    A Mutant in the ADH1 Gene of Chlamydomonas reinhardtii Elicits Metabolic Restructuring during Anaerobiosis

    Embargado hasta 01/01/2100The green alga Chlamydomonas reinhardtii has numerous genes encoding enzymes that function in fermentative pathways. Among these, the bifunctional alcohol/acetaldehyde dehydrogenase (ADH1), highly homologous to the Escherichia coli AdhE enzyme, is proposed to be a key component of fermentative metabolism. To investigate the physiological role of ADH1 in dark anoxic metabolism, a Chlamydomonas adh1 mutant was generated. We detected no ethanol synthesis in this mutant when it was placed under anoxia; the two other ADH homologs encoded on the Chlamydomonas genome do not appear to participate in ethanol production under our experimental conditions. Pyruvate formate lyase, acetate kinase, and hydrogenase protein levels were similar in wild-type cells and the adh1 mutant, while the mutant had significantly more pyruvate:ferredoxin oxidoreductase. Furthermore, a marked change in metabolite levels (in addition to ethanol) synthesized by the mutant under anoxic conditions was observed; formate levels were reduced, acetate levels were elevated, and the production of CO2 was significantly reduced, but fermentative H2 production was unchanged relative to wild-type cells. Of particular interest is the finding that the mutant accumulates high levels of extracellular glycerol, which requires NADH as a substrate for its synthesis. Lactate production is also increased slightly in the mutant relative to the control strain. These findings demonstrate a restructuring of fermentative metabolism in the adh1 mutant in a way that sustains the recycling (oxidation) of NADH and the survival of the mutant (similar to wild-type cell survival) during dark anoxic growt

    Embargado hasta 01/01/2100.El centro educativo es un lugar de aprendizaje, en el que interaccionan muchas personas y en el que surgen muchos conflictos. Estos se dan continuamente en el aula, por lo que hay que aprender a resolverlos. Así, la elección este trabajo ha venido motivado por las carencias detectadas en el alumnado. Este programa está dirigido a los alumnos de un centro de E.P.O. de Linares (Jaén), que carecen de pautas y normas de disciplina en sus hogares. Viven en un entorno sociocultural empobrecido y la inmensa mayoría de ellos pertenecen a familias desestructuradas, sin un buen referente a seguir. Sus comportamientos disruptivos no tienen consecuencias negativas en sus casas., que les permitan modular sus conductas, las cuales se ven reforzadas en muchas ocasiones. Los alumnos necesitan formarse a nivel de valores, competencias emocionales, prevención de dificultades conductuales y desarrollo de cualidades mediadoras, este aprendizaje los acompañará y será parte de sus vidas. Conforme los alumnos vayan promocionando de curso, el centro podrá contar, a largo plazo, un alumnado formado en conocimientos, destrezas, pautas y estrategias que puedan emplear en función de las necesidades que se les presenten, por lo que el número de conflictos y dificultades en el centro se reducirán. Si los alumnos desarrollan los aspectos marcados anteriormente, serán capaces de gestionar los conflictos y ayudarse a sí mismos y a los demás, serán futuras personas con una serie de destrezas sociales, capacidad de autocontrol, autonomía, autoestima, autoconocimiento y empatía hacia los demás, lo que repercutirá en la mejora de la convivencia a nivel social, y por lo tanto, una mejora a nivel cívico y moral de nuestra sociedad

Se proponen medidas para mejorar el marco institucional y jurídico, para adecuar mejor la representación y voluntad de las unidades político-territoriales y para clarificar el controvertido esquema de distribución de competencias. Papel destacado es el que corresponde a una reforma integral del Senado para convertirlo en auténtica cámara de representación territorial y de representación de las comunidades autónomas como entes con autonomía política y como partes integrantes de la voluntad general

    Labor law, economics and regulation Italy and Spain: comparing models in the European framework

