Relações culturais, cuja origem inicial reside no XIV Congresso Internacional da Asociación Internacional de Estudos Galegos. Horizontes dos Estudos Galegos e/na Lusofonia (Universidade do Minho, 17–20 de abril de 2024). Os contributos reunidos sob o título A Galiza e o mundo em português. Relações culturais exploram, de diferentes perspetivas e com foco em diversas temáticas e períodos, o quadro relacional galego-português(-Lusofonia). Para além de uma apresentação sintética dos contributos, a presente introdução oferece um breve enquadramento do percurso relacional deste objeto de estudo, de modo a contextualizar adequadamente os trabalhos selecionados, oriundos todos eles das Ciências Sociais e Humanas.Volume 39.2 of Diacrítica brings together the thematic dossier Galicia and the World in Portuguese.     Gender differences in protein expression after polystyrene nanoplastics exposure in mussels mytilus galloprovincialis

    Plastic pollution is a significant issue that the scientific community has been actively studying due to its harmful effects on aquatic ecosystems. Nanoplastics can pass through cellular barriers and enter the mussel’s bloodstream. More worryingly, they can also penetrate sperm cells and oocytes, potentially impacting their motility and resilience. Reproductive success drives a shift in population dynamics and plays a vital role in maintaining a healthy ecosystem. Proteomics helps detect protein changes caused by exposure to contaminants, such as nanoplastics, in marine organisms, providing deeper molecular-level insights into contamination-induced cellular pathways. Therefore, this study aimed to utilise a high-throughput proteomic approach to evaluate the impact of polystyrene nanoplastics (PS-NPs) on the gonads of male and female M. galloprovincialis, using a SWATH-MS analysis after 21 days of exposure to 10 μg/L of PS-NPs (50 nm). The accumulation of PS-NPs was also evaluated in male and female mussels. A comparison in protein expression of controls vs. those exposed in male and female mussel gonads and between males and females was evaluated. The findings indicate that PS-NP accumulation in male gonads alters protein expression more significantly than in females, interfering with protein synthesis, energy production, intracellular transport, and cellular homeostasis, and possibly impaired reproductive function. Female gonads exposed to PS-NPs revealed disruption in proteins associated with translation, RNA processing and signaling, ribosome biogenesis, cell cycle regulation, and stress response. Protein folding, lipid metabolism, and calcium signaling pathways were also affected, leading to oogenesis, meiotic progression, intracellular transport, and energy metabolism impairments.     Handling the heat: ocean acidification mitigates the effects of marine heatwaves on Posidonia oceanica seedlings

    Ocean acidification and marine heatwaves are key drivers of marine ecosystem changes that can interact with one another and influence marine organisms. Seagrasses, including the long-lived Posidonia oceanica that is endemic to the Mediterranean Sea, are widely distributed along coastal habitats, forming highly valuable underwater meadows. The germination and survival of the early life stages of P. oceanica are strongly affected by environmental changes. To assess the impact of warming and acidification on its future, we conducted a multifactorial experiment in which P. oceanica seedlings were grown under ocean acidification conditions for 6 months and then exposed to a seawater warming event. Seedling performance was investigated by analysing photo-physiology, antioxidant capacity, energetic metabolism, and transcriptomic profiles. A weighted gene correlation network analysis was used to integrate phenotypic plant traits with transcriptomic results to identify central genes involved in plant responses to ocean acidification and temperature exposure. Results demonstrated that prolonged ocean acidification exposure enhances P. oceanica seedling resilience to marine heatwaves. Specifically, seedlings regulated their antioxidant systems and transcriptomic machinery to better cope with thermal stress. Under current CO2 concentrations, elevated temperatures induced stress in P. oceanica seedlings, impacting photosynthesis and respiration. However, ocean acidification could mitigate the impact of warming in the future, enhancing the resilience to global stressors of P. oceanica

    The effect of pre-trip virtual reality and on-site smart device use on accessible tourism experiences

