    Climate change represents one of the most pressing challenges in the modern era, requiring a unified, equitable global response. This dissertation offers a comparative analysis of the climate change approaches employed by four key countries—India, South Africa, Germany, and the United States—each embodying a unique economic, geographical, and political context. This dissertation investigates the similarities and differences in climate finance, technology transfer, and capacity building, and Nationally Determined Contributions (NDCs) by reviewing these countries' UNFCCC submissions. The dissertation also investigates the climate-related issues that these nations brought up during UNFCCC negotiations, which took place from COP 1 to COP 28, providing insights into their negotiation positions and strategies for resolving conflicts between national interests and international climate obligations. The findings demonstrate the intricate dynamics of international climate governance, where substantial differences in national priorities, historical responsibilities, and economic capacities influence each country's contributions to global climate action. The thesis highlights that effective climate governance necessitates not only formal approaches of collaboration but also a commitment to resolving power inequalities and underlying systemic challenges that define parties' participation in global climate action. Hence, it is crucial to focus on institutional processes to address both new and existing inequities and maintain the mutually beneficial nature of cooperative frameworks. Consequently, this dissertation compares these four countries comprehensively, enhancing the understanding of obstacles and opportunities in global climate negotiations and underscoring the necessity of customized, adaptive, and collaborative solutions to tackle the climate crisis.İklim değişikliği, modern çağın en acil sorunlarından birini temsil etmekte ve eşitlikçi bir küresel müdahale gerektirmektedir. Bu tez, her biri kendine özgü ekonomik, coğrafi ve siyasi bağlamlara sahip dört önemli ülkenin (Hindistan, Güney Afrika, Almanya ve Amerika Birleşik Devletleri) iklim değişikliğine yaklaşımlarının karşılaştırmalı bir analizini sunmaktadır. Bu tez, söz konusu ülkelerin BMİDÇS başvurularını inceleyerek iklim finansmanı, teknoloji transferi ve kapasite geliştirme ile Ulusal Olarak Belirlenmiş Katkılar (NDC'ler) arasındaki benzerlik ve farklılıkları araştırmaktadır. Tez aynı zamanda bu ülkelerin COP 1'den COP 28'e kadar gerçekleşen BMİDÇS müzakereleri sırasında gündeme getirdikleri iklimle ilgili konuları araştırarak müzakere pozisyonları ve ulusal çıkarlar ile uluslararası iklim yükümlülükleri arasındaki çatışmaları çözme stratejileri hakkında içgörü sağlamaktadır. Bulgular, ulusal öncelikler, tarihsel sorumluluklar ve ekonomik kapasitelerdeki önemli farklılıkların her ülkenin küresel iklim eylemine katkılarını etkilediği uluslararası iklim yönetişiminin karmaşık dinamiklerini ortaya koymaktadır. Tez, etkili iklim yönetişiminin sadece resmi iş birliği yaklaşımlarını değil, aynı zamanda tarafların küresel iklim eylemine katılımını tanımlayan güç eşitsizliklerini ve altta yatan sistemik zorlukları çözme kararlılığını da gerektirdiğini vurgulamaktadır. Bu nedenle hem yeni hem de mevcut eşitsizlikleri ele almak ve iş birliği çerçevelerinin karşılıklı fayda sağlayan doğasını korumak için kurumsal süreçlere odaklanmak çok önemlidir. Sonuç olarak bu tez, bu dört ülkeyi kapsamlı bir şekilde karşılaştırarak küresel iklim müzakerelerindeki engellerin ve fırsatların anlaşılmasını sağlamakta ve iklim kriziyle mücadele etmek için özelleştirilmiş, uyarlanabilir ve iş birliğine dayalı çözümlerin gerekliliğinin altını çizmektedir.Ph.D. - Doctoral Progra

