    We report here that human B cells produce thymus and activation-regulated chemokine (TARC/CCL17) and macrophage derived chemokine (MDC/CCL22) if stimulated with anti-CD40 and IL-4. The production was determined by both protein and mRNA level using specific ELISA and semi-quantitative RT-PCR methods. Since the ligand of the TARC and MDC is CCR4, which is specifically expressed on Th2 type T cells, the production of these CC chemokines is likely to play important roles in the T cell and B cell interaction. Consistent with this, ovalbumin (OVA) specific IgE levels, which reflect the T-B cell interaction, are significantly correlated with the amounts of TARC and MDC in sera. Furthermore, we found that TARC and MDC levels are significantly increased in the sera obtained from patients with atopic dermatitis, and that the amounts are correlated with the severity of atopic dermatitis. Since CD40 ligand and IL-4 are produced by activated T helper cells, these results indicate that TARC and MDC produced by B cells play important roles in the production of antigen specific IgE by the T-B cell interaction and in the pathogenesis of allergic disease.departmental bulletin pape

    Interocclusal recording for removable partial denture in bilateral mandibular distal extension were evaluated for the differences of the interocclusal recording material utilized during different impression techniques. The purpose of this study is to develop interocclusal recording utilizing occlusion rims with high accuracy. The intraoral model with urethane rubber mucosa with 3 metallic spheres as measuring points was utilized. With the applied pressure (functional) and minimum pressure (anatomical) impression techniques, working casts were fabricated. Interocclusal recording was registered with the occlusion rim on the intraoral model unto the articulator by applying the interocclusal materials; paraffin wax (PW), ALUWAX (AW), impression paste (IP) and Exabite II (EB). Analyses were conducted with the three-dimensional analyzer and calculated for the amount of vertical and antero-posterior displacement between the intraoral model and the working cast. The results were that PW had the greatest displacement, especially with the functional impression technique, while AW had minimum displacement especially in the posterior direction. With the functional impression technique utilizing AW was revealed to minimize the amount of interocclusal displacement.departmental bulletin pape

    To reveal roles of neurotrophic factors in plastic changes of the primary afferent neurons following nerve injury, we investigated expression of glial cell line-derived neurotrophic factor (GDNF) as well as nerve growth factor (NGF) in a neuropathic pain model of the rat. The rats exhibited hyperalgesia and allodynia on the injured left side for at least 2 weeks after chronic constrictive injury to the sciatic nerve. Accompanied by the behavioral changes, expression of GDNF decreased in the dorsal root ganglia (DRGs) and the sciatic nerve on the injured side on the fourteenth day after the surgery. In contrast, the amount of NGF in DRGs was unchanged in spite of disturbance of NGF transport in the nerve. The present results suggest that decreased expression of GDNF takes some part in development and/or maintenance of neuropathic pain.departmental bulletin pape

    The pericentromeric region of chromosome 18, especially 18p11.2, is described as a schizophrenia susceptibility locus. We had previously cloned two novel brain-derived transcripts from this region: the gene for a second human myo-inositol monophosphatase (IMPA2) and a gene of unknown function, C18orf1. Recently, we reported a distortion of transmission of the tandem repeat marker D18S852, embedded in the 3’-untranslated region of C18orf1, in schizophrenia, using a familybased association test. A subsequent case-control study also revealed a significant association between the haplotype constructed from D18S852 and the 6409T>C polymorphism located in C18orf1 and schizophrenia. In the present study, we screened the C18orf1 gene for mutations and identified a novel single nucleotide polymorphism (SNP), -96T>C in exon 2. This SNP showed significant genotypic (P = 0.048) and allelic association (P = 0.005) with schizophrenia in a case-control study. The distributions of haplotypes defined by D18S852 and -96T>C were different between control and schizophrenia groups (P = 0.021). These findings suggest that C18orf1 or a gene nearby may contribute to the overall genetic risk for schizophrenia.departmental bulletin pape

    Purpose: To evaluate the dimensional accuracy of several impression methods including agar alginate combined impression in vivo; the marginal accuracy of stone dies was determined using a new electroformed master crown technique. Materials and Methods: Cast cores with knifeedge and chamfer margins and electroformed master crowns were fabricated for 3 patients. Five impressions were taken of each preparation, using agar alginate combined impression and silicone impression materials. Dies were made after impression. The marginal fit of the master crown on each die was analyzed by four-way analysis of variance (ANOVA) and Tukey HSD test (p<0.05). Results: The marginal fit of the master crown on the dies with chamfer margin was better than those with knife-edge margin for agar alginate combined impression. The shape of the margin did not affect the accuracy when silicone impression material was used. Conclusions: The results suggest that the agar alginate impression method is clinically acceptable for the chamfer margin, but shape of the margin may affect the dimensional accuracy of dies. The shape of the margin does not affect the accuracy of dies when silicone impression was used. Furthermore, the master crown made by electroforming technique could be useful for clinical evaluation of impression methods.departmental bulletin pape

    Nucleosome (NS)-reactive autoantibodies appear to be involved in the pathogenesis of systemic lupus erythematosus (SLE), but the mechanisms responsible for their production remain elusive. To study whether antibodies specific to NS are present prior to anti-double-stranded (ds) DNA antibodies in lupus mice, ELISA was carried out using the plates which had been confirmed to be coated with comparable amounts of oligo-NS and dsDNA. IgG antibodies reactive with NS or dsDNA were both detected as early as 6 weeks and 10 weeks of age in MRL/lpr and MRL/+ mice, respectively. Their titers increased with age. The specificity of dsDNA binding activity in these young mice was confirmed by the finding that their sera reacted not only with mammalian dsDNA but also with dsDNA from different sources including synthetic polynucleotides. On the other hand, NS binding activity in young mice could be ascribed to cross-reactivity of the anti-dsDNA antibodies, because it was completely inhibited by free dsDNA. A role for NS or chromatin in triggering the production of anti-dsDNA antibodies has recently been suggested, but the present results argue that, at least in these strains of mice, antibodies specifically recognizing conformational epitopes on NS are not produced before those specific to dsDNA.departmental bulletin pape