    This study investigates the contribution of pre-visit virtual reality (VR) experiences and the use of smart devices (SDs) in enhancing accessible tourism for visitors with hearing loss at Yim Tin Tsai Island, a rural destination in Hong Kong. A total of 163 participants, including 78 individuals with varying degrees of hearing impairment and 85 persons without reported disabilities, participated in a self-guided visit. They were randomly assigned to four groups based on whether a VR experience was provided before the visit and whether SDs were used during the visit. Post-visit focus group discussions explored challenges faced, solutions proposed, and perceptions of VR and SD applications. The findings suggest that while VR and SDs offer practical and emotional benefits, visitors with hearing loss do not perceive them as particularly effective in overcoming barriers or significantly enhancing their experiences on the island. Recommendations proposed to improve tourism experiences of visitors with hearing loss include the provision of a multi-sensory environment and information supplementing the visit in VR experiences, as well as adopting a non-intrusive and intuitive approach in presenting information on smart devices

    Identification of regulatory polymorphisms associated with breast cancer risk

    Dissertação de Mestrado, Ciências Biomédicas, Departamento de Ciências Biomédicas e Medicina, Universidade do Algarve, 2014O cancro da mama é uma das patologias oncológicas mais comuns, e causa de morte mais frequente entre as mulheres. Trata-se de uma doença complexa com fatores genéticos e não genéticos envolvidos na sua etiologia. Até ao momento, o conhecimento adquirido acerca do risco genético não explica mais do que dois terços dos casos de cancro familiares permanecendo ainda por explicar cerca de 65% a 70%. Com os recentes avanços na tecnologia de microarrays e nos estudos de associação realizados a nível de genoma inteiro (genome-wide association studies, GWAS), têm surgido evidências que sugerem que as variantes cis-reguladoras podem ser importantes para o risco de cancro da mama. Variantes cis-reguladoras são polimorfismos que regulam a expressão de genes. Podem-no fazer através da modificação de elementos reguladores como, por exemplo: afetando a ligação de fatores de transcrição em promotores e elementos intensificadores Essas variantes são frequentes na população e contribuem para a variabilidade intra e inter -espécies. Com base nestas evidências, a nossa hipótese consiste em que a maioria dos polimorfismos comuns e associados a risco ainda por identificar podem ser também cis-reguladores. Deste modo, a procura para a restante suscetibilidade genética do cancro da mama deve centrar-se nas variantes com maior potencial cis-regulador. As variantes cis-reguladoras podem ser identificadas de diferentes formas, como por exemplo através de loci de características quantitativas de expressão (expression quantitative trait loci, eQTL) e análises de expressão diferencial alélica (differential allelic expression, DAE) em indivíduos heterozigóticos. DAE é uma abordagem que compara os níveis relativos de expressão dos dois alelos do mesmo gene em indivíduos heterozigóticos, utilizando um SNP transcrito (tSNP). Esta técnica tem revelado ser bastante eficaz, uma vez que compara os níveis de transcritos dos alelos dentro do mesmo contexto celular e de haplótipos, pelo que a influência de efeitos trans é eliminada (por exemplo concentração de fatores de transcrição no núcleo). Este trabalho teve dois grandes objetivos: o primeiro, a validação da dos resultados de DAE nos polimorfismos cis-reguladores identificados num estudo anteriormente realizado pela Prof. Ana Teresa Maia; o segundo, a identificação de novos loci envolvidos no risco para o cancro da mama. No estudo para validação da DAE, foi medida a quantidade relativa da expressão dos dois alelos de 10 SNPs, previamente identificados como apresentando DAE (rs2526935; rs10503416; rs10513376; rs7600326; rs6494466; rs10016; rs13265801; rs9250; rs8097892; rs1384) e de 4 SNPs sem DAE (rs2834653; rs710945; rs1477017 e rs10521). Foram utilizadas 18 amostras de tecido mamário de indivíduos saudáveis e os níveis da DAE foram determinados utilizando a técnica de PCR em tempo real específica alélica. As distribuições