Evaluation of pseudouridine monophosphate glycosidase and snR10c candida albicans mutants
Candida albicans is an opportunistic human fungal pathogen capable of causing both superficial
and disseminated infections. C. albicans bloodstream infections continue to have high mortality rates
despite existing antifungal treatments. Therefore, it is important to continually work toward finding new
drug targets. snR10c is a C. albicans specific H/ACA box snoRNA involved in the conversion of uridine
into pseudouridine, the most abundant RNA modification. We isolated 4 mutants of snR10c that altered
the binding pocket residues. We found two of four mutant isolates also contained an additional
mutation. Isolate 1, which was heterozygous for this additional mutation had severe growth defects
under temperature- and drug-induced stress. Isolate 1 also had filamentation defects and had
significantly attenuated virulence compared to wildtype. This could suggest that snR10c also has role
outside of pseudouridylation. The study also set out to investigate pseudouridine degradation, a
metabolic process that does not occur in humans, but is predicted to occur in C. albicans. The second
aim of this study was to determine if knockout of Pug1, Tna12, and/or Gap2 changes C. albicans
virulence in the Galleria mellonella model system. We found minor growth and filamentation defects in
the strains pug1, pug1tna12, and pug1gap2. I also found tna12 had increased virulence compared to
wildtype. Our data suggests that targeting pseudouridine degradation alone may not be a suitable
therapeutic target. The third aim of this study was to design a PUG1-TurboID construct for downstream
use in identifying protein-protein interactions with Pug1. This construct has been designed and will be
integrated into the C. albicans genome to identify novel interactors. My data addresses under described
aspects of RNA metabolism in C. albicans and has identified likely distinctions between C. albicans RNA
metabolism and that of mammals.M. S
The effect of professor feedback emails on student classroom participation
This study looks at how the tone and structure of professor feedback emails affect students’
motivation and classroom engagement, using reactance theory and self-determination theory
(SDT) to explain their responses. We hypothesized that disrespectful language and disorganized
instructions, referred to as “hassle,” would trigger psychological reactance, a motivational
response to perceived threats to autonomy, leading to negative perceptions and reduced
willingness to participate. A sample of 148 college students responded to vignettes that varied in
the level of respect conveyed in the email’s tone and the clarity of the instructions. A 2 × 2
ANOVA revealed significant main effects for respect, with students who received low-respect
messages perceiving bias and reporting lower intentions to engage in class. Hassle also
influenced student responses, particularly when respect was high. These findings suggest that
subtle features of communication can activate reactance and shape students’ motivation. We
interpret these results through both reactance theory and self-determination theory, highlighting
how threats to autonomy and unmet needs for respect and competence may reduce student
engagement.M. S
Now what am I supposed to do with these?
Many works of this show stem back to the inspiration from 1800’s murano glass blowers like Toso and Salviati. Artists like Matt Urban and Jeff Mack are also heavily inspired by these techniques and traditions while being able to put modern twists on it and pass the knowledge down to others. During the height of Murano glass blowing in the 1800-1900 many of the items being made were not being used. Often made to show off or prove technical skills many of the items lacked function. Understanding why these items differ from mass manufactured counterparts dials down to understanding the history and process of these items. After understanding the process and traditions of the items it should change the perception of them when looking at the treatment by not only the average consumer but museums and collections. All of my process for this show revolves around furnace worked glass and venetian techniques of assembly through the process of furnace worked glass.B.F.A.School of Ar
An intersectional examination of cognitive test anxiety
Cognitive test anxiety (CTA) is a pervasive condition, that threatens students' learning experiences and academic success, and it increases as students matriculate in higher education (Cassady, 2004a; Cassady & Johnson, 2002; Putwain, 2007; Thomas et al., 2017b). The current study focused on the potential differences in reported levels of CTA and GPA based on race and gender. The goal was to understand how group identity played a role in the pervasiveness of cognitive test anxiety, and to explore how that may be related to academic achievement. Using Control-Value Theory as the cause of cognitive test anxiety, stereotype threat was also used to explain potential secondary causes of underperformance. Results showed significant differences in reported CTA and GPA between gender and race.M. S.M. S
Industrial renewal
With continuous changes in the global economy, buildings once vital in our
industrial economy have become obsolete. The Damen Ave site was essential
to Chicago’s history for grain storage and transportation industrialization.
Changes in society and economic movement require rapid adaptation, forcing
many historic or iconic industrial sites to be demolished or forgotten. This thesis
will explore rehabilitation as a design method for the adaptive reuse of iconic
industrial landmarks. The site on the northeast corner of 29th Street and Demon
Avenue will be a new opportunity to pull many culturally different communities
together. While each building will serve a specific purpose, the overall focus of
the site in total will be on the adaptation of the historic grain silos into residential
housing as well as a community music venue. The residential silo form was
redeveloped to minimize building disturbance and significant demolition. In
contrast, the performance center features intricate perforations to create a
continuous open volume. The silo’s curvilinear forms repeat throughout the
site in newly designed areas emphasizing their importance and unique forms.
