    Resveratrol improved kidney function and structure in malignantly hypertensive rats by restoration of antioxidant capacity and nitric oxide bioavailability

    Objective Background: The main cause of death among patients with malignant hypertension is a kidney failure. The promising field in essential and malignant hypertension therapy could be centered on the amelioration of oxidative stress using antioxidant molecules like resveratrol. Resveratrol is a potent antioxidative agent naturally occurred in many plants that possess health-promoting properties. Methods: In the present study, we investigated the therapeutic potential of resveratrol, a polyphenol with anti-oxidative activity, in NG-L-Arginine Methyl Ester (L-NAME) treated spontaneously hypertensive rats (SHR) - malignantly hypertensive rats (MHR). Results: Resveratrol significantly improves oxidative damages by modulation of antioxidant enzymes and suppression of prooxidant factors in the kidney tissue of MHR. Enhanced antioxidant defense in the kidney improves renal function and ameliorates the morphological changes in this target organ. Besides, protective properties of resveratrol are followed by the restoration of the nitrogen oxide (NO) pathway. 4) Conclusion: Antioxidant therapy with resveratrol could represent promising therapeutical approach in hypertension, especially malignant, against kidney damage

    Food Composition Data and Tools Online and Their Use in Research and Policy: EuroFIR AISBL Contribution in 2022

    Food, nutrition, and health are linked, and detailed knowledge of nutrient compositions and bioactive characteristics is needed to understand these relationships. Additionally, increasingly these data are required by database systems and applications. This communication aims to describe the contribution to databases and nutrition fields as well as the activities of EuroFIR AISBL; this member-based, non-profit association was founded to ensure sustained advocacy for food information in Europe and facilitate improved data quality, storage, and access as well as encouraging wider exploitation of food composition data for both research and commercial purposes. In addition to the description of its role and main objectives, a snapshot of EuroFIR AISBL’s activities over the years is also given using a quantitative research literature analysis approach. The focus of this communication is to provide descriptions and updates of EuroFIR’s online tools, i.e., FoodEXplorer, eBASIS, and PlantLIBRA, by highlighting the main uses and applications. Integrating food-related infrastructures and databases, following standardized and harmonized approaches, and considering interoperability and metrological principles are significant challenges. Ongoing activities and future plans of EuroFIR AISBL are highlighted, including, for instance, work within the Food Nutrition Security Cloud (FNS-Cloud) to make food, nutrition, and (food) security data more findable, accessible, interoperable, and ultimately reusable

    Clinicopathological Relevance of PAX8 Expression Patterns in Acute Kidney Injury and Chronic Kidney Diseases

    Transcription factor PAX8, expressed during embryonic kidney development, has been previously detected in various kidney tumors. In order to investigate expression of PAX8 transcription factor in acute kidney injury (AKI) and chronic kidney diseases (CKD), immunohistochemical analysis was performed. Presence, location and extent of PAX8 expression were analyzed among 31 human kidney samples of AKI (25 autopsy cases, 5 kidney biopsies with unknown etiology and 1 AKI with confirmed myoglobin cast nephropathy), as well as in animals with induced postischemic AKI. Additionally, expression pattern was analyzed in 20 kidney biopsy samples of CKD. Our study demonstrates that various kidney diseases with chronic disease course that results in the formation of tubular atrophy and interstitial fibrosis, lead to PAX8 expression in the nuclei of proximal tubules. Furthermore, patients with PAX8 detected within the damaged proximal tubuli would be carefully monitored, since deterioration in kidney function was observed during follow-up. We also showed that myoglobin provoked acute kidney injury followed with large extent of renal damage, was associated with strong nuclear expression of PAX8 in proximal tubular cells. These results were supported and followed by data obtained in experimental model of induced postischemic acute kidney injury. Considering these findings, we can assume that PAX8 protein might be involved in regeneration process and recovery after acute kidney injury. Thus, accordingly, all investigation concerning PAX8 immunolabeling should be performed on biopsy samples of the living individuals

    Interleukin-17 modulates uPA and MMP2 expression in human periodontal ligament mesenchymal stem cells: Involvement of the ERK1/2 MAPK pathway

