    Antitumor Potential of Different Treatment Approaches Using Cold Atmospheric Pressure Plasma on Oral Squamous Cell Carcinoma Models: In Vitro Study

    Background/Objectives: Cold atmospheric plasma (CAP) has shown a strong anticancer effect on a variety of tumors, presenting a new approach for the effective treatment of oral squamous cell carcinoma (OSCC), one of the most prevalent malignant neoplasms with a high mortality rate. Here, we aimed to comprehensively investigate the antitumor potential of two approaches of CAP treatment on both two-dimensional and three-dimensional OSCC cell line models, as well as to analyze whether plasma treatment enhances the sensitivity of OSCC to chemotherapy. Methods: An in-house designed plasma needle, with helium as a working gas, was used to treat the SCC-25 cell line directly or indirectly via plasma-treated medium (PTM). The antitumor effect of CAP was assessed by measuring cell viability, apoptosis, adhesion, and migration. In addition, the combined effect of PTM and cisplatin was analyzed in SCC-25 tumor spheroids, as a more complex and reliable in vitro model. Results: Both plasma treatments showed time-dependent antitumor effects affecting their viability, adhesion, and migration. The rate of apoptosis was higher after incubation with PTM and is mediated by the intrinsic pathway. By utilizing the 3D spheroid carcinoma model, we confirmed the antitumor potential of CAP and additionally demonstrated an increased chemosensitivity of PTM-treated carcinoma cells. Conclusions: The results of our study illustrate a promising avenue for the application of CAP as a therapeutic option for OSCC, either as a standalone treatment or in combination with cisplatin

    Modern agri-food systems demand crops that are climate change resilient, support environmental sustainability, enhance dietary diversity and appeal to consumers. This article examines the potential of underutilized crops in meeting these challenges. We take Lathyrus as a case study and examine it within the context of European agri-food value chains, with a particular focus on Germany and Spain. The study employs a range of methods—literature review, interviews, field observations, and consumer opinion surveys —to explore its versatility. The findings reveal that Lathyrus sativus (grass pea) offers significant benefits: it is adapted to dry climates and tolerates high rainfall. As a nitrogen fixing crop, it can play an important role in crop rotations, reducing the need for fertilization and providing an attractive alternative for organic farmers. Through appropriate processing methods, Lathyrus can enhance its beneficial nutritional characteristics. Furthermore, it is well suited for the development of innovative recipes that align with modern culinary trends and meet consumers’ expectations across different cultures. To make use of its potential, adaptive cultivation methods and value chain cooperation are needed. Along the value chain, the affordable pricing of Lathyrus-based products and clear communication of their nutritional, environmental, and social benefits prove to be most critical

    Synthesis, Characterization, HSA/DNA Binding, and Cytotoxic Activity of [RuCl2(η6-p-cymene)(bph-κN)] Complex

    A novel ruthenium(II) complex, [RuCl2(η6-p-cymene)(bph-κN)] (1), was synthesized and structurally characterized using FTIR and NMR spectroscopy. Density functional theory (DFT) calculations supported the proposed geometry and allowed for comparative analysis of experimental and theoretical spectroscopic data. The interaction of complex 1 with human serum albumin (HSA) and calf thymus DNA was investigated through fluorescence quenching experiments, revealing spontaneous binding driven primarily by hydrophobic interactions. The thermodynamic parameters indicated mixed quenching mechanisms in both protein and DNA systems. Ethidium bromide displacement assays and molecular docking simulations confirmed DNA intercalation as the dominant binding mode, with a Gibbs free binding energy of −34.1 kJ mol−1. Antioxidant activity, assessed by EPR spectroscopy, demonstrated effective scavenging of hydroxyl and ascorbyl radicals. In vitro cytotoxicity assays against A375, MDA-MB-231, MIA PaCa-2, and SW480 cancer cell lines revealed selective activity, with pancreatic and colorectal cells showing the highest sensitivity. QTAIM analysis provided insight into metal–ligand bonding characteristics and intramolecular stabilization. These findings highlight the potential of 1 as a promising candidate for further development as an anticancer agent, particularly against multidrug-resistant tumors

    Ru(II) Complexes with 3,4-Dimethylphenylhydrazine: Exploring In Vitro Anticancer Activity and Protein Affinities

