    Behavior of Broiler Chickens in Four Different Substrates: a Choice Test

    The aim of this study was to determine the selection of bedding material by broiler chickens during the rearing period and whether the choice was determinant to their performing a specific behavior. To achieve this objective, a choice test was designed. A choice pen was constructed where birds could move freely around the four selected materials (straw, wood shavings, rice hulls and sand). Chickens were introduced in this pen in four groups of eight birds, three days a week for one hour per day and group, for four weeks. The location and the activity performed by each broiler were recorded every five minutes. Results showed a preference for sand compared with the other three substrates. However, differences between the behaviors performed in each bedding material were shown mainly for resting (preferably performed on wood shavings and straw), dust bathing (on sand), pecking and scratching (on rice hulls). Other factors, such as the time of day, were also found to have effects on fighting and drinking, and changes in behavioral patterns (resting, preening, eating, standing and pecking) were also detected as broilers grew older

    Erwinia piriflorinigrans is a new pathogenic species of the bacterial genus Erwinia that has been described recently in Spain. Accurate detection and identification of E. piriflorinigrans are challenging because its symptoms on pear blossoms are similar to those caused by Erwinia amylovora, the causal agent of fire blight. Moreover, these two species share phenotypic and molecular characteristics. Two specific and sensitive conventional and real-time PCR protocols were developed to identify and detect E. piriflorinigrans and to differentiate it from E. amylovora and other species of this genus. These protocols were based on sequences from plasmid pEPIR37, which is present in all strains of E. piriflorinigrans analyzed. After the stability of the plasmid was demonstrated, the specificities of the protocols were confirmed by the amplification of all E. piriflorinigrans strains tested, whereas 304 closely related pathogenic and nonpathogenic Erwinia strains and microbiota from pear trees were not amplified. In sensitivity assays, 10(3) cells/ml extract were detected in spiked plant material by conventional or real-time PCR, and 10(2) cells/ml were detected in DNA extracted from spiked plant material by real-time PCR. The protocols developed here succeeded in detecting E. piriflorinigrans in 102 out of 564 symptomatic and asymptomatic naturally infected pear samples (flowers, cortex stem tissue, leaves, shoots, and fruitlets), in necrotic Pyracantha sp. blossoms, and in necrotic pear and apple tissues infected with both E. amylovora and E. piriflorinigrans. Therefore, these new tools can be used in epidemiological studies that will enhance our understanding of the life cycle of E. piriflorinigrans in different hosts and plant tissues and its interaction with E. amylovora

    Essential oils (EO) and short-chain fatty acids have potential antimicrobial activity in broilers. This study aimed to investigate the effect of a specific blend of EO and a combination of this blend of EO with sodium-butyrate on growth performance and Salmonella colonization in broilers. A total of 480 one-day-old male broilers were distributed into 5 treatments (8 pens per treatment and 12 birds per pen) and reared during 42 d in experimental conditions. Dietary treatments consisted of the addition of different doses of EO (0 mg/kg, control; 50 mg/kg, EO50 and 100 mg/kg, EO100) or a combination of EO with 1 g/kg of sodium- butyrate (B; EO50 + B, EOB50 and EO100 + B, EOB100) to a basal diet. All birds were orally infected with 10(8) cfu of Salmonella Enteritidis on d 7 of study. Individual BW and feed intake per pen were measured at arrival and on a weekly basis. The prevalence and enumeration of Salmonella in feces was determined per treatment at 72 h postinfection and on d 23 and 37 of study. At slaughter, cecal content and liver samples from 16 birds per treatment were cultured for Salmonella and cecal pH was measured. No differences were observed on growth performance among treatments. All fecal samples analyzed were positive for Salmonella from d 10 to the end of the rearing period. At slaughter, Salmonella contamination (positive samples) in cecum was lower in birds fed EOB50 compared with the other treatments (P < 0.05), whereas birds fed the control diet showed the highest colonization rates. The pH of the cecal content was not different among treatments. Thus, EO or its combination with sodium-butyrate did not affect growth performance. However, a clear effectiveness of these products was observed in Salmonella control, especially when low doses of EO were combined with sodium-butyrate (EOB50)

