The Activities of Antifertility Ethanol Extract Guava Leaves (Psidium Guajava L.) based on The Analysis Of Cement and Display of Immunohistochemistry Cyclooxygenase-2 In Testis Of Mice (Mus Musculus L.)
Population growth increase, without support from good economic conditions, requirer population control. One of the program to control population growth is family planning program (KB). The participation of men in family planning programs is very important. Male contraception can utilize natural materials derived from plants. One of the herb is guava leaves (Psidium guajava L.) which contains many active substances that can inhibit the process of spermatogenesis. The purpose of this study was to determine the effect of ethanol extract guava leaves to the quality (concentration, viability, morphology, and motility) of spermatozoa and the expression of Cyclooxygenase-2 (COX-2) in the testes of mice (Mus musculus L.). The research used experimental design with completely randomized sampling method. Four treatment groups, namely P0 using aquadest as a control 0,5 mL, P1 using 15 mg/25g body weight extract orally, P2 using 20 mg/25g body weight extract orally, and P3 using 25 mg/25g body weight extract orally. Data were analyzed using One Way ANOVA followed by Bonferroni Post Hoc test 5%. The results showed that ethanol extract guava leaves with a dose of 15mg/25g body weight had significantly decreased sperm quality and increased expression of COX-2 in the testes of mice. This research indicates that guava leaves have a potential to be used as a herbal contraceptive.Laju pertumbuhan penduduk yang meningkat, tanpa diimbangi dengan keadaan ekonomi yang baik, menyebabkan terjadinya ledakan penduduk. Oleh karena itu, salah satu cara yang dapat dilakukan melalui program keluarga berencana (KB). Keikutsertaan pria dalam program KB sangat penting. Sarana kontrasepsi pria dapat memanfaatkan bahan alami yang berasal dari tumbuhan. Salah satunya adalah daun jambu biji (Psidium guajava L.) yang mengandung banyak zat aktif yang dapat menghambat proses spermatogenesis. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak etanol daun jambu biji terhadap kualitas (konsentrasi, viabilitas, morfologi, dan motilitas) spermatozoa dan ekspresi Cyclooxygenase-2 (COX-2) pada testis mencit (Mus musculus L.). Penelitian ini menggunakan studi eksperimental dengan metode rancangan acak lengkap (RAL) dengan empat kelompok perlakuan, yaitu P0 sebagai kontrol diberikan aquadest 0,5mL, P1 diberikan ekstrak 15mg/25g berat badan, P2 diberikan ekstrak 20mg/25g berat badan, dan P3 diberikan ekstrak 25mg/25g berat badan yang diberikan selama 35 hari. Data yang diperoleh dianalisis menggunakan One Way Anova yang dilanjutkan dengan uji Post Hoc Bonferroni 5%. Hasil penelitian menunjukkan bahwa pemberian ekstrak etanol daun jambu biji dengan dosis 15mg/25g berat badan sudah berpengaruh terhadap penurunan kualitas spermatozoa dan meningkatkan ekspresi COX-2 pada testis mencit. Hal ini mengindikasikan bahwa daun jambu biji berpeluang untuk digunakan sebagai bahan kontrasepsi herbal
Hubungan Rasionalitas Pengobatan dan Self-care dengan Pengendalian Glukosa Darah pada Pasien Rawat Jalan di Rumah Sakit Bina Husada Cibinong
