Further Insights into Influence of Light Intensities on the Production of Long-Chain Hydroxy Fatty Acids, Fatty Diols and Fatty Alcohols in Nannochloropsis oceanica
Microalgae can modify their metabolic pathways as a response to environmental stimuli such as light, temperature, salinity, and nutrient availability, which critically influence the synthesis of lipids and other biomolecules. While extensive studies have focused on the impact of these environmental variables on the accumulation of valuable compounds such as polyunsaturated fatty acids (PUFAs) and triacylglycerols (TAGs), information on the biosynthesis of specialized metabolites, including long-chain hydroxy fatty acids (LCHFAs), long-chain diols (LCDs), and long-chain alkenols (LCAs) is scarce. These metabolites are thought to contribute to the structural integrity of cell walls in certain microalgae, such as Nannochloropsis spp. (Eustigmatophyceae), where they make up a biopolymer known as algaenan. This study investigates how varying light intensities affect the production of LCHFAs, LCDs, and LCAs in Nannochloropsis oceanica over a 12 h light/dark cycle. Our findings provide insights into the lipid biosynthetic pathways in microalgae, revealing that light strongly drives the production of LCHFAs, whereas LCDs and LCAs are less light-dependent and show more variable responses to different light intensities
Natural Deep Eutectic Solvents for PHB Recovery: Mechanistic Insights and Implications for Sustainable Downstream Processing
The growing concern over plastic pollution and the widespread presence of micro- and nanoplastics has renewed interest in polyhydroxybutyrate (PHB) as a biodegradable alternative; however, its industrial deployment remains constrained by costly recovery operations with a high environmental burden. This study examines how PHB biosynthesis and intracellular organization, physicochemical properties, and the characteristics of the producing microorganism influence the performance of conventional recovery routes, including extraction with organic solvents, alkaline/oxidative chemical digestion, and enzymatic–physical schemes coupled with mechanical disruption. Based on this foundation, quantitative data are analyzed for PHB content in bacteria, mixed microbial cultures, cyanobacteria, and microalgae, along with extraction yields, polymer purity, and solvent recyclability in processes employing chlorine-free solvents, green solvents, and hydrophobic natural deep eutectic solvents (NaDESs) formulated with terpenes and organic acids. The analysis integrates mechanistic perspectives on NaDES–cell and NaDES–PHB interactions with solvent design criteria, biorefinery configurations, and preliminary evidence from technoeconomic and life cycle assessments. The findings identify NaDES as an up-and-coming platform capable of reconciling biopolymer quality with the principles of green chemistry while delineating critical gaps in recovery efficiency, viscosity management, solvent recycling, and pilot-scale validation
Uncertainty-Aware Machine Learning for NBA Forecasting in Digital Betting Markets
This study introduces a fully uncertainty-aware forecasting framework for NBA games that integrates team-level performance metrics, rolling-form indicators, and spatial shot-chart embeddings. The predictive backbone is a recurrent neural network equipped with Monte Carlo dropout, yielding calibrated sequential probabilities. The model is evaluated against strong baselines including logistic regression, XGBoost, convolutional models, a GRU sequence model, and both market-only and non-market-only benchmarks. All experiments rely on strict chronological partitioning (train ≤ 2022, validation 2023, test 2024), ablation tests designed to eliminate any circularity with bookmaker odds, and cross-season robustness checks spanning 2012–2024. Predictive performance is assessed through accuracy, Brier score, log-loss, AUC, and calibration metrics (ECE/MCE), complemented by SHAP-based interpretability to verify that only pre-game information influences predictions. To quantify economic value, calibrated probabilities are fed into a frictionless betting simulator using fractional-Kelly staking, an expected-value threshold, and bootstrap-based uncertainty estimation. Empirically, the uncertainty-aware model delivers systematically better calibration than non-Bayesian baselines and benefits materially from the combination of shot-chart embeddings and recent-form features. Economic value emerges primarily in less-efficient segments of the market: The fused predictor outperforms both market-only and non-market-only variants on moneylines, while spreads and totals show limited exploitable edge, consistent with higher pricing efficiency. Sensitivity studies across Kelly multipliers, EV thresholds, odds caps, and sequence lengths confirm that the findings are robust to modelling and decision-layer perturbations. The paper contributes a reproducible, decision-focused framework linking uncertainty-aware prediction to economic outcomes, clarifying when predictive lift can be monetized in NBA markets, and outlining methodological pathways for improving robustness, calibration, and execution realism in sports forecasting
Application of the Extended Theory of Planned Behavior Model to Analyze Purchase Intention Determinants of Sustainable Argan Oil Among Moroccan Consumers
