    Luteolin Enhances Endothelial Barrier Function and Attenuates Myocardial Ischemia–Reperfusion Injury via FOXP1-NLRP3 Pathway

    As a natural flavonoid, the flavonoid luteolin is characterized by its powerful antioxidant and anti-inflammatory effects. While its precise mechanisms require further elucidation, existing evidence confirms its efficacy in ameliorating myocardial ischemia–reperfusion injury (MIRI). This research was designed to investigate the mechanism through which luteolin protects against MIRI. We established MIRI rat models through the ligation of left anterior descending coronary artery (LAD). To evaluate the cardioprotective effects of luteolin, echocardiographic analysis was performed, Hematoxylin and Eosin (HE) staining, and serum cardiac injury markers creatine kinase-MB (CK-MB) and lactate dehydrogenase (LDH). Cardiac vascular permeability was determined using Evans blue staining. To mimic ischemia–reperfusion injury, endothelial cells (ECs) were subjected to oxygen-glucose deprivation/reoxygenation (OGD/R) in vitro. Endothelial cell barrier function was evaluated through F-actin phalloidin staining and FITC-Dextran fluorescence leakage experiments. To elucidate the molecular mechanism, FOXP1 small interfering RNA (siRNA) and NLRP3 inhibitor MCC950 were administered. In MIRI rats, luteolin significantly improved cardiac function and preserved endothelial barrier integrity. These effects were associated with upregulation of FOXP1 and suppression of NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome. In OGD/R-treated endothelial cells, luteolin restored barrier function and cell viability. The protective effects of luteolin were abolished after FOXP1 silencing. Pharmacological NLRP3 inhibition (MCC950) mirrored luteolin’s protection. Our study indicates that luteolin enhances endothelial barrier function and attenuates MIRI via the FOXP1-NLRP3 pathway. The current study provides a potential drug for MIRI treatment

    Avoidance Behavior in Chinhai Spiny Newt Larvae: Responses to Visual and Chemical Cues from a Novel Predator

    Effective recognition of potential threats is crucial for survival in aquatic habitats, especially for amphibian larvae. As a critically endangered species, understanding how the Chinhai spiny newt (Echinotriton chinhaiensis) larvae recognize novel predators provides key scientific support for developing targeted conservation strategies. Using the American bullfrog (Lithobates catesbeiana) as a representative predator, we examined larval responses by presenting isolated visual or chemical cues, as well as visual cues from predators of differing body sizes. We measured larval avoidance and activity. Results showed that with only visual cues, larvae quickly avoided the bullfrog and significantly reduced their activity compared to controls. With only chemical cues, activity decreased significantly, but avoidance behavior did not. When both large and small bullfrogs were present, larvae avoided the larger individual significantly more. These findings demonstrate that E. chinhaiensis larvae can use visual or chemical cues to detect novel potential predators and assess risk based on size to guide their avoidance behavior. This study provides key empirical data for understanding anti-predator responses in endangered caudate amphibians and informs conservation strategies against potential threats

    Bio-Derived Cellulose Nanofibers for the Development Under Environmentally Assessed Conditions of Cellulose/ZnO Nanohybrids with Enhanced Biocompatibility and Antimicrobial Properties

    The development of eco-friendly antimicrobial materials is essential for addressing antibiotic resistance, while reducing environmental impact. In this study, bio-derived anionic and cationic cellulose nanofibers (a-CNF and c-CNF) were employed as templating matrices for the in situ hydrothermal synthesis of cellulose/ZnO nanohybrids. Physicochemical characterization confirmed efficient cellulose functionalization and high-quality nanofibrillation, as well as the formation of uniformly dispersed ZnO nanoparticles (≈10–20 nm) strongly integrated within the cellulose network. The ZnO content was 30 and 20 wt. % for a-CNF/ZnO and c-CNF/ZnO, respectively. Antibacterial evaluation against Escherichia coli and Staphylococcus aureus revealed enhanced activity for both hybrids, with c-CNF/ZnO displaying the lowest MIC/MBC values (50/100 μg/mL). Antiviral assays revealed complete feline calicivirus inactivation at 100 μg/mL for c-CNF/ZnO, while moderate activity was observed against bovine coronavirus, highlighting the role of surface charge. Cytotoxicity assays on mammalian cells demonstrated high biocompatibility at antimicrobial concentrations. Life cycle assessment showed that c-CNF/ZnO exhibits a lower overall environmental burden than a-CNF/ZnO, with electricity demand being the main contributor, indicating clear opportunities for further reductions through process optimization and scale-up. Overall, these results demonstrate that CNF/ZnO nanohybrids effectively combine renewable biopolymers with ZnO antimicrobial functionality, offering a sustainable and safe platform for biomedical and environmental applications

    Research on Landscape Enhancement Design of Street-Facing Façades and Adjacent Public Spaces in Old Residential Areas: A Commercial Activity Optimization Approach

