    The Effects of Tetrabromobisphenol A (TBBPA) on the Mussel Mytilus galloprovincialis: A Multi-Biomarker Approach

    Tetrabromobisphenol A (TBBPA) is a fire-retardant containing bromine, produced in large quantities worldwide and extensively used in several industrial products. This compound was identified as a potential contaminant of the environment, causing toxicity to organisms. However, its toxicity remains poorly understood in marine bivalves. The first objective of this work was to evaluate the impact of TBBPA on mussels (Mytilus galloprovincialis) exposed for 28 days to various concentrations of TBBPA (0, 1, 10, and 100 µg·L−1), by assessing stress biomarkers’ responses (Glutathione S-transferase, superoxide dismutase, catalase, lipid peroxidation, total antioxidant capacity, total ubiquitin, caspase-3 and acetylcholinesterase). The results showed that lower concentrations (1 and 10 µg·L−1) were efficiently detoxified, as suggested by GST activities, which were supported by the responses of the other biomarkers. The most pronounced effects were observed in animals exposed to the highest concentration of TBBPA (100 µg·L−1), suggesting oxidative stress. Additionally, significant strong correlations were found between total antioxidant capacity and some biomarkers (superoxide dismutase and lipid peroxidation), showing that processes involved in oxidative stress fighting are working to avoid cell injury. In brief, mussels’ defense mechanisms were capable of dealing with exposure to the lower concentrations tested.     Safety assessment of the substances ‘wax, rice bran, oxidised’ and ‘wax, rice bran, oxidised, calcium salt’ for use in food contact materials

    The EFSA Panel on Food Contact Materials (FCM) assessed the safety of the substances ‘wax, rice bran, oxidised’ and ‘wax, rice bran, oxidised, calcium salt’, used as additives up to 0.3% in polyethylene terephthalate (PET), polyamide (PA), thermoplastic polyurethane (TPU), polylactic acid (PLA) and poly(vinyl chloride) (PVC) in contact with all food types for long‐term storage at room temperature and below, after hot‐fill and/or heating. The substances consist of the chemical classes wax esters, carboxylic acids, alcohols and calcium salts of acids, along with an unidentified organic fraction up to w/w. Migration into 10% ethanol and 4% acetic acid was below 0.012 mg/kg for each chemical class, and about 0.001 mg/kg for the unidentified fraction. In isooctane, migration was up to 0.297 mg/kg food for wax esters, below 0.01 mg/kg food for the other chemical classes and about 0.02 mg/kg food for the unidentified fraction. The contact with dry food and food simulated by 20% ethanol were considered covered by the migration tests with aqueous simulants. Based on genotoxicity assays and compositional analyses, the constituents of the chemical classes did not raise a concern for genotoxicity. The potential migration of individual constituents or groups of chemically‐related compounds of the unidentified fraction would result in exposures below (for aqueous food) and above (for fatty food) the threshold of toxicological concern for genotoxic carcinogens. Therefore, the FCM Panel concluded that the substances are not of safety concern for the consumer, if used as additives up to 0.3% w/w in PET, PLA and rigid PVC materials and articles intended for contact with all food types except for fatty foods, for long‐term storage at room temperature and below, including hot‐fill and/or heating up to 100°C for up to 2 h

    Baseline data and associations between urinary biomarkers of polycyclic aromatic hydrocarbons, blood pressure, hemogram, and lifestyle among wildland firefighters