Diabetes melitus (DM) merupakan kelompok penyakit metabolik yang bila tidak terkendali dengan baik dapat menimbulkan komplikasi yang berbahaya. Pengobatan yang rasional pada penderita DM diduga dapat mengendalikan glukosa darah karena pasien menerima obat sesuai dengan kebutuhan klinisnya. Selain pengobatan yang rasional, penderita DM harus mampu melakukan self-care yang merupakan perawatan diri secara mandiri untuk meningkatkan kualitas hidup dan mencegah komplikasi. Penelitian ini bertujuan untuk mengetahui hubungan antara rasionalitas pengobatan dan self-care dengan pengendalian glukosa darah pasien DM tipe II di Poli Rawat Jalan Rumah Sakit (RS) Bina Husada Cibinong. Penelitian ini menggunakan metode deskriptif analitis dengan desain cross sectional. Pengambilan data dilakukan secara retrospektif maupun prospektif terhadap 109 pasien DM tipe 2 selama periode Maret-Mei 2015. Digunakan kuesioner Diabetes Self-Management Questionnaire untuk mengakses self-care. Hasil penelitian menunjukkan data demografi pasien DM tipe 2 adalah 57,80% wanita dan 42,20% pria dengan kisaran usia terbanyak yaitu 46-55 tahun (49,54%). Persentase pengobatan yang rasional cukup tinggi yaitu 61,47%, sedangkan self care pasien sebagian besar (69,72%) berada pada kriteria “baik” dan sisanya (30,28%) pada kriteria “cukup”. Pada uji korelasi Chi-square independen ditemukan tidak adanya hubungan antara rasionalitas pengobatan dan self care dengan pengendalian glukosa darah (p > 0,05). Hal ini menunjukkan kemungkinan adanya faktor lain yang dapat mempengaruhi pengendalian glukosa darah.Diabetes mellitus (DM) is a group of metabolic diseases and if it is not properly controlled can lead to dangerous complications. A rational treatment in patients with suspected diabetes can control blood glucose for patients receiving the drugs in accordance with their clinical needs. In addition to rational treatment, people with diabetes should be able to perform self-care in order to improve quality of life and prevent complications. This study aims to determine the relationship between treatment rationality and self-care with type II DM outpatients at Bina Husada Cibinong Hospital. This research uses descriptive analysis method with cross sectional design. Data were collected retrospectively and prospectively towards 109 type 2 diabetetic patients during period from March to May 2015. Diabetes Self-Management Questionnaire were used to access self-care. The results showed that the demographic data of type 2 diabetic patients was 57.80% women and 42.20% of men, with the highest age range is 46-55 years (49.54%). The percentage of rational treatment is quite high at 61.47%, while the majority of self-care patients were on the criteria of “good”(69.72%) and the rest were on the criteria of “enough” (30.28%). Chi square correlation test showed no relationship between treatment rationality and self care with blood glucose control (p> 0.05). This suggests the possibility of other factors that can affect blood glucose control
Formulasi Serum sebagai Penyembuh Luka Bakar Berbahan Baku Utama Serbuk Konsentrat Ikan Gabus (Channa striatus)
Ikan gabus (Channa striatus) diketahui dapat menyembuhkan luka karena mengandung kadar tinggi protein, asam amino esensial dan asam lemak yang berperan dalam proses penyembuhan luka. Tujuan dari penelitian ini adalah membuat serum sebagai penyembuh luka bakar yang mengandung serbuk konsentrat ikan gabus (Channa striatus) yang telah dibuat gelasi ionik dengan kitosan dan natrium tripolifosfat sebagai zat aktif. Serbuk konsentrat ikan gabus dengan kosentrasi 7,5% (formula 1), 10% (formula 2) dan 12,5% (formula 3). Selanjutnya dibuat menjadi serum dengan menggunakan kolagen dan gelatin sebagai bahan pengental. Sediaan serum yang dihasilkan dikarakterisasi in vitro dan dievaluasi secara in vivo penyembuhan luka bakar derajat dua (deep partial thickness) pada kelinci. Suspensi dan serum yang dihasilkan dikarakterisasi secara fisik maupun kimia. Hasil pengukuran suspensi formula 1, 2 dan 3 adalah sebagai berikut: ukuran partikel berturut–turut 42,67-204,23 nm, 70,81-257,11 nm, 128,86-323,68 