    With the ongoing advancement of urbanization, the renewal of old urban areas has emerged as a central front in enhancing urban quality, with street space improvement playing a pivotal role in advancing sustainable urban development. This study focuses on Chengdu, a highly urbanized megacity, employing a combination of multi-point continuous street view photography, spatial mapping, and landscape design interventions to systematically examine human activity patterns, commercial dynamics, and pathways for spatial optimization along the street-facing interfaces of old residential neighborhoods and their adjacent urban streets. The findings reveal that: (1) commercializing the street-facing façades enhances local employment opportunities; (2) window-type fences demonstrate superior adaptability by effectively balancing commercial accessibility with resident safety; and (3) a diverse mix of commercial types sustains the vitality of street-level economies in these areas. These results not only offer actionable spatial strategies for the renovation of old residential zones in Chengdu but also contribute transferable insights for urban regeneration efforts globally

    Effect of Dietary Supplementation with Rumen-Protected GABA (γ-Aminobutyric Acid) on Milk Productivity and Blood Profiles of Dairy Cattle Under Heat Stress Conditions

    This study was conducted to evaluate the effect of rumen-protected γ-aminobutyric acid (GABA) supplementation on milk productivity of lactating Holstein cows. Eighteen Holstein dairy cows (mean parity, 2.2 ± 1.0 year; mean milk yield, 34.3 ± 5.5 kg) were selected in a commercial dairy farm for the experiment. The experiment was conducted from 17 July 2024 to 11 September 2024 (56 days). Generally, THI 72 is set as a threshold since the productivity of Holstein cows starts to decrease. Animals were exposed to heat stress conditions (THI ≥ 72) during the experimental period. The basal diet was fed as a total mixed ration (TMR), and GABA was top-dressed onto the TMR. The treatments were basal diet (Control), basal diet supplemented with rumen-protected GABA 3 g/d (Treatment 1), and basal diet supplemented with rumen-protected GABA 6 g/d (Treatment 2) as a completely randomized design. Statistical significance was compared between the control and GABA treatment groups using the method of repeated measurement. Increased levels of rumen-protected GABA supplementation tended to mitigate the decline in milk yield associated with heat stress (p = 0.083). Milk fat content in the GABA supplementation groups was significantly greater than that in the control group (p = 0.036). Milk lactose content was significantly increased by GABA supplementation (p = 0.017). Blood metabolic profiles and cortisol did not differ significantly between the control and GABA supplementation groups. Activities in the GABA supplementation groups were significantly greater than those in the control group (p < 0.05). Rest and rumination times in the GABA supplementation group were significantly lower than those in the control group (p < 0.05). These results suggest that rumen-protected GABA can be a practical nutritional intervention for minimizing productivity losses in Holstein cows during periods of elevated ambient temperature

    The Effect of Apilarnil Administration on Spermiation and Semen Quality in Sterlet (Acipenser ruthenus)

    The severe decrease in natural sturgeon stocks has led to intensified efforts toward the development of sturgeon aquaculture and the application of reproductive biotechnologies to ensure sustainable production. In cultured male Acipenser ruthenus (sterlet), spermiation must be hormonally induced to obtain high-quality semen; however, reducing hormonal dosages while maintaining or improving sperm quality remains a major challenge. In this study, we investigated the effects of reduced doses of carp pituitary extract (CPE) and luteinizing hormone-releasing hormone agonist (LH-RH) combined with Apilarnil supplementation on spermiation and semen quality in A. ruthenus. Semen volume, pH, sperm concentration, total sperm output, total motility, and spermatozoa velocity were evaluated. Administering a reduced CPE dose (1.1 mg·kg−1) combined with 1 g of Apilarnil significantly increased semen volume, sperm concentration, and total sperm output, as well as improved sperm kinematic parameters (total motility and velocity) compared with the conventional hormonal protocol. Furthermore, treatments using 50% of the standard LH-RH dose supplemented with 1 g or 2 g of Apilarnil resulted in significantly higher sperm motility and velocity than observed in the control group. These results demonstrate that Apilarnil supplementation allows for a substantial reduction in exogenous hormonal doses while enhancing spermiation efficiency and semen quality in sterlet, supporting the spermiation-induction protocol for sturgeon aquaculture

    Recycled Versus Primary Aluminum in European Automotive Industry: Trends, Challenges, and Opportunities

    The automotive industry’s use of aluminum alloys continues to rise, driven by efforts to reduce vehicle weight—and thus fuel consumption—amid growing demand for larger vehicles such as SUVs, as well as the accelerating shift to electric vehicles and the expanding global vehicle fleet. These trends create major challenges for the aluminum sector. This paper provides a narrative literature review of available and published data, primarily from the period 2020–2025, examining new trends, challenges and opportunities regarding the implementation of recycled aluminum as a substitute for primary aluminum in the European automotive industry. The goal is to develop a discussion based on the answer to the following three issues: (1) What opportunities exist for increasing the production of recycled aluminum, given the imperative to conserve diminishing raw materials required for primary aluminum production? (2) What methods could enhance the obtaining of recycled aluminum over primary aluminum? (3) How might the technological barriers that hinder the wider use of recycled aluminum be overcome? This review finds that recycled aluminum availability in the EU automotive sector is improving due to rising demand for recycled material over primary aluminum—supported by a steadily growing scrap supply—alongside the development of advanced recycling strategies capable of producing high-purity recycled alloys