    Effective heritage conservation can only done by a proper understanding of construction techniques, which offers key insights into historical methods and materials. Standard restoration methods often ignore the diverse techniques originally used, which can lead to failures in preservation. In this context, the construction techniques of 19th- and 20th-century Rum Orthodox churches built during the late Ottoman period are examined within the scope of this thesis. These structures face risks from improper restoration, constant exposure to environmental conditions, and earthquakes. The purpose of the thesis is firstly to increase the knowledge about the selected eight structures and the construction techniques produced by their period and region. The knowledge gained through the thesis is necessary in case the buildings are lost in the future. For the protection of the eight case studies and similar structures, this thesis will act as a valuable resource of architectural, historical, and, most importantly, technical and material knowledge, supporting a scientifically grounded and well-reasoned approach to conservation.Kültürel mirasın etkili bir şekilde korunması, ancak tarihi yöntem ve malzemelere ilişkin önemli bilgiler sunan inşaat tekniklerinin doğru bir şekilde anlaşılmasıyla mümkündür. Standart restorasyon yöntemleri genellikle orijinal olarak kullanılan farklı teknikleri göz ardı etmekte ve bu da korumada başarısızlıklara yol açabilmektedir. Bu bağlamda, geç Osmanlı döneminde inşa edilen 19. ve 20. yüzyıl Rum Ortodoks kiliselerinin yapım teknikleri bu tez kapsamında incelenmiştir. Bu yapılar, yanlış restorasyon, sürekli çevresel koşullara maruz kalma ve deprem gibi risklerle karşı karşıyadır. Tezin amacı, öncelikle seçilen sekiz yapı ile dönemlerinin ve bölgelerinin ürettiği yapım teknikleri hakkında bilgi birikimini artırmaktır. Tez aracılığıyla edinilen bilgi, yapıların gelecekte kaybolması durumunda gereklidir.     Differentiated fatty acid allocation of Daphnia magna helped to maintain their population under food quality deterioration

    Polyunsaturated fatty acids (PUFAs) are vital to the physiological functioning of crustacean zooplankton. However, cyanobacteria blooms frequently lead to PUFA deficiencies, which poses a substantial challenge to population fitness. Therefore, we hypothesize that D. magna adapt to PUFA-deficient conditions by prioritizing PUFA allocation to somatic growth, and then to offspring during reproduction to ensure population persistence. To test this hypothesis, we applied (compound-specific) 13C labeling to compare the turnover of total carbon and certain groups of fatty acids in Daphnia magna fed with Scenedesmus bijuba for 6 days and then switching to a diet of 13C labeled Microcystis wesenbergii for 6 days (with food quality deterioration) or to a diet of 13C-labeled Scenedesmus (without food quality deterioration), respectively. Fatty acid profiles of D. magna mothers and offspring were also analyzed to reveal their PUFA allocation strategies. Life table parameters from D. magna-feeding Scenedesmus switching to Microcystis were compared with D. magna fed with only Scenedesmus or Microcystis to reveal the effect of PUFA allocation on D. magna performance. Our results showed that with food quality deterioration, D. magna exhibited a significantly lower PUFA and carbon turnover and higher offspring: mother ratios in their PUFA contents. Despite this reduced reproduction, the D. magna switching diets showed no significant different intrinsic increasing rate of populations with those fed only Scenedesmus. Meanwhile, the D. magna switching diets performed significantly better than D. magna fed only Microcystis. These results suggest that differential fatty acid allocation of consumers may serve as an adaptive strategy for population maintenance in food quality deterioration and provide ecological implications with cyanobacterial bloom management and Daphnia reproductive plasticity, which needs further explorations

    WAIT: Feature warping for animation to illustration video translation using GANs

