    Comparison of common tests performance for Mycobacterium bovis infection diagnosis in low prevalence dairy cattle herds of southern Chile

    The aim of the present study was to assess the performance of routinely used diagnostic tests to detect M. bovis infection in cattle populations with very low infection prevalence. A cross-sectional study was performed using 609 animals from 17 herds that were sampled for blood interferon, and tested with the caudal fold tuberculin test (CFT). It was planned that animals which showed CFT positive results were sent to the slaughterhouse for post mortem confirmation testing. However, not all the animals considered in the initial sampling were culled. Paired population proportions of positive results for the different diagnostic tools were compared. Apparent sensitivity and specificity was also estimated. Overall, intradermal tuberculin based diagnostics showed better performance in comparison to interferon based test, even though interferon was slightly more specific. From these results, it may be suggested that specific cut-off lines for interferon based techniques for certain areas with particular low bTB prevalence should be considered, as well as the modification of more specific antigens and the use of better combinations of PPDs if in vitro testing is intended must be taken into consideration. The uncertain role of interference bacteria that could be affecting the results of the in vitro analysis must be considered, and perhaps this could explain the apparent lack of consensus of the results obtained in Chile versus the ones obtained elsewhere. More studies must be performed in order to assess this issue

    Effect of Mycobacterium bovis infection in cattle on changes in appetite-related hormones

    There are limited studies available for appetite-related hormonal changes in bovine tuberculosis, therefore the purpose of this study was to determine the changes in appetite-related hormones (leptin, ghrelin, and nesfatin -1) of cattle with tuberculosis and to clarify their possible roles. A total of 30 cattle, comprising 15 clinically healthy cattle and 15 cattle infected with tuberculosis were enrolled in the study. The diagnosis of tuberculosis was performed by tuberculin skin test, and changes in the concentrations of ghrelin, leptin, nesfatin-1, lipid profile and muscular enzymes in cattle with tuberculosis were determined. It was found that nesfatin-1, ghrelin and muscular enzymes concentrations were higher in bovine infected with tuberculosis than in the healthy controls. On the other hand, leptin, triglyceride, low density lipoprotein (LDL), high density lipoprotein (HDL), and total cholesterol levels were all lower in the tuberculosis group when compared to the control group. In this study, we detected serum nesfatin-1 concentrations for the first time in Mycobacterium bovis infection in addition to the other known appetite-related hormones. The data suggest that increased concentrations of nesfatin-1 may play a role in weight loss in bovine tuberculosis, but there is a need for further studies to determine its exact role in the etiopathogenesis of bovine tuberculosis

    Physiological and behavioural response of two dairy cows’ genotypes during summertime in the central region of Chile

    Heat stress has been recognised as a serious problem in dairy farms. The study goal was to assess the effects of climatic conditions on physiological and behavioural responses of dairy cows in Central Chile. Data of tympanic temperature (TT), panting score, respiration rate (RR), and shade utilization of cows from two genotypes, Holstein (H) and Holstein x Montbeliarde (HM), were collected twice per day (AM/PM) during three periods of the summer season in Central Chile. Moreover, three thermal comfort indices: Comprehensive climate index (CCI), temperature humidity-index (THI), and adjusted THI were estimated using meteorological data. The hour of each day was classified as “Normal” or “Stressful” based on CCI threshold of 25 °C. Statistical analysis included ANOVA, repeated measures analysis and Chi square test (α=0.05). There was an interaction of genotype x CCI condition x period (P=0.0026) with the highest TT of both genotypes under a stressful condition within each period. In addition, interactions of genotype x hour (P<0.0001) and genotype x CCI condition (P<0.0002) were also observed. The HM cows showed greater TT than H cows in both CCI conditions. The RR was higher during the afternoon and a greater proportion of cows used shade at “Mild” and “Moderate” CCI categories (P<0.001). Both genotypes showed some degree of heat stress, but cool nights and shade seem to be enough to allow to cows’ cope with the challenging diurnal conditions observed in the summer season.     Drug resistance in parasitic helminths of veterinary importance in Chile: status review and research needs