    Two new Ru(II) complexes, mononuclear [RuCl2(η6-p-cymene)(3,4-dmph-κN)] (1) and the binuclear complex [{RuCl(η6-p-cymene)}2(μ-Cl)(μ-3,4-dmph-κ2N,N′)]Cl (2; 3,4-dmph = 3,4-dimethylphenylhydrazine), are synthesized and experimentally and theoretically structurally characterized utilizing 1H and 13C NMR and FTIR spectroscopy, as well as DFT calculations. Degradation product of 2, thus ([{RuCl(η6-p-cymene)}2(μ-Cl)(μ-3,4-dmph-κ2N,N′)][RuCl3(η6-p-cymene)] (2b) was characterized with SC-XRD. In the crystals of 2b, the cationic and anionic parts interact through N-H...Cl hydrogen bridges. The spectrofluorimetric measurements proved the spontaneity of the binding processes of both complexes and HSA. Spin probing EPR measurements implied that 1 and 2 decreased the amount of bound 16-doxylstearate and implicated their potential to bind to HSA more strongly than the spin probe. The cytotoxicity assessment of both complexes against the MDA-MB-231 and MIA PaCa-2 cancer cell lines demonstrated a clear dose-dependent decrease in cell viability and no effect on healthy HS-5 cells. Determination of the malondialdehyde and protein carbonyl concentrations indicated that new complexes could offer protective antioxidant benefits in specific cancer contexts. Gel electrophoresis measurements showed the reduction in MMP9 activity and indicated the potential of 1 in limiting the cancer cells’ invasion. The annexin V/PI apoptotic assay results showed that 1 and 2 exhibit different selectivity towards MIA PaCa-2 and MDA-MB-231 cancer cells.     A European perspective on intercultural competence and prejudice: a cross-cultural analysis

    IntroductionThis study examines the relationship between intercultural competence and subtle and blatant prejudice toward refugees in Germany, the UK, Hungary, the Czech Republic, Ukraine, and Serbia.MethodUsing the Cultural Intelligence Scale and the Multicultural Personality Questionnaire, we analyze whether measures of intercultural competence are associated with lower levels of prejudice, with a particular focus on Eastern European contexts compared to established findings from Central Europe, North America, and East Asia.ResultsResults highlight cross-cultural differences in both the overall scales and their subdimensions in their associations with prejudice.DiscussionThese findings challenge the universal applicability of measures such as the Cultural Intelligence Scale and the Multicultural Personality Questionnaire, revealing that their effectiveness varies across cultural settings. The study underscores the necessity of culturally adapted interventions to address prejudice and foster social cohesion in regions experiencing increasing migration. By advancing a culturally nuanced perspective, this research contributes to refining intercultural competence methodologies and informing targeted integration policies that address region-specific challenges

    Oral Squamous Cell Carcinoma Exosomes Upregulate PIK3/AKT, PTEN, and NOTCH Signaling Pathways in Normal Fibroblasts

    Exosomes, small extracellular vesicles secreted by various cell types, have gained significant attention in cancer investigations. Isolation and characterization of exosomes derived from DOK (dysplastic oral keratinocyte), SCC (squamous cell carcinoma) and HaCaT (normal skin keratinocyte) cell lines and microRNA profiling were conducted. Magnetic sorting was applied to obtain pure exosomes. Morphology and size were characterized by transmission electron microscopy and nanoparticle tracking analysis. Validation of membrane exosomal markers (CD9, CD63) was performed via Western blotting. MiR-21, miR-31, and miR-133 levels were analyzed in exosomes and parent cells by qPCR. Biological effects of the exosomes were tested by adding them to fibroblast cultures and determining the expression of relevant carcinogenesis markers by qPCR. Exosomes appeared as cup-shaped nano-sized particles, and there was no difference regarding particle diameter and concentration between the three types of exosomes. The oncogenic miR-21 was significantly upregulated both in SCC and SCC-derived exosomes compared to DOK and HaCaT cells and their respective exosomes. However, miR-31 unexpectedly showed the highest expression in normal cells and the lowest in HaCaT exosomes. MiR-133, the tumor suppressor miRNA, was downregulated in both SCC and DOK cells compared to normal (HaCaT) cells, while the opposite situation was observed in exosomes, with HaCaT cells showing the lowest levels of miR-133. The differences in exosome content were reflected in signaling pathway activation in exosome-treated fibroblasts, with SCC exosomes exerting the most potent effect on several cancer-related pathways, notably PIK3/AKT, PTEN, and NOTCH signaling cascades