de DAE foram comparadas aplicando um Teste t de Student (t-test), o qual compara as médias da distribuição de rácios de DAE. No total dos 10 SNPs com DAE validámos 6 SNPs que eram consistentes com os resultados observados anteriormente. Apenas um de quatro SNPs sem evidências da DAE mostrou consistência com os resultados anteriores. Para o estudo de identificação de novos loci foi aplicada uma abordagem inovadora, a qual consiste no cruzamento de dados para cancro da mama publicados nos GWAS com os nossos dados de DAE. Esse cruzamento de dados é feito de acordo com a localização cromossómica, distância física (janelas de ±250kb a partir da variante com DAE) e padrões de desequilíbrio de ligação (linkage disequilibrium, LD). Com este exercício pretendemos testar se esta abordagem pode ajuda a priorizar os loci candidatos de GWAS para validação e posteriores análises funcionais. Este cruzamento de dados permitiu identificar vários loci que contêm SNPs associados com risco para cancro da mama e SNPs com DAE. Para uma análise inicial selecionaram-se dois loci: 17q22 (TOM1L1/COX11/STXBP4) e 12q24 (AACS), com base nos cenários de DAE, LD entre o tSNP de DAE e o SNP do GWAS. Esses dois loci foram inicialmente analisados para potenciais elementos reguladores e evidência funcional (promotores, intensificadores e ligação de fatores de transcrição) nos locais onde se encontram os candidatos a variantes cis-reguladoras. Para essa análise foram utilizadas uma base de dados, RegulomeDB, e um navegador de informação genómica, Genome Browser, que contêm informações sobre hipersensibilidade à desoxirribonuclease (DNAse), TFBS, e regiões promotoras, evidências obtidas em estudos in silico e/ou in vitro para variantes codificadoras e não codificadoras. No locus 17q22, foram encontrados 12 SNPs em sobreposição com regiões que contêm marcadores para elementos funcionais, tais como promotores e intensificadores. Sugerindo que esses podem possivelmente ter um efeito funcional através da regulação da expressão do gene. Posteriormente, analisaram-se os SNPs para a estrutura de LD nessa região e para identificação dos haplótipos na população os quais podem ser responsáveis pelo aumento ou diminuição na expressão dos genes. Foram identificados quatro haplótipos comuns associados com diferenças nos níveis de expressão. No locus 12q24, o tipo de análises realizadas foram as mesmas descritas para o outro locus. Para o locus 12q24 foram encontrados 13 SNPs em sobreposição com regiões contendo elementos reguladores o que sugere um possível efeito funcional na regulação da expressão dos genes. Todos SNPs foram também analisados para a estrutura de LD e identificação de haplótipos. Com base nessa análise, foram identificados quatro haplótipos comuns associados com as diferenças nos níveis de expressão. Essas variantes candidatas a serem cis-reguladoras foram selecionadas com base em análises in silico, utilizando ferramentas para a previsão de potenciais locais de ligação de fatores de transcrição. Posteriormente, 3 SNPs foram analisadas funcionalmente in vitro, numa linha celular de cancro da mama (HCC1954). Como os genes presentes em cada um dos loci mostraram evidências para a presença de variantes cis-reguladoras, decidiu-se validar os níveis de DAE em tecido de mama normal e compará-las com os níveis de DAE no sangue. Para essa análise usou-se um tSNP para cada gene (rs17817901 no COX11 e rs7138557 no AACS). Validaram-se os resultados da DAE, no tecido da mama, e para o AACS e observou-se que esses não são comparáveis com os níveis da DAE no sangue. Os nossos resultados não suportam, portanto, o uso de sangue em estudos de DAE em substituição para o tecido da mama, na aplicação futura em estudos de predisposição ao cancro da mama. Para averiguar se a DAE em ambos os loci está realmente associada com o risco de cancro de mama, foi realizado um estudo de caso-controlo. Foi encontrada uma associação significativa neste estudo, o que sugere que DAE em ambos os loci deve ser futuramente explorado como instrumento de previsão de risco para cancro da mama. Futuramente, uma análise mais aprofundada da regulação destes genes poderá levar também à compreensão da biologia de predisposição ao tumor e contribuir para o desenvolvimento de terapias futuras, especialmente na área da medicina personalizada