    In this paper, we explore a new domain for video-to-video translation. Motivated by the availability of animation movies that are adopted from illustrated books for children, we aim to stylize these videos with the style of the original illustrations. Current state-of-the-art video-to-video translation models rely on having a video sequence or a single style image to stylize an input video. We introduce a new problem for video stylizing where an unordered set of images are used. This is a challenging task for two reasons: (i) we do not have the advantage of temporal consistency as in video sequences; (ii) it is more difficult to obtain consistent styles for video frames from a set of unordered images compared to using a single image. Most of the video-to-video translation methods are built on an image-to-image translation model, and integrate additional networks such as optical flow, or temporal predictors to capture temporal relations. These additional networks make the model training and inference complicated and slow down the process. To ensure temporal coherency in video-to-video style transfer, we propose a new generator network with feature warping layers which overcomes the limitations of the previous methods.     KOLLOİDAL KAUNTUM KUYULARI İÇİN ÇİNKO KALKOJENİT TABANLI KABUK KATMANLARININ GELİŞTİRİLMESİ

    Colloidal nanoplatelets (NPLs) possess unique optical properties and electronic structures, including narrow emission bandwidths, suppressed Auger recombination, and directional emission, which make them ideal candidates for light-emitting devices (LEDs). Among the various types of NPLs explored for LED applications, cadmium-containing core/shell structures are the most extensively studied in the literature. However, cadmium-based shells face two significant challenges. First, they are toxic, raising health and environmental concerns. Second, they exhibit quasi-type II band alignment, causing the electron wavefunction to delocalize across the entire NPL volume. This delocalization reduces the recombination probability and, consequently, the overall efficiency of the LEDs. To address these issues, we propose developing zinc chalcogenide-based shell layers for colloidal NPLs, offering critical advantages over cadmium-based counterparts. Initially, we synthesized CdSe/ZnSe core/shell NPLs that emit in the 615–630 nm range, achieving an average photoluminescence quantum yield (PLQY) of 40–50%. Furthermore, we identified an important relationship between the lateral dimensions of the CdSe core NPLs and the emission linewidths of the core/shell structures: smaller lateral dimensions of the core result in narrower emission linewidths of the core/shell NPLs. To enhance the PLQY further, we deposited an additional ZnS shell layer onto the CdSe/ZnSe core/shell NPLs. This resulted in CdSe/ZnSe/ZnS core/multi shell heterostructures with a significantly improved PLQY of 80–90%. Using these highly efficient NPLs, we successfully fabricated solution-processed NPL-based LEDs. These LEDs demonstrated an external quantum efficiency (EQE) of 3.82% and a maximum brightness of 6477 cd/m². Our findings demonstrate the potential of zinc chalcogenide-based shell layers to replace cadmium-based counterparts in optoelectronic applications, paving the way for safer and more efficient light-emitting technologies.Kolloidal nanolevhacıklar (NPL’lar), dar ışıma bant aralıkları, baskılanmış Auger birleşimi ve yönlü ışıma gibi benzersiz optik ve elektronik özelliklere sahiptir ve bu özellikler, onları ışık yayan aygıtlar (LED'ler) için ideal adaylar yapmaktadır. LED uygulamaları için araştırılan çeşitli NPL türleri arasında, kadmiyum içeren çekirdek/kabuk yapıları literatürde en çok incelenenlerdir. Ancak, kadmiyum tabanlı kabuklar iki önemli sorunla karşı karşıyadır. Birincisi, toksik olmaları sağlık ve çevre açısından endişe yaratmaktadır. İkincisi ise yarı-tip II bant hizalamasına sahip olmaları, elektron dalga fonksiyonunun tüm NPL hacmi boyunca yayılmasına neden olur. Bu yayılma, uyarılmış parçacıkların birleşme olasılığını ve dolayısıyla LED'lerin genel verimliliğini azaltır. Bu sorunları çözmek için, kolloidal NPL'ler için çinko kalkojenit tabanlı kabuk katmanları geliştirilmesini öneriyoruz. Bu kabuklar, kadmiyum tabanlı muadillerine göre