nm; nilai potensial zeta (+)16,9 mV, (+)18,3 mV, (+)8,4 mV; ketiga formula memiliki partikel berbentuk sferis. Dari hasil uji in vivo dan analisa histologi sediaan serum serbuk konsentrat ikan gabus-kitosan tripolifosfat dapat digunakan sebagai penyembuh luka bakar derajat dua dalam.Snakehead fish (Channa striatus) has been reported can be used for wound healing because contains high amount of protein, essential amino acids and fatty acids that influenced wound healing. This study was performed to formulate serum for burn wound healing contain concentrate powder of snakehead fish (Channa striatus) have been made with gelation ionic using chitosan and sodium tripolyphosphate as an active substances. Concentrate powder of snakehead fish with concentration 7.5% (formula 1), 10% (formula 2) and 12.5% (formula 3). And then formulated to serum using collagen and gelatin as thickening agent. Serum has been formulated, characterized and evaluated in vivo for burn wound healing second degree (deep partial thickness) on rabbits. Suspense and also serums has been characterized by physicochemical. The results showed that nanoparticle suspenses (formula 1, formula 2 and formula 3) have particle size in range 42.67-204.23 nm, 70.81-257.11 nm, 128.86-323.68 nm; zeta potential (+) 16.9 mV, (+) 18.3 mV, (+) 18.4 mV; all of formulas have sferichal particles. Pemodelan Molekular Enzim 3β-Hydroxysteroid Dehydrogenase Tipe 2: Pemodelan Kombinasi Homologi, Docking dan Pendekatan QSAR
A homology model of human 3β-HSD type 2 has been developed from homology modeling techniques using Phyre2 server and refi ned by ModRefi ner. The PROCHECK, QMEAN and ProSA-web online tools were carried out to evaluate the stereochemical quality of the model. The Ramachandran plot resulted from PROCHECK showed that 84.5% residues are in the most favored region, 13.7% are in the additional allowed region, 1.5% are in the generously allowed region and 0.3% are in the disallowed region. The QMEAN (Z-score) are 0.509 (-3.006) and Z-score of ProSA-web is -7.10. The negative values of protein fold energies also found in almost all sequences. Furthermore, molecular docking was also applied to validate the model using MOE. The hydrogen bonding interactions with Tyr154, Ser124, and Ser218 are found in all docked substrates as well as known inhibitors (trilostane and epostane). A dataset of azasteroid inhibitors were also docked into the substrate active site of human 3β-HSD2. These docked structures were utilized to construct corresponding docking-based QSAR equation by employing genetic algorithm (GA) statistical analysis. The contructed best QSAR equation has a robust predictive power according to its statistical parameters, hence may be applied to supersede the default scoring function provided by docking software. These results indicate that the human 3β-HSD2 model was successfully evaluated as a good model.Model homologi dari enzim 3β-HSD2 telah dikonstruksi menggunakan server Phyre2 dan dilanjutkan dengan ModRefi ner. Dataset inhibitor azasteroid juga ditambatkan ke situs aktif substrat pada enzim 3β-HSD2. Struktur yang tertambatkan digunakan untuk membangun persamaan QSAR berbasis penambatan molekuler dengan menerapkan analisis statistik genetic algorithm (GA). Persamaan QSAR terbaik yang terkonstruksi memiliki daya prediksi yang kuat sesuai dengan parameter statistiknya, sehingga dapat diaplikasikan untuk menggantikan fungsi scoring default yang disediakan oleh program MOE. Hasil ini menunjukkan bahwa model enzim 3β-HSD2 manusia berhasil dievaluasi sebagai model yang baik
Isolasi dan Identifi kasi Senyawa Kimia Zat Anti Kanker dari Daun Kopasanda (Chromolaena odorata (L.))
