Comparative Antioxidant Study of South Asian Plants Using the DPPH Assay
This study investigates the antioxidant potential of four different Indigenous South Asian plants, which include Amla (Phyllanthus emblica), Neem Leaf (Azadirachta indica), Reetha (Sapindus mukorossi), and Shikakai (Acacia concinna), using the DPPH free radical scavenging assay. Plant extracts were prepared using 95% ethanol and tested across concentrations ranging from 10-100,000 µg/mL. Ascorbic Acid was used as a standard antioxidant. The percent inhibition, IC₅₀, and AAEAC (Ascorbic Acid Equivalent Antioxidant Capacity) values were determined using UV-Vis spectrophotometry at 517 nm. The standard antioxidant ascorbic acid showed fast acting scavenging activity with an IC₅₀ of 31.5 µg/mL, and the plant extract Amla had a similar IC₅₀ of 34.5 µg/mL with an AAEAC of 0.90. Comparatively, Neem leaf, Reetha, and Shikakai had IC₅₀ values of 400, 16,819, and 11,947 µg/mL, respectively. The kinetic data showed that Ascorbic Acid reaches equilibrium and scavenges free radicals quickly. Amla’s kinetic data revealed a slower and prolonged radical scavenging activity.
