Investigation of drug use and exposure to polymer-related chemicals by wastewater and urine analysis
The aim of this thesis was to characterize exposure biomarkers and to determine the human exposure to polymer-related chemicals and drugs of abuse. Three different types of experiments were considered: in-vitro human liver metabolism studies, human biomonitoring through urine analysis, and wastewater-based epidemiology. In-vitro metabolism studies recreated the human body processes to identify potential exposure biomarkers of rubber-related chemicals. Human biomonitoring provided reliable stratified-population data and its relationship with human exposure to bisphenols. Finally, wastewater-based epidemiology allowed to monitor the use of drug of abuse and exposure to bisphenols and plasticizers. This thesis comprised Spanish and Portuguese samples and was driven by liquid chromatography hyphenated to (tandem) mass spectrometry.2025-03-2

Genome Wide Association Studies in Small-Cell Lung Cancer. A Systematic Review
Small cell lung cancer (SCLC) is one of the deadliest forms of lung cancer, but few information exists regarding the role of genetics, particularly on Genome Wide Association Studies (GWAS). The aim of the study is to explore the evidence available obtained through GWAS studies for SCLC using a systematic review. We performed a literature search in the main databases until July 31st, 2023. We included all human based studies on GWAS for lung cancer which presented results for SCLC. Only studies with participants diagnosed of SCLC with anatomopathological confirmation were included. Fourteen studies were identified; 8 studies showed a relationship between ASCL1 overexpression and SCLC, which may regulate CHRNA5/A3/B4 cluster, producing a consequent nAChR overexpression. Nine papers, including 8 of the previous, found a positive association between SNPs located in chromosome 15 and SCLC. The most important cluster of genes found is CHRNA5/A3/B4 but the mechanism for the role of these genes is unclear. Kyoto Encyclopaedia of Genes and Genome (KEGG) shows that these receptors were found to be overexpressed where nicotine, 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) and N’-Nitrosonornicotine (NNN) acts, involving different routes in SCLC carcinogenesisThis work was supported by PI15/01211 – ISCIII – co-financed FEDERS

cunha gran variabilidade espacial.
canto á presenza de contaminantes, analizáronse as
concentracións totais e dispoñibles e a bioaccesibilidade de Cu,
Pb, Zn, Cr e As, así como a presenza de PAHs en
mostras superficiais de 55 solos urbanos. Os resultados obtidos
indican a importancia do estudo dos solos urbanos, pola

Association of periodontitis with cognitive decline and its progression: Contribution of blood‐based biomarkers of Alzheimer's disease to this relationship
Aim
To assess whether periodontitis is associated with cognitive decline and its progression as well as with certain blood-based markers of Alzheimer's disease.
Materials and Methods
Data from a 2-year follow-up prospective cohort study (n = 101) was analysed. Participants with a previous history of hypertension and aged ≥60 years were included in the analysis. All of them received a full-mouth periodontal examination and cognitive function assessments (Addenbrooke's Cognitive Examination (ACE) and Mini-Mental State Examination [MMSE]). Plasma levels of amyloid beta (Aβ)1-40, Aβ1-42, phosphorylated and total Tau (p-Tau and t-Tau) were determined at baseline, 12 and 24 months.
Results
Periodontitis was associated with poor cognitive performance (MMSE: β = −1.5 [0.6]) and progression of cognitive impairment (hazard ratio [HR] = 1.8; 95% confidence interval: 1.0–3.1). Subjects with periodontitis showed greater baseline levels of p-Tau (1.6 [0.7] vs. 1.2 [0.2] pg/mL, p < .001) and Aβ1-40 (242.1 [77.3] vs. 208.2 [73.8] pg/mL, p = .036) compared with those without periodontitis. Concentrations of the latter protein also increased over time only in the periodontitis group (p = .005).
Conclusions
Periodontitis is associated with cognitive decline and its progression in elderly patients with a previous history of hypertension. Overexpression of p-Tau and Aβ1-40 may play a role in this associationThis study was partially supported by grants from the Xunta de Galicia (TS & JC: IN607A2018/3, TS: IN607D 2020/09 and IN607A2022/07), Institute of Health Carlos III (TS: PI22/00938 and CB22/05/00067) and Spanish Ministry of Science (TS: RTI2018-102165-B-I00 and RTC2019-007373-1). Anne Brontë’s The Tenant of Wildfell Hall (1848): an analysis of the novel and of its changing relevance over time
Traballo Fin de Grao en Lingua e Literatura Inglesas. Curso 2022-2023Anne Brontë is a relevant representative of female Victorian Gothic fiction. Her novels, often
overshadowed by her sisters’ success and neglected by critics, convey a strong sense of
morality and a serious approach to real life issues, such as marriage and motherhood. Anne
Brontë’s characterisation of her female protagonists, the strength of their will and their abilityto face the situations they are confronted with, grant Anne’s fiction the title of one of the pioneer’s feminist novels. In The Tenant of Wildfell Hall (1848), her most controversial novel, Anne presents themes and topics that were not only relevant for 19th century life, but also resonate in contemporary times. The aim of this project is to identify the main themes of Anne Brontë’s The Tenant of Wildfell
Hall and situate them in their Victorian context, analyse her treatment of these themes,
compare the reception of the novel at the time of publication and in the 21st century, and
show its current relevance.
The introduction will provide a historical background focusing on the sociocultural
conventions that most affected Anne as a female author. The main body of the project will
contain the analysis of The Tenant of Wildfell Hall: Chapter 1 will deal with the principal
themes addressed by Anne; Chapter 2 will study the critical reception of the novel, and
Chapter 3 will include a contrastive approach of the novel’s impact in the 19th century and
how its relevance has shifted over time. Chapter 4 will examine the current relevance of the
novel. The conclusion will focus on the attainments and resulting ideas of this project
Inhibiting HER3 Hyperphosphorylation in HER2‐Overexpressing Breast Cancer through Multimodal Therapy with Branched Gold Nanoshells
Treatment failure in breast cancers overexpressing human epidermal growth factor receptor 2 (HER2) is associated mainly to the upregulation of human epidermal growth factor receptor 3 (HER3) oncoprotein linked to chemoresitence. Therefore, to increase patient survival, here a multimodal theranostic nanoplatform targeting both HER2 and HER3 is developed. This consists of doxorubicin-loaded branched gold nanoshells functionalized with the near-infrared (NIR) fluorescent dye indocyanine green, a small interfering RNA (siRNA) against HER3, and the HER2-specific antibody Transtuzumab, able to provide a combined therapeutic outcome (chemo- and photothermal activities, RNA silencing, and immune response). In vitro assays in HER2+/HER3+ SKBR-3 breast cancer cells have shown an effective silencing of HER3 by the released siRNA and an inhibition of HER2 oncoproteins provided by Trastuzumab, along with a decrease of the serine/threonine protein kinase Akt (p-AKT) typically associated with cell survival and proliferation, which helps to overcome doxorubicin chemoresistance. Conversely, adding the NIR light therapy, an increment in p-AKT concentration is observed, although HER2/HER3 inhibitions are maintained for 72 h. Finally, in vivo studies in a tumor-bearing mice model display a significant progressively decrease of the tumor volume after nanoparticle administration and subsequent NIR light irradiation, confirming the potential efficacy of the hybrid nanocarrierE.V.-A. and I.G.-C. contributed equally to this work.
