    Determination of Suppressed & Diverted Travel Demands: İzmir Case Study

    Şehir plancıları, ulaşım mühendisleri ve politika yapıcılar etkili ulaşım sistemleri tasarlamak, politikalar oluşturmak ve yatırım stratejileri geliştirmek için halktan gelen verilere güvenirler. Bu hayati bilgilerin toplanması, seyahat talebi verilerini yakalayan hane halkı anketleri gibi yöntemlerle gerçekleştirilir. Bu verileri analiz etmenin önemli bir yönü, bastırılmış ve yönlendirilmiş seyahat taleplerini incelemeye odaklanarak, görünür talepler ile gizli talepler arasındaki farkı anlamaktır. Bu talepler, daha etkili ulaşım politikaları ve planlama stratejileri geliştirmek için anahtardır. Bu tezin temel amacı, bastırılmış ve yönlendirilmiş talepler arasındaki farkları keşfetmek için İzmir'deki seyahat taleplerini belirlemek ve analiz etmek, bunları gerçek seyahat davranışlarıyla karşılaştırmaktır. Bu araştırma, İzmir'i temsil eden yaklaşık 913 katılımcıyla yürütülen bir çevrimiçi anketten elde edilen verilerden yararlanmaktadır. Anket, hem belirtilen hem de açıklana tercih tekniklerini kullanarak mevcut seyahat davranışlarının yanı sıra bastırılmış ve yönlendirilmiş talepleri değerlendirdi. Ki-kare testi kullanılarak yapılan ilk analizler, yönlendirilmiş ve bastırılmış talepler arasında mevcut seyahat davranışlarına kıyasla önemli farklılıklar olduğunu ortaya koydu. Daha ileri kategorik veri analizi, tüm talep türlerinin mevcut seyahat tercihleriyle önemli bir ilişkiye sahip olduğunu gösterdi. Özellikle, bastırılmış ve yönlendirilmiş talepler, raylı sistemlere yönelik güçlü bir tercihle, mod seçiminde en yaygın olanıydı. Çalışma, talebin %43'ünün mod tarafından yönlendirildiğini, talebin %80'inin ise bastırıldığını buldu. Ek olarak, katılımcıların %25'i, seyahat taleplerini belirlemede en etkili faktör olarak trafik sıkışıklığını gösterdi.City planners, transportation engineers, and policymakers rely on data from the public to design effective transportation systems, create policies, and develop investment strategies. Gathering this vital information is achieved through methods such as household surveys that capture travel demand data. A crucial aspect of analyzing this data is understanding the difference between apparent demands and latent demands, with a focus on examining suppressed and diverted travel demands. These demands are key to developing more effective transportation policies and planning strategies. The primary objective of this thesis is to identify and analyze travel demands in Izmir, comparing them with actual travel behaviors to explore the differences between suppressed and diverted demands. This research draws on data from an online survey conducted with approximately 913 participants representing Izmir. The survey assessed current travel behaviors as well as suppressed and diverted demands, using both stated and revealed preference techniques. Initial analyses using the chi-square test revealed significant differences between diverted and suppressed demands compared to current travel behaviors. Further categorical data analysis showed that all demand types had a significant relationship with current travel preferences. Notably, suppressed and diverted demands were most prevalent in modal choice, with a strong preference for rail systems. The study found that 43% of the demand is diverted by mode, while 80% of the demand is suppressed. Additionally, 25% of participants cited traffic congestion as the most influential factor in determining their travel demand

    Comparison of Cell-Penetrating and Fusogenic Tat-Ha2 Peptide Performance in Peptideplex, Multicomponent, and Conjugate Sirna Delivery Systems

