Comprehensive assessment of genetic variation in native heterostylous primrose genotypes of Türkiye
Primula vulgaris L. is an essential species of ornamental plant value with attractive flowers distributed in humid and cold regions. It spreads around Giresun, Gümüşhane, Trabzon, and Rize in Türkiye’s Eastern Black Sea region. Through molecular and morphological markers, this study determined the genetic diversity and similarity levels of Primula genotypes sampled from different areas (Samsun, Trabzon, Ordu, and Giresun). Variation was determined regarding morphological characteristics, and that plant shape, flower, and leaf properties were highly variable. Molecular data support morphological features. Thirteen ISSR primers determined the dimensions of genetic diversity, and the genotypes’ similarity indexes were between 0.70 and 0.92. It is seen that there is no clustering in the two- and three-dimensional graphics created in line with the genetic parameters, and the genotypes are distributed. Also, plant characteristics were correlated with the ISSR data, and many DNA profiles were detected. This study has characterized Primula genotypes that naturally spread in the Black Sea region of Türkiye. It is a guiding study for transferring some observed properties to culture varieties
Morphological and biochemical changes with hormone and hydro- priming applications in safflower (Carthamus tinctorius L.) seedlings under salinity stress conditions
In this study, the ameliorative effects of hydro- and hormopriming applications against salt stress of safflower seeds during germination and seedling development stage were investigated. Primed (hydropriming (0, 1, 2 and 3 days) with distilled water and hormopriming for 24 hours (0, 50, 100 and 150 mg L-1 with kinetin) and nonprimed seeds were sown under control (no salt) and salt stress (0, 50, 100 and 150 mM NaCl) conditions. When priming applications under salt stress are compared with the control, germination percentage (GP), seedling lenght (SL), root length (RL), seedling fresh weight (SFW), root fresh weight (RFW), leaf relative water content (RWC), elctrolyte leakeage (EL) parameters reducing the effects of stress, however, it was determined that caused an increase in carotenoid (Car), superoxide dismutase (SOD), total phenolic compounds (TPC) parameters. Besides this, it has an inhibitory effect on the increase in proline (Pro) and malondialdehyde (MDA) content. According to the correlation analysis, significant positive correlation was determined in all parameters. A significant positive correlation was determined for hydropriming GP with SL, RL, SFW, RFW, and for hormopriming with SL, RL, SFW, RFW, total Chl, RWC and GP. According to the PCA analysis, the parameters examined in both applications it is seen that they are divided into four different groups. In conclusion, this study priming applications are compared with each other, it has been determined that hormopriming is more effective in reducing the effects of stress than hydropriming
Effect of Yupingfeng granules combined with glucocorticoid on eosinophil infiltration and inflammatory factors of lavage fluid in nasal secretion of patients with allergic rhinitis
The purpose of this study was to investigate the effect of Yupingfeng granules, a well-known traditional Chinese medication, combined with glucocorticoids on eosinophil (EOS) number and inflammatory factors in nasal secretions of patients with allergic rhinitis (AR). 76 patients with AR were randomly divided into controls (nasal inhaled corticosteroids alone) and experimental group (combined with Yupingfeng granules orally). The efficacy, symptoms, number of EOS and levels of inflammatory factors in nasal secretions were compared between the two groups after 2 months of treatment. The results showed that in the experimental group (94.73%, 0.48 ± 0.25 points), the effective rate of treatment was obviously higher and the physical sign score was clearly lower compared with controls (81.57%, 0.97 ± 0.36 points); the EOS count in the experimental group was clearly less than that in the controls; the tumor necrosis factor-α (TNF-α) (0.76 ± 0.44 ng/mL), interleukin-6 (IL-6) (11.83 ± 3.72 pg/mL), and interleukin-8 (IL-8) (17.45 ± 6.53 pg/mL) levels were evidently lower in the experimental group than in the controls (1.48 ± 0.72 ng/mL, 14.62 ± 5.19 pg/mL, 22.86 ± 9.35 pg/mL) (P < 0.05). In summary, compared with nasal inhalation of glucocorticoids alone, combined with oral administration of Yupingfeng granules, it can more significantly improve the symptoms of nasal obstruction and rhinorrhea in AR patients, reduce nasal mucosal edema, reduce the number of EOS in nasal secretions, and reduce the level of inflammatory factors, which is of positive significance for the clinical treatment of AR patients
Agro-mophological characterization, adaptational behaviour and flower abnormalities of Lilium L. cultivars in addition to a flower with stochastic flower formula
