Usefulness of Muscle Stiffness Evaluated by Ultrasonography for Detecting Muscle Change Related to Contracture in Frail Older Adults
Background: Ultrasonography can be used to evaluate the echo intensity (EI) and strain ratio (SR) to identify muscle quality and stiff-ness, respectively. EI and SR are affected by aging, frailty, and joint angle positions. We investigated the effects of aging and frailty on muscle EI and SR at different joint angle positions, and identified a useful measurement method to reflect muscle changes related to contractures in frail older adults. Methods: This study had a cross-sectional design. Twenty-two healthy young adults (young group), 22 non-frail older adults (non-frail group), and 22 frail older adults (frail group) participated in this study. The range of motion (ROM) of hip abduction, EI, and SR of the adductor longus in the zero- and full-abduction positions were measured. To investigate the effects of aging and frailty, the Kruskal–Wallis test, followed by the post-hoc Steel–Dwass test, was used. In addition, to confirm whether EI and SR in each position were useful for assessing contracture, Spearman’s correlation test was used. Results: ROM and SR in full-abduction were significantly lower in the frail group than in the other groups and lower in the non-frail group than in the young group. The SR in full- abduction (ρ = 0.73, p < .001) and in zero-abduction (ρ = 0.41, p < .001) showed strong and moderate correlation with the ROM, respectively. Conclusions: SR in full-abduction is affected by both frailty and aging and is useful for evaluating muscle stiffness related to contracture in frail older adults.Canadian Geriatrics Journal, 28(3), pp.212-220; 2025journal articl
Antitumor immune response elicited by M2 TAM-specific DDS via C-type lectin CD209b using cholesteryl pullulan nanogel as a protein drug carrier
Nagasaki University (長崎大学)博士(医学)Many cancer patients develop resistance to immunotherapy, highlighting the urgent need for novel therapeutic strategies. Various factors contribute to tumor resistance to immunotherapy, among which tumor-associated macrophages (TAMs) are critical regulators of tumor sensitivity. Therefore, combining cancer immunotherapies with drug delivery systems (DDSs) targeting TAMs has become an intriguing treatment strategy. However, the target molecules used in DDSs are limited to a few receptors expressed on TAMs. Therefore, the identification of novel target molecules for TAM-specific DDS is urgently needed. The current study evaluated the ability of a cholesteryl pullulan (CHP) nanogel to target TAMs via mDC-SIGN (CD209b). This nanogel encapsulated the cytotoxic protein drug Pseudomonas exotoxin A and was injected into a tumor-bearing mouse model. This treatment significantly reduced the abundance of CD209b-positive M2 TAMs and enhanced antitumor immune responses. Ultimately, tumor growth was suppressed, even in a low-immunogenic tumor model. Hence, CD209b is an effective target molecule for M2 TAM-specific DDSs that can be used to develop novel cancer therapies.長崎大学学位論文 学位記番号:博(医歯薬)甲第1709号 学位授与年月日:令和7年6月4日Author: Takaaki Nakatsukasa, Daisuke Muraoka, Situo Deng, Kiyoshi Yasui, Shin-ichi Sawada, Asako Shimoda, Hirokazu Matsushita, Keitaro Matsumoto, Takeshi Nagayasu, Naozumi Harada, Kazunari Akiyoshie and Hiroaki IkedaCitation: Biomaterials Science, 13, pp.2340-2350; 2025Nagasaki University (長崎大学), 博士(医学) (2025-06-04)doctoral thesi

A multicenter study of CT findings in medication-related osteonecrosis of the jaw
Medication-related osteonecrosis of the jaw is currently classified only by clinical staging, with no imaging-based qualitative classification available. To establish foundational data for a new classification method that could help guide treatment decisions and predict prognosis, this study investigated the computed tomography findings commonly observed in patients with medication-related osteonecrosis of the jaw and their frequencies. Computed tomography scans from 784 patients across 19 medical institutions were analyzed for the presence of osteolysis, sequestrum separation, periosteal reactions, mixed-type osteosclerosis, and the bone-within-bone appearance. Clinical factors associated with each finding were also examined. Osteolysis appeared as localized in 55.1% of cases, extended in 27.7%, and advanced in 13.8%. In contrast, 3.4% of patients showed no osteolysis. This non-osteolytic pattern was more common among patients treated with denosumab. Sequestrum separation was seen in 34.7% of patients, most frequently among those with osteoporosis. Periosteal reactions were observed in 5.9% of cases as the attached type, 12.4% as the gap type, and 5.6% as the irregular type. Mixed-type osteosclerosis occurred in 31.9% of cases, most often in the mandibles of patients with malignant tumors. The bone-within-bone appearance was observed in 3.1% of cases. Patients with sequestrum separation had more favorable treatment outcomes. In contrast, those without osteolysis and those with periosteal reactions, mixed-type osteosclerosis, or bone-in-bone appearance were more likely to have poorer outcomes. This multicenter study clarified the prevalence of several key computed tomography features in medication-related osteonecrosis of the jaw, highlighting their potential relevance to clinical outcomes. These findings lay the groundwork for future research into their prognostic and therapeutic implications to support the development of a new imaging-based classification system.Scientific Reports, 15(1), art. no. 40152; 2025journal articl
Stereochemical revision of brevipetins E and F: ellagitannin dimers with opposite hexahydroxydiphenoyl atropisomerism
Brevipetins E and F, ellagitannin dimers from Cleidion brevipetiolatum, were originally reported to bear both a 3,4-(Ra)-hexahydroxydiphenoyl (HHDP)-glucopyranose and a 3,4-(Ra)-HHDP-rhamnopyranose moiety. However, a detailed reevaluation of the reported electronic circular dichroism (ECD) data, corroborated by DFT-based 1H and 13C NMR chemical shift and ECD calculations, supports a stereochemical reassignment of these ellagitannins. Accordingly, the 3,4-HHDP-rhamnopyranose moiety in these compounds is reassigned to the (Sa)-atropisomeric configuration, in line with the observed reduction in Δε magnitudes relative to the constituent monomers.Organic and Biomolecular Chemistry, 23(44), pp.10214-10224; 2025journal articl
A real-time evaluation method of unstable rock risk level based on Microseismic data and CWT-CNN integrated algorithm
Geological hazards caused by unstable rocks, including rock collapse and fall, pose significant threats to both engineering productivity and the safety of residents, resulting in substantial economic losses. This study proposed a comprehensive CNN (Convolutional Neural Networks) classification recognition algorithm based on continuous wavelet analysis of microseismic monitoring data, which establishes a CNN recognition-based method for identifying risk levels of unstable rocks to predict their real-time collapse. To collect training data, fixed-base tests were conducted, and validation was performed using vibration data from freeze-thaw tests. Results of the study showed that the accuracy of the CNN classification recognition model trained with fixed-base test data reached 97.6%, and the per-second classification accuracy of vibration segments from freeze-thaw tests was above 86%. Furthermore, the study discussed the correlation between the calculated risk-level eigenvalues and safety coefficients. The CNN risk-level eigenvalues were found to be highly negatively correlated with the safety coefficients, with correlation coefficients as high as 0.8703. Finally, the study verified the superiority of the precursor identification of the risk-level evaluation method by comparing it with the safety coefficients.AIMS Environmental Science, 12(6), pp.979-998; 2025journal articl