    In this study, the performance of the cell-penetrating and fusogenic peptide, TAT-HA2, which consists of a cell-permeable HIV trans-activator of transcription (TAT) protein transduction domain and a pH-responsive influenza A virus hemagglutinin protein (HA2) domain, was comparatively evaluated for the first time in peptideplex, multicomponent, and conjugate siRNA delivery systems. TAT-HA2 in all three systems protected siRNA from degradation, except in the conjugate system with a low Peptide/siRNA ratio. The synergistic effect of different peptide domains enhanced the transfection efficiency of multicomponent and conjugate systems compared to that of peptideplexes, which was attributed to the surface configuration of TAT-HA2 peptides depending on the nature of attachment. Particularly, the multicomponent system showed better cellular uptake and endosomal escape than the peptideplexes, resulting in enhanced siRNA delivery in the cytoplasm. In addition, the presence of cleavable disulfide bonds in multicomponent and conjugate systems promoted the effective siRNA delivery in the cytoplasm, resulting in improved gene silencing activity. The multicomponent system reduced the level of luciferase expression in SKOV3 cells to 45% (+/- 4). In contrast, the conjugate system and the commercially available siRNA transfection agent, Lipofectamine RNAiMax, caused luciferase suppression down to 55% (+/- 2) at a siRNA dose of 100 nM. For the same dose, the peptideplex system could only reduce the luciferase expression to 65% (+/- 5). None of the developed systems showed significant toxicity at any dose. Overall, the TAT-HA2 peptide is promising as a siRNA delivery vector; however, its performance depends on the nature of attachment and, as a result, its surface configuration on the developed delivery system

    Development of material flow stress and damage models for 304 stainless steel

    Paslanmaz çelik 304 alaşımı üzerine yapılan önceki deneysel ve sayısal çalışmalar çoğunlukla martensitin hacim kesrine bağlı akış gerilimi davranışının belirlenmesine odaklanmıştır. Alaşımın hasar davranışı darbe ile ilgili uygulamalarda eşit derecede önemlidir. SS 304 alaşımının dinamik yükleme davranışını simüle etmek amacıyla hasar modellerinin belirlenmesi konusunda henüz sistematik bir çalışma yapılmamıştır. Bu tezde, Johnson ve Cook (JC) akış gerilimi ve JC hasar denklemlerinin parametreleri bir paslanmaz çelik 304 alaşımı için deneysel olarak belirlenmiştir. Belirlenen parametreler daha sonra bu parametreleri çıkarmak için kullanılan deneysel testlerin modellenmesiyle doğrulanmış ve kalibre edilmiştir. Sayısal modeller LS-Dyna'da uygulanmıştır. B4C kaplı ve kaplanmamış SS 304 plakalar üzerinde 800 m s-1'de gerçekleştirilen deneysel balistik testler, belirlenen model parametreleri kullanılarak Ls-Dyna'da simüle edilmiştir. Son olarak, mikroskobik çalışmalar test edilen alaşımdaki martensitik dönüşümün ve yüksek gerinim oranlarındaki adiabatik ısının martensit oluşumunun kapsamını azalttığını açıkça göstermiştir. Son olarak, martensit fraksiyonu literatürdeki denklemler kullanılarak farklı gerinim oranlarında analitik olarak tahmin edilmiştir.Previous experimental and numerical studies on stainless steel (SS) 304 alloy have been mostly focused on the determination of the flow stress behaviour as function of the volume fraction of martensite. The failure behaviour of the alloy is equally important in the impact related applications. No systematic studies have been yet performed on the determination of the damage models of SS 304 alloy in order to simulate its dynamic loading behaviour. In this thesis, the parameters of the Johnson and Cook (JC) flow stress and JC damage equations were experimentally determined for an SS 304 alloy. The determined parameters were then verified and also calibrated by modelling the experimental tests used to extract these parameters. The numerical models were implemented in LS-Dyna. The experimental ballistics tests performed at 800 m s-1 on B4C coated and uncoated SS 304 plates were further simulated in Ls-Dyna using the determined model parameters. Finally, the microscopic studies have clearly indicated the martensitic transformation in the tested alloy and adiabatic heat at high strain rates reduced the extent of the martensite formation. Finally, the fraction of martensite was predicted analytically at different strain rates using the equations from the literature

