    Seroepizootiološko-epidemiološka istraživanja i mapiranje infekcije Zapadnog Nila u Republici Srbiji

    The disease caused by the West Nile virus (WNV) has been known since 1937 when it was described for the first time in Uganda. After spreading to Europe and the Middle East, the disease has changed its primary location. Today WN infection is a significant health problem in the world. As a result of the current epizootiological and epidemiological situation in Europe studies on the occurrence of WND were introduced in Serbia, also. The investigation on the presence of WNV antibodies was intensified in the period from 2008 to 2012. In this period a total of 3618 serum samples were tested from 18 localities (2736 animal sera from 8 different species and 882 human sera samples). The method applied was gel immunodiffusion and the representative samples were confirmed by the plaque reduction test (PRNT-90). Out of the total number of samples WNV antibodies were present in 3.97% horses, 0.93% dogs, 0.31% poultry and 1.36% man. In one horse serum sample there was a positive reaction with a positive control serum, thus indicating the presence of WNV antigen. The results have confirmed that WNV antibodies are present in 9 out of 18 tested locations in the Republic of Serbia. The percentage of seropositive samples varies from 0.42% in Pozarevac (horses and humans) up to 6.45% in Novi Pazar (dogs). Out of the investigated species the highest seropositivity was recorded in horses (3.97%), and lowest in poultry (0.31%). WNV is present and widespread in the Republic of Serbia, thus enabling distribution mapping.Bolest koju izaziva virus Zapadnog Nila (WNV) je poznata još od 1937. kada je po prvi put opisana u Ugandi. Posle širenja virusa u Evropu i na Bliski istok, bolest je promenila prvobitnu lokaciju. Danas je WN infekcija značajan zdravstveni problem u svetu. Kao rezultat trenutne epizootiološko-epidemiološke situacije u Evropi, uvedena su istraživanja prisustva WNV i u Srbiji. Istraživanja prisustva antitela za WNV intenzivirana su u periodu od 2008. do 2012. godine. U ovom periodu ukupno je ispitano 3618 seruma sa 18 lokaliteta (2736 seruma životinja, od 8 različitih vrsta i 882 seruma ljudi). Korišćen je metod imunodifuzije u gelu a reprezentativni uzorak potvrđen je testom neutralizacije plaka (PRNT- 90). Od ukupnog broja uzoraka na prisustvo antitela za WNV bilo je pozitivno 3,97% konja, 0,93% pasa, 0,31% domaće živine i 1,36% ljudi. U serumu jednog konja pojavila se pozitivna reakcija sa pozitivnim kontrolnim serumom, ukazujući na prisustvo antigena WNV. Dobijeni rezultati su potvrdili da su antitela za WNV prisutna u 9 od 18 ispitivanih lokacija u Republici Srbiji. Procenat seropozitivnih varira od 0.42% u Požarevcu (konji i ljudi) do 6,45% u Novom Pazaru (psi). Od ispitivanih vrsta najveća seropozitivnost je registrovana kod konja (3,97%), a najniža kod živine (0,31%). Raspoloživi podaci ukazuju da je WNV prisutan i značajno raširen u Republici Srbiji, što je omogućilo i mapiranje njegovog širenja