    Effect of fruit thinning on fruit quality and alternate bearing of ‘Setubalense’ mandarin (Citrus deliciosa)

    Citriculture plays a major role in agriculture worldwide, especially in the Mediterranean region. However, there are limitations in citrus production. Alternate bearing can be a big problem when it is very intense. Trees produce a heavy yield one year (“on” year) and light ones the next (“off” year). During the “on” year, trees produce many small-sized fruits, while in the “off” year, they produce very few, larger fruits. The small size of the fruits is a limitation, as they are less well accepted in the market. The ‘Setubalense’ cultivar, a traditional Portuguese Mediterranean mandarin (Citrus deliciosa), is recognized for its excellent organoleptic characteristics. Despite this, the issues of alternate bearing and small fruit size are leading to the loss of this cultivar. Fruit thinning is a cultural practice involving the removal of some fruits, while leaving others. This practice can be used to manage alternate bearing and to improve fruit size. However, its impact on the plant is not well understood, whether it affects the plant as a whole or at a more localized level, such as within individual branches. To address this, two experiments were installed in August 2019. In the first experiment thinning involved removing 50% of the fruits from the entire tree. In this experiment, the fruit size and fruit quality were higher in the trees where fruit thinning was performed. In the second experiment, thinning was applied to selected branches, also removing 50% of the fruits. In this case, no significant differences were observed, neither in fruit size nor in fruit quality.     Role of ancillary techniques in the differential diagnosis of salivary gland carcinomas

The diagnosis of salivary gland carcinomas (SGC) rests mainly on histology, but immunohistochemical and molecular investigations are often necessary for differential diagnosis. This review is primarily aimed as a tool for pathologists in non-specialised head and neck hospitals who encounter a limited number of SGC annually. The use of testing an initial antibody panel, which may comprise both positive and negative expression for a suspected entity, and examples of different panels are outlined. We also focused on acinic cell carcinoma (AcCC), which is positive for DOG1 and negative for mammaglobin, whilst secretory carcinoma (SC) is positive for mammaglobin and negative for DOG1. In addition, the exclusive expression of androgen and HER2 in salivary duct carcinoma (SDC) and its use for differential diagnosis are also addressed. This review also highlights the particularities of mucoepidermoid carcinoma (MEC) and its negativity for S100 and SOX10, which distinguishes it from some of its mimics. In laboratories with limited access to antibodies for SGC, we recommend inclusion of mammaglobin. The use of molecular techniques for the diagnosis of MEC (MAML2), SC (ETV6), adenoid cystic carcinoma (MYB), and AcCC (NR4A3) is discussed.     Medication reconciliation in primary care: Practices, knowledge and attitudes in the Lisbon and Tagus Valley Health Region

    Introdução: Apesar da relevância da reconciliação da medicação para a continuidade dos cuidados, atualmente, não existe informação relativa às práticas, conhecimentos e atitudes dos médicos de família nacionais sobre este assunto. Este estudo teve como objetivos caracterizar os procedimentos formais de reconciliação da medicação em vigor na Região de Saúde de Lisboa e Vale do Tejo, bem como a perceção dos médicos de família desta região sobre o que sabem, como pensam e como praticam a reconciliação da medicação.Métodos: Foi conduzido um estudo observacional, transversal e descritivo, utilizando duas unidades de observação: unidades de cuidados de saúde primários (estudo 1) e médicos de família (estudo 2) da Região de Saúde de Lisboa e Vale do Tejo. A recolha de dados realizou-se através de dois questionários de autopreenchimento, disponibilizados online.Resultados: Participaram 89 unidades de cuidados de saúde primários e 208 médicos de família (taxas de resposta de 31% e 12%, respetivamente). Verificou-se que apenas uma em cada quatro unidades (n = 22/89) tinha um procedimento formal de reconciliação da medicação em vigor. Entre as unidades com procedimento formal, existe variabilidade em alguns parâmetros, ainda que todos incluam os médicos. Mais de 70% (n = 150; 72,1%) dos médicos referiram ter contacto prévio com o termo ‘reconciliação da medicação’, e metade (n = 104; 50,0%) referiu realizar reconciliação da medicação em mais de 75% das consultas após alta hospitalar. Não se identificaram diferenças relativamente à frequência com que os médicos praticam reconciliação da medicação após alta hospitalar em função da sua idade, sexo, tipo de unidade onde trabalham e volume de consultas. A maioria dos médicos (n = 155; 74,5%) inclui no processo os três passos preconizados pela Direção-Geral da Saúde à data, e resolve as discrepâncias sem contactar o médico hospitalar (n = 168; 88,8%). Os médicos reconhecem a importância da reconciliação da medicação (mais de 95% estão de acordo/totalmente de acordo), ainda que o nível de concordância quanto à sua responsabilização sobre esta prática seja inferior. Conclusão: A proporção de unidades de cuidados de saúde primários com procedimento formal de reconciliação da medicação é baixa. Apesar dos médicos de família da Região de Saúde de Lisboa e Vale do Tejo valorizarem a reconciliação da medicação, não a incluem em todas as consultas após internamento hospitalar. A comunicação entre níveis de cuidados e a padronização dos processos são áreas com potencial de melhoria, com vista ao objetivo de promover a segurança da utilização dos medicamentos, centrada no doente