    The design and optimization of multiscale hybrid nanocomposite structures for vibration and buckling behavior

    Bu tezde, otomotiv, havacılık ve uzay sanayi gibi endüstrilerde yaygın olan geleneksel sentetik karbon ve cam elyaf takviyeli kompozit yapılara alternatif olarak çok fazlı hibrit doğal fiber takviyeli nanokompozit yapılar sunulmaktadır. Alternatif tasarımların kritik burkulma yükünü, doğal frekansını ve yapısal güvenlik faktörünü maksimize etmek için Differential Evolution, Simulated Annealing, ve Nelder-Mead stokastik optimizasyon yöntemleri kullanılmıştır. Fiber hacim oranı, fiber oryantasyon açısı ve her tabakadaki Karbon Nanotüplerin (CNT) veya Grafen Plaketlerin (GPL) hacim içeriği eş zamanlı olarak tasarım değişkenleri olarak kullanılmıştır. CNT veya GPL ile güçlendirilmiş matrislerin etkili malzeme özellikleri, nanodolgu maddelerinin kümelenme etkileri göz önünde bulundurularak Modifiye Halpin-Tsai denklemleri ve karışım kuralı kullanılarak hesaplanmıştır. Çok fazlı ara hibrit fiber tabakalı nanokompozit yapıların vibrasyon, burkulma ve hasar analizleri, hem analitik yöntemler (Navier çözümü ile Birinci Derece Kayma Deformasyon Teorisi (FSDT) ve Klasik Laminasyon Teorisi (CLT)) hem de Sonlu Elemanlar Metodu (FEM) kullanılarak yapılmıştır. Maksimum mekanik özellik ve minimum ağırlık ve maliyet için çok amaçlı optimizasyon problemleri, geleneksel kompozit yapılara çevre dostu, hafif ve düşük maliyetli alternatifler önerebilmek amacıyla Ceza Fonksiyonu yaklaşımı kullanılarak stratejik bir şekilde gerçekleştirilmiştir. Ayrıca, otomotiv endüstrisinde potansiyel gelecekteki uygulamalar için optimum nanokompozit tahrik mili tasarımları, kalınlık boyunca kümelenme etkileri dahil olmak üzere eşit olmayan şekilde dağılmış fiber ve CNT dağılımına sahip hibrit Karbon/Keten/CNT takviyeli yapılarla önerilmiştir. Genel sonuçlar, doğal fiberlerin GPL veya CNT ile optimize edilmesinin, mühendislik yapılarında sadece çevresel sürdürülebilirlik açısından değil, aynı zamanda ağırlık, maliyet, frekans ve burkulma özelliklerine dayalı kompozit malzeme tasarımının performansı açısından da avantajlar sağladığını göstermiştir.This thesis presents multiscale hybrid natural fiber-reinforced nanocomposite structures as viable alternatives to the traditional synthetic carbon and glass fiber composites commonly used in industries like automotive, aviation, and aerospace. These alternatives were created using stochastic optimization methods—Differential Evolution, Simulated Annealing, and Nelder-Mead algorithms—to optimize critical buckling load, fundamental frequency, and factor of safety, while reducing weight and cost. A broad range of design variables were employed, including fiber volume fraction, stacking sequences, and the volume content of Carbon Nanotubes (CNTs) or Graphene Platelets (GPLs) in each layer. The effective material properties of matrices reinforced with CNTs or GPLs were determined using the Modified Halpin-Tsai equations and the rule of mixtures, accounting for the agglomeration effects of the nanofillers. Vibration, buckling, and failure analyses of multiphase hybrid fiber-reinforced nanocomposite structures were performed using both analytical methods (Navier's solution with First-order Shear Deformation Theory (FSDT) and Classical Laminated Theory (CLT)) and the Finite Element Method (FEM). A multi-objective optimization problem was strategically executed using the Penalty Function approach to propose optimal eco-friendly, lightweight, and cost-effective alternatives to conventional composite materials, aiming for maximum mechanical response with minimal weight and cost. Additionally, optimal nanocomposite driveshaft designs were proposed for future automotive applications, featuring hybrid Carbon/Flax/CNT structures with non-uniform fiber and CNT distribution, accounting for agglomeration effects. The results indicated that optimizing natural fibers with GPLs or CNTs in engineering structures offers substantial benefits, enhancing both environmental sustainability and composite material performance in terms of weight, cost, frequency, and buckling properties