    The increasing development of anthelmintic resistance (AR) in parasites of livestock is threatening animal health and production worldwide. In Chile, studies evaluating the field efficacy of anthelmintics and the detection of AR have been performed since the 2000s, but until now, no previous attempt has tried to systematise the available information. This article reviews general concepts about AR in helminths of veterinary importance, methods for diagnosis of AR and summarises the published reports of AR in Chile. Anthelmintic resistance in Chile has been reported in gastrointestinal nematodes of horses (benzimidazole resistance) and ruminants (sheep and cattle, macrocyclic lactone and benzimidazole resistance). However, these cases involved a limited number of selected farms and no further conclusions can be made of the status of parasite drug resistance at a regional or national level. No published cases of AR in Fasciola hepatica have been reported in Chilean livestock, but human infections with triclabendazole-resistant F. hepatica have been described in patients with previous consumption of watercress or untreated water from marshes grazed by livestock. Given the zoonotic potential and endemic nature of F. hepatica in Chile, it is urgent to determine the extent of liver fluke resistance. Current research gaps of the situation of AR in Chile and suggestions for the performance of laboratory and field studies are further discussed

    Effect of immunocastration on behaviour and blood parameters (cortisol and testosterone) of Holstein bulls

    To evaluate the effect that immunocastration has on behaviour, testosterone and cortisol levels of feedlot Holstein bulls, 720 intact animals aged between 7 and 8 months, weighing 232±1.19 kg were randomly assigned to two treatments: immunocastration using the Bopriva vaccine and a placebo (360 animals per treatment). The bulls were slaughtered at day 239 of treatment. Animals were vaccinated on days 1, 21, 101, and 181, and on those same days testosterone levels were measured; while cortisol, glucose and creatin kinase measurements were done on day 181 and during exanguination at slaughter. Sexual, aggressive and social behaviours were evaluated and it was found that intact bulls showed a higher average of head butts, mounting, threats, flehmen sign and sniffing (P<0.05), no differences were found for vocalisations, lowering of the head and grooming (P>0.05). Testosterone levels in intact bulls remained at 0.47ng/mL throughout the study, however, by day 181 differences (P<0.05) were observed in immunised bulls, with values of 0.22ng/mL. At slaughter, testosterone levels were 0.21 ± 0.06 ng/mL in immunocastrated bulls and 0.54 ± 0.06 ng/mL in the placebo group. The use of immunocastration with Bopriva has shown to be effective to reduce testosterone, sexual and aggressive behaviours on Holstein bulls

    Humoral immune response of pregnant goats to two Corynebacterium pseudotuberculosis bacterin formulations

    Caseous lymphadenitis is a chronic suppurative bacterial disease caused by Corynebacterium pseudotuberculosis, and it has a high prevalence among small ruminants. Different vaccine preparations have been employed to control this disease. The adjuvant used in the formulation is a critical factor for the improvement of vaccine efficacy. No commercial vaccines are available in Argentina. The aims of the present work were to compare the specific antibody response against a C. pseudotuberculosis whole cell vaccine formulated with a classical and an immunoestimulant adjuvant in pregnant goats, and to assess the transfer of vaccine-specific antibodies from the goat to the offspring through colostrum. Twenty three pregnant goats were vaccinated. The animals were randomly allocated to three groups: control, aluminum hydroxide Al(OH)3 adjuvant and a new cage-like particle adjuvant (ISPA). Antibodies (IgG) against this antigen were measured by ELISA. A robust humoral immune response was detected in vaccinated animals, with production of specific IgG. No significant differences were found between the vaccines formulated with different adjuvants. High levels of specific antibodies were detected in colostrum samples from both immunised groups, which were passively transferred from goats to offprings. No adverse local reactions in the vaccines injection sites were detected

    Probiotics on performance, intestinal morphology and carcass characteristics of broiler chickens raised with lower or higher environmental challenge

    This study aimed to evaluate the effect of probiotics on performance, intestinal morphology and carcass characteristics of broiler chickens housed on lower or higher environmental challenge. Three hundred male Cobb chicks were distributed into four groups in completely randomised design with treatments arranged in a 2 × 2 factorial scheme to evaluate effects of two diets (with or without probiotics) and two environmental conditions (lower or higher challenge), totaling four treatments with five replications with 15 birds per box. Probiotics were added on diets and were composed of Lactobacillus acidophilus, Bacillus subtilis, Bifidobacterium bifidum and Enterococcus faecium. The environment with lower challenge was made up of new wood shavings used as litter, low bird density (8 birds/m2) and daily-cleaned bell drinkers. The environment with higher challenge was made up of re-used wood shavings used as litter (after three broods of broilers), bird density of 16 birds/m2 with bell drinkers cleaned every two days. No significant interaction between diet and environmental challenge was found for any of the variables evaluated. Performance, slaughterhouse variables and chemical carcass composition were not affected by the use of probiotics. Chicks receiving diets without probiotics had lower intestinal crypt depth (267.1 vs. 316.6 ?m, P=0.0068). Birds raised in the environment with higher challenge decreased feed intake (4,660 vs 5,020 g, P=0.0422), weight gain (2,610 vs 2,810 g, P= 0.0054), drumstick and thigh yield (21.98 vs 24.14 %, P=0.0354), and increased crypt depth (325.2 vs 258.5 μm P=0.0009). In conclusion, the probiotic does not promote satisfactory improvements, regardless of the environmental challenge