    Common ragweed (Ambrosia atremisiifolia) is one of the most frequent causes of pollen-induced allergic reactions both in humans and dogs. It has not been defined yet, what is the major allergen(s) to which most dogs allergic to ragweed show a positive result on intradermal skin test (IDST). In the present study sensitization to Ambrosia artemisiifolia pollen allergens in dogs with atopic dermatitis was examined with both in vivo and in vitro tests, including IDST and serum allergen specific IgE test. Detection of specific-IgE antibodies against ragweed allergens by immunoblotting in the sera of allergic dogs was optimized, as well. Dogs that were positive, as judged by IDST reactions to ragweed pollen allergens, also had alergen specific IgE antibodies in their sera. Results indicate that major allergens of A. artemisifolia pollen in dogs are Amb a 1 and Amb a 2. Further characterization of ragweed allergens is needed before they could potentially be used in intradermal testing or allergen immunotherapy in affected dogs. Also, we evaluated new Favrots diagnostic criteria for canine atopic dermatitis in dogs allergic to Ambrosia atremisiifolia pollen. It might be concluded that proposed criteria are of great assistance for seting up suspected diagnosis of canine atopic dermatitis, after ruling out other pruritic dermatoses.Kratka ambrozija (Ambrosia artemisiifolia) je jedan od najčešćih uzročnika alergijskih reakcija izazvanih polenom kod ljudi i pasa. Još uvek nije definisano koji je glavni alergen (i), na koji, većina pasa alergičnih na polen ambrozije, ispoljava pozitivnu reakciju na intradermalnom testu. U ovoj studiji je ispitana senzibilizacija na polen ove biljke kod pasa sa simptomima atopijskog dermatitisa in vivo i in vitro testovima, uključujući intradermalni test i dokazivanje prisustva alergen specifičnih IgE antitela u serumu. Optimizovani su uslovi za detekciju IgE specifičnih antitela iz seruma pasa alergičnih na polen ambrozije imunoblot tehnikom. Psi koji su imali pozitivnu reakciju na polen ove biljke na intradermalnom testu, takođe su imali specifična IgE antitela u serumu. Dobijeni rezultati ukazuju da su glavni alergeni Ambrosia artemisiifolia kod pasa Amb a 1 i Amb a 2. Neophodna je dalja karakterizacija alergena ambrozije kako bi se oni mogli primeniti pri rutinskom intradermalnom testiranju ili u alergen specifičnoj imunoterapiji obolelih pasa. Takođe je razmatrana i validnost Favrotovih dijagnostičkih kriterijuma kod pasa alergičnih na polen ambrozije. Može se zaključiti da su predloženi kriterijumi od velike pomoći u postavljanju suspektne dijagnoze atopijskog dermatitisa pasa, nakon isključenja drugih pruritičnih dermatoza

    Epidemija pandemijskog gripa H1N1 u Vojnoj gimnaziji

    Background/Aim. The first cases of the pandemic pH1N1 influenza virus infection was observed in the United States and Mexico in April 2009 and the first laboratory confirmed case in Serbia was registered in June 2009. The aim of this paper was to report on the investigation of the first confirmed outbreak of the 2009 pandemic H1N1 influenza in Serbia and to describe the clinical and epidemiologic findings from this investigation. Methods. Descriptive and analytical epidemiological methods were used. Data were collected from medical records of the Military School students and epidemiological questionnaire. Pandemic H1N1 infection was initially confirmed by the RT-PCR assay in nasopharyngeal and oropharyngeal swabs and subsequently by the complement fixation test in serum samples. Results. The attack rate of acute respiratory illness was 70.8% (204/288). Pandemic H1N1 virus infection was confirmed in 44 of 82 tested cases of acute respiratory illness (53.7%) The most common clinical manifestations of pandemic influenza H1N1 were fever (88.6%), cough (61.4%), malaise (38.6%), runny nose (36.4%), headache (29.6%), sore throat (20.5%) and muscle pain (15.9%). Conclusion. The findings from this investigation suggest that pandemic H1N1 influenza in a high military school was widespread but did not cause severe illness.Uvod/Cilj. Prvi slučajevi pandemijske infekcije virusom influence pH1N1 ustanovljeni su u Americi i Meksiku u aprilu 2009. a u Srbiji prvi laboratorijski potvrđen slučaj registrovan je u junu 2009. godine. Cilj rada bio je da se prikaže istraživanje prve dokazane epidemije pandemijske influence H1N1 u Srbiji i opišu klinički i epidemiološki nalazi iz ovog istraživanja. Metode. Primenjen je deskriptivni i analitički epidemiološki metod. Izvor podataka bila je medicinska dokumentacija učenika Vojne gimnazije i epidemiološki upitnik. Infekcija pandemijskim virusom H1N1 prvo je dokazana pomoću RT-PCR u nazofaringealnim i orofaringealnim brisevima, a zatim i reakcijom vezivanja komlementa u uzorcima seruma. Rezultati. Stopa javljanja akutnog respiratornog oboljenja iznosila je 70.8% (204/288). Infekcija pandemijskim H1N1 virusom potvrđena je kod 44 od 82 testiranih slučajeva akutnog respiratornog oboljenja (53,7%). Najčešće kliničke manifestacije pandemijske influence H1N1 bile su povišena temperatura (88,6%), kašalj (61,4%), malaksalost (38,6%), rinitis (36,4%), glavobolja (29,6%), gušobolja (20,5%) i bol u mišićima (15,9%). Zaključak. Rezultati ovog istraživanja ukazuju da je pandemijska influenca H1N1 bila raširena u Vojnoj gimnaziji, ali da nije izazvala teške forme oboljenja

