Evaluation of the antioxidant and anti-inflammatory activity in macroalgae asparagopsis armata and asparagopsis taxiformis
Master’s in Quality for Analytical Laboratories (EMQAL)In recent times, natural compounds have become widely employed in the treatment of human illnesses and health conditions such as cancer, neurological disorders, multiple sclerosis, diabetes, atherosclerosis, arthritis, and cardiovascular diseases all around the world. Plants have been the principal source of numerous pharmaceutical agents throughout history. However, in this modern era, much emphasis has been placed on the amazing biodiversity of life in the marine environment, which has proven to be an extraordinary repository of novel bioactive compounds with diverse structural and chemical properties, as well as a source of inspiration for new drug discovery. In the never-ending quest for novel safe and effective anti-inflammatory and antioxidant compounds beneficial to human health, this research focused on evaluating the ability of two distinct species of the macroalgae Asparagopsis spp (Asparagopsis taxiformis, as a gametophyte and as a sporophyte, and Asparagopsis armata as a gametophyte) to contribute for the treatment of anti-inflammatory diseases by assessing their antioxidant and anti-inflammatory activities.
Ethanol, ethyl acetate and hexane were employed as solvents for the extraction of bioactive compounds from Asparagopsis spp. These solvents cover a wide range of polarity indices with polarity ranging from 0.1 for hexane, 4.4 for ethyl acetate, and 5.2 for ethanol to maximize the spectrum of extracted compounds. In this study, the best extraction yields were obtained with ethanol across all species of Asparagopsis with yields of 12.4% in A. armata, 9.0% in A. taxiformis as a gametophyte and 6.8% in the same as a sporophyte. Hexane yielded the least amount of extracts across all the species with yields of 0.4% in A. armata, 1.1% and 1.0% in A. taxiformis as a gametophyte and sporophyte respectively. Ethyl acetate showed yields of 2.4% in A. armata and 2.5% and 1.8% in A. taxiformis gametophyte and sporophyte respectively.
Antioxidant properties were evaluated using tests such as ABTS, DPPH, iron & copper chelating activity, total phenolic content, and iron reducing activity while anti-inflammatory analysis was conducted specifically analysing for COX-2 inhibition. Results obtained suggests that Asparagopsis spp contain several bioactive compounds that exhibit good antioxidant abilities especially with metal chelating activities. Activity results in the DPPH assays were rather low (below 50% activity) across all species. For the ABTS assay, only the ethanolic extract of A. armata showed activity above 50% with an IC50 value of 0.24 mg/mL. The best results for copper chelating activity were observed in the hexane extracts of A. taxiformis sporophyte with an IC50 of 0.12 mg/mL. This was followed by the hexane extract of A. armata with an IC50 of 0.28 mg/mL. The best iron chelating activity was seen in the hexane extract of A. taxiformis sporophyte once again with an IC50 of 0.09 mg/mL. The ethyl acetate extracts of A. armata and A. taxiformis sporophyte also showed IC50 values of 0.25 and 0.24 mg/mL respectively. The iron reducing antioxidant assay showed good results for ethyl acetate extracts across all species with IC50 values of 0.07 mg/mL recorded of A. taxiformis sporophyte, 0.16 mg/mL for A. armata and 0.34 mg/mL for the gametophyte life stage of A. taxiformis. These extracts were observed to show activities higher than the gallic acid positive control.
The macroalgae also demonstrated tremendous potential for COX-2 inhibition activity at an extract concentration of 1 mg/mL for all species (especially the hexane extracts showing over 50% activity across species) with the highest activity of 98.9 ± 0.4% observed in A. taxiformis gametophyte hexane extracts. Ethanol extract from A. taxiformis gametophyte also showed an activity of 79.1 ± 1.8% to take the second place whereas the hexane extract from A. taxiformis as a sporophyte showed inhibitory activity of 72.3 ± 5.1% taking the third spot. These activities compared to the ibuprofen positive control that was used at a concentration of 5 μg/mL and showed an activity of 55.0 ± 13.2% demonstrates that even as crude extracts Asparagopsis spp have great anti-inflammatory potentials and therefore in their highly refined states, extracts from Asparagopsis spp may be very competitive against other commercially known anti-inflammatory compounds on the market.