önemli avantajlar sunmaktadır. İlk olarak, 615–630 nm aralığında ışıma yapan CdSe/ZnSe çekirdek/kabuk NPL'leri sentezledik ve %40–50 civarında ışıma verimliliği elde ettik. Ayrıca, CdSe çekirdek NPL'lerinin yatay boyutları ile çekirdek/kabuk yapıların ışıma bant aralıkları arasında önemli bir ilişki belirledik: Daha küçük yatay boyutlara sahip çekirdek yapıları kaplandığında, çekirdek/kabuk NPL'lerinin daha dar ışıma bant aralıklarına yol açmaktadır. Işıma verimliliğini daha da artırmak için, CdSe/ZnSe çekirdek/kabuk NPL'lerine ek bir ZnS kabuk katmanı kapladık. Bu sayede, ışıma verimliliği %80–90'a önemli ölçüde artırılmış CdSe/ZnSe/ZnS çekirdek/çoklu kabuk heteroyapılarını sentezlemiş olduk. Bu yüksek ışıma verimliliğine sahip olan NPL'leri kullanarak, çözelti tabanlı NPL bazlı LED'leri başarıyla ürettik. Bu LED'ler, %3,82 harici kuantum verimi (EQE) ve 6477 cd/m² maksimum parlaklık sergiledi.     Synergistic hybrid nanostructures for Ultra-Sensitive photoelectrochemical detection of hepatocellular carcinoma ctDNA

    In recent years, the integration of quantum dots (QDs) and upconverting nanoparticles (UCNPs) has emerged as a promising strategy in biomedical research, leveraging QDs’ efficient down-conversion and UCNPs’ remarkable up-conversion capabilities. This synergy has attracted significant attention for its potential in biomedical applications, particularly in sensitive detection systems. Here, we report the development of a highly sensitive photoelectrochemical (PEC) employing a hybrid nanostructure comprising CdSe QDs and NaYF4:Yb,Er UCNPs. This sensor was specifically tailored for the detection of circulating tumor DNA (ctDNA) with mutations associated with hepatocellular carcinoma (HCC) using synthetic DNA probes. The CdSe QDs and NaYF4:Yb,Er UCNPs were synthesized and integrated using Triton X-100, a non-ionic surfactant. Comprehensive characterization confirmed the successful formation of the hybrid nanostructure, crucial for optimizing sensor performance. Our PEC platform effectively addressed the challenge of detecting single-nucleotide mismatches in HCC ctDNAs, demonstrating superior sensitivity compared to conventional electrochemical methods. Notably, the system exhibited excellent linearity across a broad concentration range (400 aM to 200 pM), surpassing traditional electrochemical approaches. Key to its clinical relevance, the PEC sensor achieved a remarkable limit of detection (2.32 aM) and limit of quantification (15.61 aM), underscoring its potential for early-stage cancer diagnosis as well as very good recovery from spiked samples. This capability is pivotal in overcoming current diagnostic limitations, such as the asymptomatic nature of early-stage HCC and the need for more accurate biomarkers. Our study highlights the promising role of hybrid nanostructures in enhancing biosensor sensitivity and response times, offering a pathway towards improved early detection and management of HCC

    Radical changes in the seismic design philosophy based on 2023 Kahramanmara,s earthquakes observations in Türkiye

    The two simultaneous earthquakes occurred in 2023 leading to over 50 000 casualties in more than 250 thousand collapsed and heavily damaged buildings in T & uuml;rkiye. Despite modernizing the seismic code regularly, it has not been possible for T & uuml;rkiye to mitigate the seismic risk. Two important and new observations were made in the aftermath of 2023 earthquakes: (i) Structural wall systems exhibited superior performance with no record of collapse. (ii) Several thousand buildings constructed after 2000 collapsed despite better seismic codes. This paper presents the background of the radical modifications for minimum structural wall amounts and maximum interstory drift ratio limits. First, the site observations for the performance of buildings with structural walls are presented. Based on seismic assessment of more than 30 000 existing building plans, the relationship between maximum interstory drift demand and wall-column area ratio is examined. The proposed limits are compared to the risk levels of existing buildings for validation

