Copper Binding and Subsequent Aggregation of α-Synuclein Are Modulated by N-Terminal Acetylation and Ablated by the H50Q Missense Mutation
The Parkinson’s disease-associated protein α-synuclein exhibits significant conformational heterogeneity. Bacterially expressed α-synuclein is known to bind to copper, resulting in the formation of aggregation-prone compact conformations. However, in vivo, α-synuclein undergoes acetylation at its N-terminus. Here the effect of this modification and the pathological H50Q mutation on copper binding and subsequent conformational transitions were investigated by electrospray ionization–ion mobility spectrometry–mass spectrometry. We demonstrate that acetylation perturbs the ability of α-synuclein to bind copper and that the H50Q missense mutation in the presence of N-terminal acetylation prevents copper binding. These modifications and mutations prevent the formation of the most compact conformations and inhibit copper-induced aggregation

Data associated with 'Origin of terminal voltage variations due to self-mixing in terahertz frequency quantum cascade lasers'
We explain the origin of voltage variations due to self-mixing in a terahertz (THz) frequency quantum cascade laser (QCL) using an extended density matrix (DM) approach. Our DM model includes a full calculation of scattering between quantum states as well as coherent transport from miniband states into the upper lasing level and allows calculation of both the current–voltage (I-V) and optical power characteristics of the QCL under optical feedback. The model is applied to an exemplar 2.6 THz bound-to continuum QCL that is typical of those used in optical feedback interferometry. Feedback from an external cavity is included in the model through a change in cavity loss, to which the gain of the active region is clamped. The variation of intra-cavity field strength necessary to achieve gain clamping, and the subsequent change in bias required to maintain a constant current density through the heterostructure is then calculated. Strong enhancement of the self-mixing voltage signal due to non-linearity of the I–V characteristics in the analyzed THz QCL device is predicted and confirmed experimentally
Data from: Effects of an acute bout of aerobic exercise on immediate and subsequent three-day food intake and energy expenditure in active and inactive pre-menopausal women taking oral contraceptives
oai:shurda.shu.ac.uk:1This study examined the effects of an acute bout of exercise of low-intensity on food intake and energy expenditure over four days in women taking oral contraceptives. Twenty healthy, active (n = 10) and inactive (n = 10) pre-menopausal women taking oral contraceptives completed two conditions (exercise
and control), in a randomised, crossover fashion. The exercise experimental day involved cycling for one hour at an intensity equivalent to 50% of maximum oxygen uptake and two hours of rest. The control condition comprised three hours of rest. Participants arrived at the laboratory fasted overnight; breakfast was standardised and an ad libitum pasta lunch was consumed on each experimental day. Participants kept a food diary to measure food intake and wore an Actiheart to measure energy expenditure for the remainder of the experimental days and over the subsequent 3 days. There was a condition effect for absolute energy intake (exercise vs. control: 3363 ± 668 kJ vs. 3035 ± 752 kJ; p = 0.033, d = 0.49) and relative energy intake (exercise vs. control: 2019 ± 746 kJ vs. 2710 ± 712 kJ; p < 0.001, d = −1.00) at the ad libitum lunch. There were no significant differences in energy intake over the four days in active participants and there was a suppression of energy intake on the first day after the exercise experimental day
compared with the same day of the control condition in inactive participants (mean difference = −1974 kJ;
95% CI −1048 to −2900 kJ, p = 0.002, d = −0.89). There was a group effect (p = 0.001, d = 1.63) for freeliving
energy expenditure, indicating that active participants expended more energy than inactive participants during this period. However, there were no compensatory changes in daily physical activity energy expenditure. These results support the use of low-intensity aerobic exercise as a method to induce a short-term negative energy balance in inactive women taking oral contraceptives

The present study sets out to explore an apparent increase in participation in local organisations reported in the 2004 survey and to validate respondents' survey responses. The study assesses whether it is possible to design a new participation question which offers better value for money i.e. facilitates a similar level of understanding of the types of groups respondents are involved in and better captures their understanding of social participation, yet is sufficiently short to make it a viable option for local surveys
Data from: Investigation of factors influencing the hydrolytic degradation of single PLGA microparticles
Abstract Poly lactide-co-glycolide (PLGA) is an important polymer matrix used to provide sustained release across a range of active pharmaceutical ingredients (APIs) and works by hydrolytic degradation within the body, thereby releasing entrapped drug. Processing and sterilisation can impact on the morphology and chemistry of PLGA therefore influencing the hydrolysis rate and in turn the release rate of any entrapped API. This paper has looked at the effect of supercritical carbon dioxide (scCO2) processing, gamma irradiation, comonomer ratio and temperature on the hydrolysis of individual PLGA microparticles, using a combination of Attenuated Total Reflectance-Fourier Transform Infrared (ATR-FTIR) imaging, Scanning Electron Microscopy (SEM), Differential Scanning Calorimetery (DSC) and Gel Permeation chromatography (GPC) to facilitate a better understanding of the physiochemical factors affecting the hydrolysis rate. This work has shown that scCO2 processing influences hydrolysis rates by increasing the porosity of the PLGA microparticles, increasing the lactide comonomer ratio decreases hydrolysis rates by reducing the hydrophilicity of the PLGA microparticles and increasing the gamma irradiation dose systematically increases the rate of hydrolysis due to reducing the overall molecular weight of the polymer matrix via a chain scission mechanism. Moreover this work shows that ATR-FTIR imaging facilitates the determination of a range of physicochemical parameters during the hydrolysis of a single PLGA microparticle including water ingress, water/polymer interface dimensions, degradation product distribution and hydrolysis rates for both lactide and glycolide copolymer units from the same experiment
Generation of reproducible fingermarks and application to fingermark dating
Optical images of fingerprints generated by manual deposition and by the Reed-Stanton press rig are reported. In particular 3 sets of images are collected. Set 1, Set 2 and Set 3. In all cases, pre-inked strips were used to produce visible prints and plain white paper was employed as deposition surfaces. Fingerprint Images were captured as jpeg files on the Visual Spectral Comparator (VSC4CX, Foster & Freeman, Evesham, UK). Images were uploaded to this data collection as jpeg files as they were created in this format. Set 1 is a collection of images of a depletion series of 5 “manually deposited fingerprints” and another of 5 fingerprints generated by the press rig; Set 2 is a collection of 4 fingerprints generated by the press rig at a time interval of 1 week; Set 3 is a collection of 10 “manually deposited replicate fingerprints” and 10 replicate fingerprints generated by the Reed-Stanton press rig. Another set of data collected under the folder name of "Ageing data" contains MALDI MS Imaging raw data of silicon stamped fingermarks imaged after ageing for 0, 1, 4 and 8 days. Data may be opened and processed using the freeware software Biomap