    Unmixing-Based Fusion of Hyperspatial and Hyperspectral Airborne Imagery for Early Detection of Vegetation Stress

    Many applications require a timely acquisition of high spatial and spectral resolution remote sensing data. This is often not achievable since spaceborne remote sensing instruments face a tradeoff between spatial and spectral resolution, while airborne sensors mounted on a manned aircraft are too expensive to acquire a high temporal resolution. This gap between information needs and data availability inspires research on using Remotely Piloted Aircraft Systems (RPAS) to capture the desired high spectral and spatial information, furthermore providing temporal flexibility. Present hyperspectral imagers on board lightweight RPAS are still rare, due to the operational complexity, sensor weight, and instability. This paper looks into the use of a hyperspectral-hyperspatial fusion technique for an improved biophysical parameter retrieval and physiological assessment in agricultural crops. First, a biophysical parameter extraction study is performed on a simulated citrus orchard. Subsequently, the unmixing-based fusion is applied on a real test case in commercial citrus orchards with discontinuous canopies, in which a more efficient and accurate estimation of water stress is achieved by fusing thermal hyperspatial and hyperspectral (APEX) imagery. Narrowband reflectance indices that have proven their effectiveness as previsual indicators of water stress, such as the Photochemical Reflectance Index (PRI), show a significant increase in tree water-stress detection when applied on the fused dataset compared to the original hyperspectral APEX dataset (R-2 = 0.62, p 0.1). Maximal R-2 values of 0.93 and 0.86 are obtained by a linear relationship between the vegetation index and the resp., water and chlorophyll, parameter content maps

    Frequently asked questions about in vivo chlorophyll fluorescence: practical issues

    The aim of this educational review is to provide practical information on the hardware, methodology, and the hands on application of chlorophyll (Chl) a fluorescence technology. We present the paper in a question and answer format like frequently asked questions. Although nearly all information on the application of Chl a fluorescence can be found in the literature, it is not always easily accessible. This paper is primarily aimed at scientists who have some experience with the application of Chl a fluorescence but are still in the process of discovering what it all means and how it can be used. Topics discussed are (among other things) the kind of information that can be obtained using different fluorescence techniques, the interpretation of Chl a fluorescence signals, specific applications of these techniques, and practical advice on different subjects, such as on the length of dark adaptation before measurement of the Chl a fluorescence transient. The paper also provides the physiological background for some of the applied procedures. It also serves as a source of reference for experienced scientists

    Total phenolics, organic acids, sugars and antioxidant activity of mandarin (Citrus clementina Hort. ex Tan.): Variation from rootstock

    The objective of this study was to evaluate and compare 'Clemenules' mandarin on fourteen rootstocks: 'Carrizo' citrange, 'Swingle' citrumelo, 'Cleopatra' mandarin, Macrophylla, Volkameriana, C-35 and eight new hybrids rootstocks developed at the Instituto Valenciano de Investigaciones Agrarias in Valencia, Spain. Some organoleptic and nutritive parameters related to fruit quality (total soluble solids, total acidity, organic acids and sugars), the content of bioactive compounds (total phenolics) as well as the total antioxidant activity (TAA) due to hydrophilic (H-TAA) and lipophilic (L-TAA) compounds have been determined in Clementine juices. Results showed that rootstock plays an important role in determining the organoleptic and nutritive quality, as well as the concentration of bioactive compounds. Ripening index ranged from 16.9 to 24.2 and total acidity from 5.19 to 8.37 g citric acid L-1. The major organic acids detected were citric and tartaric acids and they varied among rootstocks too. Sucrose was the main sugar for all rootstocks. The antioxidant activity and total phenolic results were affected considerably by the rootstock. Given the differences on bioactive compounds concentration and antioxidant activity between rootstocks, these parameters should be included as an important factor in future breeding program to obtain new scion-stock combinations with enhanced bioactive compounds.     Comparison of statistical models in a meta-analysis of fungicide treatments for the control of citrus black spot caused by Phyllosticta citricarpa

