    Glucocorticoid-catecholamine interplay within the composite thymopoietic regulatory network

    This paper highlights the multiple putative thymic and extrathymic points of intersection and interaction between glucocorticoids (GCs) and catecholamines (CAs)-the end-point mediators of the major routes of communication between the brain and the immune system-in the context of intricate thymic T cell-developmental tuning. More specifically, we discuss in detail findings indicating that adrenal GCs can influence thymopoiesis by adjusting directly and/or indirectly (through modulation of pituitary and local ACTH synthesis) not only thymic GC synthesis, in a cell type-specific manner, but also thymic CA bioavailability (via altering CA outflow from sympathetic nerve endings and local CA synthesis), beta and alpha(1)-adrenoceptor (AR) expression, and/or AR-mediated intracellular signal transduction in thymic cells. In addition, this short review points to GC-and CA-sensitive stages along the multistep T cell-developmental journey and the possible effects of altered GC, and consequently CA signaling, on thymopoietic efficiency

    Molecular characterization of Pseudomonas syringae isolates from fruit trees and raspberry in Serbia

    Infection of fruit trees by Pseudomonas syringae is a potentially serious problem that may limit the establishment and sustained productivity of pome and stone fruit orchards in Serbia. To estimate possible diversity of Pseudomonas syringae fruit trees strains, we collected a set of strains in several areas of Serbia. The samples were taken from infected orchards with raspberry, plum, cherry, sour cherry, peach, pear and apple trees. Genetic diversity of P. syringae strains isolated from fruit trees was determined by using SpeI macrorestriction analysis of genomic DNAs by pulsed-field gel electrophoresis (PFGE) and REP-PCR. Molecular analysis showed that most of isolates had unique profiles, with the exception of isolates from plum and cherry that displayed profiles identical to each other and similar to P. syringae pv. morsprunorum. The study presented here clearly demonstrates the discriminative power of molecular techniques in enabling a detailed analysis of the genetic variations between strains of P. syringae from different pome and stone fruit hosts in Serbia

    It is widely accepted that the progressive decline in the efficiency of thymopoiesis, starting at puberty, is causally related to increasing concentrations of gonadal hormones. In addition, thymic level of catecholamines, which act, via β- adrenoceptors, as tonic inhibitors of thymopoiesis, increases around puberty. There is a pile of evidence that mature immune cells (T-lymphocytes, macrophages and dendritic cells) as well as different types of epithelial cells, synthesize catecholamines. Furthermore, it has been shown that in various tissues male gonadal hormones affect catecholamine synthesis and β-adrenoceptor expression. These findings imposed the question whether thymocytes and/or thymic stromal cells synthesize catecholamines, and, if so, whether testicular hormones influence the synthesis of catecholamines and/or expression of β2-adrenoceptors, and possibly, consequently, the modulatory action of catecholamines on thymopoiesis. Considering all the aforementioned, the dissertation was aimed to: 1) explore whether thymocytes and/or thymic stromal cells in adult male rats synthesize catecholamines, and to define the types of thymic cells expressing β2-adrenoceptor; 2) examine whether orchidectomy at the age of 30 days, when rat thymus is the most sensitive to testicular hormone deprivation, affects catecholamine levels and β2- adrenoceptor expression in the thymus of adult rats, and, if so, 3) determine whether the modulatory effects of catecholamines on thymopoiesis differ in orchidectomized and non-orchidectomized animals. It has been found that thymocytes, thymic epithelial cells and macrophages in adult rats contain and synthesize catecholamines. In addition, it was shown that subsets of these cells also express the β2-adrenoceptors. Orchidectomy at the age of 30 days decreased the concentration of noradrenaline in the thymus of adult animals by diminishing the density of noradrenergic nerve fibers and catecholamine-containing thymic stromal cells and their noradrenaline content...Danas je široko prihvaćeno mišljenje da je progresivno smanjenje efikasnosti timopoeze, koje počinje u pubertetu, kauzalno povezano sa porastom koncentracije polnih hormona. Pored toga, i koncentracija kateholamina, toničnih inhibitora timopoeze posredstvom β-adrenergičih receptora, se u timusu povećava, počevši od pubertetnog perioda. Poznato je da zrele ćelije imunskog sistema (T-limfociti, makrofage i dendritske ćelije), i različiti tipovi epitelnih ćelija, imaju sposobnost sinteze kateholamina. Konačno, u različitim tkivima, muški polni hormoni utiču na sintezu kateholamina i ispoljavanje β-adrenergičkih receptora. Ova saznanja su nužno nametnula pitanje da li timociti i/ili ćelije strome timusa poseduju sposobnost sinteze kateholamina, i, ukoliko je odgovor potvrdan, da li i u kojim tipovima ćelija timusa hormoni testisa utiču na sintezu kateholamina i/ili ispoljavanje β2-adrenergičkog receptora i time, moguće, na modulatorno delovanje kateholamina na proces timopoeze. Imajući u vidu sve prethodno navedene nalaze, postavljeni su ciljevi ove disertacije: 1) da se utvrdi da li timociti i/ili ćelije strome timusa odraslih mužjaka pacova sintetišu kateholamine, i da se precizno definišu tipovi ćelija u ovom organu koji ispoljavaju β2-adrenergički receptor, 2) da se ispita da li orhidektomija u uzrastu od 30 dana, kada je timus pacova najosetljiviji na efekte kastracije, utiče na koncentraciju kateholamina i ispoljavanje β2-adrenergičkog receptora u timusu odraslih pacova, i, ako je odgovor potvrdan, 3) da se utvrdi da li se modulatorno delovanje kateholamina na timopoezu razlikuje kod orhidektomisanih i neorhidektomisanih životinja. Nađeno je da timociti, timusne epitelne ćelije i makrofage sadrže i sintetišu kateholamine. Osim toga, subpopulacije ovih tipova ćelija, takođe, ispoljavaju β2- adrenergički receptor. Orhidektomija u uzrastu od 30 dana je smanjila koncentraciju noradrenalina u timusu odraslih životinja, usled smanjenja gustine noradrenergičkih nervnih vlakana i ćelija strome timusa koje sadrže kateholamine i sadržaja noradrenalina u njima. Pored toga, u timusu ovih životinja je detektovano i smanjenje ekspresije iRNK za β2-adrenergički receptor, kao i površinske gustine ovog receptora na membrani timocita..