    Effect of diet on live weight and egg weight of backyard hens during the rainy season

    The objective was to determine the effect of diet on live weight (LW) and egg weight (EW) of backyard hens (BH) during the rainy season in the Bajío region of the state of Michoacán, Mexico. Seventeen municipalities were sampled, where 101 BH (six hens/municipality) were captured and weighed and 101 eggs (six eggs/municipality) were harvested and weighed. The crops of the captured hens were removed Post-sacrifice, the organic content/crop (OCC) was classified and weighed by components, to later perform chemical compositional analysis. Data were analysed using generalised linear models and the differences between municipalities were obtained by the method of least squares means. The weight of the OCC (36.4 ± 22.4 g) was affected by the municipality (P<0.001), but not by the LW of the BH (P>0.05). Commercial feed (8.1 ± 6.0 g), grains: maize and sorghum (13.9 ± 13.5 g) kitchen waste (1.5 ± 2.9 g), herbaceous (0.6 ± 0.9 g) and insects (0.3 ± 0.7 g) were found in the crop. According to the commercial feed (COF) component, two feeding systems (FS) were identified: traditional FS, without COF and nontraditional FS, with COF. The diets of both FS were similar (P>0.05) in nutritional composition and do not meet the nutritional requirements of the hens. The LW (1.567 ± 0.316 kg) and EW (51.3 ± 1.0 g) of the BH cannot be completely attributed to the diet consumed during the rainy season

    Cell block sensitivity for immunohistochemical detection of cytokeratin 5, oestrogen and progesterone receptors in canine primary mammary carcinoma

    Mammary carcinomas are relatively common ailments among female canines aged around 10 years old, presenting an important morbidity with an average survival of five years. The cytoinclusion technique is frequently employed in human medicine as the investigative method of choice as it quickly provides resources for the determination of the correct therapeutic response, however, the effectiveness of the technique in canines remains understudied in veterinary medicine. This study aims at evaluating the degree of correlation with immunohistochemical marking for cytokeratin 5 (CK5), oestrogen and progesterone receptors (ER and PR) between the cytoinclusion and the histopathology technique in mammary carcinomas. Twenty-five samples of mammary carcinoma, both for the cytoinclusion and histopathological techniques were submitted for histological processing; microscope slides were created for hematoxylin-eosin (HE) staining and the immunohistochemical technique (IHC) was assessed for the ER, PR and CK5 receptors. Through the HE staining, we reached a concordance rate of 100% between the cytoinclusion and the histopathological analysis in the diagnosis of carcinomas. The immunohistochemical assay presented sensitivity of 85.71%, 95.45% and 100% and Cohen’s kappa of 0.78, 0.84 and 0.95 for ER, PR and CK5, respectively, as well as 100% specificity and P<0.01 for all three markers. Therefore, cytoinclusion represents an accessible, fast and low-cost method, offering high sensitivity for the cytomorphological and immunohistochemical diagnosis of mammary carcinoma in female canines

    Absence of molecular evidence of Leptospira spp. in urine samples collected from rodents captured in Yucatán, México

    Leptospira spp. is a spirochete bacteria, causal agent of leptospirosis, zoonotic disease endemic in México that represents a serious public health and veterinary problem. Rodents are recognised as the most important reservoirs of this bacteria, which is transmitted mainly through direct or indirect contact with the Leptospira spp. excreted in the urine of infected individuals. Theaim of this study was to evaluate the circulation of Leptospira spp. in urine samples of wild and synanthropic rodents from Yucatán, México. Eighty-four rodents were captured in the community of Cenotillo, Yucatán. Twenty-six urine samples were collected from the bladder and were used in the total DNA extraction. The identification of Leptospira spp. was intended through the polymerase chain reaction test in its endpoint variant. No evidence of Leptospira spp. was found in the urine samples. It is necessary to use other tissues for the identification of Leptospira spp., before concluding that the rodents used in the present study are not reservoirs of this bacteri