Total phenolic content analysis (TPC) conducted on the extracts confirmed the presence of phenolic compounds in all the species of Asparagopsis evaluated. The highest levels of TPC were observed in the ethyl acetate extracts of all the species. Levels in the ethyl acetate extracts ranged from 1.50 ± 0.12 mg-GAE/g for A. armata, 1.19 ± 0.03 mg-GAE/g for A. taxiformis gametophyte to 0.61 ± 0.07 mg-GAE/g for A. taxiformis sporophyte. The relatively higher phenolic content observed in the ethyl acetate extracts suggests that most of the phenolic compounds present in Asparagopsis spp may be phenolic compounds of lower polarity such as tocopherols and tocotrienol which have been demonstrated to be well extracted by ethyl acetate. In general, the levels of total phenolic compounds present in Asparagopsis spp were determined to be relatively low compared to other macroalgae in literature. Despite the relatively low phenolic contents demonstrated in Asparagopsis spp it still showed good antioxidant and anti-inflammatory activities thus the antioxidant activity observed in the crude extracts were not solely dependent on the phenolic compounds but rather to a greater extent other bioactive compound(s) that may be present in the crude extracts of Asparagopsis spp.
Statistical analysis in the form of ANOVA at a 95% Confidence Interval (alpha = 0.05) and Tukey multiple comparison test (P<0.05) were performed on the results obtained for all the antioxidant and anti-inflammatory tests conducted. The analysis revealed significant differences between the Asparagopsis species (A. taxiformis and A. armata) considered in this study and most importantly between the life stages of A. taxiformis as a gametophyte and a sporophyte.
Multivariate data analyses in the form of principal component analysis (PCA) and cluster analysis (CA) were carried out between the antioxidant assay, metal chelating assays, COX-2 anti-inflammatory assay, and total phenolic content assay. Results of the analysis revealed that the solvent type used for extraction had a great influence on the antioxidant performance of the extracts. The analysis also explored the similarities between the tests and identified a good positive correlation between copper chelating activity test and COX-2 anti-inflammatory test. This observation is of great importance because COX-2 inhibition test is noted to be time consuming, complex, and very expensive to carry out. Hence in the event of a study where a large number of samples are to be tested it becomes challenging and puts a great strain on not only human resources, but financial resources and time as well. However, copper chelating activity (CCA) is comparatively simple and economical with time and funds hence could be a great way of pre-screening samples to determine which samples are likely to show good results with COX-2 analysis and hence reduce sample size which will eventually save time and other resources.
Overall, findings presented from this research work suggests that the macroalgae Asparagopsis spp demonstrates high anti-inflammatory and antioxidant potentials for use in the pharmaceutical and nutraceutical industries.Nos últimos tempos, os compostos naturais tornaram-se amplamente utilizados no tratamento de doenças humanas e condições de saúde como o cancro, perturbações neurológicas, esclerose múltipla, diabetes, aterosclerose, artrite, e doenças cardiovasculares em todo o mundo. As plantas têm sido a principal fonte de numerosos agentes farmacêuticos ao longo da história. Contudo, nesta era moderna, tem sido dada muita ênfase à espantosa biodiversidade da vida no ambiente marinho, que provou ser um extraordinário repositório de novos compostos bioativos com diversas propriedades estruturais e químicas, bem como uma fonte de inspiração para a descoberta de novos medicamentos. Na busca incessante de novos compostos anti-inflamatórios e antioxidantes seguros e eficazes e benéficos para a saúde humana, esta investigação centrou-se na avaliação da presença de compostos bioativos em duas espécies distintas da macroalga Asparagopsis (Asparagopsis taxiformis, como gametófito e esporófito, e Asparagopsis armata, como gametófito), avaliando as suas capacidades antioxidantes e anti-inflamatórias.