    Impact and size selectivity of fishing gears used in estuarine crab fisheries

    Crustacean fisheries have expanded in the last decade due to the high economic value of the species. However, estuarine crustacean fisheries remain poorly studied compared with marine crustacean fisheries. In Portugal, the European green crab ( Carcinus maenas) may become increasingly important for the fishing industry and seafood production in general, especially in the current context of overexploitation of estuarine traditional fisheries resources. The unknown ecological impact of crab fishing gear and its size-selectivity characteristics constrain gear regulations by fisheries authorities. We developed an integrated study over three years to analyse a C. maenas fishery in three Portuguese estuarine systems aiming to: 1) describe the fishing characteristics and modus operandi of the fishing gears; 2) analyse gear selectivity and catch rates; 3) analyse environmental impact of fishing gears; 4) Contribute to supporting better regulations for the fishery. Two types of crab fishing gear are used, box traps and drop nets. Daily catches of green crabs varied by gear, system, and month, with the highest catches observed in box traps during the warmest months. Selectivity studies indicate that a mesh size of 18 mm in box traps and 30 mm in drop nets would allow crabs larger than the minimum landing size (40 mm carapace width) to be targeted while maximising catch rates. However, regardless of the mesh size, the catch from both gears will need to be sorted to exclude juveniles from the catch. The bar spacing, which allows crabs to be sorted according to the minimum landing size, is 17 mm. Gears present none (drop nets) or low by-catch (220 g or 23 individuals/40 box traps), which was promptly discarded, indicating a low impact on estuarine communities.     Reasons for living and depressive symptomatology in young adults with and without suicide attempts: a moderated mediation approach

    Introduction Suicide is a public health problem worldwide, being the fourth leading cause of death in young adult population (15-29 years). Therefore, it is important to identify its risk and protective factors, and how they interact to develop more effective interventions. The present study aims to analyze the relation between depressive symptoms, hopelessness (risk factors) with reasons for living (protective factors) and suicidal ideation in young adults with and without previous suicide attempts.Methods A sample of 845 Portuguese young adults answered an online form which assessed suicidal ideation, hopelessness, depressive symptoms, and reasons for living. Mean comparisons, correlations, and moderated mediation analysis were computed.Results Individuals with a history of suicide attempt showed higher mean levels in risk factors, compared to individuals without suicide attempt. When no history of suicide attempt was present, ideation was positively and significantly correlated to depression and hopelessness, and negatively correlated to the reasons for living. Higher levels of depressive symptomatology predict lower levels of reasons for living. Mediation and moderation relations were assessed in a model and the history of suicide attempts showed a relevant role.Conclusions The evidence found in this study reinforce that reasons for living may play a preponderant role in suicidal ideation, that is as a negative mediator and therefore it can act as a protective factor.UIDB/PSI/04345/202