    Synthesis of composite filling materials for non-scratch paints and coatings

    Modern boyalar ve kaplamaların mekanik ve fiziksel özelliklerinin geliştirilmesi, otomotiv, inşaat ve denizcilik gibi belirli endüstriyel uygulamalar için oldukça önemlidir. Bu araştırma, dayanıklılığı ve çizilme direncini artırmayı hedefleyen, boya ve kaplamalarda kullanılmak üzere silika (SiO₂) ve bor-silika kompozitleri (Boron-SiO₂) içeren kompozit dolgu malzemelerinin geliştirilmesine odaklanmaktadır. Çalışma, bu dolguların diğer katkı maddeleriyle birlikte koruyucu kaplamaların mekanik ve işlevsel özelliklerini nasıl geliştirebileceğini incelemektedir. Silika nanoparçacıklarının iyi bilinen kimyasal stabilitesi, sertliği ve yüksek yüzey alanından yola çıkarak, bu çalışmada kullanılan üretim tekniği Stöber sentez yöntemidir. Bu teknik, partikül boyutu, morfolojisi ve dağılımı üzerinde hassas kontrol sağlar ve kaplama özelliklerinin yüksek bir doğrulukla ayarlanmasına olanak tanır. Ayrıca, boyalar/kaplamalar için çizilme direncini daha da geliştirmek amacıyla Boron-SiO₂ kompozitlerinin sentezi gerçekleştirilmiştir. Silika matrisine bor eklenmesi, malzemenin mekanik dayanımını ve dayanıklılığını artırmış, böylece onu koruyucu uygulamalar için son derece uygun hale getirmiştir. Boron-SiO₂ parçacıklarının özelliklerini optimize etmek için kontrollü içerik ekleme yöntemleri dahil olmak üzere çeşitli sentez teknikleri incelenmiştir. Elde edilen kompozit nanoparçacıklar, SEM, STEM, TEM, FTIR ve ICP-MS gibi karakterizasyon teknikleriyle birlikte parçacık yapısını incelemek için ek testlere tabi tutulmuştur. Çizilme direnci performans testleri, Boron-SiO₂ kompozitleri ile doldurulmuş kaplamaların, üretim yöntemine bağlı olarak geleneksel silika dolgulu kaplamalara göre önemli ölçüde daha iyi performans gösterdiğini ortaya koymuştur.The development of mechanical and physical properties in modern paints and coatings are highly relevant to specific industrial applications, such as automotive, construction, and marine industries. This research focuses on developing composite filler materials comprising silica (SiO₂) and boron- silica composites (Boron-SiO₂) for use in paints and coatings, aiming to enhance durability and scratch resistance. The study investigates how these fillers, together with other additives, can improve the mechanical and functional properties of protective coatings. Starting with the well-known chemical stability, hardness, and high surface area of silica nanoparticles, the production technique used in this study is the Stöber synthesis method. This technique allows for precise control over particle size, morphology, and distribution, enabling adjustments to coating characteristics with a high degree of accuracy. Furthermore, the synthesis of Boron-SiO₂ composites is performed to further improve scratch resistance of paints/coatings. The addition of boron to the silica matrix enhanced its mechanical strength and durability, making it highly suitable for protective applications. Various synthesis techniques, including controlled contents additon methods, were investigated to optimize the properties of Boron-SiO₂ particles. The resulting composite nanoparticles were then characterized using SEM, STEM, TEM, FTIR, and ICP-MS, along with additional tests to examine particle structure. The scratch resistance performance tests revealed that coatings filled with Boron-SiO₂ composites perform significantly better than conventional silica-filled coatings, depending very much on the method of boron-silica composite nanoparticles were produced

