    Parentage Assignment in Striped Catfish (Pangasianodon hypophthalmus) Based on Microsatellite Marke

    In this study, five microsatellite markers from two multiplex PCR sets were used to determine the pedigree of five striped catfish (Pangasianodon hypophthalmus) families that came from a breeding program in Vietnam. The number of alleles per locus and polymorphic information content (PIC) were 4 to 7 and 0.551-0.803, respectively. In the parentage analysis based on five microsatellites, 62.7% of offspring could be allocated unambiguously to their parental pairs using the perfect mismatch setting, and assignment rate improved to 89.3% after a single mismatch was allowed. Parentage analysis when only four microsatellites were used (a locus with a high frequency of null alleles was excluded) increased the assignment rate to 68% for a perfect mismatch and to 90% for assignment with a single mismatch. Results here confirm that a DNA markerbased approach to parentage assignment in P. hypophthalmus will be useful in genetic improvement programs for this important aquaculture species in Vietnam.

    Comparative Analysis of Immune Function of Three Wild Mandarin Fish Populations in Yuanjiang River Basin

    Mandarin fish Siniperca chuatsi (Basilewsky) is a unique species in East Asia and has very important ecological and economic value. There are significant differences in immune function between different populations of Mandarin fish. However, up to now, the comparative study of immune indices of different populations of wild mandarin fish has been scarce. In the present study, the transcription level of Immunoglobulin M (IgM) and blood serum immunologically related enzyme activity of three wild Mandarin fish populations from Wuqiangxi reservoir, Lingjintan reservoir, and Taoyuan reservoir of Yuanjiang river basin were detected. In head kidney, spleen and in the liver of Mandarin fish, the transcription level of IgM was highest in Taoyuan reservoir followed by Lingjintan reservoir, and was lowest in Wuqiangxi reservoir. The catalase activity of Mandarin fish was highest in Taoyuan reservoir followed by Wuqiangxi reservoir, and was lowest in Lingjintan reservoir. Total number of white blood cells, lysozyme activity, acid phosphatase activity, superoxide dismutase (SOD) activity, and alkaline phosphatase activity of Mandarin fish were in line with immunoglobulin M gene expression, which were highest in Taoyuan reservoir followed by Lingjintan reservoir, and lowest in Wuqiangxi reservoir. This indicated that the immune function of Mandarin fish from Taoyuan reservoir was strongest, and these fish had the greatest resistance to harmful invaders such as parasite, bacteria, and virus compared to Lingjintan reservoir group and Wuqiangxi reservoir group. The results of this study provide a theoretical basis for disease-resistant breeding and germplasm optimization in mandarin fish breeding production, and may be valuable in future researc

    Dietary Iron Requirements of Adult GIFT Tilapia (Oreochromis niloticus)

    Dietary iron requirement of adult Nile tilapia (Oreochromis niloticus), genetically improved farmed tilapia (GIFT strain), was determined in this study through a 77 day feeding trial. Diets with six concentrations of iron (45.55 - the control group, 94.91, 193.62, 292.34, 391.05, and 489.77 mg iron/kg diet) from ferrous sulfate were formulated and hand-fed to adult GIFT (initial body weight 174.58±3.49g). The results indicated that no fish died in all groups. Weight gain ratio, feed efficiency rate, as well as specific growth rate decreased in relation to increasing dietary iron levels, and reached the lowest when supplemented with 489.77 mg iron/kg diet. Crude body fat content showed an increasing trend in relation to increasing dietary iron levels. The iron content in the body, vertebrae, and liver significantly increased with dietary iron levels which reached up to 391.05 mg iron/kg diet. The number of red blood cells, the hemoglobin content and the packed cell volume was highest in 94.91 mg iron/kg group. In conclusion, dietary iron made no obvious improvement on growth performance, but improved hematopoietic function of adult Nile tilapia, reared in freshwater. We found that no additional iron needs to be added to the adult Nile tilapia (174.6–558.1 g) feed based on the growth, whereas broken-line regression analysis were used to determine the optimum iron requirements for maximum hemoglobin levels, the dietary iron requirement for hemoglobin was 120.94 mg iron/kg diet with ferrous sulfate as a source of iron