Isolation and identifi cation of chemical compound as anticancer from the leaves of Kopasanda (Chromolaena odorata L.) had been done. Extraction was conducted successfully with n-hexane, ethylacetate and ethanol by maceration, and by refl ux for water solvent. Fractionation of ethylacetate extract by column chromatography (SiO2). n-hexane-ethylacetate = 10:1 ~ 1:1; ethylacetate). n-hexaneethylacetate = 5:1) gave one pure isolated fraction fr. 5-3 with inhibition activity against leukemia cell line L1210 at concentration 1.515 ppm. Chemical structure determination was done based on spectral data interpretation of Infra Red, 1D NMR (1H, 13C) and 2D NMR (H-H COSY, HMQC and HMBC) and mass spectra (LC-MS). Pure isolated fraction fr. 5-3 was deducted as methyl ether naragenine.Isolasi dan identifi kasi senyawa kimia sebagai antikanker dari daun Kopasanda (Chromolaena odorata L.) telah dilakukan. Ekstraksi dilakukan secara berturut-turut dengan n-heksan, etilasetat, dan etanol secara maserasi kecuali dengan air dilakukan secara refl uks. Fraksinasi ekstrak etilasetat dengan kromatografi kolom (SiO2,: i). n-heksan-etilasetat = 10:1 ~ 1:1; etilasetat, ii). n-heksan-etilasetat = 5:1) memberikan satu isolat murni fr. 5-3 yang mempunyai aktivitas daya hambat terhadap sel leukemia L1210 sebesar 1,515 bpj. Penentuan struktur kimia dilakukan berdasarkan interpretasi data spektra infra merah, RMI 1D (1H, 13C) dan RMI 2D (H-H COSY, HMQC dan HMBC) dan spektra masa (LC-MS). The Role of Fennel Infusion on Estrous Cycle and Follicles Development of White Rats
This study aims to describe the estrous cycle and follicles development in female rats given infusion of fennel fruit that known to have phytoestrogen compound. Twenty five of female rats were used in this research and were divided into 5 groups. Group I was negative control group (NC), given 1 mL/100g bw of distilled water, group II was positive control group (PC), given 0.0045 mg/100g bw of ethinyl estradiol, groups III, IV and V were treatment groups (TI, T2 and T3), given fennel infusion at 36.5; 73 and 146 mg/100g bw respectively. All treatments were conducted every morning for 20 days by oral route. Changes of vaginal epithelium were observed through vaginal swabs previously stained with Giemsa stain. Histopathological examination of ovarian swere examined to reveal follicles development. Results showed that fennel fruit infusion extended the duration of estrous and metestrous phases, while shortened the proestrous and diestrous phases. Eventhough the longest estrous phase was found in T3 group, there was no significant effect on the lengthening of estrous cycle. Formulasi Sediaan Tablet Ekstrak Sambung Nyawa (Gynurae procumbens (Lour).Merr) sebagai Kandidat Antidiabetes
Ekstrak daun sambung nyawa (Gynura procumbens (Lour.) Merr.) telah terbukti secara in vitro berkhasiat sebagai antidiabetes karena mampu menghambat enzim α-glukosidase. Penelitian ini bertujuan untuk memformulasikan ekstrak etanol 70% daun sambung nyawa menjadi sediaan tablet yang memenuhi parameter mutu fisik dan uji in vivo. Ekstrak kental daun sambung nyawa diformulasikan menjadi 3 formula dengan variasi pengisi: kalsium fosfat dibasa (formula I), laktosa (formula II) dan mikrokristalin selulosa (formula III) dengan metode granulasi basah. Selanjutnya, dilakukan uji penghambatan α-glukosidase, formula yg mempunyai aktivitas teraktif dilakukan uji aktivitas secara in vitro dan in vivo. Hasil evaluasi mutu fisik dari ketiga formula tablet yaitu organoleptik: warna hijau, rasa pahit dan berbau khas; keseragaman bobot: 510,26; 506,61 dan 508,08 mg; kekerasan tablet: 6,2; 5,6 dan 5,2%; friabilitas: 0,14; 0,13 dan 0,15%; waktu hancur: 6 menit 50 detik, 7 menit 49 detik dan 7 menit 39 detik dan hasil uji aktivitas penghambatan enzim α-glukosidase: 76,35; 67,13 dan 78,14%. Pada pengujian ini digunakan akarbose sebagai kontrol positif dengan penghambatan sebesar 88,49%. Dapat disimpulkan bahwa ketiga