    The epithelial alteration in interstitial pneumonias is one of the repair processes at the sites of disease activity. Regenerative epithelial cells may participate in remodeling of the lung. To determine the phenotype of regenerative epithelial cells in usual interstitial pneumonia (UIP) and nonspecific interstitial pneumonia (NSIP), the expression of Clara cell 10KD protein (CC10), cytokeratin (CK) 14 and 17, surfactant apoprotein (SP)-A, KL-6/MUC1, transforming growth factor (TGF) β2 were examined in 25 patients with UIP, 9 patients with NSIP and normal lung tissues from 10 patients with lung cancer. In honeycomb lesions of UIP, non-ciliated columnar cells mainly expressed CC10, cuboidal cells expressed CC10, CK17, CK14 and SP-A in descending order. Fibroblastic foci are covered by CK17, CK14, CC10, and a few SP-A positive flattened or cuboidal cells. Regenerative epithelium in NSIP mainly comprised cuboidal cells expressing SP-A, CC10 and CK17. KL-6 was more remarkably expressed in cuboidal and non-ciliated columnar cells both in UIP and NSIP. Expression of TGFβ2 was observed in cuboidal and flattened epithelium. In severe fibrotic areas, CC10 expressing cells were more prominent, while SP-A positive cells were more prominent in less fibrotic areas. Regenerative epithelial cells in remodeling area in UIP may be derived from bronchiolar basal cells and Clara cells, while most of those in NSIP may be derived from type II pneumocytes. The different origin of regenerative epithelium may reflect the severity and extent of the injury and the degree of consequent fibrosis in UIP and NSIP.departmental bulletin pape

    Rats were subjected to unpredictable chronic stress (UCS), which was composed of 3 cycles of 7 kinds of stress for 21 days. Rats given UCS exhibited a depressive state in behavioral tests such as emergence tests and forced swim tests. Administration of cyclosporine-A (CsA), an immunosuppressive drug, gave rise to antidepressant effect in rats under the UCS, but not in stress-free rats. In other words, CsA shortened both the latency time in emergence tests and the immobility time in forced swim tests in rats given UCS. Analysis of brain tissue by HPLC revealed that CsA caused a significant increase in NE, 5-HT and 5-HIAA levels in the cortex of UCS treated rats, but treatment with either UCS or CsA alone resulted in the opposite effect. Comparing the data of monoamines and their metabolites in the brain, cascades may be different between CsA and imipramine, although both of them showed antidepressive effect in behavioral tests.departmental bulletin pape

    CD23 has roles in proliferation, antigen uptake and presentation, and the generation of IgE. Signals through IL-4R and CD40 stimulate transcription of CD23 in B cells and are necessary for immunoglobulin class switch (IgCS). The same signals induce nuclear translocation of Ku, which is also required for IgCS, in human resting B cells, suggesting that these signaling pathways are connected. We examined the regulation of CD23 gene, and located the minimal promoter at-132+80 region. A pair of 188bp inverted repeats inhibited its activity. The intronic region including EBV responsive element (EBVRE) and the surrounding sequence, required the gene specific promoter to enhance the reporter gene activity. Western blotting and FACS analysis using subclones of DND39 B cells infected with recombinant EBV, revealed that CD23 upregulation did not necessarily correlate with EBNA 2 and LMP 1 expression. Although the specific binding of Ku to EBVRE was not demonstrated, dominant negative Ku80 suppressed IL-4 + anti-CD40-driven CD23 expression. These results suggest that Ku is involved in gene regulation as a signal transducer and gene enhance. Detailed analysis of CD23 gene regulation would lead to a better understanding of disorders such as allergy and lymphoproliferation.departmental bulletin pape

    Spaceflight and bed rest induce loss of bone mass. A number of in vivo and in vitro studies have been conducted to clarify the mechanisms, however,the results have been conflicting. The purpose of this study was to investigate the effects of gravity unloading on proliferation, phenotypes, and apoptosis of normal human osteoblastic cells in the presence of 1α,25-dihydroxyvitamin D3. We used a vector-averaged gravity condition generated by clinostat rotation to simulate gravity unloading. Clinostat rotation did not affect the cell proliferation. On the first day, the mRNA levels for osteocalcin, ALP, CBFA1, VDR, RANKL, and OPG were reduced by clinostat rotation to 21%, 65%, 62%, 52%, 43%, and 54% of control, respectively. ALP activity was decreased to 75% of control. On the second day, the mRNA levels for osteocalcin and RANKL were reduced to 77% and 61% of control, respectively. The decreased VDR mRNA level might be responsible for the reduction for mRNA levels for osteocalcin, RANKL, and OPG. Clinostat rotation increased the pro-apoptotic index (Bax/Bcl-2 ratio) but did not induce apoptosis due to the simultaneous upregulation of the anti-apoptotic XIAP. Reduction of osteoblast responsiveness to 1α,25-dihydroxyvitamin D3 might be involved in osteopenia that is induced by gravity unloading.departmental bulletin pape