The global demand for argan oil has grown considerably in recent years, creating economic opportunities while raising concerns about ecosystem degradation and the sustainability of production systems. To support long-term viability, several initiatives have promoted environmentally friendly practices and fair value-chain models. However, the effective market integration of these initiatives depends on understanding consumer behavior and preferences toward sustainable products. This study aims to identify the determinants influencing consumers’ purchase intention for sustainable argan oil using an extended framework of the Theory of Planned Behavior (TPB). A structural equation modeling approach was applied to analyze responses from adult consumers with a minimum education level of secondary education. The results show that consumer attitude, perceived behavioral control, and willingness to pay have significant positive effects on purchase intention, while ecological literacy exerts an indirect influence through attitude, social norms, perceived behavioral control, and willingness to pay. In contrast, ecological literacy has no significant direct impact. These findings improve the understanding of behavioral mechanisms underlying green product consumption and offer insights into designing marketing strategies that align with sustainability values and promote responsible consumer choices
Cereal-Based Derivatives as Functional Fortifiers of Sourdough Bread: Nutritional, Antioxidant and Quality Insights
Fortifying sourdough bread with functional ingredients is an effective strategy to enhance nutritional value, bioactive content, and sensory quality. The novel strain Lactiplantibacillus paracasei SP5 was incorporated into mother sponges together with cereal-based derivatives, such as trahanas or delignified wheat bran (2% and 5% w/w), and their effects on sourdough bread functionality were compared with a control bread without supplementation. Sourdough bread containing 5% w/w trahanas exhibited the highest lactic acid bacteria (10.4 log cfu/g), approximately 22% higher than the control, and the strongest fermentation activity, leading to acetic acid levels 27% higher than the control. This sourdough bread sample also had 73% higher total phenolic content (112.1 mg GAE/100 g) and approximately 27% higher antioxidant activity compared to control bread. In addition, phytic acid degradation exceeded 91%, representing a 16% higher reduction than the control, thereby improving mineral bioavailability. Shelf-life was also markedly extended, in terms of mould and rope spoilage compared with the control. Wheat bran-supplemented breads showed moderate improvements, with LAB counts, antioxidant activity, and phytic acid reduction about 10–25% higher than the control. Overall, these findings demonstrate that L. paracasei SP5 in combination with nutrient-rich, low-cost cereal-based derivatives can significantly enhance the technological, nutritional, and bioactive properties of sourdough breads. This approach provides a simple, industrially feasible strategy for producing functional breads with improved health-promoting and preservative attributes
Natural and Synthetic Peptides as Alternatives to Antibiotics in Intestinal Infections—A Review
Background/Objectives: Antimicrobial peptides (AMPs), evolutionarily conserved components of innate immunity characterized by their broad-spectrum efficacy and minimal resistance development, are increasingly recognized as promising therapeutic candidates. This review aims to integrate current knowledge concerning natural and synthetic antimicrobial peptides and their therapeutic effectiveness in addressing gastrointestinal infections. Methods: A literature review was performed, evaluating recent peer-reviewed studies on AMPs. The research concentrated on their molecular mechanisms of action, antimicrobial spectrum, and their interactions with standard antibiotics. More in detail, the peptide classes examined herein included defensins, cathelicidins, histatins, and various natural peptides such as lactoferricin, protamines, RegIII, and hepcidin, along with synthetic analogs like WR12, D-IK8, MSI-78, and IMX942. Results: Natural AMPs demonstrated significant antimicrobial and immunomodulatory effects against Escherichia coli, Klebsiella pneumoniae, Salmonella spp., and Shigella spp. Beyond direct antimicrobial activity, antimicrobial peptides act as integrated anti-infective agents not only by modulating host–microbiota interactions, but also preserving epithelial barrier integrity, and limiting inflammation, thereby offering a multifaceted strategy to control gastrointestinal infections. On the other hand, synthetic peptides showed improved stability, reduced cytotoxicity, and synergistic interactions with antibiotics, which suggests that they could be used either alone or in combination with other treatments. Conclusions: AMPs constitute a promising category endowed with anti-infective activity, especially for therapy of intestinal diseases, which is attributed to their distinctive anti-infective mechanisms, immune-modulating characteristics, and a relatively low propensity for resistance development compared to conventional antibiotics. However, more clinical trials and improvements to their formulation are needed to translate promising in vitro results into reliable patient outcomes
A Conjoint Analysis of Consumer Preferences on Shiitake Mushrooms: A Case Study of the Republic of Korea
Shiitake mushrooms (Lentinula edodes) are widely consumed as a key health food in the Republic of Korea. However, they face declining production value and consumption, necessitating a shift from production-focused research to an understanding of consumer demand. The aim of this study was to quantify Korean consumers’ trade-offs among key shiitake attributes and to derive actionable marketing strategies to expand domestic consumption. We conducted an online survey (n = 500) to quantify consumer utility for four key attributes: cap size (two levels), cap color (two levels), origin (two levels: domestic (Korean) and imported (Chinese)), and price (four levels per 500 g). The results identified price as the most important attribute (relative importance = 46.41%), followed by origin (19.85%), cap color (17.10%), and cap size (16.64%). Utility analysis (part-worths) revealed a distinct dual preference: consumers value both low-priced shiitake (KRW 4000 (USD 2.9)/500 g) for personal consumption and high-priced options (KRW 13,000 (USD 9.5)/500 g) for gifting. Consumers showed a clear preference for dark-colored caps, while the aggregate-level utility difference between origin levels was small. A Logit model simulation indicated the highest predicted shares for profiles priced at KRW 13,000 (15.9%) and KRW 4000 (15.7%), consistent with a polarized value–premium structure. These findings indicate that Korean producers should adopt a dual strategy: developing low-cost products to stimulate general consumption while simultaneously marketing high-quality, dark-colored, domestically produced shiitake as premium gift items, thereby establishing effective food choice strategies in a competitive market. Although the empirical setting is the Republic of Korea (with ‘Chinese’ included only as an imported-origin level representing the main foreign competitor), the findings speak to broader specialty-food contexts where import competition and dual-purpose purchasing (everyday use vs. gifting) shape attribute trade-offs