formula memenuhi syarat mutu fisik tablet dan mampu menghambat enzim α-glukosidase. Formula III merupakan sediaan teraktif. Hasil uji in vivo pada hari ke-14 dengan dosis penggunaan 3x2 tablet per hari dapat menurunkan kadar glukosa dalam darah.Sambung nyawa leaf extract (Gynura procumbens (Lour.) Merr.) which have in vitro efficacious as antidiabetic that it can inhibit the enzyme α-glucosidase. This study aims to formulate a 70% ethanol extract of sambung nyawa leaves into a tablet dosage form that meets the quality parameters of physical and in vivo tests. Condensed extract of sambung nyawa leaves is formulated into 3 formulas with variations of filler: dibasic calcium phosphate (Formula I), lactose (Formula II) and microcrystalline cellulose (Formula III) by wet granulation method. Furthermore, the formula tested α-glucosidase inhibition, a formula which has the most activity tested in vivo. The results of physical evaluation of all formulas: organoleptic is green, bitter taste and distinctive smell; average weight uniformity: 510.26; 506.61 and 508.08 mg; tablet hardness: 6.2, 5.6 and 5.2%; friability: 0.14, 0.13 and 0.15%; desintegration time: 6 minutes 50 seconds, 7 minutes and 49 seconds and 7 minutes 39 seconds and inhibitory activity of α-glucosidase enzyme: 76.35, 67.13 and 78.14%. In this test, acarbose is used as a positive control with % inhibition is 88.49%. It can be concluded that physical quality tablet of all formulas are within the acceptable standard and is able to inhibit the enzyme α-glucosidase. Formula III is the most active. The results of in-vivo test on the 14th day after the administration at a dose tablet use 3x2 tablet per day can lower blood glucose levels
Isolasi Senyawa Flavonoid dari Ekstrak Metanol Daun Katuk (Sauropus androgynus (L.) Merr), Euphorbiaceae
Katuk (Sauropus androgyrus (L.) Merr) is one of the plants used for medicinal purposes. It has been carried out the research to isolate and to identify flavonoid compounds in the methanol extract of katuk leaves using paper chromatography and spectrophotometry ultraviolet-visible. The results demonstrated that light spectrum of uv-visible in the presence of a reagent gave an isolate NB-3 which was a compound suspected of flavonol with OH group in positions 5,7,4 ‘and o-di OH groups in the position of the A-ring (6,7 or 7,8) whrereas NB-4 isolate was suspected as flavonoids with OH groups at 5.7 and the prenil groups in position 6, methylation or glycosylation at OH group in a position 3 and o-di OH groupsin position of the a-ring (6,7 or 7,8).Katuk (Sauropus androgynus (L.)Merr) adalah salah satu tumbuhan yang digunakan untuk tujuan pengobatan. Telah dilakukan penelitian terhadap daun katuk meliputi isolasi dan identifikasi golongan senyawa flavonoid dari ekstrak metanol daun katuk (Sauropus androgyrus (L.)Merr) menggunakan metode kromatografi kertas dan diidentifikasi secara spektrofotometri ultraviolet-cahaya tampak. Hasil spektrum uv-cahaya tampak dengan menggunakan pereaksi geser diperoleh isolat NB-3 yang diduga merupakan senyawa flavonol dengan gugus OH pada posisi 5,7,4’ serta gugus o-di OH pada cincin A (6,7 atau 7,8), dan isolat NB-4 yang diduga sebagai senyawa flavon dengan gugus OH pada 5,7, dengan gugus prenil pada posisi 6, metilasi atau glikosilasi pada gugus OH posisi 3 serta gugus o-di OH pada cincin A (6,7 atau 7,8)
Pemodelan Molekul, Sintesis dan Penentuan Aktivitas Antineoplastik 1-(4-Trifluorometilbenzoiloksi)Urea
