    Ameliorating Effect Of Quercetin On Ethanol-Induced Liver Injury Via Targeting RISC Machinery

    Background: Over the past few decades, increased alcohol consumption has had deleterious effects on human health. Alcoholic fatty liver disease (AFLD) is becoming a major global challenge, as the currently approved drugs for AFLDs are subject to several side effects. This has broadened the scope of the use of natural compounds as therapeutics. Recent advances in nutraceuticals as therapeutics have shed light on flavonoids such as Quercetin. It is a natural antioxidant of multiple dietary origins and has been extensively studied for its beneficial role as an anti-inflammatory and anti-cancer agent. Objective: Based on this framework, in the proposed study, we investigated the therapeutic role of Quercetin in Ethanol-induced liver damage using the Swiss Albino mice model and the hepatic cell line HepG2. Methodology: WST-1 assay was performed to access the effect of Quercetin on cell proliferation. The impact of Ethanol on the body and liver weights of mice was measured, and liver injury was determined by H&E staining and TMS. The mRNA expression levels of inflammatory genes (TNF-α, IL-6, and IL-1β) and SND1, a significant unit of the RNA-induced silencing complex (RISC), were analyzed. The liver enzyme levels were also measured. Results: Our experimental results showed that HepG2 cells treated with ethanol had a lower proliferation rate, which was later mitigated by treatment with quercetin. In the mice model, a considerable reduction in body weight was detected after ethanol treatment. Conversely, there was a significant elevation in liver weight and enzyme activity. All of these effects were ameliorated by Quercetin treatment. Immunohistochemistry data revealed an improvement in the inflammation and fibrosis characteristics in liver tissues of the Quercetin-treated group. Decreased expression of inflammatory markers and SND1 levels were also observed in the Quercetin-treated group. Conclusion: Based on our results it may be concluded that Quercetin demonstrated hepatoprotective activity in both ethanol-treated HepG2 cell line and ethanol-induced liver injury in mice model.     Cerebral venous thrombosis (CVT) is a rare complication encountered in pregnancy. Symptoms of CVT include headache, limb paresis, seizures , disturbed consciousness , visual disturbances. We present our 25 years old pregnant patient who presented with unsteadiness of gait , weakness and numbness of all four limbs, vomiting headache , giddiness due to which it was difficult for her to perform her daily activities. After the differential diagnostic examinations, we recognised cerebral venous thrombosis. After anti-coagulant treatment , the symptoms resolved. We would like to convey that such symptoms in pregnant women we should always think of CVT

    A Rare case of Combined Immunodeficiency Syndrome with Otomastoiditis, Parotitis & facial nerve palsy

    This article presents a case study of a 4-month-old female baby with combined immunodeficiency syndrome complicated by bilateral otomastoiditis, right-sided parotitis, and facial nerve palsy. The patient presented with symptoms of fever, cold, earache, and ear discharge. Clinical examination revealed redness and swelling in the post-auricular region, along with bloody discharge from the right ear. Further investigations, including blood tests, culture sensitivity, ultrasound scans, and imaging studies, confirmed the diagnosis. The patient was treated with appropriate antibiotics, antifungals, and supportive care. The article discusses the challenges in diagnosing and managing primary immunodeficiency diseases (PIDs), emphasising the importance of early recognition and appropriate treatment. It also highlights the association between recurrent parotitis and immunological factors and the need for further investigations to study this condition. Additionally, the article emphasises the significance of Ig replacement therapy in managing PID patients with significant antibody production disorders. Otolaryngologists play a crucial role in identifying and managing PID patients, contributing to improved outcomes and quality of life for these individuals. Overall, this case underscores the importance of considering combined immunodeficiency syndromes in the differential diagnosis of paediatric patients presenting with recurrent infections, chronic otitis media, and parotitis, and highlights the need for further research in this area

    Availability of Essential Medicines in Healthcare Facilities Offering Maternal and Reproductive Healthcare Services in Nigeria

    Background: The availability of essential medicines is important in preventing maternal and child deaths. This study assessed the availability of essential medicines in healthcare facilities offering maternal and reproductive healthcare services in Nigeria. Method: This study was cross-sectionally designed using structured interviewer-administered survey questions and site inspections. The study was conducted across all States in Nigeria except three states (Borno, Adamawa, and Yobe) that had security challenges. The survey was carried out between May and July 2019. The data were analysed with IBM-SPSS version 25.0. Results: The mean availability of 17 essential medicines was 58.8%. South-East has a very low availability of 49.1%, North East, South West, and South-South had availability below 60% whereas North-West (71.8%) and North-central (60.2%) had higher availability (
