Reducing microglial lipid load enhances β amyloid phagocytosis in an Alzheimer’s disease mouse model
Macrophages accumulate lipid droplets (LD) under stress and inflammatory conditions. Despite the presence of LD-loaded macrophages in many tissues, including the brain, their contribution to neurodegenerative disorders remains elusive. This study investigated the role of lipid metabolism in Alzheimer’s disease (AD) by assessing the contribution of LD-loaded brain macrophages, including microglia and border-associated macrophages (BAMs), in an AD mouse model. In particular, activated CD11c+ microglia, localized proximal to beta-amyloid (Aβ) plaques, had a pronounced lipid-associated gene signature and a higher LD load than other microglial subtypes. Having observed that elevated intracellular LD content correlated inversely with microglial phagocytic activities, we subsequently inhibited LD formation specifically in CX3CR1+ brain macrophages using an inducible transgenic mouse model. Reducing the lipid load of microglia and CX3CR1+ BAMs improved phagocytic and efferocytic capacities and significantly reduced Aβ deposition in the brain. Therefore, mitigating LD accumulation in microglia and BAMs provides new perspectives for the treatment of AD

Wastewater surveillance (WWS) has been globally recognised to be a useful tool in quantifying SARS-CoV-2 RNA at the community and residential levels without biases associated with case-reporting. The emergence of variants of concern (VOCs) have given rise to an unprecedented number of infections even though populations are increasingly vaccinated. This is because VOCs have been reported to possess higher transmissibility and can evade host immune responses. Results in this study corroborate clinical incidence of the Omicron lineages in Singapore and indicate minimal silent circulation prior to January 2022. The subsequent simultaneous spread of both variant lineages followed strategic relaxation of safe management measures upon meeting nationwide vaccination goals

This paper proposes an optical reflecting surface (ORS)-assisted free space optics (FSO) communication system based on the optical space shift keying (OSSK) technique. Closed-form expressions for the probability density function (PDF) and moment generating function (MGF) of the system's signal-to-noise ratio (SNR) are derived using the Malaga turbulence model. Upper bounds for the average bit error rate (BER) and lower bounds for ergodic capacity are also obtained, with diversity gain calculated through asymptotic BER.