    Submerged breakwaters as a measure to reduce wave overtopping at vertical seawalls

    Kıyı kentlerindeki hızlı kentleşme ve artan nüfus, kentsel alanlar üzerindeki baskıyı artırmakta. Bu durum küresel ısınmanın tetiklediği iklim değişikliğiyle daha da şiddetlenmektedir. İklim değişikliğinin en belirgin etkilerinden biri olan deniz seviyesinin yükselmesi, özellikle fırtına koşullarıyla birleştiğinde kıyı koruma yapılarının etkinliğini azaltarak kentsel bölgelerin taşkın riskini artırmaktadır. Bu bağlamda, dalga aşmasını azaltmak için batık bir dalgakıran çözüm olarak kullanılabilir. Bu çalışma, İYTE Hidrolik Laboratuvarında, 122M553 kodlu TÜBİTAK projesi kapsamında gerçekleştirilen 2 boyutlu, küçük ölçekli deneysel bir araştırma olup, dikey deniz duvarlarında dalga aşmasını azaltmada batık dalgakıranların etkinliğini incelemektedir. Batık dalgakıranların, dalga aşma debisini %90'a kadar azaltarak etkin bir tamamlayıcı kıyı savunma sistemi olarak kullanılabileceği gösterilmiştir. Batık dalgakıranın etkinliği, dalgaların aşması üzerindeki etkisini ölçen bir indirgeme katsayısı (sub) ile ifade edilir. Bu indirgeme katsayısı, dalgakıran yüksekliği ve kret genişliği gibi batık dalgakıranın geometrik parametreleri ile dalga yüksekliği, dalga boyu ve su derinliği gibi hidrodinamik koşullardan da etkilenir. Bu parametrelerin etkisi dalga aşmasındaki azalmanın belirlenmesinde önemli bir rol oynar. 128 farklı deney testi kullanılarak, dalga aşmasındaki azalma niceliksel olarak belirlenmiştir. Daha sonra batık dalgakıranın tasarım parametrelerinin bir fonksiyonu olarak ifade edilmiş azaltma faktörü tanımlanmıştır. Bu indirgeme katsayısı, yaygın olarak kabul gören EurOtop (2018) dalga aşması tahmin formülüne entegre edilerek, batık dalgakıranın dikey deniz duvarı önüne yerleştirilmesi durumunda, dalga aşma deşarjının tahmin edilmesini sağlar. Ayrıca bu deneysel çalışmada, literatürde önerilen dalga iletim formüllerinin etkinliği, bu çalışmada üretilen veri seti ile karşılaştırılarak değerlendirilmiştir.Rapid urbanization and population growth in coastal cities are increasing pressure on urban areas. This pressure is fed by climate change caused by global warming. Sea level rise, one of the most significant impacts of climate change, increases flood risk of urban areas by reducing the effectiveness of coastal protection structures, especially during storms. Therefore, submerged breakwater can be used as a solution to reduce wave-overtopping. This study is 2D, small-scale experimental research carried out in IZTECH Hydraulics Laboratory within the scope of TUBITAK project 122M553 and investigates the performance of submerged breakwaters in reducing wave overtopping on vertical seawalls. It is shown that submerged breakwaters can be used as an effective coastal defense system by reducing the overtopping discharge by up to 90%. The effectiveness of the submerged breakwater is expressed through a reduction factor (sub), which quantifies its impact on wave overtopping. Reduction factor is influenced by geometric parameters of the submerged breakwater, such as breakwater height and crest width, as well as hydrodynamic conditions including wave height, wavelength, and water depth. Based on 128 different experimental tests, relationship between these parameters and wave overtopping reduction is quantified, and the reduction factor is formulated as a function of submerged breakwater's design parameters. Reduction factor is then integrated into widely accepted EurOtop (2018) wave overtopping prediction formula to provide an estimate of wave overtopping discharge in the presence of a submerged breakwater. Furthermore, the effectiveness of wave-transmission formulas proposed in literature is evaluated by comparing them with the data set generated in this study

