Ferroptosis Inhibition Enhances Osteoblast Activity: The Role of Liproxstatin-1 and Coenzyme Q10
Ferroptosis, a form of regulated cell death triggered by lipid peroxidation, is implicated in various degenerative diseases and bone regeneration. In this study, we hypothesized that the ferroptosis inhibitors Liproxstatin-1 (Lip-1) and Coenzyme Q10 (CoQ10) play a dual role in protecting cells against ferroptotic damage and promoting osteogenic differentiation in MC3T3-E1 cells. Erastin-induced ferroptosis significantly reduced cell viability and increased lipid peroxidation, as evidenced by BODIPY™ 581/591 C11 staining. Both Lip-1 and CoQ10 decreased lipid peroxidation and restored cell viability, particularly at early treatment points. Post-treatment recovery experiments showed that both agents reversed erastin-induced damage, with Lip-1 having a stronger and more sustained effect. ALP activity assays on day 14 revealed dose-dependent increases with Lip-1 and moderate stimulation with CoQ10, indicating additional osteoinductive properties. Moreover, cell density affected sensitivity to lipid peroxidation, with higher cell densities providing protection through antioxidant pooling. These results highlight CoQ10 and Lip-1 as promising candidates for bone tissue engineering, as they offer protection against ferroptosis and promote osteoblast differentiation. This study investigates the issues related to shelter environments during the planning and actual use stages, based on shelter operation manual guidelines for each prefecture and a survey of disaster victims from the July 2020 heavy rain. It identifies problems in shelter conditions, such as limited space and insufficient environmental improvements, especially when many evacuees are staying together. During the initial and deployment phases, shelters are planned with specific details, but dissatisfaction arises due to overcrowding and poor conditions. In the stabilization and withdrawal phases, while shelter environments are somewhat improved based on the manual, there are no concrete improvement plans beyond the stabilization phase. Enantioselective Synthesis of 2‐Substituted 2‐Hydroxymethylaziridines by Copper‐Catalyzed Sulfonylative Desymmetrization of 2‐Amino‐1,3‐Diols
An enantioselective protocol for the synthesis of 2-hydroxymethylaziridines with a tetrasubstituted carbon stereocenter have been established through the copper-catalyzed asymmetric desymmetrization of 2-substituted 2-amino-1,3-diol derivatives by monosulfonylation. The present transformation consists of a reaction sequence involving the enantioselective monosulfonylation of 2-amino-1,3-diols and the intramolecular ring closure of the resultant monosulfonylated products, which provides an efficient and straightforward entry to optically active 2-substituted 2-hydroxymethylaziridines. The potential synthetic utility of the present reaction is exemplified by the derivatization of the obtained aziridines into a variety of nitrogen-containing molecules such as amino alcohols and functionalized aziridine derivatives with high enantiopurities.Asian Journal of Organic Chemistry, 14(3), art. no. e202400643; 2025journal articl
Primary ciliary dyskinesia in a Japanese woman caused by a novel RSPH4A variant
Primary ciliary dyskinesia (PCD) is a rare genetic disorder characterized by ciliary dysfunction. We report the case of a 50-year-old Japanese woman with chronic cough, sinusitis, hearing loss, and bronchiectasis. High-speed video analysis of the nasal mucosa revealed reduced ciliary beat frequency and amplitude. Electron microscopy revealed normal cilia mixed with cilia lacking central microtubules. Genetic testing identified a homozygous RSPH4A variant (NM_001010892.3: c.1484C > A). Pre-coagulation of surface water for mitigating membrane fouling during submerged nanofiltration
Membrane fouling becomes a challenge in nanofiltration (NF) when pre-filtration, such as microfiltration or ultrafiltration, is not performed. This study aimed to identify the potential of pre-coagulation for minimizing membrane fouling during submerged NF membrane treatment. Pre-coagulation (i.e., rapid and slow mixing) before submerged NF treatment was performed by adding optimized doses of ferric chloride (FeCl3) or polysilicate iron (PSI). The FeCl3-based pre-coagulation only slightly reduced membrane fouling compared to that without pre-coagulation, with a transmembrane pressure (TMP) increase of 9 kPa compared to 18 kPa over 12 days. In contrast, PSI-based pre-coagulation achieved a smaller TMP increase (7 kPa) compared to filtration without pre-coagulation (26 kPa) over 15 days. Interestingly, considerable deposition of foulants was observed on the surface of both NF membranes. Regardless of pre-coagulation, foulants on the membrane surface were removed using one-wipe sponge cleaning, fully restoring membrane permeance. Membrane characterization indicated that FeCl3 or PSI pre-coagulation resulted in a fouling layer comprising iron, which had low hydraulic resistance. On the normalised energy dissipation rate in homogeneous isotropic turbulence
The Reynolds number dependence of the normalised energy dissipation rate Cϵ=ϵL/u3 is studied, where ϵ is the energy dissipation rate, L is the integral length scale and u is the root-mean-square velocity. We present the derivation of the exact relationship between the normalised energy dissipation rate and integrated form of the Kármán–Howarth equation in homogeneous isotropic turbulence. The present mathematical formulation is valid for both forced and decaying turbulence. The discussion of Cϵ is developed under the assumption that the term resulting from the nonlinear energy transfer appearing in Cϵ is constant at sufficiently high-Reynolds-number turbulence. The fact that the integrated term originating from nonlinear energy transfer is constant plays the role of a lower bound in Cϵ, implying that the energy dissipation rate is finite in high-Reynolds-number turbulence. Furthermore, the origin of the non-equilibrium dissipation law could be the imbalance between u and dL/dt, the influence of external forces, or both. In decaying turbulence with forced turbulence as the initial condition, the imbalance between u and dL/dt causes the non-equilibrium dissipation law.