    Introduction: Available literature has found an association between firefighting and pathologic pathways leading to cardiorespiratory diseases, which have been linked with exposure to polycyclic aromatic hydrocarbons (PAHs). PAHs are highlighted as priority pollutants by the European Human Biomonitoring Initiative in occupational and non-occupational contexts. Methods: This cross-sectional study is the first to simultaneously characterize six creatinine-adjusted PAHs metabolites (OHPAHs) in urine, blood pressure, cardiac frequency, and hemogram parameters among wildland firefighters without occupational exposure to fire emissions (> 7 days), while exploring several variables retrieved via questionnaires. Results: Overall, baseline levels for total OHPAHs levels were 2 to 23-times superior to the general population, whereas individual metabolites remained below the general population median range (except for 1-hydroxynaphthalene+1-hydroxyacenaphtene). Exposure to gaseous pollutants and/or particulate matter during work-shift was associated with a 3.5-fold increase in total OHPAHs levels. Firefighters who smoke presented 3-times higher total concentration of OHPAHs than non-smokers (p < 0.001); non-smoker females presented 2-fold lower total OHPAHs (p = 0.049) than males. 1-hydroxypyrene was below the recommended occupational biological exposure value (2.5 μg/L), and the metabolite of carcinogenic PAH (benzo(a)pyrene) was not detected. Blood pressure was above 120/80 mmHg in 71% of subjects. Firefighters from the permanent intervention team presented significantly increased systolic pressure than those who performed other functions (p = 0.034). Tobacco consumption was significantly associated with higher basophils (p = 0.01–0.02) and hematocrit (p = 0.03). No association between OHPAHs and blood pressure was found. OHPAHs concentrations were positively correlated with monocyte, basophils, large immune cells, atypical lymphocytes, and mean corpuscular volume, which were stronger among smokers. Nevertheless, inverse associations were observed between fluorene and pyrene metabolites with neutrophils and eosinophils, respectively, in non-smokers. Hemogram was negatively affected by overworking and lower physical activity.     Rainfall events and daily mortality across 645 global locations: two stage time series analysis

    Objective: To examine the associations between characteristics of daily rainfall (intensity, duration, and frequency) and all cause, cardiovascular, and respiratory mortality. Design: Two stage time series analysis. Setting: 645 locations across 34 countries or regions. Population: Daily mortality data, comprising a total of 109 954 744 all cause, 31 164 161 cardiovascular, and 11 817 278 respiratory deaths from 1980 to 2020. Main outcome measure: Association between daily mortality and rainfall events with return periods (the expected average time between occurrences of an extreme event of a certain magnitude) of one year, two years, and five years, with a 14 day lag period. A continuous relative intensity index was used to generate intensity-response curves to estimate mortality risks at a global scale. Results: During the study period, a total of 50 913 rainfall events with a one year return period, 8362 events with a two year return period, and 3301 events with a five year return period were identified. A day of extreme rainfall with a five year return period was significantly associated with increased daily all cause, cardiovascular, and respiratory mortality, with cumulative relative risks across 0-14 lag days of 1.08 (95% confidence interval 1.05 to 1.11), 1.05 (1.02 to 1.08), and 1.29 (1.19 to 1.39), respectively. Rainfall events with a two year return period were associated with respiratory mortality only, whereas no significant associations were found for events with a one year return period. Non-linear analysis revealed protective effects (relative risk 1) with extreme intensities. Additionally, mortality risks from extreme rainfall events appeared to be modified by climate type, baseline variability in rainfall, and vegetation coverage, whereas the moderating effects of population density and income level were not significant. Locations with lower variability of baseline rainfall or scarce vegetation coverage showed higher risks. Conclusion: Daily rainfall intensity is associated with varying health effects, with extreme events linked to an increasing relative risk for all cause, cardiovascular, and respiratory mortality.     Temporal change in minimum mortality temperature under changing climate: A multicountry multicommunity observational study spanning 1986-2015