    Evanescent field-based optical force techniques for the manipulation of spherical and non-spherical nanoparticles

    Bu tez, sönümlenen dalgaların fiziksel mekanizmalarını ve optik kuvvetlerin nanoparçacık manipülasyonundaki etkinliğini teorik analizler ve simülasyonlar yoluyla incelemektedir. İki ve dört Gauss kaynağı kullanan sistemler, yenilikçi parçacık manipülasyonu için faz modülasyonu ile zenginleştirilmiş sönümlenen alanlar oluşturmak üzere kullanılmıştır. Asimetrik sönümlenen alanlara uyumluluğu, doğruluğu ve hesaplama verimliliği nedeniyle, optik kuvvet hesaplamalarında Ayrık Dipol Yaklaşımı (Discrete Dipole Approximation - DDA) yöntemi tercih edilmiştir. Tezin önemli bir odak noktası, faz modülasyonuna dayalı olarak tanımlanan ve değerlendirilen modüller —Holder, Centrifugal, Side-Puller ve Rotator— aracılığıyla parçacık hizalama, yer değiştirme ve ayrıştırma süreçleridir. Her bir modül, parçacık manipülasyonunda benzersiz avantajlar sunmuştur: Holder modülü, parçacıkları yüksek yoğunluk gradyanlarına sahip bölgelerde sabitlerken, Centrifugal modülü parçacıkları kanal kenarlarına yönlendirmiştir. Side-Puller modülü parçacıkları belirli bir yönde hareket ettirirken, Rotator modülü açısal hareketi kontrol etmiştir. Bu modüllerin bir arada kullanılması, kontrol mekanizmalarını geliştirerek daha verimli filtreleme sistemleri sağlamıştır. Tezde ayrıca sıvı akışı ile optik kuvvetlerin birleştirildiği senaryolar incelenmiş ve bu kombinasyonların partikül ayrıştırılmasındaki etkinliği değerlendirilmiştir. Farklı boyutlardaki küresel partiküllerin ayrıştırılması, optik ve sürükleme kuvvetlerinin dengesi üzerinden analiz edilmiştir. Küp ve küresel partikül hareketleri incelenerek geometrik farklılıkların optik kuvvetler üzerindeki etkisi gösterilmiş, ardından eşit hacimdeki küre ve elipsoidal partiküller üzerindeki çalışmalarla partikül geometrisinin ayrışma dinamikleri üzerindeki etkisi detaylandırılmıştır. Bu çalışma, optik kuvvetlerin nanopartikül manipülasyonunu nasıl optimize ettiğini ve sönümlenen dalgaların sunduğu benzersiz avantajlar sayesinde gelişmiş filtreleme ve kontrol mekanizmaları sağladığını ortaya koymaktadır. Önerilen faz modülasyon sistemleri ve DDA yöntemi, hem küresel hem de küresel olmayan nanopartiküllerin kontrolü ve ayrıştırılması için yenilikçi bir çerçeve sunmaktadır. Çalışmanın bulguları, biyoteknoloji, nanoteknoloji ve mikroskobik akışkan dinamiği gibi alanlarda optik kuvvet temelli uygulamaların geliştirilmesi için önemli bir potansiyel taşımaktadır.This thesis examines the physical mechanisms of evanescent waves and the effectiveness of optical forces in nanoparticle manipulation through theoretical analysis and simulations. Systems with two and four Gaussian sources were used to generate evanescent fields, enriched through phase modulation for innovative particle manipulation. The Discrete Dipole Approximation (DDA) method, chosen for its accuracy and computational efficiency, was instrumental in studying non-spherical particles due to its adaptability to asymmetric evanescent fields. A major focus is the definition and evaluation of phase-modulated modules — Holder, Centrifugal, Side-Puller, and Rotator—for particle alignment, displacement, and separation. Each module offered unique advantages in particle manipulation; the Holder module stabilized particles in regions with high-intensity gradients, while the Centrifugal module directed particles outward to the channel edges. The Side-Puller module guided particles in a specific direction and the Rotator module controlled angular motion. Cascading these modules enhances control and enables efficient filtering mechanisms. The thesis also investigates scenarios combining fluid flow with optical forces, evaluating their effectiveness in particle separation. The separation of spherical particles of varying sizes was analyzed based on the balance between optical forces and drag forces. The study of cubic and spherical particle movements demonstrated how geometric differences influence optical forces. Subsequently, the examination of particles with equal volumes, such as spheres and ellipsoids, provided insights into how particle geometry affects separation dynamics. This study demonstrates that optical forces refine nanoparticle manipulation, enabling advanced filtering and control through the unique properties of evanescent waves. The proposed phase modulation schemes and the DDA method provide a novel framework for controlling and separating both spherical and non-spherical nanoparticles. The findings highlight the potential for developing optical force-based applications in fields such as biotechnology, nanotechnology, and microscopic fluid dynamics