This experiment was undertaken to determine the adaptation of Lilium L. cultivars to North-Western part of Türkiye and to define morphologic plant growth characteristics at aboveground part, including flower life and flower abnormalities. The bulbs of the three Lilium L. cultivars, ‘Profundo’, ‘Valverde’, and ‘Vonq’, were sown to perlite in pots in June, 2021, and evaluated for emergence and flowering rates, stem and plant height, stem and flower diameter, leaf and flower number, flower abnormality rates, flower fragrance degree and flower life in 2022, after one year adaptation without removing the previous sown bulbs. As the results of the cultivars differences these were determined that, at the adaptive year; the emergence rates varied between 60.42-89.58%, flowering rates changed between 43.75-87.50%, stem heights changed between 40.63-48.25 cm, plant heights changed between 52.50-59.75 cm and these data showed statistically importance. In addition to the data in the adaptive year; stem diameters, leaf numbers, flower numbers flower fragrance degree and flower longevities also showed statistical importance between cultivars. The flower abnormality rates were found between 8.3-25% in cultivars although no statistical results. Interestingly a striking, stochastic case also was observed from a flower with 8 tepals, 8 stamen and 4 lobed stigma at ‘Valverde’ contrary to well-known flower formula. It has been concluded that ‘Profundo’ has more remarkable result in terms of longevity of flower compared to the other two varieties while ‘Valverde’ and ‘Vonq’ shared the same group with some precedence in most of the agro-morphologic characteristics e.g. plant height, stem height and stem diameter. Lilium L. ‘Vonq’ showed that the highest adaptive capacity among the cultivars with the highest emergence rate (89.58) in all sowed bulbs and flowering rate (87.50) in all emerged bulbs under the study conditions in the open-air
Investigating the metagenomics of the bacterial communities in the rhizosphere of the desert plant Senna italica and their role as plant growth promoting factors
Natural microbial communities associated with desert plants are found in soils that face nutrient deficiencies and extreme environments, including salinity and drought. In this study, 16S rRNA metagenomic sequencing was used to screen and identify bacterial assemblies associated with the desert plant Senna italica, obtained from diverse soil samples located in the Asfan region, northeast of Jeddah, Saudi Arabia. Several studies found Senna italica as a valuable medicinal plant for treating different diseases; however, a few studies were done on its association with bacterial communities under drought conditions. This study aimed to identify bacterial communities present in the drought soil environment of the Senna italica plants. To approach our goals, we applied metagenomic techniques, discovering a new bacterial strain beneficial for biotechnological applications. Our results showed that the analysis of the 16S rRNA sequences at the taxonomic phylum level detected 15 phyla of bacterial populations in the soil samples. The most prevalent was kept for further research. Our findings demonstrated that rhizospheric bacteria may be used as indicators of plant growth rate and survival ability in hostile environments. Studying the soil microbiome's taxonomic, phylogenetic, and functional diversity will facilitate identifying new candidates for biological agents that can be used to improve agricultural and industrial processes
Effect of viticultural practices on yield and volatile composition of ‘Cabernet Sauvignon’ grapes
‘Cabernet Sauvignon’ is a vigorous grapevine that develops herbaceous aromas in grapes. Canopy management practices can impact grapevine physiology, and as consequence the photosynthesis, fruit yield, and grape composition, and improve the quality of the fruit. Leaf removals and apical leaf removals on grapevines have been shown to reduce herbaceous aromas. However, little is known, about the effect of the use of girdling on the concentration of volatile compounds in grapes. Thus, the objective of this study was to evaluate the effect of basal leaf removal (BLR), girdling, and girdling plus cutting the apical regions of the canopy (girdling + CAC), on the concentration of volatile compounds of ‘Cabernet Sauvignon’ grapes. Fruit yield, chemical composition, and volatile compounds were analysed in all treatments. Girdling + CAC showed a reduction in grape yield, and total acidity, however, increases in total soluble solids, and fruity aromas were observed in comparison to BLR and the control (no-canopy treatment). Controls showed the highest concentration of volatile compounds related to herbaceous and unripe fruit characteristics aromas. On the other hand, BLR showed increases in volatile compounds in the grapes related to floral character. These results suggest that grapevine canopy management modifies the chemical composition of ‘Cabernet Sauvignon’ grapes, and girdling techniques and increase the fruity aromas
Incompatible pollen tubes in the quince style and their impact on fertilization success
Self-incompatibility presents one of the challenges in modern fruit production. It can be correlated with a lower yield of self-incompatible cultivars which also demand the planting of the pollinisers. The aim of this work was to investigate the phenomenon of incompatibility and its influence on fertilization success in quince (Cydonia oblonga Mill.), using the cultivars ‘Leskovacka’, ‘Vranjska’, ‘Morava’, ‘Pazardzijska’, ‘Hemus’, ‘Asenica’, ‘Portugal’ and ‘Triumph’. Incompatible pollen tubes were determined by using fluorescence microscopy. In two types of pollination (self-pollination and open pollination) pollen tubes showed signs of incompatibility, mostly in the upper third of the style. The most common sign of incompatibility is the formation of swelling at the tip of a pollen tube. Also, sometimes twisted, bifurcated, and short and thickened pollen tubes along their entire length are formed. The incompatibility was significantly more pronounced in the self-pollination than in the open pollination variant in all tested cultivars. The highest number of incompatible pollen tubes in both pollination types was present in the cultivar ‘Pazardzijska’, while the lowest number was present in the cultivars ‘Leskovacka’ and ‘Vranjska’. The appearance of incompatibility affected the degree of fertilization in quince. The results showed that only ‘Leskovacka’ and ‘Vranjska’ are self-compatible, while other studied cultivars (‘Morava’, ‘Pazardzijska’, ‘Hemus’, ‘Asenica’, ‘Portugal’ and ‘Triumph’) are self-incompatible. The results provide a good background for the future research of reproductive biology and also for adequate management of the quince orchards