    Meta-analysis has been recognised as a powerful method to synthetize existing published data from different studies through a formal statistical analysis. Several statistical models have been proposed to evaluate the effectiveness of treatments against plant diseases using meta-analysis, but the sensitivity of the estimated treatment effects to the model chosen has not been investigated in detail in the context of plant pathology. In this paper, four different statistical models were defined to analyse fungicide control trials with binary outcomes. These models were used to conduct a meta-analysis on the effectiveness of fungicide treatments against citrus black spot, a fungal disease caused by the quarantine pathogen Phyllosticta citricarpa. The models differed in the assumption made on the variability of the treatment effect (constant or variable between experimental plots) and in the method used for parameter estimation (classical or Bayesian). Odds ratios were estimated for two groups of fungicides, copper compounds and dithiocarbamates, widely applied for CBS control using each model in turn. Classical and Bayesian statistical models led to similar results, but the estimated treatment effectiveness and their associated levels of uncertainty were sensitive to the assumption made about the variability of the treatment effect. Estimated odds ratios were different depending on whether the treatment effect was assumed to be constant or variable between experimental plots. The size of the confidence intervals was underestimated when the treatment effect was assumed constant while it was variable in reality. Because of the strong between-plot variability, the 90 % percentiles of the odds ratios were much higher than the point estimates, and this result revealed that, in some plots, treatment effectiveness could be much lower than expected. Based on our results, we conclude that it is not sufficient to calculate point estimates of odds ratio when the between-plot variability of the treatment effect is strong and that, in such case, it is recommended to compute the predictive distributions of the odds ratio

    Antifungal activity of sodium propylparaben alone or in combination with low doses of imazalil against Penicillium decay on citrus fruit

    The performance of postharvest treatments with sodium propylparaben (SPP), alone or combined with low doses of the fungicide imazalil (IMZ), against citrus green (GM) and blue (BM) molds was evaluated on several citrus species and cultivars artificially inoculated with Penicillium digitatum and P. italicum, respectively, and incubated at 20 °C or cold-stored at 5 °C. Effectiveness of 100 mM SPP dips at 20 °C for 60 s was higher on oranges than on mandarins, with GM and BM incidence reductions of up to 60–90 % after 7 days at 20 °C. Irrespective of citrus cultivar and storage condition, SPP generally improved the curative action of 25 μl l−1 IMZ to control Penicillium molds. In additional tests, 100 mM SPP dips at 20 °C for 60 s only prevented GM on ‘Valencia’ oranges inoculated 24 h after treatment when combined with IMZ. It can be concluded that postharvest SPP treatments show promise as an effective alternative to be considered in citrus postharvest disease control programs

    Aphid management

    Belonging to the superfamily Aphidoidea, aphids are Hemiptera insects, many of the species causing economic damage to most cultivated plants. This occurs directly, when they suck the sap and weaken the plants, or indirectly through the transmission of several plant diseases, especially of a viral nature. Aphids cause this plant damage using their biter-sucker mouth, as all the Hemiptera (MIYAZAKI, 1987b). The labium has been modified, taking the shape of a wide canal mouth (rostrum or proboscis) that encloses four thin stylets, which are developed from the two maxillae and the two mandibles, forming an elongated filament, which contains two ducts. The insect, after piercing the plant with its stylets, will inject the saliva through one of the ducts (the salivary canal) in order to dissolve the sap, while the other duct (the food canal) is used for absorbing the sap once it has been dissolved. This feeding system explains the Hemiptera capacity (and particularly, the aphid capacity) to transmit viruses or other type of pathogens: when they suck the fluids of a sick plant, the insects acquire the virus and later, when they inject the sap into a healthy plant, they infect it with the virus acquired