Etanol, acetato de etilo e hexano foram utilizados como solventes para extração de compostos bioativos de Asparagopsis spp. Estes solventes cobrem uma vasta gama de índices de polaridade com polaridade variando entre 0,1 para o hexano, 4,4 para o acetato de etilo, e 5,2 para o etanol para maximizar o espectro de compostos extraídos. Neste estudo, os melhores rendimentos de extração foram realizados com etanol em todas as espécies de Asparagopsis com rendimentos de 12,4% em A. armata, 9,0% em A. taxiformis como gametófito e 6,8% esporófito. O hexano originou a menor quantidade de extratos em todas as espécies com rendimentos de 0,4% em A. armata, 1,1% e 1,0% em A. taxiformis como gametófito e esporófito, respetivamente. O acetato de etilo apresentou rendimentos de 2,4% em A. armata e 2,5% e 1,8% em A. taxiformis como gametófito e esporófito respetivamente.
As propriedades antioxidantes foram avaliadas usando testes tais como ABTS, DPPH, atividade quelante do ferro e cobre, conteúdo fenólico total, e atividade redutora do ferro, enquanto que a análise anti-inflamatória foi conduzida especificamente para a inibição da enzima Ciclooxigenase-2 (COX-2). Os resultados obtidos sugerem que as espécies de Asparagopsis testadas contêm vários compostos bioativos que exibem boas capacidades antioxidantes, especialmente com atividades quelantes de metais. Os resultados da atividade nos ensaios DPPH foram bastante baixos (abaixo de 50%) em todas as espécies. Para o ensaio ABTS, apenas o extrato etílico de acetato de etilo de A. armata mostrou atividade acima de 50% com um valor de IC50 de 0,24 mg/mL. Os melhores resultados para a atividade quelante do cobre foram observados nos extratos hexanos de A. taxiformis (esporófito) com um valor IC50 de 0,12 mg/mL. Seguiu-se o extrato de hexano de A. armata com um IC50 de 0,28 mg/mL. A melhor atividade quelante do ferro foi observada no extrato hexano de A. taxiformis (esporófito) mais uma vez com um IC50 de 0,09 mg/mL. Os extratos de acetato de etilo de A. armata e A. taxiformis (esporófito) também mostraram valores de IC50 de 0,25 e 0,24 mg/mL, respetivamente, no teste de quelação do ferro. O ensaio antioxidante redutor de ferro mostrou bons resultados para extratos de acetato de etilo em todas as espécies com valores de IC50 de 0,07 mg/mL registados de A. taxiformis (esporófito), 0,16 mg/mL para A. armata e 0,34 mg/mL para a fase de vida do gametófito de A. taxiformis. Estes resultados mostram que os extratos brutos demonstram um potencial bastante bom para a redução do ferro.
As macroalgas também demonstraram um enorme potencial de inibição da atividade da COX-2 com uma concentração de extrato de 1 mg/mL para todas as espécies (especialmente os extratos de hexano que mostram mais de 50% de atividade). O valor mais alto (98,9 ± 0,4% de inibição) foi observada para os extratos de A. taxiformis (esporófito) hexano. O extrato de etanol de A. taxiformis gametófito mostrou também uma atividade de 79,1 ± 1,8%, enquanto o extrato de hexano de A. taxiformis como esporófito mostrou uma atividade inibitória de 72,3 ± 5,1%. Estas atividades em comparação com o controlo positivo do ibuprofeno que foi utilizado numa concentração de 5 μg/mL e mostrou uma atividade de 55,0 ± 13,2% demonstra que mesmo como extratos brutos, Asparagopsis spp, têm grandes potenciais anti-inflamatórios e, portanto, nos seus estados altamente refinados, os extratos de Asparagopsis spp. podem ser muito competitivos contra outros compostos anti-inflamatórios comercialmente conhecidos no mercado.