    Background: The minimum mortality temperature (MMT) or MMT percentile (MMTP) is an indicator of population susceptibility to nonoptimum temperatures. MMT and MMTP change over time; however, the changing directions show region-wide heterogeneity. We examined the heterogeneity of temporal changes in MMT and MMTP across multiple communities and in multiple countries. Methods: Daily time-series data for mortality and ambient mean temperature for 699 communities in 34 countries spanning 1986-2015 were analyzed using a two-stage meta-analysis. First, a quasi-Poisson regression was employed to estimate MMT and MMTP for each community during the designated subperiods. Second, we pooled the community-specific temporally varying estimates using mixed-effects meta-regressions to examine temporal changes in MMT and MMTP in the entire study population, as well as by climate zone, geographical region, and country. Results: Temporal increases in MMT and MMTP from 19.5 °C (17.9, 21.1) to 20.3 °C (18.5, 22.0) and from the 74.5 (68.3, 80.6) to 75.0 (71.0, 78.9) percentiles in the entire population were found, respectively. Temporal change was significantly heterogeneous across geographical regions (P < 0.001). Temporal increases in MMT were observed in East Asia (linear slope [LS] = 0.91, P = 0.02) and South-East Asia (LS = 0.62, P = 0.05), whereas a temporal decrease in MMT was observed in South Europe (LS = -0.46, P = 0.05). MMTP decreased temporally in North Europe (LS = -3.45, P = 0.02) and South Europe (LS = -2.86, P = 0.05). Conclusions: The temporal change in MMT or MMTP was largely heterogeneous. Population susceptibility in terms of optimum temperature may have changed under a warming climate, albeit with large region-dependent variations.What this study adds: This study examines the geographical heterogeneity in the temporal changes in minimum mortality temperature (MMT) across 34 countries (699 communities) over the 30-year period spanning 1986–2015. This study attempts to investigate both the pooled evidence and the heterogeneity in the temporal change in MMT on a global scale. Our findings suggest an overall increase in MMT globally over the 30 years, with certain country- or region-specific trends displaying both upward and downward movements without discernible patterns. O presente instrumento tem três grandes objetivos: 1) Averiguar a concretização do Quadro de Avaliação e Responsabilização e do Plano de Atividades de 2023, nomeadamente, avaliar a estratégia assumida através dos seus objetivos estratégicos, verificando o grau de realização dos objetivos operacionais que foram propostos nestes documentos.     Phenotypic and genotypic characterization of Salmonella Typhi and Paratyphi isolates in Portugal, between January 1994 and May 2024

    Os serotipos Typhi e Paratyphi causam infeções sistémicas graves, sendo crucial a administração de antibioterapia adequada. No entanto, o aumento crescente da taxa de multirresistência aos antibióticos, especialmente no caso de Salmonella Typhi, dificulta a escolha dos antibióticos a administrar. Em Portugal, os casos de febre tifoide e paratifoide são raros. Com este estudo pretendeu-se identificar e caracterizar os perfis de resistência aos antibióticos de isolados de Salmonella Typhi e Salmonella Paratyphi A, B e C, entre janeiro de 1994 e maio de 2024, e avaliar a sua proximidade genética. Após confirmação do serotipo, 86 isolados foram submetidos a testes de suscetibilidade aos antibióticos e sequenciação total do genoma. Neste estudo, 60,5% dos isolados foram resistentes a pelo menos um antibiótico, dos quais 15,4% eram multirresistentes. Entre 1994-2016 todos os isolados foram suscetíveis, mas a partir de 2017 foi possível observar uma tendência crescente na resistência aos antibióticos, especialmente para S. Typhi. A resistência à pefloxacina foi a mais frequente, seguida da resistência ao ácido nalidíxico, ampicilina, trimetoprim, sulfametoxazol e cloranfenicol. Dos diversos genes de resistência identificados, destaca-se a presença do gene blaCTX- M-15 num isolado de Salmonella Typhi de 2020, resistente a antibióticos de importância crítica. Foram identificados 11 pequenos clusters. Apesar dos serotipos tifoides e paratifoides não serem endémicos em Portugal, é essencial manter uma vigilância laboratorial contínua, de forma a caracterizar os isolados circulantes em Portugal e detetar potenciais surtos.Typhi and Paratyphi serovars are responsible for severe systemic infections that require appropriate antimicrobial therapy. However, the increasing rate of multidrug resistance, particularly in Salmonella Typhi, complicates therapeutics. In Portugal, typhoid and paratyphoid fever cases are rare. This study aimed to identify and to characterize the antibiotic resistance profiles of Salmonella Typhi and Salmonella Paratyphi A, B, and C isolates from 1994 to May 2024, and to assess their genetic proximity. Af ter serotyping confirmation, 86 isolates underwent antimicrobial susceptibilit y testing and Whole Genome Sequencing. In this study, 60.5% of the isolates were resistant to at least one antibiotic, of which 15.4% were multidrug-resistant. Between 1994 and 2016, all isolates were susceptible, but an increasing trend in antibiotic resistance was observed af ter 2017, par ticularly in S. Typhi. Resistance to pefloxacin was most frequent, followed by nalidixic acid, ampicillin, trimethoprim, sulphamethoxazole, and chloramphenicol. Among the resistance genes identified, blaCTX-M-15 gene was found in a S. Typhi isolate from 2020, resistant to critically impor tant antibiotics. Eleven small clusters were identified. Although typhoid and paratyphoid serotypes are not endemic in Por tugal, continuous laborator y surveillance is essential to characterize circulating isolates and to detect potential outbreaks.info:eu-repo/semantics/publishedVersio