    Embargado hasta 01/01/2100.The work here presented wishes to propose a critical and reasoned reflection about the relationship between labor law and economics in a continuously changing international scenario. The theme can certainly be inserted among the “classic” ones, because it faces one of the most fascinating issues labor law – as a subject – places when it projects itself outside its natural conceptual perimeter. The research object is in fact based on a critical reflection around the vexata quaestio of the relationship between the juridical-labor law sphere and the economic dimension of reference. More in detail, in this general framework, we carry out a research which thrusts down its roots in an organic analysis of the theoretical positions of law and labor economics, in order to develop a synergic argument which can possibly be advantageous in both research ambits. With the present work, we wish therefore to test the holding of the relationship between the two spheres of knowledge considered, also in the perspective of the elaboration of hermeneutic contributions useful for a possible re-conceptualization of labor law, partially imposed by the morphological change of the socio-economic contexts of reference. Following a logical sequence, the present work is structured in five conceptually autonomous chapters, which however permeate each other and are conceived in a indissoluble unitary dimension in order to guarantee systematic coherence to the research. In detail, moving from a careful reflection about the “crisis” of labor law considered by itself and in its interaction with economics, attention is placed on the intrinsically conflictual and dualistic nature of the subject, in its being a projection of the pair “capital/work”. After some unavoidable methodological considerations, useful for an analytical-conceptual reflection, we then highlight the elements of interest deriving from the comparison between and the balancing of economic and social rationality, economic factuality and juridical “evaluation”. From a methodological point of view, the logical and scientific assumption of the research is the firm conviction that only through a systemic approach, characterized by a strong comparative and multidisciplinary framework, it is possible to carefully analyze the current structure and configuration of the relationship between labor law and economics in order to outline in particular the boundaries of future perspectives of development. A clear reconstruction of a suitable method to rationalize the dialectic process between cognitive openness and juridical reconceptualization is indeed inescapable. The use of the comparison – contextualized and teleologically addressed to give the work an appreciable hermeneutic contribution – is thus considered the privileged, functional research method. The labor law systems compared – as shown in the body of the present work – are those of Italy and Spain, because of the proximity of the regulatory paths explored from time to time and of the convergence resulting also from recent reforms. Hence, looking at the paradigmatic institutes of the impact of the economic sphere on juridical regulations, the concrete relationship between economics and labor law is in particular considered with specific reference to the worker’s tasks (and demotion/deskilling), also as a consequence of the recent legislative reforms, which have been introduced in the two legal systems object of comparison. In addition, special consideration is reserved to the concept of “flexibility”, to the specular notion of “security”, and to the boundaries of the concept of “flexicurity”, in the scenario of a European labor law undergoing a deep change in the search of a possible new balance. Exactly the search of a new adjustment between the different interests involved in present-day labor law relationships finds in an axiological framework of values the natural landing of the research path here briefly presented. In the conclusions of the present work, we propose some targeted reflections about the urgent need to “return” to the principles and the values which have represented the essence of the subject, yesterday as today. Labor regulation, in relation to economic efficiency and to the requests of deregulation coming from the market, cannot leave aside the rediscovery of the table of values of reference and the balancing of the different interests involved

    Impact of Energy Storage Systems Value-Added Options

    Embargado hasta 01/01/2100.Thermal Energy Storage (TES) technologies are becoming important due to their significance in the implementation of renewable energies in buildings and in industry. They also increase the energy efficiency of our systems, thus, contributing to the decrease of CO2 emissions and climate change mitigation. Furthermore, the assessment and the optimization of TES technologies within the environmental and energy systems, in proper applications, bridge the gap and drive the transition towards sustainable goals. This chapter starts with an Introduction to the TES technologies, in Section 1, and the fundamentals of the Thermal Energy Storage and the cycling process, in Section 2. Then it provides insightful information from the three kinds of TES technologies, namely the sensible, latent and thermo-chemical, in Section 3, 4 and 5 respectively. In each category, the analysis of the critical mechanisms is followed by a description of role of the materials and the interaction of the key properties in various applications. A brief conclusion with the state of the art advances is presented at the end of each section. Section 6 is dedicated in hybrids applications, where two or more kinds of technologies are combined in order to enhance the output, while Section 7 depicts the role of assessment with regards to the choice of technology. Section 8 concludes with a summary of the findings and investigates further research development

