    Pro gradu -tutkielma käsittelee kahden valtion välisen rajan sulkeutumisen vaikutuksia raja-alueella asuvien ihmisten elämään. Rajan sulkeutumiseen liittyy tässä tapauksessa Suomen itsenäistyminen emämaa Venäjästä, näiden valtioiden välinen poliittinen jännittyneisyys ja vasta itsenäistyneen Suomen sisäpoliittinen sekasorto. Tutkimuskysymyksenä on, miten Suomen ja Venäjän välisen rajan avonaisuus ja sen sulkeutuminen vaikuttivat rajalla asuvien elämään elinkeinon sekä kulttuurisen ja sosiaalisen elämän näkökulmasta. Tarkemmin tutkielma avaa perifeerisen Kuhmoniemen alueen asukkaiden kokemia muutoksia, ja vertailee niitä raja-alueella olevien keskuksien muutoksiin. Keskuksina toimivat Karjalankannaksen isot kunnat, kuten Terijoki. Tutkielma tarkastelee laajemmin, miten pääkaupungista johdettu itsenäistyminen, irtaantuminen aiemmasta emämaasta sekä uusi sisä- ja ulkopolitiikka vaikuttavat periferiassa asuvien elämään. Tutkielman aikarajaus ylittyy 1900-luvun alkuvuosista ennen Suomen itsenäistymistä 1920-luvun puoliväliin asti. Aineistona toimii vuosien 1884–1935 välillä syntyneiden kuhmolaisten haastattelut, jotka on kerännyt Erkki Ala-Könnin tutkimusryhmä vuonna 1975. Tutkimusryhmän teettämistä 90 kuhmoniemeläisen haastattelusta tutkimuksessa käytetään noin 70:ää, jotka ovat valikoituneet haastatteluissa käsiteltyjen aihealueiden mukaan. Aineiston analyysissä käytetään muistitietotutkimuksen menetelmiä, joiden avulla tarkastellaan aikavälin sekä muistamisen politiikan vaikutusta muisteluiden sisältöihin ja luotettavuuteen. Tutkielman keskeisin tulos aineistojen perusteella on se, että rajan sulkeutuminen vaikutti etenkin negatiivisesti raja-alueen ihmisten elämään sekä elinkeinon että sosiaalisen ja kulttuurisen elämän näkökulmasta. Muistitietoaineistosta nousee kuitenkin esille, että ihmiset pystyvät ajan kuluessa sopeutumaan muutokseen omien toimiensa sekä valtion hankkeiden avuilla. Tutkimustulos tuo esille sen, että poliittisia toimia johtavan keskuksen tulisi ottaa huomioon päätöksiä tehdessä periferia-alueiden olosuhteet ja ominaisuudet, jotta muutoksien vaikutukset olisivat helpommin nähtävissä

    Rate-limiting Steps in Transcription Initiation are Key Regulatory Mechanisms of Escherichia coli Gene Expression Dynamics