    Revealing Spatiotemporal Characteristics of Global Seismic Thermal Anomalies: Framework Based on Annual Energy Balance and Geospatial Constraints

    Thermal anomalies serve as potential earthquake precursors and are crucial for understanding the mechanisms underlying seismogenic mechanisms and geodynamic perturbations. To address the limited understanding of the polarity evolution of thermal anomalies, we developed a dynamic spatiotemporal adaptive framework to quantify global thermal anomaly responses. Four parameters—the coefficient of determination (R2), spatiotemporal uncertainty (SU), temporal–spatial uncertainty ratio (TSUR), and spatiotemporal correlation coefficient (SCC)—were established to characterize the spatiotemporal patterns of thermal anomaly responses. Additionally, the Anomaly Emphasis Proximity (AEP) was introduced to identify statistically significant thermal anomaly events. The results indicate that the spatiotemporal evolution of thermal anomalies exhibits a transition from pre-earthquake mixed anomalies (both positive and negative) to post-earthquake unipolar anomalies (TIB decreased from 92% to 49%), accompanied by pronounced sea–land differentiation (SST increased from 0.3% to 98.7%). The AEP reveals significant thermal anomaly clustering highly consistent with earthquake activity (e.g., the 2008 Mw 8.0 Wenchuan earthquake in the Qinghai–Tibet Plateau), showing strong correlations in structurally active regions (e.g., SCA and SWS; FDR < 18.5%, STCW > 3.7%) but weaker ones in stable regions (e.g., CNA and ECA). Overall, this framework significantly enhances the robustness and reliability of seismic thermal anomaly detection

    Cinobufagin as a Potential Intervention Against Liver Cancer—A Comprehensive Review

    Liver cancer remains a significant global health challenge, with hepatocellular carcinoma (HCC) being the most prevalent form. Despite advancements in treatment, high recurrence rates and the limited efficacy of conventional therapies highlight the need for novel interventions. Cinobufagin (CB), a bufadienolide extracted from the parotid secretion of Bufo gargarizans and B. melanostictus, has emerged as a promising compound with multiple antitumor mechanisms. This comprehensive review assesses the current evidence regarding CB and its containing medicine, cinobufacini, in liver cancer models. Cinobufacini is a traditional Chinese medicine extract, whereas CB refers specifically to one of its active components. The pharmacodynamic actions of CB include induction of apoptosis, DNA damage, inhibition of proliferation and migration, and modulation of key oncogenic pathways such as PI3K/Akt/mTOR, Akt/ERK, and AURKA-mTOR-eIF4E. Additionally, CB disrupts tumor metabolism and induces oxidative stress. Preclinical studies, both in vitro and in vivo, demonstrate significant antitumor efficacy. However, concerns remain regarding CB’s toxicity profile at high doses. This review emphasizes the therapeutic potential of CB in HCC treatment and advocates for further translational research to optimize its clinical applicability, dosage, and safety

    Non-Invasive Assessment of Water-Based Gel Cleaning on a Capogrossi Oil Painting Using NMR-MOUSE

    This study investigates water-based gel and gel-like cleaning treatments on Superficie 553, an oil painting on canvas by Giuseppe Capogrossi, using portable NMR to assess their impact. The objective was to evaluate the effects of four cleaning systems composed of a buffer solution released in free form and combined with xanthan gum, a cross-linked silicone polymer gel, and an agar gel matrix. Two distinct NMR experiments were conducted. The first involved the acquisition of 1H depth profiles to detect the distribution of the cleaning solution within the painted layer and the thickness variations resulting from cleaning procedures. The second employed the acquisition of relaxation times, facilitating the investigation of molecular mobility within the organic components of the paint layer. NMR results indicated that the agar gel system caused negligible structural changes, whereas the silicone gel induced rigidification, and the other systems permanently increased molecular mobility. These measurements provided insights into alterations in the dynamic behavior of the polymerized oil. A key strength of this investigation lies in the direct application of diagnostic methods on Superficie 553, made possible by the non-invasive nature and portability of the NMR-MOUSE system. Additionally, portable FTIR was used to detect residues and obtain chemical information, confirming that the silicone gel left detectable residues and identifying the agar gel as the most conservative cleaning method. This enabled in situ analysis of the original artwork without sampling or relocation—a crucial advantage given the difficulty of replicating the complex physicochemical conditions of historical paint surfaces under laboratory constraints. Such real-time, on-site monitoring ensured an authentic evaluation of the treatment effects, preserving the integrity of the artwork throughout the conservation process