    Plain Legal Language as accessibility tool for translators: an empirical study at a phraseological level

Likewise, for any legal translator, understanding legal language will be a crucial first step in overcoming any communication barrier. In light of this consideration, the purpose of this study is to compare the original Charter of the United Nations with the plain-language version adapted to the same language, paying particular attention to differences in phraseology. By conducting this study, we will have an opportunity to examine the linguistic simplification strategies applied by this international organization, taking into consideration the factors highlighted by plain language as a means of making legal language more communicatively accessible

    Familias ON. Gestión responsable de los dispositivos móviles en el hogar

    Embargado hasta 01/01/2100La digitalización del hogar reta a las familias a gestionar el consumo interno de contenidos audiovisuales, pues cada vez más los menores usan “dispositivos móviles con acceso a Internet fuera del control de los padres o adultos de referencia” (López-Sánchez y García del Castillo, 2017, p.108). Las investigaciones han puesto sobre la mesa los riesgos derivados del uso de Smartphones y Tablets por parte de los menores. Algunos de estos riesgos son la posibilidad de geolocalización, recepción masiva de publicidad de diferente contenido, introducción de códigos maliciosos y acceso al control del dispositivo, vulneración de la privacidad, phubbing, grooming, sexting, ciberbullying, extorsión con fotografías o material gráfico de los menores, acceso de pederastas a la vida del niño o la niña, abuso de menores, consumo de pornografía, desarrollo de comportamientos adictivos asociados al uso problemático de estos dispositivos, al tiempo medio de conexión a los dispositivos en sí mismos, a las redes sociales y a Internet, entre otros. El reto no es afrontado por igual en cada familia, las características del modelo familiar y las competencias de los padres y madres provocan que los resultados de la gestión que se realiza en el hogar sean totalmente diferentes de unas familias a otras. Aquellas familias que han percibido la importancia de sus funciones como padres y madres y su repercusión en el desarrollo de los menores se encontrarán en mejor disposición para dar respuesta a las exigencias de la sociedad digital. Sin embargo, existen otras familias que requieren de una “ayuda” suplementaria en el ejercicio de su parentalidad

    Anaesthetic and cardiorespiratory effects of a constant rate infusion of fentanyl in isoflurane-anaesthetized sheep

To determine the anaesthetic and cardiorespiratory effects of a constant rate infusion of fentanyl in sheep anaesthetized with isoflurane and undergoing orthopaedic surgery. Study design. Prospective, randomised, ‘blinded’ controlled study. Animals. Twenty healthy sheep (weight mean 41.1 ± SD 4.5 kg). Methods. Sheep were sedated with intravenous (IV) dexmedetomidine (4 μg kg−1) and morphine (0.2 mg kg−1). Anaesthesia was induced with propofol (1 mg kg−1 minute−1 to effect IV) and maintained with isoflurane in oxygen and a continuous rate infusion (CRI) of fentanyl 10 μg kg−1 hour−1 (group F) or saline (group P) for 100 minutes. The anaesthetic induction dose of propofol, isoflurane expiratory fraction (Fe’iso) required for maintenance and cardiorespiratory measurements were recorded and blood gases analyzed at predetermined intervals. The quality of recovery was assessed. Results were compared between groups using t-tests or Mann–Whitney as relevant. Results. The propofol induction dose was 4.7 ± 2.4 mg kg−1. Fe’iso was significantly lower (by 22.6%) in group F sheep than group P (p = 0). Cardiac index (mean ± SD mL kg−1 minute−1) was significantly (p = 0.012) lower in group F (90 ± 15) than group P (102 ± 35). Other measured cardiorespiratory parameters did not differ statistically significantly between groups. Recovery times and recovery quality were statistically similar in both groups. Conclusions and clinical relevance. Fentanyl reduced isoflurane requirements without clinically affecting the cardiorespiratory stability or post-operative recovery in anaesthetized sheep undergoing orthopaedic surgery