Reclaimed steel from existing building structures and machinery will emphasize
thoroughfares and points of gathering, creating moments to reflect on the
historical and industrial significance of the site.Thesis (B. Arch.)College of Architecture and Plannin
I am my own art collection
In this paper, I talk about my influences and significances through my art career and the way they have driven me creatively through the semester. I touch on subjects including my past experiences and emotions, major events that have impacted my life, pieces of both illustrated and animated media that I have taken inspiration from regarding style and color, as well as digging a little into color psychology to explain color choices I made when choosing inks to use in my works. I go on further to explain the significance of sharing my personal experiences and what I want to show off to the audience and those who view my work, sending out a message that there are moments that help make up who I am as a person and why I strive to be an artist. I then talk about future plans and reflect back on my series and my work as an artist and what is next for me moving forwards. My exhibition statement comes up next followed by my exhibition list that showcases the collection and titles of the six pieces I created.B. A.School of Ar
Should we return to normal? A phenomenological study of the COVID-19 experiences of student affairs employees and their impact on job satisfaction and role retention
This dissertation includes a phenomenological study of the experiences of student affairs
professionals in non-faculty roles during the COVID-19 pandemic. Utilizing semi-structured
interviews, this study gathers information on the experiences of these individuals and establishes
common themes present in the data. The data are used to answer the research question of how the
lived experiences of these professionals during the COVID-19 pandemic influenced their overall
career trajectory. The results show that, while COVID-19 was overall a negative experience,
there were some positives that contributed to increased work/life balance and the creation of
professional boundaries for student affairs professionals. However, this is not true for those
working in housing and residence life during the pandemic. Their experiences were widely
negative and differed from those of all other participants.D. Ed
Mundane monuments
In every city, many cheaply built, small-scale commercial buildings were
constructed for a singular purpose and now sit vacant or underutilized. They
either fade into the background of our minds or stick out like a sore thumb. How
can the transformation of everyday architecture re-establish the ordinary as a
significant community asset and inspire future utilization of the mundane? After
a defined inventory of commonplace architecture, I selected six underutilized
sites in Anderson, Indiana, whose exterior character, site conditions, and use
uniquely reflect the city’s dialect. Located off major roads throughout Anderson,
each host’s previous function was preserved; however, an addition to each
building defines a new architectural language and allows for programmatic
diversification throughout the city. By extracting and exaggerating aspects of the
host building, this thesis explores how everyday architecture’s existing program,
formal language, material, or site situation can inform a new elevated presence.
A new identifiable architectural language embedded in the mundane buildings
of a city elevates the quality of place while acknowledging a consistent narrative
and systematic approach to the transformations. This thesis demonstrates
that the ordinary qualities of a community’s previous existence can influence a
newfound purpose and meaning.Thesis (B. Arch.)College of Architecture and Plannin
Traditional Chinese medicine (TCM) psychotherapy in contemporary China: a qualitative study of TCM practitioners' perspectives
Traditional Chinese medicine (TCM) is a distinct system with a profound history and a comprehensive scholarship. TCM posits a different understanding of the human body, mind, and mental health disorders compared to Western medicine. Research is limited on the application of TCM to mental health treatment, and few studies have examined TCM practitioners’ views about using TCM to treat mental health disorders. With the rise of Western psychotherapy in China, the resulting need for indigenizing Western theories and practices, and the emergence of the discipline of TCM psychology, interest has grown in exploring TCM when treating Chinese individuals with mental health disorders. Fourteen TCM practitioners in mainland China completed semi-structured interviews focused on their conceptualization and treatment of mental health disorders. A team of six coders used thematic analysis to code the interview data, generating 14 themes. Results suggested the interviewees employed knowledge and theories from TCM and multiple other disciplines to understand the mental health disorders of their Chinese clients. Findings also indicated that, when working with their clients, interviewees generated diverse and individualized treatment plans depending on their clients’ presenting concerns, applied a wide range of interventions, and weighed various factors (e.g., their time and capacity to assist patients, patients’ needs and preferences, and advantages and disadvantages of various treatment approaches) in the provision of their services. Interviewees’ understanding and treatment of mental health disorders reflected medical pluralism, revealed crucial issues in the current therapeutic milieu, and unveiled a host of sociocultural changes in contemporary Chinese society. Results have implications for developing indigenous therapeutic approaches and culturally relevant treatment programs in China, training TCM practitioners, and creating and implementing health care policies in contemporary China. Additionally, findings have implications for further research strategies about TCM psychotherapy and integrative medicine. Strengths and limitations of the study are articulated.Ph.D
Change or die
Change or Die is a feminist's perspective on women's rights and abortion rights. Drawing from my own lived experiences and inspirations from other female figures in my life, I created this series of oil paintings to confront the audience with motifs regarding female genitalia and references to past political movements. Creating abstractions and ambiguity within symbols of fertility and natural elements to cultivate her message. A woman's control over her body is essential to her fundamental rights and freedoms.B.F.A.School of Ar