    Rana alergijska reakcija na metilprednizolon sa tolerancijom drugih kortikosteroida

    Introduction. In spite of the wide usage of corticosteroids for the treatment of a plethora of diseases, sometimes they can induce immediate hypersensitivity reactions, which are however uncommon. Case Outline. We report a case of immediate allergic reaction induced by intravenous methylprednisolone given before operation for surgical repair of an arm contracture as a sequel of burns, which the child had tolerated a month before. Six weeks later the patient repeated the anaphylactic reaction during skin testing to methylprednisolone. In addition, basophile activation test with methylprednisolone (BAT) was positive. Conclusion. This case report describes a patient who experienced intraoperative anaphylaxis and anaphylactic reaction induced by skin testing. This is the first report on induction of both anaphylactic reactions by methylprednisolone in the same child. Clinical findings, positive BAT and positive skin tests with methylprednisolone imply that the child developed type-I hypersensitivity. The lack of cross-reactivity with other corticosteroids emphasizes that the reactions were caused by the steroid molecule.Uvod. Uprkos širokoj primeni kortikosteroida u lečenju od različitih bolesti, oni ponekad mogu izazvati ranu alergijsku reakciju. Prikaz bolesnika. Kod dvanaestogodišnjeg dečaka došlo je do rane alergijske reakcije izazvane intravenskom primenom metilprednizolona neposredno pre hirurške intervencije, tačnije, korekcije kontrakture šake koja se javila kao komplikacija opekotine. Mesec dana pre pojave alergijske reakcije dete je primalo metilprednizolon i dobro ga podnosilo. Šest nedelja posle operacije ponovo se javila anafilaktička reakcija tokom kožnog testiranja metilprednizolonom. Primenjen je i test aktivacije bazofila (BAT) ovim lekom, čiji je nalaz bio pozitivan. Zaključak. Ovo je prvi prikaz dve vrste anafilaktičke reakcije izazvane metilprednizolonom kod iste osobe. Klinička slika, pozitivni nalaz BAT i pozitivne kožne probe na metilprednizolon pokazuju da se kod deteta razvio prvi tip hipersenzitivne reakcije. Nedostatak unakrsne reaktivnosti s ostalim kortikosteroidima ukazuje na to da je alergijska reakcija izazvana steroidnim molekulom

    Molecular and immunological characterization of Mus a 5 allergen from banana fruit

    Scope Banana fruit has become an important cause of fruit allergy in the recent years. Among the five registered IUIS allergens, Mus a 1 and Mus a 2 have been characterized in detail. In this study, molecular characterization and evaluation of the allergenic properties of beta-1,3-glucanase from banana (Musa acuminata), denoted as Mus a 5, were performed Methods and results: The gene of Mus a 5 was cloned and sequenced. The obtained cDNA revealed a novel Mus a 5 isoform with an open reading frame encoding a protein of 340 amino acids comprising a putative signal peptide of 28 amino acid residues. By MALDI-TOF analysis Mus a 5 isolated from banana fruit revealed a molecular mass of 33 451 +/- 67 Da. Two Mus a 5 isoforms (pI 7.7 and 8.0) were detected by 2D immunoblot with an identical N-terminal sequence. By mass fingerprint, 76 and 83% of the primary structure was confirmed for the two mature Mus a 5 isoforms, respectively. IgE reactivity to Mus a 5 was found in 74% of patients sensitized to banana fruit. Upregulation of basophil activation markers CD63 and CD203c was achieved with Mus a 5 in a concentration-dependent manner. Conclusion: Mus a 5 is a functional allergen and a candidate for the component-resolved allergy diagnosis of banana allergy