Evaluation of Veterinary Prescription of Gastroprotectants in Dogs in Spain
The overprescription of gastroprotectants, in particular acid suppressants in dogs, is of increasing concern in veterinary medicine. There have been specific guidelines published to document the appropriate use of this class of drugs; however, the injudicious use of gastroprotectants continues to be a concern and is often not evidence-based. The primary objective of the present study was to evaluate the veterinary prescription of gastroprotectants for dogs in Spain. A survey employing a snowball recruitment effect was distributed among small animal medicine veterinarians practicing in Spain. A total of 265 veterinarians participated in the survey. Proton pump inhibitors (PPIs) were found to be the most commonly prescribed gastroprotectant utilised by 50.6% of the participants. Veterinarians with fewer years of clinical experience and those focusing on the fields of internal medicine, emergency, and anaesthesia were more likely to adhere to evidence-based guidelines in their prescribing practices. Those who prescribed gastroprotectants less frequently tended to rely on PPIs and on international consensus guidelines. Although the main indications in which Spanish veterinarians used gastroprotectants was supported by scientific evidence, the injudicious administration of this class of drugs for disorders lacking robust scientific evidence or recommendations was well documented
Transcriptomic Analysis Identifies Acrolein Exposure-Related Pathways and Constructs a Prognostic Model in Oral Squamous Cell Carcinoma
Acrolein, a highly reactive environmental toxicant widely present in urban air and tobacco smoke, has been implicated in the development of multiple malignancies. In oral tissues, chronic acrolein exposure induces oxidative stress, inflammation, and genetic mutations, all of which are closely linked to the development of oral squamous cell carcinoma (OSCC). Although accumulating evidence indicates a strong association between acrolein exposure and OSCC, its prognostic significance remains poorly understood. In this study, we analyzed transcriptome data to identify differentially expressed genes (DEGs) between tumor and adjacent normal tissues, and screened acrolein-related candidates by intersecting DEGs with previously identified acrolein-associated gene sets. Functional alterations of these genes were assessed using Gene Set Variation Analysis (GSVA), and a protein–protein interaction (PPI) network was constructed to identify key regulatory genes. A prognostic model was developed using Support Vector Machine–Recursive Feature Elimination (SVM-RFE) combined with LASSO-Cox regression and validated in an independent external cohort. Among the acrolein-related DEGs, four key genes (PLK1, AURKA, CTLA4, and PPARG) were ultimately selected for model construction. Kaplan–Meier analysis showed significantly worse overall survival in the high-risk group (p < 0.0001). Receiver operating characteristic (ROC) curve analysis further confirmed the strong predictive performance of the model, with area under the curve (AUC) values of 0.72 at 1 year, 0.72 at 3 years, and 0.75 at 5 years. Furthermore, the high risk score was significantly correlated with a ‘cold’ immune microenviroment, suggesting that acrolein-related genes may modulate the tumor immune microenvironment. Collectively, these findings highlight the role of acrolein in OSCC progression, suggesting the importance of reducing acrolein exposure for cancer prevention and public health, and call for increased attention to the relationship between environmental toxicants and disease initiation, providing a scientific basis for public health interventions and cancer prevention strategies
Serum Protein Signatures for Breast Cancer Detection in Treatment-Naïve African American Women Using Integrated Proteomics and Pattern Analysis
Breast cancer is the leading cause of cancer-related mortality in African American (AA) women. In this study we evaluated the serum proteomic profile of AA women with breast cancer using an integrated proteomic framework with multivariate pattern analysis. Using 2D-DIGE, thousands of serum protein spots were detected across 33 gels; 46 spots met criteria for presence, statistical significance, and differential expression. Proteins from the spots were identified by MALDI-TOF/TOF and matched in curated databases, highlighting serum biomarkers including ceruloplasmin, alpha-2-macroglobulin, complement component C3 and C6, alpha-1-antitrypsin, alpha-1B-glycoprotein, alpha-2-HS-glycoprotein and haptoglobin-related protein. LC–MS/MS analysis revealed 163 differentiating peptides after imputing and filtering 286 peptides. These were evaluated using cumulative distribution function (CDF) analysis, a nonparametric method suited for limited sample sizes. Peptide patterns were explored with Random Forest, showing concordance with CDF. The model achieved an AUC of 0.85 at the peptide level. This workflow identified differentiating proteins (CERU, A2MG, CO3, VTDB, HEMO, APOB, APOA4, CFAH, CO4A, AACT, K1C10, ITIH2, ITIH4), highlighting CERU, A2MG, and CO3 with overexpression and reproducible identification across platforms.
