BMP2 downregulates urokinase-type plasminogen activator via p38 MAPK: Implications in C2C12 cells myogenic differentiation
Bone morphogenetic protein (BMP)2 strongly affects the differentiation program of myoblast cells by inhibiting myogenesis and inducing osteogenic differentiation. In turn, extracellular matrix (ECM) proteinases, such as urokinase-type plasminogen activator (uPA), can influence the fate of muscle stem cells by participating in ECM reorganization. Although both BMP2 and uPA have antagonistic roles in muscles cells differentiation, no connection between them has been elucidated so far. This study aims to determine whether BMP2 regulates uPA expression in the myogenic C2C12 cell line and its impact on muscle cell fate differentiation. Our results showed that BMP2 did not modify C2C12 cell proliferation in a growth medium or myogenic differentiation medium. Although BMP2 inhibited myogenesis and induced osteogenesis, these effects were achieved with different doses of BMP2. Low concentrations of BMP2 blocked myogenesis, while a higher concentration was needed to induce osteogenesis. Reduced uPA expression was noticed alongside myogenic inhibition at low concentrations of BMP2. BMP2 activated p38 MAPK signaling to inhibit uPA activity. Furthermore, ectopic human uPA expression reduced BMP2′s ability to inhibit the myogenic differentiation of C2C12 cells. In conclusion, BMP2 inhibits uPA expression through p38 MAPK and in vitro myogenesis at non-osteogenic concentrations, while uPA ectopic expression prevents BMP2 from inhibiting myogenesis in C2C12 cells
The Metabolic Features of Tumor-Associated Macrophages: Opportunities for Immunotherapy?
Besides transformed cells, the tumors are composed of various cell types that contribute to undesirable tumor progression. Tumor-associated macrophages (TAMs) are the most abundant innate immune cells in the tumor microenvironment (TME). Within the TME, TAMs exhibit high plasticity and undergo specific functional metabolic alterations according to the availability of tumor tissue oxygen and nutrients, thus further contributing to tumorigenesis and cancer progression. Here, we review the main functional TAM metabolic patterns influenced by TME, including glycolysis, amino acid, and fatty acid metabolism. Moreover, this review discusses antitumor immunotherapies that affect TAM functionality by inducing cell repolarizing and metabolic profiles towards an antitumoral phenotype. Also, new macrophage-based cell therapeutic technologies recently developed using chimeric antigen receptor bioengineering are exposed, which may overcome all solid tumor physical barriers impeding the current adoptive cell therapies and contribute to developing novel cancer immunotherapies
Influence of Higenamine on Exercise Performance of Recreational Female Athletes: A Randomized Double-Blinded Placebo-Controlled Trial
The aim of this study was to determine the ergogenic effects and the safety profile of a one-component higenamine supplement in female recreational athletes. Twelve recreational female basketball players (age 29–41 years, oxygen consumption (VO2max) > 30 ml⋅kg–1⋅min–1, with training > 5 h wk–1) were randomized either to the higenamine group, or to the placebo group for 3 weeks. In order to determine ergogenic effects and safety profile of higenamine administration, we assessed the following variables before and after 3 weeks of supplementation: anthropometric parameters, resting metabolic rate (RMR), exercise testing variables, serum free fatty acids (FFAs), blood pressure, enzyme activity, urea, lipid profile, and complete blood count. There were no differences between groups in anthropometric parameters, including basal metabolic rate (BMR), RMR and body fat [p = 0.706 (Cohen’s d 0.223), p = 0.169 (Cohen’s d 0.857), and p = 0.223 (Cohen’s d 0.750), respectively], FFAs [0.43 ± 0.03 vs. 0.54 ± 0.23, p = 0.206 (Cohen’s d 0.540)], neither significant differences in cardiopulmonary parameters after the intervention period. Furthermore, all measured outcome variables in the safety assessment were not significant, with values remaining stable during the intervention period for participants in both groups. This is the first study to document the effects and the safety profile of higenamine-based dietary supplements at a specified dose in female recreational athletes. Our data indicate that 21-day of supplementation with 75 mg higenamine would not result in improving cardiopulmonary exercise fitness and weight loss in female recreational athletes. Moreover, supplementation with 75 mg higenamine is safe and well-tolerated in younger recreational female athletes

Short-Term Consumption of Pomegranate Juice Alleviates Some Metabolic Disturbances in Overweight Patients with Dyslipidemia
Pomegranate juice (PJ) has potential positive effects in patients with metabolic disturbances due to a high content of polyphenols. The objective of this study was to evaluate effects of a 2-week consumption of dietary doses of PJ on blood pressure, lipid metabolism, and oxidative stress markers in overweight patients with dyslipidemia. Twenty-four patients, 8 males and 16 females, 40-60 years of age, with established overweight and dyslipidemia were randomly assigned into intervention group, who consumed 300 mL of PJ daily for 2 weeks, or control group. After 2 weeks of juice intake, intervention group had significantly lower diastolic blood pressure, low-density lipoprotein cholesterol, aminotransferase, and activity of glutathione peroxidase. Furthermore, patients who consumed PJ had reduced percentage of docosahexaenoic acid (22:6n-3, DHA) in plasma phospholipids and increased estimated activity of stearoyl-CoA desaturase. In erythrocytes, we found a significant increase in the levels of dihomo-gamma- linolenic acid (20:3n-6, DGLA) and DHA, as well as in estimated activity of Delta 6 desaturase, and a decrease in estimated activity of Delta 5 desaturase. These results show that even a short-term consumption of dietary doses of PJ exerts beneficial effects and affects lipid metabolism in overweight patients with dyslipidemia
Metabolically healthy obesity: is there a link with polyunsaturated fatty acid intake and status?