    Periodontal disease is a chronic infection of periodontal tissue characterized by extracellular matrix (ECM) degradation due to increased expression of plasminogen activators and matrix metalloproteinases (MMPs) and various proinflammatory cytokines, including interleukin (IL)-17. Successful regeneration of damaged periodontal tissues depends on the proper functionality of periodontal ligament mesenchymal stem cells (PDLMSCs), especially the production of extracellular matrix proteases. We investigated the influence of IL-17 on ECM remodeling through modulation of urokinasetype plasminogen activator (uPA) and MMP2/MMP9 expression in human PDLMSCs at mRNA, protein and activity levels using by RT-PCR, Western blotting and zymography, respectively. Investigation of the involvement of MAPKs in these processes in PDLMSCs was determined by Western blotting, as well as by utilizing specific p38 and MEK1/2 inhibitors. Our results show that IL-17 activates MAPK signaling in PDLMSCs. Moreover, IL-17 had no effect on MMP9 expression, but it stimulated uPA and MMP2 gene and protein expression in PDLMSCs through the activation of the ERK1/2 MAPK signaling pathway. The obtained data suggest that IL-17 contributes to ECM degradation in the periodontal ligament by stimulating uPA and MMP2 expression and activity in PDLMSCs. This information is important for understanding periodontal disease development and defining future directions of its treatment

    Neutrophil Death in Myeloproliferative Neoplasms: Shedding More Light on Neutrophils as a Pathogenic Link to Chronic Inflammation

    Neutrophils are an essential component of the innate immune response, but their prolonged activation can lead to chronic inflammation. Consequently, neutrophil homeostasis is tightly regulated through balance between granulopoiesis and clearance of dying cells. The bone marrow is both a site of neutrophil production and the place they return to and die. Myeloproliferative neoplasms (MPN) are clonal hematopoietic disorders characterized by the mutations in three types of molecular markers, with emphasis on Janus kinase 2 gene mutation (JAK2V617F). The MPN bone marrow stem cell niche is a site of chronic inflammation, with commonly increased cells of myeloid lineage, including neutrophils. The MPN neutrophils are characterized by the upregulation of JAK target genes. Additionally, MPN neutrophils display malignant nature, they are in a state of activation, and with deregulated apoptotic machinery. In other words, neutrophils deserve to be placed in the midst of major events in MPN. Our crucial interest in this review is better understanding of how neutrophils die in MPN mirrored by defects in apoptosis and to what possible extent they can contribute to MPN pathophysiology. We tend to expect that reduced neutrophil apoptosis will establish a pathogenic link to chronic inflammation in MPN

    Knowledge, Attitudes, and Practices on Tick-Borne Encephalitis Virus and Tick-Borne Diseases within Professionally Tick-Exposed Persons, Health Care Workers, and General Population in Serbia: A Questionnaire-Based Study

    This study assessed the level of knowledge, attitudes, and practices (KAP) regarding tick-borne encephalitis virus (TBEV) and tick-borne diseases (TBDs) among different groups of people in Serbia. Professionally tick-exposed persons (PTEPs), health care workers (HCWs), and the general population (GP) were subjected to an anonymous, voluntary, online questionnaire using Microsoft Forms. A total of 663 questionnaire responses were collected (February–March 2021), while 642 were included in the analysis. The significant difference in knowledge in TBDs existed between GP and PTEPs, and HCWs (p < 0.001). The perception of risk-to-tick exposure and TBDs was generally high (42.4 (95% CI: 33.6–51.2) within GP, 44.9 (95% CI: 35.8–53.9) within PTEPs and 46.2 (95% CI: 38.0–54.5) within HCWs), while fear was low (13.7 (95% CI: 7.9–19.5) within GP, 12.6 (95% CI: 7.3–19.9) within PTEPs, and 13.5 (95% CI: 7.4–19.5) within HCWs). Protective practices differed across groups (F (2639) = 12.920, p < 0.001, η2 = 0.039), with both PTEPs (t = 3.621, Cohen d = 0.332, p < 0.001) and HCWs (t = 4.644, Cohen d = 0.468, p < 0.001) adhering to more protective practices than the GP, without differences between PTEPs and HCWs (t = 1.256, Cohen d = 0.137, p = 0.421). Further education about TBDs in Serbia is required and critical points were identified in this study

