    Pls1 Is a Peroxisomal Matrix Protein with a Role in Regulating Lysine Biosynthesis

    Peroxisomes host essential metabolic enzymes and are crucial for human health and survival. Although peroxisomes were first described over 60 years ago, their entire proteome has not yet been identified. As a basis for understanding the variety of peroxisomal functions, we used a high-throughput screen to discover peroxisomal proteins in yeast. To visualize low abundance proteins, we utilized a collection of strains containing a peroxisomal marker in which each protein is expressed from the constitutive and strong TEF2 promoter. Using this approach, we uncovered 18 proteins that were not observed in peroxisomes before and could show their metabolic and targeting factor dependence for peroxisomal localization. We focus on one newly identified and uncharacterized matrix protein, Ynl097c-b, and show that it localizes to peroxisomes upon lysine deprivation and that its localization to peroxisomes depends on the lysine biosynthesis enzyme, Lys1. We demonstrate that Ynl097c-b affects the abundance of Lys1 and the lysine biosynthesis pathway. We have therefore renamed this protein Pls1 for Peroxisomal Lys1 Stabilizing 1. Our work uncovers an additional layer of regulation on the central lysine biosynthesis pathway. More generally it highlights how the discovery of peroxisomal proteins can expand our understanding of cellular metabolism

    Associated Newspapers Criticised by UK Court of Appeal for “Lack of Clear Focus” in Its Meghan Markle Judgment

    The commentary argues that the Duchess of Sussex’s, Meghan Markle’s, privacy claim against the publishers of the Mail on Sunday and online, Associated Newspapers Ltd (ANL), was a courageous attempt to demonstrate via the courts that freedom of the press and its claim to the public interest can go too far. She was also successful in her copyright claim, in that the Mail’s misuse of private information, namely her letter to her father, Thomas Markle, in August 2018, which had largely been copied by the newspaper verbatim, breached copyright. ANL could not claim fair dealing in a publication of a letter which had been handwritten in private and sent to Meghan’s estranged father. The CA held that the original trial judge, Warby J, was right in delivering a summary judgment in February 2021 and that there would be no public trial now

    Adoption and influence of robotic process automation in beef supply chains

    This paper aims to critically examine the potential barriers to the implementation and adoption of Robotic Process Automation (RPA) in the beef supply chain. The beef supply chain has been challenging due to its complex processes, activities, and management. The beef industry has relied heavily on the human workforce in the past; however, RPA adoption allows automating tasks that are repetitive and strenuous in nature to enhance beef quality, safety and security. There are considerable potential barriers to RPA adoption as organisations have not focused on trying to eliminate them due to various reasons. Previous studies lack knowledge related to potential barriers to RPA adoption, so this creates a research gap and requires attention. Methods: Statistical data and information are extracted using secondary data relevant to RPA adoption in the beef supply chain. A business process model is formed which uses values or variables using existing statistical data and information. Simulation of the process model is carried out using Simul8 software and analyses of different scenarios help in choosing the best approach for RPA adoption. Results: The results have identified the potential barriers in RPA adoption through the simulation process thus ensuring RPA performs with more potential. Analysis of ‘what-if’ scenarios allow organisational and employee-level improvements along with enhancing RPA’s accuracy. Conclusion: The process model is a generic model for use in real-life scenarios and can be modified by organisations according to their own business needs and requirements. The study contributes in theoretical and practical aspects as it allows decision-makers to adopt RPA in a robust manner and adds to scientific knowledge by identification of potential barriers to RPA adoption

    Degree apprenticeship end-point assessment dilemma

    A growing number of learners join HE institutions through degree apprenticeships with a strong emphasis on both on-the-job and off-the-job training, with apprentices sharing time between lecture theatres and the workplace. In addition to meeting the degree requirements, the completion of degree apprenticeships requires passing a work-based end-point assessment (EPA). EPAs often include a capstone project that is equivalent to a project or dissertation and plays a crucial role in degree apprenticeships, but their execution is not without its problems. This paper identifies common challenges for EPA projects, including academia-business goal misalignment, external factors, confidentiality and commercial sensitivity, and gaps between expectations and experience. Consequently, a set of recommendations is proposed to mitigate the identified challenges

    Group interactions modulate critical mass dynamics in social convention

    How can minorities of individuals overturn social conventions? The theory of critical mass states that when a committed minority reaches a critical size, a cascade of behavioural changes can occur, overturning apparently stable social norms. Evidence comes from theoretical and empirical studies in which minorities of very different sizes, including extremely small ones, manage to bring a system to its tipping point. Here, we explore this diversity of scenarios by introducing group interactions as a crucial element of realism into a model for social convention. We find that the critical mass necessary to trigger behaviour change can be very small if individuals have a limited propensity to change their views. Moreover, the ability of the committed minority to overturn existing norms depends in a complex way on the group size. Our findings reconcile the different sizes of critical mass found in previous investigations and unveil the critical role of groups in such processes. This further highlights the importance of the emerging field of higher-order networks, beyond pairwise interactions

    Perception of semi-autonomous intelligent vehicles such as smart powered wheelchairs

    This paper investigates perceptions of what semi-autonomous vehicles such as Smart powered-wheelchairs are and whether perception has changed. Information about semi-autonomous vehicles (especially powered-wheelchairs) was analysed to investigate how concepts and their benefits are perceived. Some common understanding is found, but there is uncertainty about whether autonomous/semi-autonomous implies automatic/un-manned. There is a tendency to distinguish between semi-autonomous and un-manned but that distinction does not appear to be made in the minds of everyday people. Implications of this are discussed