The aim of this study was to compare dietary intake and status of polyunsaturated fatty acids (PUFA) in plasma and erythrocyte phospholipids metabolically healthy and unhealthy, and obese and nonobese persons. Metabolic health status in 171 participants was defined according to criteria for metabolic syndrome. Obese and nonobese metabolically unhealthy persons (MUHO and MUHNO) had higher energy intake of n-6 PUFA (7.82 +/- 1.03 and 7.49 +/- 0.86) and lower intake of n-3 PUFA (0.60 +/- 0.12 and 0.62 +/- 0.11) compared to obese and nonobese metabolically healthy persons (MHO and MHNO) (5.92 +/- 0.63 and 5.72 +/- 0.67; 1.20 +/- 0.07 and 1.22 +/- 0.09, respectively) and a higher n-6/n-3 PUFA ratio. The plasma level of n-6 PUFA was lower in the MUHO and MUHNO groups (38.49 +/- 3.71 and 38.53 +/- 2.19) compared to MHNO (40.90 +/- 2.43), while n-3 PUFA status was lower in obese than in nonobese persons (3.58 +/- 0.79 and 3.50 +/- 1.02 vs. 4.21 +/- 0.80 and 4.06 +/- 1.15). The MHO group had a higher eicosapentaenoic/arachidonic acid ratio and estimated desaturase (SCD16, D6D) and elongase activity in plasma phospholipids compared to MHNO. The low intake of n-3 PUFA is directly associated with metabolic risk factors. These results indicated that obesity is closely associated with low levels of n-3 PUFA in plasma phospholipids, suggesting that dietary modifications including n-3 PUFA supplementation appear to be suitable therapeutic strategy in obese persons
Is There a FADS2-Modulated Link between Long-Chain Polyunsaturated Fatty Acids in Plasma Phospholipids and Polyphenol Intake in Adult Subjects Who Are Overweight?
Dietary polyphenols promote cardiometabolic health and are linked with long-chain polyunsaturated fatty acids in plasma phospholipids (LC-PUFA). The FADS2 polymorphisms are associated with LC-PUFA metabolism and overweight/obesity. This 4-week study examined the link between polyphenol intake, FADS2 variants (rs174593, rs174616, rs174576) and obesity in 62 overweight adults (BMI gt = 25), allocated to consume 100 mL daily of either: Aronia juice, a rich source of polyphenols, with 1177.11 mg polyphenols (expressed as gallic acid equivalents)/100 mL (AJ, n = 22), Aronia juice with 294.28 mg polyphenols/100 mL (MJ, n = 20), or nutritionally matched polyphenol-lacking placebo as a control (PLB, n = 20). We analyzed LC-PUFA (% of total pool) by gas chromatography and FADS2 variants by real-time PCR. Four-week changes in LC-PUFA, BMI, and body weight were included in statistical models, controlling for gender and PUFA intake. Only upon AJ and MJ, the presence of FADS2 variant alleles affected changes in linoleic, arachidonic, and eicosapentaenoic acid (EPA). Upon MJ treatment, changes in EPA were inversely linked with changes in BMI (beta= -0.73, p = 0.029) and weight gain (beta= -2.17, p = 0.024). Only in subjects drinking AJ, the link between changes in EPA and anthropometric indices was modified by the rs174576 variant allele. Our results indicate the interaction between FADS2, fatty acid metabolism, and polyphenol intake in overweight subjects
Surveillance of foodborne parasitic diseases in Europe in a One Health approach
In 2012, WHO/FAO ranked 24 foodborne parasites (FBP) using multicriteria decision analysis (MCDA) to provide risk assessors with a basis for prioritising control of highly ranked FBP on the global level. One conclusion was that ranking may differ substantially per region. In Europe, the same methodology was used to rank FBP of relevance for Europe. Of the 24 FBP, the top-five prioritised FBP were identified for Europe as Echinococcus multilocularis, Toxoplasma gondii, Trichinella spiralis, E. granulosus, and Cryptosporidium spp., all of which are zoonotic. The objective of the present study was to provide an overview of surveillance and reporting systems in Europe for these top five prioritised FBP in the human and animal populations, to identify gaps, and give recommendations for improvement. Information on the surveillance systems was collected from 35 European countries and analysed according to the five different regions. For most FBP, human surveillance is passive in most countries and regions in Europe and notification differs between countries and regions. Adequate surveillance programmes for these FBP are lacking, except for T. spiralis, which is notifiable in 34 countries with active surveillance in susceptible animals under EU directive. Although human and animal surveillance data are available for the five prioritised FBP, we identified a lack of consistency in surveillance and reporting requirements between national experts and European bodies. Recommendations for improved surveillance systems are discussed
