A simple and versatile degradable polymeric capsule for sustained delivery of bioactive molecules

    No full text
Traditional fabrication methods for delivery vehicles often rely on thermal or solvent processing, which can pose challenges for heat- and solvent-sensitive bioactive molecules, such as proteins and peptides. These molecules may denature or lose their biological activity, ultimately reducing their therapeutic efficacy. This study proposed a simple and versatile biodegradable perforated capsule for sustained delivery of bioactive molecules. Poly (lactic-coglycolic acid) (PLGA) capsules with side perforations were fabricated using a compressionmolding method. Teicoplanin, mixedwith either phosphate-buffered solution, Pluronic F-127 hydrogel, or hyaluronic acid, was then filled into the core of the capsules. The in vitro release rates of teicoplanin were characterized over a 30-day period using an elution method and a high performance liquid chromatography (HPLC) assay. The experimental results demonstrated that the biodegradable perforated capsules released high concentrations of teicoplanin, exceeding the minimum inhibitory concentration (MIC), in vitro for 30 days. Furthermore, the high viscosity of the Pluronic F-127 hydrogel and hyaluronic acid prolonged the antibiotic’s release period. This novel capsule design holds significant promise for delivering active biomolecules, including drugs, proteins, and peptides, for long-term therapeutic applications.補正完畢DE

High-Level Scale Bioproduction of Prodigiosin Using a Novel Designed Culture Medium Completely Based on Organic By-Products and its Potential Anti-Alzheimer Effect: An Experimental and Virtual Study

    No full text
The red microbial pigment - prodigiosin has attracted much attention in research due to its multiple bioactivities, which can be applied in various fields. Production prodigiosin with an eco-friendly strategy is currently an interesting and prioritized research topic. This study introduces an innovative production approach utilizing a novel, eco-friendly culture medium system that completely replaces expensive commercial nutrients with a combination of low-cost organic byproducts (shrimp shell powder and groundnut oil cake). This new approach yielded a remarkably high prodigiosin concentration (7520 mg/L) in an exceptionally short cultivation time (8 h) during large-scale fermentation. Beyond sustainable production, this research identified a novel therapeutic potential for prodigiosin in managing Alzheimer's disease via its ability to block cholinesterase enzymes, which are key targets in Alzheimer's therapy. Besides the experimental approach, computational simulations were used to predict the drug-likeness, pharmacokinetics, and safety profiles of prodigiosin in two forms (neutral and cationic). Notably, molecular docking and DFT analysis revealed effective interactions of prodigiosin with cholinesterase enzymes, with the cationic form showing a more promising capacity for protein target interaction. This study provided scientific insights into the eco-friendly cultural medium for prodigiosin production, its anti-cholinesterase activity, and potential properties through virtual simulations.補正完畢TW

Red phosphorus grafted high-index (116) faceted anatase TiO2 for Z-scheme photocatalytic pure water splitting

    No full text
Red phosphorus (RP) is an emerging visible-light-responsive photocatalyst, yet the rapid charge recombination has limited photocatalytic hydrogen production activity. In this work, a Z-scheme heterostructure with RP nanolayer coated on high-index (116) faceted anatase TiO2 nanoparticles (TiO2@RP) is designed and fabricated via chemical vapor deposition. Compared with pristine TiO2 and RP, the optimized TiO2@RP Z-scheme heterostructure exhibits a significantly boosted photocatalytic activity for pure water splitting, with hydrogen evolution rate reaching 12.9 µmol·h−1, under simulated solar light irradiation. The strong interfacial interaction and staggered band alignment between (116) faceted TiO2 and RP result in the formation of built-in electric field, which can drive the directional charge migration from the conduction band (CB) of TiO2 to the valance band (VB) of RP under light irradiation, with photoelectrons and holes of high redox ability maintained at the CB of RP and the VB of TiO2, respectively. This well-designed heterostructure greatly promotes photogenerated charge separation and migration via a direct Z-scheme charge transfer pathway.補正完畢DE

Elevating urban sustainability: An intelligent framework for optimizing water-energy-food nexus synergies in metabolic landscapes

    No full text
As global urbanization accelerates, harmonizing water, energy, and food (WEF) resources within urban contexts is pivotal for sustainable development. This study introduces the Intelligent Urban Metabolism Framework (IUMF) for synergizing WEF dynamics, with a focus on socio-technological linkages and environmental concerns arising from climate change. Through a pioneering fusion of system dynamics simulation, machine learning surrogate, metaheuristic optimization, and multi-criteria decision making techniques, IUMF offers a transformative approach to resource management under climate uncertainty. Leveraging comprehensive data sourced from Taipei, Taiwan, this study demonstrates noteworthy enhancements in WEF nexus synergies, including a 9% boost in water supply, an 8% rise in energy benefits, and a significant 13.8% increase in food production. The cases corresponding to the best solutions under the scenario depicting a wet year and high solar radiation intensity would attain the largest benefits: 873 million m3 of water supply (water sector), 90.3 million USD of power benefits (energy sector), and 79 million kg of food production (food sector). These advancements are achieved while reducing computational runtime from 20 h to 30 min. By fostering a user-friendly interface and embracing an intelligent framework, IUMF catalyzes urban sustainability efforts. Our study highlights the potential of intelligent frameworks in addressing complex urban challenges and guiding the evolution of resource-efficient systems and offers a blueprint for a more resilient and sustainable urban future.補正完畢GB

Hybrid Genetic Algorithm to Parameter Identification of Structural Systems with Added-Damping-and-Stiffness Device

    No full text
The structural parameters may be deviated from the design values due to the yielding or the fatigue of the material strength. In this regard, the dynamic characteristics may also be changed due to the damage of the structure. In order to realize the dynamic behavior of structural systems, we can determine the dynamic models and parameters by system identification techniques. The method proposed for the parameter identification of nonlinear systems in this paper is the hybrid genetic algorithm combining Wen’s model as the restoring force model for each story shear. The process of exploring this algorithm is demonstrated by the simulated single-degree-of-freedom system. Finally, the method was applied to the three-story structural models with added-damping-and-stiffness devices mounted on the shaking table. The ground motion records used for these models are time histories of El Centro earthquake adjusted to different intensities. The results showed that the D20H100 model of added-damping-and-stiffness devices experienced slight damage when subjected to the excitation of the El Centro earthquake with PGA values equal to 500 gal and received more damage when the excitation increased to 600 gal.補正完畢PO