    Vitamin A5: Evidence, Definitions, Gaps, and Future Directions

    With the emergence of a new vitamin concept—vitamin A5—it is essential to first clarify the basic definition of vitamins, particularly vitamin A. This article summarizes the foundational concepts and definitions of vitamins with particular relevance to the discovery, establishment, and categorization of new vitamin concepts. Vitamin A5 was discovered 80 years after the last vitamin was identified. It serves as an umbrella term for the dietary precursors 9-cis-β,β-carotene and 9-cis-13,14-dihydroretinol for the endogenous activator of the nuclear hormone receptor RXR, 9-cis-13,14-dihydroretinoic acid. However, several questions arise: Which criteria are typically used to identify a substance as a vitamin? How does vitamin A5 fit into the sometimes misleading definition of vitamin A? This review summarizes key findings and provides a comprehensive assessment of the current understanding, concluding that (a) vitamin A5 is a newly identified micronutrient that plays an important role in the prevention of diet-related diseases and (b) vitamin A5 is an important micronutrient that provides a plausible, mechanistic explanation for why a Western lifestyle diet low in vegetables and especially leafy vegetables can lead to a high prevalence of Western-lifestyle diseases, particularly neurological diseases and poor mental health

    Physiological Differences in Cardiorespiratory and Metabolic Parameters Between Football Players from Top- and Mid-Ranked Teams in the Serbian Super League

    This study investigated physiological differences in cardiorespiratory and metabolic performance parameters between professional football players from top- (TR) and mid-ranked teams (MR) in the Serbian Super League. A total of 55 male outfield players (TR: n = 29; MR: n = 26) were assessed in March 2022 using a maximal multistage treadmill protocol and lactate analysis. The key cardiorespiratory variables included maximum oxygen uptake (VO2max), heart rate at the anaerobic threshold (HR AT), and recovery heart rate metrics, while the metabolic variables focused on lactate concentrations and efficiency indices. The results indicate that the TR players achieved significantly lower HR AT values (162 ± 10.26 vs. 168.77 ± 7.28 bpm; p = 0.017) and demonstrated superior second-minute recovery (%Re 2′: 66.62 ± 14.08% vs. 34.53 ± 9.13%, p < 0.001). In contrast, the MR players exhibited higher VO2max (62.65 ± 4.48 vs. 60.06 ± 3.29 mL/kg/min; p = 0.017) and greater cardiorespiratory efficiency scores. The lactate parameters were comparable between the groups, except for the metabolic efficiency index (Index ME), which were favorable among the TR players (p = 0.011). These findings highlight that while MR players possess higher aerobic capacity, TR players demonstrate superior physiological recovery and metabolic control, reflecting adaptations to different tactical demands and match intensities. The results offer practical implications for individualized training design and performance monitoring in elite football settings

    A Prevalence Study on Anoplocephala spp. in Serbian Horses: Navigating Diagnostic Challenges and Understanding Infection Risks

    Anoplocephala spp. are common equine tapeworm species in Europe, frequently found in grazing horses. Anoplocephala perfoliata is the most pathogenic, clinically significant species associated with gastrointestinal disorders, particularly colic, and can have a fatal outcome in some horses. The aim of this study was to determine the infection prevalence of Anoplocephala spp. in Serbia and to identify relevant risk factors. A total of 173 horses from various regions were tested using a combination of diagnostic methods: coprological analysis via combined sedimentation–flotation and the Mini-FLOTAC technique, as well as serological testing using a commercial ELISA test. The overall prevalence was 38.7%, with a higher number of positive cases being identified by serology. It was demonstrated that coprology and serology are complementary approaches for prevalence studies. Infection risk factors included high stocking density, free-roaming status and co-infection with nematodes, while lower rainfall and temperate climate were associated with reduced risk of tapeworm infection. These findings highlight the challenges of Anoplocephala spp. detection and provide insight into the epidemiology of equine tapeworm infection in Southeastern Europe