Reliability of Sprint Force-Velocity-Power Profiles Obtained with KiSprint System
This study aimed to assess the within-and between-session reliability of the KiSprint system for determining force-velocity-power (FVP) profiling during sprint running. Thirty (23 males, 7 females; 18.7 ± 2.6 years;) young high-level sprinters performed maximal effort sprints in two sessions separated by one week. Split times (5, 10, 20 and 30 m), which were recorded with a laser distance meter (a component of the KiSprint system), were used to determine the horizontal FVP profile using the Samozino’s field-based method. This method assesses the FVP relationships through estimates of the step-averaged ground reaction forces in sagittal plane during sprint acceleration using only anthropomet-ric and spatiotemporal (split times) data. We also calculated the maximal theoretical power, force and velocity capabilities and the slope of the FV relationship, the maximal ratio of horizontal-to-resultant force (RF), and the decrease in the RF (DRF). Overall, the results showed moderate or good to excellent within-and be-tween-session reliability for all variables (ICC > 0.75; CV < 10 %), with the exception of FV slope and DRF that showed low rel-ative reliability (ICC = 0.47-0.48 within session, 0.31-0.33 be-tween-session) and unacceptable between-session absolute reliability values (CV = 10.9-11.1 %). Future studies are needed to optimize the protocol in order to maximize the reliability of the FVP variables, especially when practitioners are interested in the FV slope and DRF. In summary, our results question the utility of the sprint-based FVP profiling for individualized training pre-scription, since the reliability of the FV slope and D RF variables is highly questionable
Scale Characteristics of Intercultural Competence Measures and the Effects of Intercultural Competence on Prejudice
Due to proceeding globalization processes, involving a rise in mobility and international interdependencies, the frequency and relevance of intercultural contact situations increases. Consequently, the ability to deal effectively with intercultural situations is gaining in importance. However, the majority of studies on measures of intercultural competence focuses on Western Europe and the United States or cultures of the Far East. For the present study, previously understudied Eastern European (former communist) cultures were included, by sampling in Hungary, Serbia, and the Czech Republic, in addition to (the Central or Western European country) Germany. Thus, this study enabled comparisons of scale characteristics of the cultural intelligence scale (CQS), the multicultural personality questionnaire (MPQ), as well as the blatant and subtle prejudice scales, across samples from different cultures. It was also examined how the CQS and MPQ dimensions are associated with prejudice. To analyse scale characteristics, the factor structures and measurement invariances of the used instruments were analyzed. There were violations of configural measurement invariance observed for all of these scales, indicating that the comparability across samples is limited. Therefore, each of the samples was analyzed separately when examining how the CQS and MPQ dimensions are related to prejudice. It was revealed that, in particular, the motivational aspect of the CQS was statistically predicting lower prejudice. Less consistently, the MPQ dimensions of open-mindedness and flexibility were statistically predicting lower prejudice in some of the analyses. However, the violations of measurement invariance indicate differences in the constructs' meanings across the samples from different cultures. It is consequently argued that cross-cultural equivalence should not be taken for granted when comparing Eastern and Western European cultures