A análise do conteúdo fenólico total (TPC) realizada nos extratos confirmou a presença de compostos fenólicos em todas as espécies de Asparagopsis avaliadas. Os níveis mais elevados de TPC foram observados nos extratos de acetato de etilo de todas as espécies. Os níveis de TPC nos extratos de acetato de etilo variaram de 1,50 ± 0,12 mg-GAE/g para A. armata, 1,19 ± 0,03 mg-GAE/g para A. taxiformis (gametófito) a 0,61 ± 0,07 mg-GAE/g para A. taxiformis (esporófito). O conteúdo fenólico relativamente mais elevado observado nos extratos de acetato de etilo sugere que a maioria dos compostos fenólicos presentes na Asparagopsis spp. podem ser compostos fenólicos não polares, tais como tocoferóis e tocotrienol, que se demonstrou serem bem extraídos pelo acetato de etilo. Em geral, os níveis de compostos fenólicos totais presentes nas espécies de Asparagopsis foram determinados como sendo relativamente baixos em comparação com outras macroalgas da literatura. Apesar dos teores fenólicos relativamente baixos demonstrados em Asparagopsis spp, ainda apresentava boas atividades antioxidantes e anti-inflamatórias que poderão não depender apenas dos compostos fenólicos mas sim, em maior medida, de outros compostos bioativos que possam estar presentes nos extratos brutos de Asparagopsis spp.
A análise estatística sob a forma de ANOVA com um Intervalo de Confiança de 95% (alfa = 0,05) e o teste de comparação múltipla Tukey (P<0,05) foram realizados com base nos resultados obtidos para todos os testes antioxidantes e anti-inflamatórios realizados. A análise revelou diferenças significativas entre as espécies de Asparagopsis (A. taxiformis e A. armata) consideradas neste estudo e, mais importante, entre as fases de vida de A. taxiformis como um gametófito e um esporófito.
Foram realizadas análises de dados multivariados na forma de análise de componentes principais (PCA) e análise de agregados (CA) entre o ensaio de antioxidantes, ensaios de quelantes metálicos, ensaio de anti-inflamatórios COX-2, e ensaio de teor fenólico total. Os resultados da análise revelaram que o tipo de solvente utilizado para extração teve uma grande influência no desempenho antioxidante dos extratos. A análise também explorou as semelhanças entre os testes e identificou uma boa correlação positiva entre o teste de atividade quelante do cobre e o teste anti-inflamatório COX-2. Esta observação é de grande importância porque o teste de inibição do COX-2 é demorado, complexo e muito caro. No entanto, a atividade quelante de cobre (CCA) é comparativamente simples e económica, pelo que poderia ser uma ótima forma de pré-seleção de amostras para determinar quais as amostras suscetíveis de mostrar bons resultados na inibição da COX-2 e, consequentemente, reduzir o tamanho da amostra, o que acabará por poupar tempo a outros recursos.
Globalmente, os resultados apresentados neste trabalho de investigação sugerem que as macroalgas Asparagopsis spp demonstram elevados potenciais anti-inflamatórios e antioxidantes para utilização nas indústrias farmacêutica e nutracêutica
Impact of elexacaftor/tezacaftor/ivacaftor on the small airways in cystic fibrosis
Objective: To evaluate the impact of the elexacaftor/tezacaftor/ivacaftor (ELX/TEZ/IVA) combination on the small airways in adults with cystic fibrosis (CF), a genetic disorder that primarily affects the respiratory system, leading to progressive lung disease. In CF, the small airways play a critical role, contributing to chronic symptoms such as cough, sputum production, and dyspnea. Methods: This was a single-center, retrospective observational study of adults with CF treated with ELX/TEZ/IVA for 12 months. We compared the patients who were homozygous for the F508del mutation of the CF transmembrane conductance regulator (CFTR) gene with those who were heterozygous for that mutation, in terms of lung function outcomes (FEV1, FEF25-75%, and the RV/TLC ratio) and the extent of non-homogeneous ground-glass opacity. Among the patients within the cohort, the same parameters were evaluated separately in those who had advanced lung disease and in those who had previously undergone CFTR modulator therapy. Results: There was a significant post-treatment improvement in lung function, with a median increase of 0.42 L/s in the FEF25-75% (p < 0.001) and a 5% reduction in the mean RV/TLC ratio (p < 0.001). There was a trend toward a higher improvement the F508del homozygous patients. A significant reduction in non-homogeneous ground-glass opacity was observed in 79.5% of the patients. Among the patients with advanced lung disease, there were notable post-treatment improvements in all of the parameters assessed. Conclusions: Our results highlight the positive impact that ELX/TEZ/IVA treatment can have on small airway function in patients with CF, with potential benefits even for those with advanced lung disease. Further research is needed in order to evaluate the long-term effects of this treatment and its relationship with patient-reported outcomes
The genomics of t’ef and finger millet domestication and spread