To design new drugs, physical-chemical characteristics of drug molecules can be predicted by in silico test before drugs are synthesized. Ribonucleotide reductase is the main target or receptor of antineoplastic compounds such as hydroxyurea (HU) and its derivatives like 1-(4-trifluoromethylbenzoyloxy)urea or 4-CF3BOU. This compound forms a complex with crystal structure of ribonucleotide reductase I enzyme, which is 2EUD. The hydrogen bond and bond energy in the form of rerank score from both complexes was calculated with Molegro program. Theoretically, compound activity is indicated by rerank score. The compound whose rerank score is small is predicted to have greater activity. The activity of 4-CF3BOU was found to be greater than HU. The reaction mechanism of synthesis 4-CF3BOU was the substitution of nucleophilic hydroxyl group from HU to carbonyl group of 4–trifluoromethylbenzoyl chloride (4-CF3BCl). Purity test was conducted using TLC and melting point. Structure identification was performed based on the spectra of UV-VIS, FT-IR, H/C-NMR and MS. In this study, 4-CF3BOU was discovered to have antineoplastic activity with the IC50 value of 82.37 μg/mL and was tested towards HeLa cells. On the other hand, HU had the IC50 value of 430.21 μg/mL. The antineoplastic activity of 4-CF3BOU was greater than HU.Untuk merancang obat baru, sifat fisikokimia molekul obat dapat diprediksi sebelum disintesis dan dimurnikan melalaui uji in silico. Ribonukleotida reduktase merupakan target utama atau reseptor dari senyawa antineoplastik seperti hidroksiurea (HU) dan turunannya, yaitu 1-(4-trifluorometilbenzoiloksi)urea atau 4-CF3BOU. Senyawa ini membentuk kompleks dengan struktur kristal enzim ribonukleotida reduktase I yaitu 2EUD. Jumlah ikatan hidrogen dan energi ikatan berupa rerank score dari kompleks keduanya dihitung dengan program Molegro. Secara teoretis, aktivitas senyawa ditunjukkan oleh rerank score. Senyawa yang mempunyai rerank score kecil diprediksi aktivitasnya besar. Senyawa 4-CF3BOU diprediksi memiliki aktivitas lebih besar dibanding HU. Mekanisme reaksi sintesis 4-CF3BOU adalah masuknya gugus nukleofilik hidroksil dari HU pada gugus karbonil dari 4–trifluorometilbenzoil klorida (4-CF3BCl). Uji kemurnian dilakukan dengan menggunakan KLT dan penentuan titik lebur. Identifikasi struktur dilakukan berdasarkan spektra UV-VIS, FT-IR, H/C-RMI, dan MS. Dalam penelitian ini, 4-CF3BOU telah berhasil disintesis dan setelah diuji aktivitas antineoplastiknya terhadap sel HeLa mempunyai IC50 sebesar 82,37 μg/mL, sedangkan HU mempunyai IC50 430,21 μg/mL. Mangosteen peel contains xanthone as antioxidant, useful as prooxidant balancer. The activity of xathone can be found from the number of NK cells that will increase the production of IFN-γ cytokine. The human experimental research has been done by Randomized Control Group Pretest-Post Test Design, 20 patients sample for each group. Mann Whitney test was used to determine the diff erences between the group 1 & group 2. The Spearman’s Rho Test was used to determine the correlation between the NK cells number and IFN-γ level. There are no signifi cant diff erences between NK cell number (p:0.911) and IFN-γ level (p:0.588) in the pre and post test examination after have been given mangosteen peel extract. There are no signifi cant correlations between NK cell number and IFN-γ level in each group (p: 0.074). There are no eff ects of giving mangosteen peel extract on the number of NK cell and IFN-γ level to HIV patients.Kulit manggis mengandung xanthone sebagai antioksidan yang, dibutuhkan tubuh sebagai penyeimbang prooxidant (reduxing radicals, oxidizing radicals, carbondentered, sinar UV dan metal) yang ada di lingkungan manusia. Aktivitasnya dapat dilihat dari jumlah Sel NK. Pengaruh jumlah Sel NK akan memacu peningkatan sitokin IFN-γ. Penelitian eksperimental pada manusia, dengan Randomized Control Group Pretest-Post Test Design, sampel sebanyak 20 pada masing-masing kelompok. Penelitian ini menggunakan Mann Whitney U Test untuk menentukan perbedaan antara kelompok perlakuan dan placebo, serta Spearman’s Rho Test untuk menguji korelasi antara jumlah sel NK dan kadar INF-γ. Tidak terdapat perbedaan bermakna antara jumlah sel NK (p:0,911) dan kadar IFN-γ (p:0,588) dan tidak terdapat korelasi antara sel NK dan kadar IFN- γ (p: 0,074) pada penderita HIV pada pemeriksaan sebelum dan sesudah diberikan ekstrak kulit manggis (Garcinia mangostana).