İki tür arasındaki yapısal benzerlikler, Elastik Ağ-Yapı Modeli, Temel Bölge Tarama Analizi (ESSA), DrugRep, CB-Dock2 ve Pharmit dahil olmak üzere çeşitli hesaplama teknikleri kullanılarak analiz edildi. Bu tezde sanal tarama çalışmalarından yüksek bağlanma afinitesine sahip üç aday molekül tespit edildi.This thesis investigates potential allosteric binding sites in the human and parasitic glucokinase enzymes to facilitate species-specific therapeutic design. The structural similarities between the two species were analyzed using various computational techniques, including the Elastic Network Model, Essential Site Scanning Analysis (ESSA), DrugRep, CB-Dock2, and Pharmit. These methods successfully identified all known catalytic and allosteric sites in human glucokinase, supporting the accuracy of the findings. Notably, an alternative allosteric site in T. cruzi was proposed for targeted drug design. The ESSA analysis of T. cruzi, employing cut-off values of 7 Å, 10 Å, and 13 Å, revealed the known active binding site and identified a potential allosteric site characterized by significant residues such as R216, Y221, H247, Q250, K281, Y285, and R288. This emphasizes the necessity of using multiple cut-off values to capture subtle structural features. Additionally, the analysis of human glucokinase in both free and complex forms elucidated critical amino acids involved in glucose interaction while revealing that the identified region in T. cruzi does not correspond to any binding site in human glucokinase, highlighting unique structural adaptations between the two enzymes. Virtual screening and docking studies were conducted on the potential binding site in T. cruzi using the DrugRep server, further advancing the understanding of species-specific interactions for therapeutic development. Three candidate molecules with high binding affinities were detected from virtual screening studies in this thesis

    1,2-Diboranes With Strong Donor Substitutes: Synthesis, Ovicidal and Larvicidal Effect on Important Vector Species

    Novel control products are needed in the control of important insects like mosquitoes which are developing resistance to insecticides and larvicides currently in the market. Boron compounds have been demonstrated to exhibit antibacterial and anticancer effects. 1,2-diboranes with a long history and importance in boron chemistry have been described. These compounds are synthesized from reactions of 1,2-dichlordiborane derivatives with lidium amides (ArNHLi/Et2NLi, etc.). In addition to the three previously synthesized diborane compounds, five novel 1,2-diborane compounds were synthesized in good yield using the same method for the first time. The structures of the novel derivatives were characterized by nuclear magnetic resonance spectroscopy, and the molecular structure of one of them (2a) was also demonstrated using single crystal X-ray diffraction. In this preliminary study, the ovicidal and larvicidal effects of new 1,2-diamino-1,2-diborane derivatives against Aedes aegypti and Aedes albopictus eggs and larvae were investigated for the first time. Of these, 2a and 2e showed the highest ovicidal activity against both species, while 7, 4 and 2d showed particularly high larvicidal activity. Some 1,2-diborane derivatives were found to be significantly toxic, with LC50 values ranging from 14,930 to 27,975 mu g/mL. Some derivatives (6, 2a, 2c) were less effective against mosquito larvae. 1,2-Diborane derivatives have high ovicidal and larvicidal effects on mosquitoes and are therefore potential candidates for the development of new larvicides. Further studies are needed to evaluate its mode of action and safety. Understanding their mode of action against mosquito development is crucial to optimizing their use and reducing the potential development of resistance. Their potential effects on other mosquito species and non -target organisms need to be investigated