    Immunoproteomic characterization of Ambrosia artemisiifolia pollen allergens in canine atopic dermatitis

    Canine atopic dermatitis (CAD) is an immune system disorder that affects 10-15% of the canine population. Short ragweed (Ambrosia artemisiifolia) pollen represents one of the major seasonal sources of allergenic pollen proteins in Europe, particularly in the Pannonian valley of the Balkan region. In Serbia, about 66% of atopic dogs showed a positive intradermal skin test with its pollen extract, which is second to house dust mites. Therefore, characterization of Ambrosia artemisiifolia pollen components, in terms of defining major and minor allergens that induce clinically manifested allergic reaction in dogs, is important for valid diagnosis and efficient therapy. This study has, for the first time, characterized and identified major Ambrosia artemisiifolia allergens in CAD, using an immunoproteomic approach. To assess the prevalence of specific IgE in electrophoretically separated ragweed pollen proteins, individual reactivity of sera from dogs with CAD was analyzed and compared to the reactivity of sera from healthy dogs in the non-reducing conditions, which were found optimal for specific canine IgE detection. A specific IgE band (38 kDa) was recognized as the most dominant allergen in CAD, occurring in 81% of positive dog's sera. 2-D immunoblotting followed by a mass spectrometry peptide fingerprint analyses with pooled canine and human atopic sera, revealed that 38 kDa major Ambrosia atremisiifolia allergens in CAD were all five isoallergens of the Amb a 1 group (antigen E), including the previously named Amb a 2 (antigen K). In contrast to canine sera, human atopic sera also recognized lower mass allergens such as the 13 fragment of Amb a 1 and profilins (Amb a 8 variants). The most prominent ragweed proteins in CAD, represent, as in humans, variants of all five isoallergens of the Amb a 1 group (pectate lyase): Amb a 1.0101 and its natural variant E1XUL2, Amb a 1.0202, 1.0304, 1.0402 and the natural variant of Amb a 1.0501, E1XUM0, as well as the a fragment of pollen allergen Amb a 1.0201.     In Vitro and In Vivo Uptake Study of Escherichia coli Nissle 1917 Bacterial Ghosts: Cell-Based Delivery System to Target Ocular Surface Diseases

    PURPOSE. For the successful topical administration of drugs or vaccines to treat ocular surface diseases, efficient and well-tolerated delivery systems/carriers for conjunctival delivery are crucial in the development of new treatment strategies. The present study investigated the efficiency of internalization of bacterial ghosts (BGs) produced from probiotic Escherichia coli Nissle 1917 (EcN) by human conjunctival epithelial (HCjE) cell line, the EcN BGs cytotoxicity for HCjE cells, and in vivo uptake of EcN BGs by conjunctival guinea pig epithelial cells. METHODS. The uptake of EcN BGs by HCjE cells was analyzed by laser scanning microscopy and flow cytometry. Immunohistochemistry was used to localize the EcN BGs in the guinea pig conjunctival tissue. Cytotoxicity of EcN BGs on HCjE cells was evaluated by measurement of LDH. RESULTS. Laser scanning microscopy and flow cytometry revealed that EcN BGs internalization by HCjE cells was time-and dose dependent. No cytotoxic effect on HCjE cells was observed after EcN BGs inoculation for 30 and 120 minutes, as well as 24 hours. In addition, the uptake of EcN BGs was detected in the conjunctival cells after in vivo administration of EcN BGs into the eye of the guinea pig. CONCLUSIONS. The findings that EcN BGs are nontoxic and effectively internalized in vitro by human and in vivo by guinea pig conjunctival cells comprise an important contribution to the future use of BGs as a system for conjunctival delivery of drugs and vaccines, either to treat or prevent ocular surface diseases

    The influence of aging and estradiol to progesterone ratio on rat macrophage phenotypic profile and NO and TNF-alpha production