    The aim of this study is to redefine the relationship between art and architecture, focusing on their in-between. Site-specificity is introduced as a critical spatial strategy that extends beyond definitions of art, medium, practice, site, space, and architecture. Therefore, it becomes an investigative tool that can magnify the site-art relationship, which would help to analyze shifts, transformations, and mutations of terms, practices, and spaces. The aim is to look into the terms and conditions of critical spatial practice in the expanded field that site-specificity defines, while creating a rich dialogue with existence of contemporary art spaces. The study approaches the history of site-specific art and contemporary art spaces transnationally, shifting the focus from isolated national narratives to practices and spaces that operate within complex layered networks, circulations of art, artists, curators, exhibitions, and architecture. Bridging art history and architecture on this transnational basis, the thesis examines exhibitions such as Gar (1995), Anı/Bellek (1991, 1993), İstanbul Yaya Sergisi (2002), and the 4th İstanbul Biennial (1995), combining archival analysis, oral history studies, and visual analysis. By tracing the transformations of art and space in the Turkish context, the study aims to fill a critical gap in scholarship and contribute a new perspective to global discussions on the spatiality of contemporary art vis-à-vis the concepts of liminality, sedentariness, mobility, and place-making.Bu çalışmanın amacı, sanat ve mimarlık arasındaki ilişkiyi yeniden tanımlamaktır, özellikle mekân ve sergilenen nesne kalan alana odaklanmaktadır. Mekâna özgüllüğün; sanat, araç, uygulama, yer, mekân ve mimarlık tanımlarının ötesine geçen eleştirel bir mekansal strateji olarak tanıtılması amaçlanmaktadır. Bu nedenle, yer-sanat ilişkisini büyütebilen bir araştırma aracı haline gelerek terimlerin, uygulamaların ve mekanların değişimlerini, dönüşümlerini ve mutasyonlarını analiz etmeye yardımcı olur. Amaç, mekâna özgüllüğün tanımladığı genişletilmiş alanda, eleştirel mekân pratiğinde şart ve koşullarını incelemek ve çağdaş sanat mekanlarının varlığında zengin bir diyalog oluşturmaktır. Çalışma, mekânsallığa özgü sanatın ve çağdaş sanat mekânlarının tarihine ulusötesi bir yaklaşımla yaklaşır ve odak noktasını izole ulusal anlatılardan, karmaşık ve katmanlı bağlantılar, sanat, sanatçılar, küratörler, sergiler ve mimarinin dolaşımı içinde işleyen uygulamalara ve mekânlara kaydırır. Bu ulusötesi sanat tarihi ve mimariyi birbirine bağlayan tez, Gar (1995), Anı/Bellek (1991, 1993), İstanbul Yaya Sergisi (2002) ve 4.     In this research, I question how feminist acts of resistance, specifically Feminist Night Marches in Istanbul, produce alternative spatial and temporal conditions within the striated architectures of contemporary urban environments. To establish this process of inquiry, I borrow the concepts of the smooth and striated space that Gilles Deleuze and Félix Guattari coined in their infamous work “A Thousand Plateaus” (1987) to investigate how feminist bodies disrupt, negotiate, and reclaim urban space through movement, rhythm, and collective presence. In this regard, this thesis argues that feminist night marches transform urban space not simply by occupying it, but by what Elizabeth Grosz (2001) terms “thinking from the outside”—mobilizing bodily forces that unsettle the striated logics of urban architecture and territorial control. Rather than seeing architecture and urban space as fixed forms, Grosz invites us to consider space as dynamic, shaped by the intensities and trajectories that pass through it. In the gendered dimension of urban life, I focus my research on the input that cities are not neutral socio-spatial urban terrains. Their streets and buildings have always been shaped by histories of power, control, and exclusion. Anchored in the conceptual framework of Deleuze and Guattari’s smooth