    Avaliação da sensibilidade aos fármacos em tuberculose (método molecular e fenotípico), resultados do PNAEQ (2018-2022)

    A deteção das resistências permite adotar um esquema terapêutico adequado contribuindo desta forma para o aumento do sucesso terapêutico e para a diminuição da mortalidade e das taxas de recidiva. Os métodos moleculares permitem a rápida deteção das mutações mais frequentes que conferem resistências e por isso a sua utilização é recomendada pela Organização Mundial de Saúde (OMS) e pela Direção Geral da Saúde (DGS). O método fenotípico permite confirmar os resultados dos testes moleculares e testar outros fármacos importantes para definir o esquema terapêutico adequado a cada caso. A participação em programas de avaliação externa da qualidade permite aos laboratórios que determinam a sensibilidade aos fármacos em tuberculose avaliarem os seus resultados e, se necessário, corrigirem procedimentos.     Evaluating a training programe: European Health Information Training Programme - EHITP

Disponível em: https://www.gacetasanitaria.org/es-pdf-X0213911123036410?local=trueBackground: The InfAct (Information for Action) is a Joint Action of the European Commission’s 3rd Health Programme with the main goal to build an infrastructure of a health information system for a stronger European Union and to strengthen its core elements. The InfAct Joint Action was developed along 36 months and structured in 10 work packages. Portugal co-led the Work Package 6 (WP6) of this project, which included the development of the proposal of a flagship capacity building programme - the European Health Information Training Programme - and its evaluation. The evaluation objectives included: to evaluate the adequacy of the training programme to the health information needs in the European Member States; to identify possible changes regarding the participants selection process, the training activities and the pedagogical project; and to contribute to the understanding of the potential of the programme to add to available offers in learning on the topics of Public Health information, on the capacity building and behavioural changes in Public Health activities which can be attributed to the course, and of the potential of the programme to contribute to the alignment of health information criteria and procedures between the European Member States. Methods: The evaluation process was developed using an observational descriptive study design using a mixed methodological approach with both document analysis and primary data collected by questionnaires and interviews analysis. Mixed quantitative and qualitative data collection methods and analysis were used. Results The proposal of the European Health Information Training Programme seemed adequate to the formative needs and capacities in line with the work performed by the InfAct project. In what concerns about its main thematic areas, it was also aligned with the areas identified in the previous formative needs and capacities mapping. The participants selection process proposed seemed, in general, adequate. The potential of the European Health Information Training Programme proposal to learning, capacity building and behavioral changes at work attributable to the course was considered positive, as well as the potential to the alignment of health information criteria and procedures between European Union Member States. Discussion In general, we found high consistency between the results obtained from data collected by the techniques used. However, different suggestions for improvement were outlined by the evaluation study population.Project '801553 InfAct' which has received funding from the European Union’s Health Programme (2014-2020). InfAct (Information for Action) is a JA on health information (HI). HI is essential to build up country specific and cross-country knowledge. HI is the basis to inform national health policies, to steer population health investment decisions, to assess the effectiveness of different approaches to enhance population health through promotion, prevention, treatments and care.     Functional studies of APOB variants the experience of the Portuguese Familial Hypercholesterolemia Study

    Familial hypercholesterolemia (FH) is clinically characterized by increased levels of circulating LDL cholesterol leading to premature coronary heart disease. It can be caused by variants in LDLR, APOB, and PCSK9 genes. APOB variants are responsible for 5-10% of the FH cases, p.(Arg3527Gln) being the most common. Only recently the whole gene has been sequenced due to Next Generation Sequencing, increasing the variant spectrum of APOB and with it the number of variants that need to be functionally assessed. We aimed to characterize novel APOB variants identified in patients included in the Portuguese FH Study to confirm if they are the genetic cause of hypercholesterolemia. To better analyze these variants, we also create a database with all APOB rare variants found up to date in the Portuguese FH Study. The functional study of 5 variants is ongoing. To access if these variants affect apoB:LDL receptor binding, LDL from index cases and relatives with the variants was separated using sequential ultracentrifugation, and proliferation assays were performed with U937 cells.     The “5th International Conference on Food Contaminants: Challenges on exposure assessment, health impact and sustainability of food systems”(ICFC2023) took place between 4 and 6th september and was held at the Instituto Agronómico de Campinas, Brazil. This event, under the responsibility of INSA, was held in a hybrid format, in a close collaboration with CESAM, University of Aveiro and the Faculty of Animal Science and Food Engineering, University of S. Paulo, with the support of FAPESP and SCBTA. The event took place for the first time outside Portugal with the motto “Let's Open Horizonts” with the aim of contributing to strengthening collaboration between Portugal and Brazil in the field of food safety and human health. ICFC2023 featured four sessions that included topics such as the occurrence of contaminants, assessment of human exposure and health impact, sustainability of food systems and collaborative networks and their impact on society. These sessions included a plenary lecture by EFSA on food sustainability, food safety and human health, nine presentations by guest speakers from Europe and Brazil, 17 oral communications by participants and two round tables, with three brief communications each.     SRC/RAC1 Pathway and other NIS Plasma Membrane Interactors in NIS Regulation

    The Sodium/Iodide Symporter (NIS) is responsible for the uptake of iodide into thyroid follicular cells, which is required for thyroid hormone production. NIS is present in most differentiated thyroid carcinoma (DTC) cells, allowing radioactive iodine (RAI) to be used to eliminate any residual disease after thyroidectomy. However, in advanced DTC, a significant proportion of metastatic lesions stop taking up iodide and become refractory to RAI therapy. This impairment of iodide uptake is mainly caused by defective functional expression of NIS, due to dysregulated transcriptional and posttranscriptional events. In this presentation, part of a Symposium entitled “Thyroid Cancer Basic: New Molecules for Better Treatments”, I will talk about how we discovered that signaling by the small GTPase RAC1 plays a crucial role in regulating NIS expression, both at the transcriptional and posttranscriptional levels, and how this might impact RAI therapy in advanced DTC.N/

    Free PMC article: https://www.ncbi.nlm.nih.gov/pmc/articles/PMC10716728/Following a request from the European Commission, EFSA was asked to deliver a scientific opinion on the safety and efficacy of zeolites (≥ 50%) obtained from Neapolitan Yellow Tufa as a technological additive (functional group: anticaking) for all animal species. The additive is specified to contain not less than 50% of zeolites, namely phillipsite, chabazite and analcime. Neapolitan Yellow Tufa originates from the volcanic activity of Campi Flegrei, Italy. According to the conventional risk assessment, due to a lack of adequate data, the safety of the additive for the target species cannot be established. Based on current knowledge, there is no indication of substantial absorption of the components of the additive and, therefore, of concern for the consumer. The additive poses a risk by inhalation. It is not irritant to the skin. The Panel cannot conclude on the eye irritancy and on the dermal and respiratory sensitisation potential of the additive. As no suitable data in line with the requirements of the Guidance on risk assessment of nanomaterials were provided by the applicant, the potential risks associated with the presence of nanoparticles for the target species, the consumer and the user could not be assessed. The additive is safe for the environment. The additive is considered to be efficacious in feedingstuffs for all animal species at 20,000 mg/kg complete feed.info:eu-repo/semantics/publishedVersio