    In all living organisms, the “blueprints of life” are documented in the genetic material. This material is composed of genes, which are regions of DNA coding for proteins. To produce proteins, cells read the information on the DNA with the help of molecular machines, such as RNAp holoenzymes and a factors. Proteins carry out the cellular functions required for survival and, as such, cells deal with challenging environments by adjusting their gene expression pattern. For this, cells constantly perform decision- making processes of whether or not to actively express a protein, based on intracellular and environmental cues. In Escherichia coli, gene expression is mostly regulated at the stage of transcription initiation. Although most of its regulatory molecules have been identified, the dynamics and regulation of this step remain elusive. Due to a limited number of specific regulatory molecules in the cells, the stochastic fluctuations of these molecular numbers can result in a sizeable temporal change in the numbers of transcription outputs (RNA and proteins) and have consequences on the phenotype of the cells. To understand the dynamics of this process, one should study the activity of the gene by tracking mRNA and protein production events at a detailed level. Recent advancements in single-molecule detection techniques have been used to image and track individually labeled fluorescent macromolecules of living cells. This allows investigating the intermolecular dynamics under any given condition. In this thesis, by using in vivo, single-RNA time-lapse microscopy techniques along with stochastic modelling techniques, we studied the kinetics of multi-rate limiting steps in the transcription process of multiple promoters, in various conditions. Specifically, first, we established a novel method of dissecting transcription in Escherichia coli that combines state-of-the-art microscopy measurements and model fitting techniques to construct detailed models of the rate-limiting steps governing the in vivo transcription initiation of a synthetic Lac-ara-1 promoter. After that, we estimated the duration of the closed and open complex formation, accounting for the rate of reversibility of the first step. From this, we also estimated the duration of periods of promoter inactivity, from which we were able to determine the contribution from each step to the distribution of intervals between consecutive RNA productions in individual cells. Second, using the above method, we studied the a factor selective mechanisms for indirect regulation of promoters whose transcription is primarily initiated by RNAp holoenzymes carrying a70. From the analysis, we concluded that, in E. coli, a promoter’s responsiveness to indirect regulation by a factor competition is determined by its sequence-dependent, dynamically regulated multi-step initiation kinetics. Third, we investigated the effects of extrinsic noise, arising from cell-to-cell variability in cellular components, on the single-cell distribution of RNA numbers, in the context of cell lineages. For this, first, we used stochastic models to predict the variability in the numbers of molecules involved in upstream processes. The models account for the intake of inducers from the environment, which acts as a transient source of variability in RNA production numbers, as well as for the variability in the numbers of molecular species controlling transcription of an active promoter, which acts as a constant source of variability in RNA numbers. From measurement analysis, we demonstrated the existence of lineage-to-lineage variability in gene activation times and mean transcription rates. Finally, we provided evidence that this can be explained by differences in the kinetics of the rate-limiting steps in transcription and of the induction scheme, from which it is possible to conclude that these variabilities differ between promoters and inducers used. Finally, we studied how the multi-rate limiting steps in the transcription initiation are capable of tuning the asymmetry and tailedness of the distribution of time intervals between consecutive RNA production events in individual cells. For this, first, we considered a stochastic model of transcription initiation and predicted that the asymmetry and tailedness in the distribution of intervals between consecutive RNA production events can differ by tuning the rate-limiting steps in transcription. Second, we validated the model with measurements from single-molecule RNA microscopy of transcription kinetics of multiple promoters in multiple conditions. Finally, from our results, we concluded that the skewness and kurtosis in RNA and protein production kinetics are subject to regulation by the kinetics of the steps in transcription initiation and affect the single-cell distributions of RNAs and, thus, proteins. We further showed that this regulation can significantly affect the probability of RNA and protein numbers to cross specific thresholds. Overall, the studies conducted in this thesis are expected to contribute to a better understanding of the dynamic process of bacterial gene expression. The advanced data and image analysis techniques and novel stochastic modeling approaches that we developed during the course of these studies, will allow studying in detail the in vivo regulation of multi-rate limiting steps of transcription initiation of any given promoter. In addition, by tuning the kinetics of the rate-limiting steps in the transcription initiation as executed here should allow engineering new promoters, with predefined RNA and, thus, protein production dynamics in Escherichia coli

    Implementation of a Radio Frequency Fingerprint Detector Based on GNSS Signals

    Geolocation is one of the most significant manifestations of the current development of information technologies and it is used for multiple applications, such as mobile networks, military systems, or in the stock market. For that reason, it is important to verify the source of this type of signals, as they could be susceptible to being tricked by spoofing attacks, namely fake transmitters. This thesis is based on the development of a GNSS signal type classifier based on radio frequency (RF) fingerprinting methods that will determine if a signal belongs to an authorized transmitter or if it comes from a non-authorized GNSS signal generator/repeater. First, a total of 620 signals have been recorded in lab environments, follows: 40 different scenarios of real GNSS signal (with antennas located on the roof of the university) and 580 scenarios of the generated signal (using a GNSS signal generator). Each of the scenarios contains different types of signals (different GNSS constellations and/or bands, different satellites, etc.). Then, using a MATLAB-based simulator, the recorded signal is read, a certain time-frequency transform is applied (in this case the discrete Wavelet Transform), and an image of the wavelet transform of each sample is saved. These images include the features of the signal's RF fingerprinting. Next, a machine learning algorithm called SVM, also designed in MATLAB, is used. This algorithm classifies two or more different signal classes, and finally evaluate the classification accuracy. We used 80% of the images in each category for training and the remaining 20% for testing. Finally, a confusion matrix is obtained showing the accuracy obtained by the SVM algorithm in the testing phase. The analysis of the results has shown that the SVM classification algorithm can be a very effective model for the identification of GNSS transmitters through the use of fingerprinting features. It has been observed that when the Spectracom scenario is configured with more than one satellite, accuracy is lower compared to being configured with only one. This is because the signal obtained when more than one satellite is configured is more similar to the signal obtained from the antenna in comparison to the single satellite configuration, and for that reason, SVM has more difficulty in classifying it correctly. Another observation is that accuracy is also reduced when more than two categories are classified at the same time compared to a binary classification. Despite this, the accuracy is very high in the scenarios used, with 99.47% being the lowest value obtained and 100% the highest. Therefore, this implementation of RF fingerprinting methods is very promising in the context of determining whether a signal belongs to the actual GNSS satellite constellation or to a signal generator with a high level of accuracy