    The phenotype and function of tissue macrophages substantially depend on the cellular milieu and biological effector molecules, such as steroid hormones, to which they are exposed. Furthermore, in female rats, aging is associated with the altered macrophage functioning and the increased estrogen level is followed by a decrease in that of progesterone. Therefore, the present study aimed to investigate the influence of estradiol/progesterone balance on rat macrophage function and phenotype throughout whole adult lifespan. We ovariectomized rats at the late prepubertal age or at the very end of reproductive lifespan, and examined the expression of ED2 (CD163, a marker of mature resident macrophages related to secretion of inflammatory mediators) on peritoneal macrophages and their ability to produce TNF-alpha and NO upon LPS-stimulation at different age points. In addition, to delineate direct and indirect effects of estrogen, we assessed the in vitro influence of different concentrations of 17 beta-estradiol on LPS-induced macrophage TNF-alpha and NO production. Results showed that: ( a) the low frequency of ED2(high) cells amongst peritoneal macrophages of aged rats was accompanied with the reduced TNF-alpha, but not NO production; (b) estradiol level gradually increased following ovariectomy; (c) macrophage ED2 expression and TNF-alpha production were dependent on estradiol/progesterone balance and they changed in the same direction; (d) changes in estradiol/progesterone balance differentially affected macrophages TNF-alpha and NO production; and (e) estradiol exerted pro-inflammatory and anti-inflammatory effects on macrophages in vivo and in vitro, respectively. Overall, our study discloses that estradiol/progesterone balance contributes to the fine-tuning of rat macrophage secretory capacity, and adds to a better understanding of the ovarian steroid hormone role in the regulation of macrophage function, and its significance for the age-associated changes in innate immunity.     Kiwifruit (Actinidia deliciosa) extract shows potential as a low-cost and efficient milk-clotting agent

    Actinidin, a cysteine protease accounting for more than 50% of the soluble proteins in kiwifruit pulp, has shown promise as a milk-clotting agent. In this study, the potential use of kiwifruit pulp extract as a clotting agent was investigated. It was shown that three kiwifruit extracts made from the pulp of a single fruit have significantly different levels of active actinidin, depending on the extraction buffer employed. Kiwifruit extract prepared at pH 5.0 had the best milk-clotting properties, with a nearly 30% better ratio of clotting activity to proteolytic activity than purified actinidin. This extract produced a casein coagulum clearly separated from the whey proteins, and was shown to be stable at room temperature for up to two months. This extract has the potential to be employed as an efficient and low-cost milk-clotting agent in the production of dairy products

Second, longevity statistics will be reviewed: Why do the most scientifically advanced countries have such low (comparatively) life expectancies? Scientific knowledge expands exponentially each decade, whereas there have been no significant advances in our knowledge, government, economics, politics, anti-corruption, and so forth since the dawn of history. What can we expect in the future? Will the human species outlive the cockroach? Can we expect to get closer to that theoretical asymptote of 120 years of human life? Will this ceiling ever be lifted?     The effect of single-pulse transcranial magnetic stimulation and peripheral nerve stimulation on complexity of EMG signal: fractal analysis

    The aim of this study was to examine whether single-pulse transcranial magnetic stimulation (spTMS) affects the pattern of corticospinal activity once voluntary drive has been restored after spTMS-induced EMG silence. We used fractal dimension (FD) to explore the 'complexity' of the electromyography (EMG) signal, and median frequency of the spectra (MDF) to examine changes in EMG spectral characteristics. FD and MDF of the raw EMG epochs immediately before were compared with those obtained from epochs after the EMG silence. Changes in FD and MDF after spTMS were examined with three levels of muscle contraction corresponding to weak (20-40 %), moderate (40-60 %) and strong (60-80 % of maximal voluntary contraction) and three intensities of stimulation set at 10, 20 and 30 % above the resting motor threshold. FD was calculated using the Higuchi fractal dimension algorithm. Finally, to discern the origin of FD changes between the CNS and muscle, we compared the effects of spTMS with the effects of peripheral nerve stimulation (PNS) on FD and MDF. The results show that spTMS induced significant decrease in both FD and MDF of EMG signal after stimulation. PNS did not have any significant effects on FD nor MDF. Changes in TMS intensity did not have any significant effect on FD or MDF after stimulation nor had the strength of muscle contraction. However, increase in contraction strength decreased FD before stimulation but only between weak and moderate contraction. The results suggest that the effects of spTMS on corticospinal activity, underlying voluntary motor output, outlast the TMS stimulus. It appears that the complexity of the EMG signal is reduced after spTMS, suggesting that TMS alters the dynamics of the ongoing corticospinal activity most likely temporarily synchronizing the neural network activity. Further studies are needed to confirm whether observed changes after TMS occur at the cortical level