    Programa Nacional de Rastreio Neonatal: relatório 2023

    Relatório de atividades do Programa Nacional de Rastreio Neonatal (PNRN) relativo ao ano de 2023, elaborado pela Comissão Executiva do Programa. O documento refere todos os casos detetados, bem como os Centros de Tratamento de apoio aos doentes e a prevalência ao nascimento das doenças rastreadas, entre outra informação estatística. Do presente relatório de desenvolvimento do Programa, destaca-se o seguinte: - Em 2023, as 27 doenças integradas no painel do Programa Nacional de Rastreio Neonatal (Hipotiroidismo Congénito, Fibrose Quística, Drepanocitose (Anemia de Células Falciformes e 24 Doenças Hereditárias do Metabolismo) foram rastreadas de uma forma sistemática, em 85.764 recém-nascidos (RN) e identificados 136 doentes com uma média de idade de início de tratamento de 9,6 dias de vida; - No ano transato, teve início o estudo-piloto para o rastreio neonatal da Atrofia Muscular Espinal em 100.000 recém-nascidos que foi concluído no final deste ano com uma prevalência ao nascimento de 1:14.515; - A taxa de cobertura nacional mantém-se próxima dos 100%, o que constitui um excelente indicador de aceitação da população a este programa nacional de saúde pública; - Desde o início do Programa, foram rastreados 4.224.550 recém-nascidos e identificados 2.678 casos positivos; - Pela qualidade dos seus indicadores, número de patologias rastreadas, tempo médio de início de tratamento e taxa de cobertura a nível nacional, considera-se que é, de facto, um Programa de grande eficácia clínica e epidemiológica. Criado em 1979, este programa de saúde pública, conhecido como “teste do pezinho”, tem como objetivo primário o rastreio neonatal de doenças raras, de forma a evitar a evolução da patologia rastreada, através do diagnóstico pré-sintomático e da orientação para implementação precoce de terapia adequada. A Unidade de Rastreio Neonatal, Metabolismo e Genética do DGH, que funciona no Centro de Saúde Pública Doutor Gonçalves Ferreira do INSA no Porto, é o braço laboratorial do Programa, sendo composta pelo Laboratório Nacional de Rastreios, Laboratório de Metabolismo e Laboratório de Genética Molecular. Após 44 anos de atividade, o PNRN continua a apresentar um elevado padrão de qualidade e desenvolvimento que lhe confere um prestígio nacional e internacional por todos reconhecido e respeitado, mantendo uma trajetória e um dinamismo de que são sinais, para além dos indicadores de eficácia clínica e epidemiológica, o elevado nível de produção científica, a implementação de normas internacionais de qualidade e o alargamento do rastreio a novas entidades nosológicas

