland-Altman analysis revealed a significant difference between the two annotation programs. The assessment of the data material revealed several limitations in terms of image quality and environment; some horses made accurate annotation more difficult due to their coat colour or markings. Body points such as the nose or tail were better annotated by both programs compared to the withers. Overall, "Coco Annotator" showed a higher hit rate for correctly annotated images across all body points than "Loopy" and performed better in this study within the framework of the thesis.Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Ziel dieser Arbeit ist es, die zwei Videotracking Programme „Coco Annotator“ und „Loopy“ auf ihre Treffgenauigkeit der Setzung vordefinierter Körperpunkte zu vergleichen. Der Einsatz an künstlicher Intelligenz wird in vielen Teilen der Veterinärmedizin bereits regelmäßig angewendet, Beispiele dafür sind die automatisierte Erkennung von Körperpositionen oder im Bereich des „Pain Scorings“. Für diese Diplomarbeit sind Videos von sechs hospitalisierten Pferden auf der Veterinärmedizinischen Universität Wien verwendet worden, die mithilfe eines Modells des jeweiligen Programms automatisch annotiert wurden. Nach Setzung der Körperpunkte durch KI-Modelle in den Videos, sind jeweils 2000 Bilder pro Pferd manuell überprüft worden und die Körperpunkte „Nase“, „Widerrist“, „Schweif“, „linke“ und „rechte Vorderextremität“ hinsichtlich mehrerer Kriterien überprüft worden. Die Punkte konnten dabei als „true positive“, „true negative“, „false positive“ und „false negative“ gelabelt werden. Die Auswertung der richtigen Bilder ergab eine höhere Trefferquote des Programms „Coco Annotator“ als bei „Loopy“, die Anzahl der falsch annotierten Bilder ist bei „Loopy“ höher gewesen. Die Bland-Altman Analyse ergab, dass es einen signifikanten Unterschied zwischen beiden Annotierungsprogrammen gibt. Die Beurteilung des Datenmaterials zeigte mehrere Limitationen hinsichtlich der Bildqualität und der Umgebung, manche Pferde erschwerten eine korrekte Annotation durch ihre Fellfarbe oder Abzeichen. Körperpunkte wie die „Nase“ oder der „Schweif“ wurden bei beiden Programmen besser annotiert als der Widerrist. „Coco Annotator“ zeigte über alle Körperpunkte hinweg eine höhere Trefferquote an richtig annotierten Bildern als „Loopy“ und hat bei dieser Studie im Rahmen der Diplomarbeit besser abgeschnitten
Ablation of Vitamin D Signaling in Cardiomyocytes Leads to Functional Impairment and Stimulation of Pro-Inflammatory and Pro-Fibrotic Gene Regulatory Networks in a Left Ventricular Hypertrophy Model in Mice
The association between vitamin D deficiency and cardiovascular disease remains a controversial issue. This study aimed to further elucidate the role of vitamin D signaling in the development of left ventricular (LV) hypertrophy and dysfunction. To ablate the vitamin D receptor (VDR) specifically in cardiomyocytes, VDRfl/fl mice were crossed with Mlcv2-Cre mice. To induce LV hypertrophy experimentally by increasing cardiac afterload, transverse aortic constriction (TAC) was employed. Sham or TAC surgery was performed in 4-month-old, male, wild-type, VDRfl/fl, Mlcv2-Cre, and cardiomyocyte-specific VDR knockout (VDRCM-KO) mice. As expected, TAC induced profound LV hypertrophy and dysfunction, evidenced by echocardiography, aortic and cardiac catheterization, cardiac histology, and LV expression profiling 4 weeks post-surgery. Sham-operated mice showed no differences between genotypes. However, TAC VDRCM-KO mice, while having comparable cardiomyocyte size and LV fibrosis to TAC VDRfl/fl controls, exhibited reduced fractional shortening and ejection fraction as measured by echocardiography. Spatial transcriptomics of heart cryosections revealed more pronounced pro-inflammatory and pro-fibrotic gene regulatory networks in the stressed cardiac tissue niches of TAC VDRCM-KO compared to VDRfl/fl mice. Hence, our study supports the notion that vitamin D signaling in cardiomyocytes plays a protective role in the stressed heart