The Northern Highlands of Ethiopia and Eritrea (NHE) were a centre for food production in Africa, hosting one of the earliest agriculture-based complex societies on the continent. The NHE's geographical connections with the Arabian Peninsula, and Nilotic cultures led to the cultivation of southwest Asian crops and African native domesticates in its territory. Additionally, the NHE were also the domestication centre for crops like t'ef (Eragrostis tef (Zucc.) Trotter) and finger millet (Eleusine coracana L. Gaertn L.), after well-adapted local wild plants. Considering the paucity of the archaeobotanical record in the region and food remains' preservation issues, in this study, we aim to investigate the domestication and spread of t'ef and finger millet using genomics and interpreting the results in the light of archaeological proxies. Our data confirmed Eragrostis pilosa and Eleusine coracana subsp. africana as the sole wild progenitors of t'ef and finger millet, respectively. T'ef was initially domesticated in the NHE before spreading into southern Ethiopia and eastwards into southern Arabia. First catalogs of humpback and fin whales in the Algarve region
Baleen whales such as humpback (Megaptera novaeangliae) and fin whales (Balaenoptera physalus) are among the most highly migratory marine species. After experiencing severe population declines during the whaling era, both species are now showing signs of recovery. However, their complex movement patterns, wide-ranging distribution, and the absence of clearly defined population boundaries—particularly in the North Atlantic—pose challenges for monitoring and effective conservation efforts. While certain areas are recognized as key migratory corridors, the role of the southern Iberian coast, including the Algarve region in south of Portugal, remains largely unexplored. This thesis presents the first two photo-identification (photo-ID) catalogs of humpback and fin whales in the Algarve, establishing baseline data for this understudied region. The humpback whale chapter documents occurrences collected from 2009 to 2024 in collaboration with whale-watching companies and the Algarve Stranded Network (RAAlg). Fluke photo-ID allowed for the identification of 13 unique individuals. Comparison with the North Atlantic Humpback Whale Catalog identified matches with individuals previously sighted in distant locations, including Cabo Verde, the Ligurian Sea, and Scotland. Entanglement, particularly among juveniles, emerged as a recurrent anthropogenic threat. The fin whale chapter analyzes opportunistic data collected from 2020 to 2024, namely photo-ID and GPS boat tracks, resulting in a catalog of 86 unique individuals. Spatial capture-recapture modeling estimated a local population of 1,476 individuals suggesting that a large part of the population hasn’t been identified yet. Sightings peaked in spring and were often associated with feeding behavior, suggesting that the Algarve may serve as a seasonal foraging ground. However, low re-sighting rates across years suggest a transient rather than a resident use of the area. These findings highlight the ecological relevance of the Algarve for migratory baleen whales and underline the value of photo-ID and collaborative data collection with whale-watching companies. The two catalogs provide essential references for future research and inform conservation strategies in Portuguese waters.A conservação de espécies marinhas migratórias, como as baleias de barbas, exige um conhecimento aprofundado sobre a sua distribuição, ecologia e ameaças. Estas espécies percorrem vastas áreas oceânicas, cruzando fronteiras internacionais e utilizando múltiplos habitats ao longo do ano. Apesar da sua importância ecológica e do interesse global pela sua conservação, muitas regiões do Atlântico Norte continuam sub-amostradas, o que dificulta a implementação de medidas eficazes de gestão. A costa sul de Portugal, particularmente o Algarve, é uma dessas regiões. Apesar da crescente atividade de observação de cetáceos na região, onde poucos estudos científicos se dedicaram a documentar a ocorrência regular de baleias de barbas, a sua ocorrência, os seus padrões de migração e as potenciais ameaças antropogénicas que enfrentam. Esta tese pretende colmatar essa lacuna, estabelecendo os primeiros catálogos de foto-identificação regionais de duas espécies emblemáticas de baleias de barbas: a baleia-de-bossa (Megaptera novaeangliae) e a baleia-comum (Balaenoptera physalus). O primeiro manuscrito desta dissertação é dedicado à baleia-de-bossa e documenta dados recolhidos entre 2009 e 2024. Durante este período, foram registadas 17 ocorrências ao longo da costa do Algarve, dos quais 14 envolveram avistamentos no mar e 3 corresponderam a indivíduos encontrados mortos na costa. Os dados foram recolhidos em colaboração com empresas locais de observação de cetáceos e a Rede de Arrojamentos do Algarve (RAAlg). A identificação individual baseou-se na análise das barbatanas caudais utilizando padrões de pigmentação e