    The mechanism of nonsense-mediated mRNA decay and its players

    Dissertação de Mestrado em Biologia Molecular e Genética, apresentada à Faculdade de Ciências da Universidade de Lisboa, 2024.Luísa Loison, Departamento de Genética Humana, INSA.Data do grau: 24-10-2024.Nonsense-mediated mRNA decay (NMD) is a post-transcriptional surveillance mechanism harbouring two functions: identification and degradation of transcripts containing premature translation-termination codons (PTC), preventing deleterious effects in the cell; and downregulation of mRNAs in response to cellular needs, maintaining the quality of gene expression. One-third of gene mutations in human genetic diseases, including cancer, are due to nonsense mutations or frameshift that result in transcripts harbouring nonsense codons and can be eliminated by NMD. The several factors involved in this mechanism may act in diverse ways depending on the set of transcripts to be regulated, contributing to the branching of this pathway. Cytoplasmic DIS3 exosome independent 3′-5′ exoribonuclease 2 (DIS3L2) has been reported as one of the factors to induce NMD targets decay. Therefore, this study aims to enlighten how DIS3L2 functions in NMD: i) analyse the correlation between the distinct branches of NMD and cervical and uterus cancer; ii) investigate which branch guides DIS3L2-mediated degradation; and iii) test which region of the NMD/DIS3L2-targets mediate DIS3L2 degradation through a system of hybrid-genes. For our first aim, we detected no correlation between any of the branches of NMD and uterus and cervical cancer. Each factor acts independently. In our second objective, we analysed the mRNA expression of five transcripts, but none displayed a significant alteration in their expression to infer a correlation between DIS3L2 and a particular NMD branch. Relatively to the last aim, we successfully cloned three out of the four constructs but due to time constrictions we could not continue. Nonetheless, further testing is needed to better understand this mechanism and how transcript degradation is mediated, including the diverse factors needed for its activation, which might be the key to future advanced therapeutic strategies.A expressão génica nos seres vivos tem como objetivo a transferência da informação proveniente dos ácidos desoxirribonucleicos (DNA) para RNA mensageiro (mRNA) e, a partir deste, proceder à síntese proteica. Por ser um processo complexo, é preciso haver uma regulação contínua de forma a impedir a passagem de erros. O decaimento de RNA mensageiro mediado por codões nonsense (NMD) é considerado um de vários mecanismos de vigilância celular, tendo duas funções: 1) identificação e degradação de transcritos que contêm codões de terminação prematura da tradução (PTC), evitando a produção de proteínas truncadas que podem ter efeitos deletérios na célula e 2) regulação de mRNAs fisiológicos (codificantes de proteínas funcionais) em resposta às necessidades celulares, mantendo a qualidade da expressão génica. Acredita-se que este mecanismo influencia os níveis de expressão de aproximadamente 10 % dos transcritos em eucariotas, estando implicado em vários processos biológicos que moldam o respetivo programa de expressão génica. Além disso, acredita-se que um terço das mutações encontradas em doenças genéticas humanas, incluindo o cancro, resultam em transcritos portadores de PTC, que podem ser eliminados por NMD. No NMD, há vários fatores envolvidos que podem atuar de diferentes maneiras, dependendo do conjunto de transcritos a regular, contribuindo para a diversificação deste mecanismo em dois modelos principais: NMD dependente do complexo da junção exão-exão (EJC) e NMD independente de EJC. Adicionalmente, vários estudos têm observado a ramificação deste mecanismo aquando da depleção de uma das proteínas intervenientes, havendo a utilização de caminhos alternativos pelos outros fatores para que se dê a cascata de reações. Por conseguinte, acredita-se que o modelo da via de NMD dependente de EJC se possa diversificar, por sua vez, em duas vias: uma, dependente da proteína upframeshift 3B (UPF3B) e da proteína do gene 3 candidato à suscetibilidade ao cancro (CASC3); e outra, dependente da UPF2 e da proteína de ligação a RNA com domínio 1 rico em serina (RNPS1). Ambas RNPS1 e CASC3 são proteínas associadas ao EJC. No modelo de NMD independente de EJC, há atuação da UPF3B em associação com UPF2 sem o envolvimento de EJC. De uma forma geral, após reconhecimento do transcrito, o mecanismo de NMD é ativado, havendo interação do fator UPF1 com os fatores de terminação da tradução 3 e 1 (eRF3 e eRF1), associados ao complexo proteico de tradução, e, ainda, com o supressor de efeito morfológico na genitália 1 (SMG1), formando o complexo SURF. Este complexo irá interagir de forma variável, dependendo dos