    Distinct fatty acid redistribution and textural changes in the brain tissue upon the static magnetic field exposure

    We observed different outcomes upon the subacute exposure to the 128 mT highly homogeneous static magnetic field (SMF) when its orientation was (i) aligned with the vertical component of the geomagnetic field; (ii) in the opposite direction. We employed the fatty acids (FA) composition and digital image analyses (DIA) to provide insights into the underlying processes and examine the possible weak SMF effects. Swiss-Webster male mice were whole-body exposed for 1 h/day over five days. Brain tissue’s thin liquid chromatography resulted in brain FA composition, indicating a possible sequence of changes due to the SMF exposure. Quantitative DIA accurately assessed different image parameters. Delicate textural changes were revealed in the group where pathohistological or biochemical alterations have not been detected. DIA-based biological markers seem to be very promising for studying delicate tissue changes, which results from the high sensitivity and wide availability of DIA

    Effects of constant anodal tDCS, oscillatory tDCS, and tACS on short-term associative memory

    Different transcranial electric stimulation (tES) techniques showed promise for noninvasive neuromodulation of memory functions. However, previous studies yielded inconsistent effects, while only a few studies directly compared the effects of different types of stimulation. Thus, here we aim to directly contrast three types of tES in the enhancement of short-term associative memory (STAM), namely, constant anodal transcranial direct current stimulation (tDCS), oscillatory transcranial current stimulation (otDCS), and transcranial alternating current stimulation (tACS). Forty healthy right-handed volunteers (25 females, M = 25.15 ± 3.66 years) participated in the cross-over sham-controlled experiment. Participants underwent four experimental sessions in counterbalanced order (at least 7 days apart) in which they received: tDCS (1.5mA), otDCS at individual theta frequency (ITF, 4-8Hz) (1.5mA ± 0.5mA), tACS at ITF (0±1mA), or sham over the left posterior parietal cortex (P3, 10-20 International EEG system). The current was applied for 20min with 30s ramp-up and ramp-down periods. In the sham condition, the current was applied only at the beginning and at the end in a 30s rump up/down fashion. STAM was assessed during the stimulation, 3-4 minutes after the onset of the protocol. The STAM task was designed in four parallel forms and consisted of digit-color association sequences in which single-digits (0-9) were presented sequentially on the cards of different colors. Participants were instructed to try to remember each digit-color association. The sequence length varied between 3 (low-demand) to 5 stimuli (high-demand) and the sequences were presented in pre-randomized order. At the end of each sequence, the participants were presented with a cue i.e., one of the previously seen color cards and they needed to recall the digit that was presented on a given card. Results showed that relative to sham tDCS improved STAM in low- [F(1,39) = 7.39, p = .010, ηp2 = .16] as well as high-demand sequences [F(1,39) = 4.71, p = .036, ηp2 = .11], while otDCS [F(1,39) = 4.79, p = .035, ηp2 = .11] and tACS [F(1,39) = 4.42, p = .042, ηp2 = .10] exclusively improved performance for high-demand sequences. The results indicate that different stimulation protocols potentially facilitate different cognitive processes. Namely, the effects of constant tDCS on STAM appear to be mediated by boosting the lower-level attention processes while the oscillatory protocols seem to directly promote associative binding

    Suplementacija magnezijuma i status gvožđa kod studentkinja - studija intervencije