    Antimicrobial Activity of Bacillus sp Natural Isolates and Their Potential Use in the Biocontrol of Phytopathogenic Bacteria

    Screening of 203 Bacillus sp. natural isolates for antimicrobial activity against phytopathogenic bacteria showed that 127 tested strains inhibit at least one sensitive strain, which illustrates their potential use as biocontrol agents. Among them, 104 isolates showed significant antagonism against Xanthomonas oryzae pv. oryzae, and only one of these (VPS50.2) synthesizes bacteriocin. An additional screening tested whether 51 isolates contained genes involved in the biosynthesis of lipopeptides of the iturin and surfactin classes. Results show that 33 isolates harbour the operon for iturin biosynthesis, and six of them carry the sfp gene, responsible for the biosynthesis of surfactin. Lipopeptide purification from the supernatant of isolate SS12.9 (identified as B. subtilis or B. amyloliquefaciens) was performed using ethyl acetate extraction, ultrafiltration and reversed phase HPLC. Mass spectrometry analysis confirmed that isolate SS12.9 produces a substance of the iturin class with potential for biocontrol of X. oryzae pv. oryzae

    Examination of antimicrobial potential in natural isolates of lactobacillus casei/paracasei group

    Cilj ove studije je izučavanje antimikrobnog potencijala 52 prirodna izolata vrste L. casei/paracasei. Učestalost gena koji kodiraju BacSJ (bacSJ2-8/bacSJ2-8i genski klaster), acidocin 8912 (acdT), ABC-transporter (abcT) i pomoćni protein (acc) su takođe izučavani. Genski klaster bacSJ2-8/bacSJ2-8i prisutan je kod 49 (94.23%), a acdT kod 41 (78.85%) od 52 testirana soja. Četrdeset sojeva (76.92%) poseduje oba analizirana gena. Interesantno je da samo 17 sojeva (32.69%) koji poseduju bacSJ2-8/bacSJ2-8i genski klaster i/ili acdT gen proizvode bakteriocine. Soj L. paracasei BGNK1-62 poseduje bacSJ2-8/bacSJ2-8i genski klaster, ali ne proizvodi bakteriocin BacSJ što je verovatno posledica nedostatka abcT i acc gena. Nakon transformacije soja BGNK1-62 konstruktom pA2A koji poseduje abcT i acc gene ostvarena je proizvodnja bakteriocina BacSJ. Osim toga, utvrđeno je da soj L. paracasei BGGR2-66 proizvodi nov bakteriocin označen kao BacGR, koji je biohemijski okarakterisan, a određena je i njegova N-terminalna sekvenca.The aim of this study was to investigate the antimicrobial potential of 52 natural isolates of Lactobacillus casei/paracasei. The incidence of relevant genes encoding BacSJ (bacSJ2-8/bacSJ2-8i gene cluster), acidocin 8912 (acdT), ABC-transporter (abcT) and accessory protein (acc) was also studied. These genes were found to be widespread amongst the analyzed L. casei/paracasei strains. The bacSJ2-8/bacSJ2-8i gene cluster was present in 49 (94.23%) and acdT in 41 (78.85%) of the 52 tested strains. Forty of these strains (76.92%) harbored both analyzed genes. Interestingly, only 17 strains (32.69%) with the bacSJ2-8/bacSJ2-8i gene cluster and/or the acdT gene showed bacteriocin production. Strain L. paracasei BGNK1-62 contained the bacSJ2-8/bacSJ2-8i gene cluster, but did not produce bacteriocin BacSJ possibly due to absence of the abcT and acc genes. Hence, these genes were introduced into BGNK1-62 by transformation with constructed plasmid pA2A, after which BacSJ was produced. In addition, it was found that L. paracasei BGGR2-66 produced new bacteriocin designated as BacGR that was biochemically characterized and its N- terminal sequence was determined

    Studija prevalencije antitela u Vojvodini (Srbija) nakon pandemije gripa A-(H1N1)v 2009. godine