características morfológicas únicas. Treze indivíduos únicos foram identificados com sucesso, e as suas imagens foram processadas e comparadas internamente e com o catálogo do Atlântico Norte (North Atlantic Humpback Whale Catalog — NAHWC). Três destes indivíduos foram avistados anteriormente em regiões distintas do Atlântico Norte: nas proximidades das Ilhas de Cabo Verde, no mar de Ligúria (Itália) e nas águas costeiras de Gairloch (Escócia). Estes resultados confirmam a conectividade migratória entre o Algarve e importantes áreas de reprodução e alimentação da população do Atlântico Norte. A distribuição temporal dos avistamentos revelou um padrão sazonal bem definido, com maior frequência na primavera (maio-junho) e no início do outono (setembro-outubro). Estes períodos coincidem com os picos de produtividade primária, levando ao incremento de toda a cadeia trófica, e em particular as baleias de barbas. Alguns indivíduos foram reavistados em locais distintos da costa algarvia com poucos dias de intervalo, indicando movimentações locais e possível uso transitório da área para alimentação. Um dos indivíduos (Mn110) foi registado três vezes em seis dias consecutivos na zona de Sagres, enquanto outro (Mn106) foi observado em Faro, Lagos e Sagres no espaço de 12 dias. Estes comportamentos reforçam a hipótese de que o Algarve poderá funcionar como uma “stopover” migratória estratégica. Os três registos de indivíduos arrojados apresentavam interações com atividades pesqueiras. Mn101, encontrado em Tavira, foi identificado como um jovem macho que anteriormente havia sido avistado nas águas da Ilha da Boavista (Cabo Verde) dois meses antes, confirmando não só a ligação entre as duas regiões como também o elevado risco que crias e juvenis enfrentam ao longo das suas rotas migratórias. Estes episódios de interação com artes de pesca sugerem que a mortalidade não natural, causada por impactos antropogénicos, pode estar sub-estimada nesta região, onde a pesca artesanal com redes de emalhar e armadilhas é amplamente praticada. O segundo manuscrito foca-se na baleia-comum e constitui o primeiro estudo sistemático sobre esta espécie na região de Faro. Os dados foram recolhidos entre 2020 e 2024 através de excursões de observação de cetáceos, com registo de imagens fotográficas e trajetos GPS das embarcações. Foi criado um catálogo com 86 indivíduos únicos, com base nas características morfológicas da barbatana dorsal, padrões de coloração e marcas específicas. Estes dados foram posteriormente analisados através de um modelo de captura-recaptura (spatial explicit IV capture-recapture — SECR), o que permitiu estimar uma população local de 1.476 indivíduos. A distribuição sazonal dos avistamentos mostrou um pico claro na primavera, particularmente entre março e maio, coincidindo com períodos de elevada produtividade primária. Durante este período, foram frequentemente observados comportamentos de alimentação e agregações mistas com outras espécies, como o golfinho-comum (Delphinus delphis), o roaz-corvineiro (Tursiops truncatus) e a baleia-anã (Balaenoptera acutorostrata). Este tipo de interação sugere que a área de Faro pode servir como um local de alimentação temporária, utilizado durante a sua migração. Apesar da abundância estimada ser relativamente elevada, a taxa de recaptura inter-anual foi baixa, o que indica que a maioria dos indivíduos observados provavelmente utiliza a região de forma transitória, e que a maioria da população ainda está por ser identificada. No entanto, alguns indivíduos foram reavistados dentro do mesmo ano, reforçando a possibilidade de fidelidade sazonal ao local. Do ponto de vista metodológico, a dissertação demonstra a eficácia da utilização de plataformas de oportunidade, como as embarcações de observação de cetáceos, para recolha de dados científicos em regiões com escassez de dados científicos. Em síntese, esta dissertação fornece os primeiros dados estruturados sobre a presença, os movimentos e as ameaças que afetam baleia-de-bossa e baleias-comuns na região do Algarve. Descrevem-se pela primeira vez a ligação do Algarve com outros locais no Atlântico Norte, os padrões sazonais de uso do habitat, e as interações com artes de pesca. Estas conclusões destacam a relevância ecológica da região do Algarve para as baleias de barbas e sublinham o valor da foto-identificação e da recolha colaborativa de dados com empresas de observação de cetáceos. Os dois catálogos fornecem referências essenciais para futuras investigações e contribuem para o desenvolvimento de estratégias de conservação nas águas portuguesas
“Roads that walk”: the Tagus River, the scene of encounters in the year 1000 BC
From the border, when it enters Portuguese territory, to the estuary area, the Tagus is approximately 230 km long. Upstream, in its international stretch, it is a river that runs narrowly and embedded in the plateaus of the Hesperic Massif, while downstream, after the mouth of the Zêzere, it becomes a plain river, entering the Cenozoic Basin and ending in a wide inland estuary near Lisbon.