    Abstract Background: Literature data indicate the benefit of magnesium (Mg) supplementation. The aim of this study was to examine the effect of short-term Mg supplementation on iron status in healthy female participants. Methods: One hundred healthy female students of the University of Belgrade - Faculty of Pharmacy participated the study during eleven intervention days. Students ingested Mg preparations with the same dose of the active substance. The analysis included the measurement of serum iron, unsaturated iron binding capacity (UIBC), total iron binding capacity (TIBC), total Mg (tMg), ionized Mg (iMg), complete blood count, met-, carboxy- and oxy-haemoglobin (metHgb, COHgb, O2Hgb). Transferrin concentrations and percentage of transferrin saturation (SAT) were calculated manually. The association among the analyzed biochemical parameters was examined using polynomial regression. A principal component analysis (PCA) was used for the evaluation of interdependence between the analyzed parameters. Results: A statistically significant trend for change in O2Hgb (%) by tertiles of iMg concentrations was found (P = 0.029). Serum tMg reached significant positive correlation with the SAT at concentration levels greater than 0.9 mmol/L, after 11 days of intervention (R2=0.116). Ionized Mg in a concentration higher than 0.6 mmol/L is positively correlated with SAT and serum Fe (R2=0.214; 0.199, respectively). PCA revealed variability of 64.7% for two axes after 11 days. Conclusions: Mg supplementation leads to an improvement in the certain iron status parameters even in individuals with optimal levels of these indices. However, caution should be exercised when supplementing Mg, and laboratory monitoring of the interaction is required.Uvod: Literaturni podaci ukazuju na benefit suplementacije magnezijumom (Mg). Cilj ove studije bio je da se ispita uticaj kratkotrajne suplementacije Mg na status gvožđa kod zdravih žena. Metode: Sto zdravih studentkinja Univerzitet u Beogradu - Farmaceutskog fakulteta je učestvovalo u istraživanju tokom jedanaest dana intervencije. Studenti su uzimali preparate Mg sa istom dozom aktivne supstance. U serumu je određivano gvožđe, nezasićen kapacitet vezivanja gvožđa (UIBC), ukupan kapacitet vezivanja gvožđa (TIBC), ukupan Mg (tMg), jonizovni Mg (iMg), kompletna krvna slika, met-, karboksiioksihemoglobin (metHgb, COHgb, O2Hgb). Transferin i saturacija transferina (SAT) su izračunati ručno. Povezanost analiziranih biohemijskih parametara je ispitana pomoću polinomalne regresije. Za procenu međuzavisnosti između analiziranih parametara korišćena je analiza glavnih komponenti (PCA). Rezultati: Utvrđen je statistički značajan trend promene O2Hgb (%) po tertilima koncentracija iMg (P = 0,029). Ukupan Mg je dostigao značajnu pozitivnu korelaciju sa SAT pri koncentracijama većim od 0,9 mmol/L, nakon 11 dana intervencije (R2 = 0,116). Jonizovani Mg u koncentraciji većoj od 0,6 mmol/L pozitivno korelira sa SAT i gvožđem (R2 = 0,214; 0,199, redom). PCA analizom je pokazana varijabilnost od 64,7% za dve ose nakon 11 dana. Zaključak: Suplementacija Mg dovodi do poboljšanja određenih parametara statusa gvožđa čak i kod pojedinaca sa optimalnim nivoima ovih parametara. Međutim treba biti oprezan pri suplementaciji Mg, a dodatno je neophodno i laboratorijsko praćenje ovih interakcija

    Lactate content, total polyphenols and antioxidant activity of selected commercial yogurts from the Serbian market

    One of the most popular dairy products worldwide is yogurt, which flavor is highly influenced by lactic acid content. Yogurts with fruit preparations contain polyphenols, secondary plant metabolites with proven antioxidant properties. As there is no data regarding neither lactate nor polyphenol content in commercial yogurts in Serbia, this study aimed to determine lactate and polyphenol levels, as well as antioxidant activity in selected yogurts. A total of 15 plain and 5 fruit yogurts with strawberry preparations were analyzed. Lactate contents were from 0.83 to 1.33%. Three plain yogurts in plastic containers differed in lactate content. There was no difference in lactate content among the same plain yogurts packed in plastic and Tetra Pack containers. Fruit yogurts with strawberry preparations differed in total polyphenol content in the range from 6.84 to 29.11mg GAE/100 g and antioxidant properties were determined by reducing power test (from 0.22 to 0.79) and DPPH assay (from 28.13 to 87.23%), while there was no difference regarding lactate. Our results provided new information about the levels of lactate, total polyphenols and antioxidant activity of selected commercial yogurts available on the Serbian market