    Investigation of Coupling Methods for Floating Offshore Wind Turbines and Their Application for an Innovative Floating Platform

    The search for more sustainable energy sources to replace fossil fuels has encouraged the development of several energy sectors, including floating offshore wind energy. Growth in this area focuses on research in platforms for aero-hydro-servo-elastic codes such as OpenFAST and aero-hydro coupling methods. In this thesis, the innovative floating platform, which was previously designed for 300 kW and 5 MW capacities, has been modified in terms of dimensions and features to be suitable for analysis with the 5 MW NREL reference wind turbine. The study examined coupling methods for spar, semi-submersible, common in the literature, and innovative platforms. This involved determining natural periods and damping ratios to see the system responses and behaviors of turbines and platforms, free decay tests, and various loads, including steady and turbulent wind conditions in regular, irregular wave conditions. The analysis provided several conclusions, including the verification of coupling methods, comparative evaluations of different platforms, and investigations into the behaviors of the innovative platform under flexible and rigid configurations. In this context, open-source programs like Capytaine and NEMOH and the commercial program AQWA produce results similar to WAMIT's OpenFAST outputs. The F2A coupled method also shows strong correlation and consistency with these results. Furthermore, while the innovative platform with mooring lines achieves higher values than other platforms, its pitch angle is significantly lower, which is crucial for system stability, fatigue, and extreme loads. This reduces loading on components such as the tower blade and yaw bearing.Fosil yakıtların yerini alacak daha çevreci enerji kaynaklarına olan ilgi arttıkça, çeşitli enerji alanları da devreye girmektedir. Bunlardan biri ve giderek artan potansiyeli ile dikkat çeken deniz üstü yüzer rüzgâr enerjisi, önemli araştırma konularından biri olarak platformun OpenFAST gibi aero-hidro-servo-elastik programlara entegrasyonu veya benzer aero-hidro kuvvetlerinin bir arada verilmesi yöntemlerinin incelenmesini kapsamaktadır. Bunun sonucunda, akuple yöntemlerinin doğrulanması, platformların birbirleriyle karşılaştırılması, yenilikçi platformun esnek ve rijit yapısındaki farklı davranışlar incelenmiştir. Bu bağlamda, OpenFAST'a girdi olarak hidrodinamik katsayı sağlayan açık kaynaklı Capytaine ve NEMOH gibi programların yanı sıra AQWA gibi ticari programların, WAMIT gibi yaygın olarak kullanılan ticari programın OpenFAST çıktılarıyla benzer sonuçlar verdiği görülmüştür. Ayrıca, F2A akuple yönteminin de bu sonuçlarla oldukça tutarlı olduğu gözlemlenmiştir. Ayrıca, platformlar karşılaştırıldığında, bağlama halatlarındaki kuvvetlerin özgün platformda diğer platformlara göre daha yüksek olmasına rağmen, özellikle sistemin kararlılığı açısından önemli bir kriter olan platformun yunuslama açısının önemli ölçüde daha düşük olması sebebiyle, kule, kanat ve sapma yatağı gibi bileşenler üzerinde diğer platformlara kıyasla daha az yük oluştuğu hesaplanmıştır
