    Stormwater Management Planning for Sustainable and Affordable Housing Development

    Bridgewater Township, in collaboration with the Regional Sustainable Development Partnerships (RSDP) and the University of Minnesota’s St. Anthony Falls Laboratory (SAFL), initiated a stormwater management project to incorporate sustainable stormwater practices into a new 120-unit housing development. The primary objective was to evaluate and recommend stormwater control measures (SCMs) that would effectively manage runoff, improve water quality, and integrate into the community’s development plan. In addition, this project represented an opportunity to incorporate stormwater management at the beginning of the conceptual plan phase as opposed to later stages of final design. Ong's Work and to Selected Related Scholarly Work," I highlight the life and work of the American Jesuit scholar Walter J. Ong (1912-2003; Ph.D. in English, Harvard University, 1955) of Saint Louis University, the Jesuit university in the City of St. Louis, Missouri (USA), where, over the years, i took five courses from him. In the present review essay, I also identify five recurring themes in Ong's work. Then for each of those five themes, I first list certain relevant publications by Ong, chronologically arranged by date of publication, and then I also list selected related scholarly work, alphabetically arranged by the author's last name. In chapter 1, I examine whether reducing the number of firms in public equity markets harms investors, and how much has the value firms can get from going public changed in the past few decades? I develop a dynamic supply and demand model of the firm entry to and exit from public markets to relate firm benefits from being public to firm characteristics. Firms face a dynamic discrete choice problem on whether to be in public markets, with the benefits of being public a function of their characteristics, demand elasticities for their characteristics, and various regulatory and cost of capital changes. My structural analysis allows me to not only break down the causes of the transformation in U.S. public equity markets, but also say what the consequences of them have been for firms and investors. I find that investors would have had slightly higher excess returns but no change in their portfolio's Sharpe ratio if firms behaved as they did before Sarbanes-Oxley. I further find that a private firm's implied option value of going public has fallen by over half since the pre-Sarbanes era. The reduction is mostly caused by an increase to fixed costs of being a public company in the post-Sarbanes era. In the second chapter, I build on traditional models of dynamic oligopoly which assume perfectly frictionless capital markets. This assumption can be relaxed to show that introducing financing constraints can lead to counterintuitive outcomes. When financing constraints are introduced to a standard model of dynamic oligopoly with mergers, the steady state equilibrium probability of monopoly decreases, as opposed to the intuitive expectation that it would increase. This increase is in fact observed in an environment lacking these financial frictions. This result demonstrates that consideration of financing frictions is essential for models of antitrust policy and its effects on welfare outcomes.Stanek, Leo. (2025).     Generating exogenic neural cells through blastocyst complementation for neurodegenerative diseases

Advisor: Walter Low. 1 computer file (PDF); x, 202 pages.Neurodegenerative diseases burden individuals, their families, and the economy. The progressive symptoms and neuropathology affect cognitive skills and daily activities. The only available treatments alleviate symptoms, but do not stop or change the course of disease progression. Therapeutics are rather limited for neurodegenerative diseases. Cellular transplantation to treat neurodegenerative diseases has shown promise as a therapeutic, but a novel source of cells is needed. Fetal/embryonic tissue is fraught with ethical concerns and many stem cell derived cells behave differently than primary cell types. This dissertation seeks to bridge some of these gaps by characterizing and quantifying cell types found within chimeric brains. Insights from this research have the potential to inform neurodegenerative therapeutics, deliver new information of cell survival and differentiation, and elucidate some interspecies chimeric differences. We reviewed the current state of cellular transplantation, relevant therapeutic neuronal and non-neuronal types, interspecies chimeric barriers, intraspecies and interspecies chimeras, and generation of chimeric brains. Our study of E12.5 intraspecies mouse-mouse chimeric brains identified, characterized, and quantified serotonergic, dopaminergic, and cholinergic precursors derived from donor cells. Microglia-like and macrophage-like cells derived from donor cells were also observed and quantified. To better understand development of these cells, E16.5 intraspecies mouse-mouse chimeric brains were generated. To support interspecies chimera characterization, we generated E12.5 interspecies rat-mouse chimeric brains, and began to characterize and quantify cell types. Dopaminergic precursors, microglia-like and macrophage-like cells were identified cells in these interspecies chimeras. To demonstrate that a targeted developmental niche could be made, we developed and tested single guide RNA to target a PU.1 gene that is important for microglia development. This research seeks to improve our understanding of cell types and proportions of those cell types generated in wild type intraspecies and interspecies chimeras. It also highlights new methods, like automated imaging, for quantifying cell types within these chimeric animals. This research also provides the foundational steps for making a new PU.1 knockout model to be used with blastocyst complementation.Strell, Phoebe. (2025).     The fungi and bacteria associated with three tree-killing beetles: from new species to complex communities.

Advisors: Robert Blanchette, Kathryn Bushley. 1 computer file (PDF); xv, 180 pages.Fungi and bacteria can play critical roles in the life cycles of bark and woodboring beetles, influencing tree colonization and, in some cases, negatively impacting tree health. This dissertation investigates the microbial communities of two bark beetles and one woodboring beetle. The eastern larch beetle (Dendroctonus simplex) is a native bark beetle to North America that has undergone an unprecedented outbreak over the past two decades, impacting trees on over 90% of the tamarack (Larix laricina) forests within Minnesota. Prior to this work, its gallery-associated fungal communities were poorly characterized. This study demonstrated that Grosmannia americana is highly associated with the eastern larch beetle galleries in Minnesota. Additionally, four new species of Ophiostomatales are described and a new classification for one of the eastern larch beetle associates is proposed. The red turpentine beetle (Dendroctonus valens) is also a native bark beetle to North America and acts mostly as a secondary pest of pines throughout the continent, rarely killing healthy trees. A community-level analysis of the fungal and bacterial communities associated with the red turpentine beetle adults and their galleries found that red pine (Pinus resinosa) and white pine (P. strobus) host different microbial communities. Additionally, a rich diversity of Ophiostomatales were isolated from red turpentine beetle environments in the Great Lakes region, where Leptographium terebrantis was the most isolated species. Finally, the emerald ash borer (Agrilus planipennis), an invasive woodboring beetle in North America, has caused extensive damage to ash (Fraxinus spp.) over the past two decades, threatening an entire genus of trees. This dissertation studies how the fungal and bacterial communities in ash trees change during an emerald ash borer infestation. As the emerald ash borer attack progressed, fungal species richness declined. Additionally, most of the microbial species found in the emerald ash borer galleries were not detected in the phloem of trees without the emerald ash borer, however, the abundances of the latent microorganisms were higher in the galleries than the non-latent microorganisms. Overall, the findings in this dissertation contribute to the understanding of the fungi and bacteria associated with tree-killing beetles both on a microbial community-level and individual fungal species-level.Mann, Andrew.