    Politiikan medioitumisesta käytyyn keskusteluun nojaten ja sitä laajentaen tutkimuksessa teoretisoidaan esiin kolme journalistisen julkisuuden rakentumista ja merkitystä poliittiselle toiminnalle määrittelevää ulottuvuutta. Kulttuuriset jäsennykset määrittelevät julkisuuden asiasisältöä. Institutionaaliset käytännöt määrittelevät yhtäältä journalismin tapaa rakentaa julkisuutta politiikan toimintaympäristöksi, toisaalta poliittiseen toimintaan osallistuvien toimijoiden suhtautumista julkisuuteen. Järkeilyn tyyli määrittelee julkisen argumentaation luonnetta. Teoreettiseen jäsennykseen nojaten ympäristöpolitiikka tunnistetaan tutkimuksessa erityiseksi politiikan medioitumisen kontekstiksi, jossa median ja politiikan välisen suhteen rakentumista määrittelee omanlaisensa kulttuuristen jäsennysten, institutionaalisten käytäntöjen ja järkeilyn tyylin yhteen kietoutuminen. Tutkimuksen empiirisessä osassa tarkastellaan kolmeen suomalaiseen ympäristökiistaan liittyvää sanomalehtijulkisuutta. Tutkimuksen kohteena ovat kiistat Lievestuoreen sellutehtaan käynnistämisestä (1971), Kuusamon yhteismetsän omistamien metsien suojelusta (1994) ja Talvivaaran kaivoksen jätealtaan vuodosta (2012). Kulttuurisia jäsennyksiä tarkastellaan erittelemällä kiistoihin liittyvien sanomalehtitekstien sisällöllisiä elementtejä sekä niihin kytkeytyviä puhetapoja ja diskursseja. Institutionaalisia käytäntöjä tarkastellaan erittelemällä journalismin uutiskehyksiä, kerrontaa ja mielipiteenilmaisua sekä uutisoinnissa esiintyneiden toimijoiden julkisuuskäytäntöjä. Järkeilyn tyyliä tarkastellaan erittelemällä kiistämistä ja siihen liittyviä perusteita. Tutkimuksen perusteella median ja politiikan välisen suhteen rakentumista ympäristöpolitiikan toimintakentällä määrittelee ympäristönsuojelun kulttuurisen painoarvon merkittävä vahvistuminen 1970-luvulta nykypäivään. Tutkituissa tapauksissa journalismin kehykset, kerronta ja mielipiteenilmaisu virittyivät pitkälti tämän kulttuurisen muutoksen mukaisesti. Ympäristökiistojen tapahtumapaikkoihin kiinnittyvän paikallisjulkisuuden ja valtakunnallisen julkisuuden välillä vaikuttaisi tässä suhteessa olevan kuitenkin myös selkeitä eroja. Toimijoiden näkökulmasta ympäristön pilaamisen oikeuttamisesta journalistisessa julkisuudessa on tullut hankalampaa ja ympäristönsuojelun merkityksen suorasta kiistämisestä riskialtista. Järkeilyn tyylissä ei tutkituissa tapauksissa havaittu vastaavanlaisia eroja. Julkisuus ei muuttunut konfliktihakuisemmaksi. Ympäristöön liittyvän tiedon, ympäristönsuojelun arvon tai vilpillisyyteen liittyvien epäilysten merkitys kiistoperusteena vaihteli tapauksittain. Teos on saatavilla painettuna mm. Booky.fi ja Adlibris -verkkokirjakaupoista.Drawing on the debate on mediatization of politics this study identifies cultural meanings, institutional practices and styles of argumentation as three theoretical dimensions that illuminate both the construction of journalistic publicity and its potential uses in political action. Based on the theoretical model, environmental politics is identified as a specific context for mediatization. In environmental politics, the interplay of cultural meanings, institutional practices and styles of argumentation define the construction and use of journalistic publicity in a way that differs from other fields of politics. The empirical part of this study analyzes the newspaper coverage of three Finnish environmental conflicts from 1971, 1994 and 2012. The theoretical model is transformed into an analytical framework that focuses on discursive construction of the conflicts (cultural meanings), journalistic frames, narration and opinion-giving in the coverage of the conflicts and the publicity practices adopted by actors during the conflicts (institutional practices) and legitimation of arguments (styles of argumentation). According to this study, the relation between media and politics in the field of environmental politics is defined by the increasing cultural significance of environmental protection from the 1970s to the present. In the newspaper coverage of the three environmental conflicts studied, changes on the frames, narration and opinion-giving of journalism resonated with this cultural change. However, the significance of environmental protection in the framing, narration and opinion-giving of journalism was greater in the national newspaper studied compared to the local newspapers selected based on their proximity to the conflict sites. Styles of argumentation appeared to be more case dependent. The increasing cultural significance of environmental protection did not have a clear effect on how arguments were legitimated