    Introduction. The seroprevalence study was performed in Vojvodina during May and June 2010 in order to asses the effects of the 2009 pandemic influenza A(H1N1)v epidemic on herd immunity. It was a part of the Serbian Ministry of Health funded nationwide study. Objective. Prevalence of antibodies against 2009 pandemic influenza A(H1N1)v was determined in a 1% sample of the population monitored for influenza-like illness and acute respiratory infections in Vojvodina through sentinel surveillance system. Methods. The study sample involved a total of 1004 inhabitants of Vojvodina. The control group consisted of randomly selected and age-adjusted 1054 sera collected in the pre-pandemic period. Sera were tested by the reaction of hemagglutination inhibition using influenza A/California/7/2009 (H1N1) antigen in dilution from 1:8 to 1:256. Antibody titers ≥1:32 and ≥1:8 were considered protective and diagnostic, respectively. Results. The differences between control and study sera in all age groups were significant for both diagnostic ≥1/8 and protective titres ≥1/32 of hemagglutination inhibition antibodies (chi square test, p lt 0.001). The highest percentage of seropositive subjects was registered in the age group 15-19 years followed by children aged 5-14 years. Both diagnostic and protective titres were about twice higher in the vaccinated as compared to the non-vaccinated group. There were no statistically significant differences in seroprevalence between seven districts of Vojvodina. Conclusion. The 2009 pandemic influenza A(H1N1)v epidemic significantly influenced the herd immunity in our population regardless of low immunization coverage with highest immunity levels in adolescents aged 15-19 years and with similar herd immunity levels in all the regions in the province six months after the outbreak.Uvod. Studija prevalencije antitela izvedena je u Vojvodini tokom maja i juna 2010. godine, kako bi se procenili efekti epidemije izazvane pandemijskim virusom gripa A­(H1N1)v iz 2009. godine na imunitet stanovništva. Studija je bila sastavni deo nacionalne studije koju je finansiralo Ministarstvo zdravlja Republike Srbije. Cilj rada. Prevalencija antitela protiv pandemijskog virusa gripa A­(H1N1)v je utvrđivana na jednoprocentnom uzorku populacije praćene sentinelnim nadzorom nad oboljenjima sličnim gripu i akutnim respiratornim infekcijama u Vojvodini radi procene imuniteta stanovništva Vojvodine. Metode rada. Ispitivanjem su obuhvaćena 1.004 stanovnika Vojvodine čiji serum je dat na analizu (studijska grupa). Kontrolnu grupu činio je uzorak seruma iz prepandemijskog perioda 1.054 nasumično odabrane osobe sličnog uzrasta. Serološko ispitivanje vršeno je reakcijom inhibicije hemaglutinacije antigenom virusa gripa A/Kalifornija/7/2009 (H1N1). Serumi su testirani u razblaženju od 1:8 do 1:256. Titar antitela u razblaženju većem od 1:32 smatrao se zaštitnim titrom, a u razblaženju većem od 1:8 dijagnostičkim. Rezultati. Utvrđena je visoko statistički značajna razlika (p lt 0,001) u prevalenciji antitela između studijske i kontrolne grupe, kako u dijagnostičkom (≥1:8), tako i u zaštitnom titru (≥1:32) hemaglutinin-inhibirajućih antitela. Najveći procenat seropozitivnih ispitanika otkriven je u dobnoj grupi 15–19 godina, a zatim u grupi 5–14 godina. Prevalencija antitela i u dijagnostičkom i u zaštitnom titru bila je dva puta veća kod vakcinisanih osoba u odnosu na nevakcinisane. Nije utvrđena statistički značajna razlika u seroprevalenciji između pojedinih okruga Vojvodine. Zaključak. Utvrđen je visok kolektivni imunitet, bez teritorijalnih razlika, prema pandemijskom virusu gripa A­(H1N1)v iz 2009. godine uprkos slabom obuhvatu stanovništva imunizacijom.     Identification and molecular characterization of Chryseobacterium vrystaatense ST1 isolated from oligomineral water of southeast Serbia

    The isolation and molecular characterization of bacterial strains isolated from water sources in the Vlasina Mountain in southeast Serbia, confirmed the presence of a new species Chryseobacterium vrystaatense ST1. This Gram- negative species showed an extremely low level of biochemical reactivity in biochemical tests. The gene for 16S rRNA was amplified by PCR using universal primers and sequenced. Comparison of 16S rRNA gene sequence and phenotypic features indicated that the isolate ST belonged to Chryseobacterium vrystaatense. A BLAST search of sequenced 1088 nucleotides of the 16S rRNA gene with all sequences deposited in the NCBI collection showed the highest similarity (98%) with the strain Chryseobacterium vrystaatense sp. nov., designated as strain R-23533. The very high homology of these two strains allowed classification of our strain at the species level, but some differences indicate, and indirectly confirm, that the isolate ST is an authentic representative. On the basis of these results, we could conclude that Chryseobacterium vrystaatense ST was for first time isolated in Serbia, which is particularly important when one bears in mind that there are only three sequences of this species deposited in the NCBI collection