For the period in question, between the 13th and 9th centuries BC, the characteristics of the river were different: the ancient Tagus estuary was wider and deeper, as a result of less silting, which began in Mesolithic times and has not stopped until today.
The communities, fully hierarchical and through their elites, established contacts, traveled, produced and exchanged goods of various natures, which circulated between sometimes very distant regions. It is in this particular case that it makes perfect sense to look at the river as a true “road that walks”

The post-glacial communities of the Tagus Valley: an example of successful adaptation to climate change
The Muge shell middens, located approximately 60 kilometers upstream from the current Tagus estuary, are an important testament to the adaptive capacity of human communities to the climate changes that occurred at the beginning of the Holocene, about 11,700 years ago, a period that marked the end of the last great glaciation.
Recent studies indicate that, about 8,000 years ago, these estuarine areas were already partially flooded by the sea, like the Tagus estuary, creating humid environments rich in natural resources, including fish, molluscs and a great diversity of terrestrial fauna. These conditions attracted groups of Mesolithic hunter-gatherers, who began to intensively exploit these resources

A dissertação consiste no acompanhamento do processo de criação do ColecTA – Colectivo Teatral do Algarve, enquanto investigadora e enquanto observadora participante insider no processo de criação da peça teatral Quando a sul o azul luar. Com base no dossier genético constituído pelos diferentes agentes durante o processo de criação (registo fotográfico, registo vídeo, anotações, figurinos, escritos dramatúrgicos, etc) e no fabrico do meu material de observação e de acompanhamento do processo (anotações, entrevistas, registos fotográficos e de vídeo dos ensaios, etc), procura-se fazer uma descrição densa do processo de criação em questão, de forma a estudar, graças à identificação dos traços saturados ou lacunares e à operacionalização das escolhas efetuadas, as funções do jogo do coro na criação dramatúrgica e cénica. Espera-se que este estudo contribua não só para um conhecimento mais aprofundado da prática artística teatral em torno de projetos coletivos como o projeto ColecTA – Colectivo Teatral do Algarve, recentemente lançado na região algarvia, mas também para a compreensão de novas práticas colaborativas e de novas formas de criação artística no campo teatral, ancoradas na herança cultural regional.The dissertation consists of monitoring the creation process of ColecTA – Colectivo Teatral do Algarve, as a researcher and as an insider participant observer in the
process of creating the theatrical play Quando a sul o azul luar. Based on the genetic dossier created by the different agents during the creation process (photographic
records, video records, notes, costumes, dramaturgical writings, etc.) and on the production of my observation and process monitoring material (notes, interviews, photographic and video records of rehearsals, etc.), the aim is to provide a dense description of the creative process in question, in order to study, thanks to the
identification of saturated or gapped features and the operationalization of the choices made, the functions of the game of choir in dramaturgical and scenic creation.
It is hoped that this study will contribute not only to a deeper knowledge of theatrical artistic practice around collective projects such as the ColecTA – Colectivo Teatral do
Algarve project, recently launched in the Algarve region, but also to the understanding of new collaborative practices and new forms of artistic creation in the theatrical field, anchored in regional cultural heritage