and striated space, this thesis provides an interdisciplinary spatial theory—including Elizabeth Grosz’s notion of “thinking from the outside” and Henri Lefebvre’s rhythmanalysis— weaving together diverse theoretical perspectives to deepen the comprehension of feminist acts of resistance in urban space.Bu araştırmada, feminist direniş eylemlerinin—özellikle İstanbul’daki Feminist Gece Yürüyüşleri’nin—günümüz kent ortamlarının çizgisel (striated) mimarileri içinde nasıl alternatif mekânsal ve zamansal koşullar ürettiğini sorguluyorum. Bu sorgulama sürecini kurmak için Gilles Deleuze ve Félix Guattari’nin A Thousand Plateaus (1987) adlı eserlerinde ortaya koydukları striated (çizgisel) ve smooth (akışkan) mekân kavramlarından yararlanarak, feminist bedenlerin hareket, ritim ve kolektif varlık yoluyla kent mekânını nasıl bozduğunu, yeniden müzakere ettiğini ve sahiplendiğini inceliyorum. Bu bağlamda tez, feminist gece yürüyüşlerinin kent mekânını yalnızca işgal ederek değil, Elizabeth Grosz’un (2001) “dışarıdan düşünme” (thinking from the outside) olarak adlandırdığı biçimde dönüştürdüğünü öne sürmektedir. Bu dönüşüm, kentsel mimarinin çizgisel mantığını ve mekânın hiyerarşik düzenlenişini sarsan bedensel kuvvetlerin seferber edilmesiyle gerçekleşmektedir. Grosz, mimariyi ve kentsel mekânı sabit ve kapalı formlar olarak değil, içinden geçen yoğunluklar ve yönelimlerle şekillenen dinamik yapılar olarak kavramsallaştırmamızı önerir. Kent yaşamının toplumsal cinsiyetle kurduğu ilişkiye odaklanan bu tez, şehirlerin nötr ya da tarafsız mekânsal düzenlemeler sunmadığını vurgular. Sokaklar ve yapılar tarih boyunca iktidar ilişkileri, denetim stratejileri ve dışlayıcı mekânsal politikalarla biçimlendirilmiştir.     Endometrial cancer (EC) is one of the most common gynecologic malignancies, with 420.242 new cases and 97.704 deaths reported worldwide in 2022. EC comprises two major subtypes, endometrioid (EEC) and non-endometrioid (NEC), which differ in prognosis and therapeutic response. Thus, subtype-specific biomarkers are crucial for precise diagnosis, prognosis, and treatment stratification. This study aims to verify previously reported potential biomarkers for EC as an essential phase in the biomarker development pipeline and to evaluate their potential diagnostic and prognostic value. For this purpose, multiple-reaction-monitoring (MRM) method specific to nine proteins, namely transgelin (Q01995), peptidyl prolyl cis-transisomerase (Q00688), cyclophilin A (P62937), macrophage capping protein (P40121), pyruvate kinase M1/M2 (P14618), glutathione S-transferase P (P09211), manganese-superoxide dismutase (P04179), cytosol aminopeptidase (P28838), and complement component C4A (P0C0L4), was developed with stable isotopic-labeled (SIL) peptides. Method performance was evaluated through linearity, limit-of-detection, instrument stability, and reprodubility of sample preparation by following European Medicines Agency guideline. Unique peptides of nine proteins were monitored in tissue samples from EEC, NEC, and healthy groups following protein extraction and proteolytic digestion. Seven proteins displayed significant dysregulation. Down-regulation of Q01995 is a recurring event in malignancies, serving as a potential biomarker of cancer progression. Conversely, P14618, P40121, Q00688, P28838 were up-regulated, reflecting hallmarks of cancer. However, down-regulation of P0C0L4 and up-regulation of P09211 are not typically observed across cancers. Importantly, P09211 was the only protein showing subtype-specific elevation in EEC compared to NEC, highlighting its prognostic value. Yedi proteinde anlamlı regülasyon göstermiştir. Q01995’in aşağı regülasyonu, malignitelerde tekrarlayan bir olaydır ve kanser sürecinin potansiyel biyobelirteci olabileceğini göstermektedir. Buna karşılık, P14618, P40121, Q00688 ve P28838 yukarı regülasyon göstermiştir ve kanserin karakteristik özelliklerini yansıtmaktadır. Ancak, P0C0L4’ün aşağı regülasyonu ve P09211’in yukarı regülasyonu kanserlerde genellikle gözlenmemektedir.