In vitro germination and regeneration of Senna artemisioides, a valuable leguminous ornamental shrub
Τhis research was carried out at the prospect of introducing new ornamental species in the floriculture industry concerning the use of valuable plants. A leguminous species native to Australia was the object of the present study. Seeds of Senna artemisioides established in a private botanical garden were chosen as the starting material for both germination and in vitro studies. Hot water treatment at 90 °C, for 10 min was applied to interrupt the seed-coat dormancy. Incubation was conducted in vitro in Petri dishes, at seven different temperatures ranging from of 5.0 to 35.0 °C, involving three separate treatments of incubation at 5.0 °C, for 0, 30 and 60 days. The seeds germinated at 25 °C (56.0%). Node explants derived from produced seedlings were cultured in vitro onto Murashige and Skoog media (MS), hormone free (Hf) or containing 6-Benzyladenine (BA). The establishment of initial cultures was successful being higher on MS supplemented with BA at the range 0.1-1.0 mg L-1 media. The multiplication phase followed on MS media containing the same range of a total of four cytokinins widely used in in vitro experimentation, i.e BA, Kinetin (KIN), N6-Isopentenyladenine (2IP) and Zeatin (ZEA). 1-Naphthaleneacetic acid (NAA) was tested in conjunction with the higher concentration of the four cytokinins. MS media supplemented with BA at 0.5 mg L-1 led to a satisfactory proliferation rate; 2.4 shoots/explant were formed. Rooting phase had moderate efficiency, being higher for media with 4.0 mg L-1 IBA. Acclimatisation was successful for all rooted plantlets, promising an effective method for exploitation of this new ornamental species of high value.
Characterization of palm date varieties (Phoenix dactylifera L.) growing in Saudi Arabia: Phenotypic diversity estimated by fruit and seed traits
In order to determine the variation and the degree of diversity among the most well-known Saudi date palm (Phoenix dactylifera L.), this study applied various widely detectible fruit and seed features. The properties of the fruit and seeds were described using ten phenotypic traits. Eighteen date palm varieties from six production sites were used in this study (Ḥaʼil, Al-Madina, Al-Hassa, Al-Qassim, Kharaj, Najran). The data was analysed by Pearson r correlation. The principal components analysis (PCA) and UPGMA clustering were used to analyse the data set. According to PCA, the results showed significant variation among the analysed varieties. Our data shows that seed ratio varies among all varieties. The mean seed weight ratio varies between 4 and 13%. Varieties ‘Raziz’, ‘Lubab’ and ‘Wasily’ demonstrate higher seed ratio (over 10%). Whereas, Fankha depicts a 5 and 4% fresh and dry seed ratio. The statistical analysis indicates that the seed ratio in all 18 varieties is comparable in fresh and dry fruits. The result suggests variation among the numerous features due to dissimilarities and heterogeneity. However, the obtained results also propose the clustering and grouping of closely related features, e.g., weights of fresh and dry fruits. Eventually, it is suggested to conduct additional research on Saudi date palms utilizing more phenotypic traits in order to have a better understanding of the pack of morphological descriptors
Meiotic behavior of two grapevine somatic mutants with ornamental potential
‘Niagara Steck’ and ‘Niagara Variegada’ are two grapevine varieties that arose spontaneously as somatic mutants of ‘Niagara Rosada’. Berries characterize ‘Niagara Steck’ at young stages exhibiting a remarkable brown reticulated structure on the epidermis which develops into a brown-bronze russet-like structure as the berries ripe. The mature berries have strongly acidic flesh. ‘Niagara Variegada’ shows notable variegation in leaves and berries. Meiotic studies were carried out to ascertain if the mutant phenotypes could be related to any change in chromosome number, structure or other irregularity. Floral buds of both varieties at different developmental stages were collected and fixed for cytological analyses. Chromosome counts at diakinesis/metaphase I showed n=19 bivalents for both varieties. Univalent, trivalent, tetravalent chromosomes or chain configuration were not recorded ruling out the possibility of any alteration in the chromosome number or structure in both mutant varieties. However, low percentages of meiotic irregularities were recorded such as chromosome stickiness, laggards, non-oriented chromosomes, precocious chromosome segregation, tetrads with one microcyte, microspore fusion, and tetrad degeneration. The total percentage of abnormalities was higher in ‘Niagara Variegada’ (19.68%) than in ‘Niagara Steck’ (14.40%) which may have contributed to a lower percentage of pollen fertility (79.80%) when compared to ‘Steck’ (90.74%). The aforementioned varieties can be propagated by hardwood cuttings and constitute an interesting option for ornamentation of home backyards, patios, and gardens with the advantage that ‘Niagara Variegada’ bearing sweet edible berries