fatores envolvidos na reação e, consequentemente, do ramo. Independentemente destes acontecimentos, a interação destes fatoresirá formar o complexo de indução de decaimento (DECID) e, consequentemente, a fosforilação da UPF1 pela SMG1, conduzindo ao recrutamento das SMG6 ou SMG5/7, que desencadeiam a degradação do transcrito. Alguns estudos identificaram que uma das enzimas envolvidas nesta última etapa é a exoribonuclease 3'-5' independente do exossoma – DIS3L2. Esta enzima está associada a vários processos celulares como a divisão celular e carcinogénese, assim como ao desenvolvimento de síndrome de Perlman. O presente estudo visa esclarecer como a proteína DIS3L2 atua no NMD e a sua relação com os vários ramos deste mecanismo. No nosso primeiro objetivo, pretendeu-se analisar a correlação entre os diferentes ramos do NMD e a sua expressão nos cancros do útero e cólo do útero. Para tal, efetuou-se uma análise bioinformática de dados transcritómicos disponíveis publicamente na plataforma Xena, comparando a expressão de cada um dos transcritos que codificam para proteínas especificas dos diferentes ramos de NMD — UPF2, RNPS1, UPF3B e CASC3 — em tecido saudável e cancerígeno, no útero e no cólo do útero. Detetámos a sobreexpressão do mRNA de RNPS1 e UPF3B em tecidos do cancro de cólo do útero e do útero. Contrariamente, observou-se níveis baixos de mRNA de CASC3 nas amostras destes dois cancros, comparativamente com os respetivos níveis em amostras saudáveis. Apesar de não podermos concluir a sua função nestes tecidos, estes três fatores poderão ter um grande potencial clínico como possíveis biomarcadores de cancro do cólo do útero e do útero e como alvos terapêuticos. Quanto à expressão de UPF2, embora com diferenças estatisticamente significativas nos seus níveis de mRNA, a gama em que se encontram os valores observados é muito semelhante entre células saudáveis e cancerígenas nos tecidos de cólo do útero e de útero e, consequentemente, não podemos concluir o efeito de UPF2. Com base nestas análises, não se detetou nenhum efeito conjunto a nível de ramos de NMD, sendo que fatores que constituam o mesmo ramo obtiveram expressões de mRNA diferentes consoante tecido saudável e cancerígeno. Era de esperar que fatores que constituíssem o mesmo ramo tivessem efeitos semelhantes aquando da análise da sua expressão transcricional, mas não foi isso que se observou. Desta forma, não se pode concluir que no cólo do útero e no útero, haja uma atuação especifica de um ramo ou outro do NMD, mas sim que cada fator se comporta de maneira diferente e, consequentemente, podem influenciar o próprio mecanismo a atuar positiva ou negativamente na tumorigénese. O segundo objetivo do estudo teve como intuito investigar em qual dos ramos de NMD, DIS3L2 funciona, em particular, perceber se há uma associação especifica entre DIS3L2 e os ramos de NMD dependentes de UPF2 e UPF3B (independente de EJC) ou dependentes de UPF2 e RNPS1. Para tal, selecionou-se, com base nos resultados obtidos em Garcia-Moreno et al. e na literatura disponível, cinco transcritos-alvo cuja modulação é específica de DIS3L2 e de um fator de cada ramo de NMD (UPF2, RNPS1 e UPF3B). De seguida, procedeu-se ao silenciamento (KD) singular e combinado de três dos fatores envolvidos no NMD — UPF2, RNPS1 e UPF3B — e de DISL32 na linha celular HeLa. Após a avaliação da eficiência de KD, analisou-se o nível de expressão dos transcritos-alvo por transcrição reversa seguida de reação em cadeia da polimerase (RT-PCR) semiquantitativa em todas as condições de KD efetuadas. Os respetivos níveis de expressão de mRNA observados por eletroforese foram quantificados, mas os valores obtidos apresentaram grandes discrepâncias entre replicados, pelo que as diferenças não foram estatisticamente significativas, o que invalidou a interpretação biológica. Na verdade, este estudo carece da otimização de diversas condições como a baixa eficiência de silenciamento que se verificou em alguns dos ensaios e a análise da expressão dos transcritos feita por RT-PCR, técnica pouco rigorosa, o que pode ter causado a incongruência observada nos resultados dos ensaios. Assim sendo, não se conseguiu determinar se DIS3L2 atuava em associação com algum dos ramos de NMD. Relativamente ao terceiro objetivo, este estudo visou testar que região genómica de transcritos-alvo de NMD é responsável pelo acionamento da degradação por DIS3L2. Para tal, recorreu-se a um sistema de construções génicas contendo diferentes combinações das regiões genómicas 5’ e 3’ não traduzidas (UTR) e codificante do gene GADD45A (alvo conhecido de NMD e DIS3L2), clonadas às do gene beta ()-globina (HBB) (resistente ao NMD e à DIS3L2), com o intuito de formar quatro genes-híbridos e, sequencialmente, analisar os seus níveis de expressão. Esta análise teve como base resultados anteriores no grupo, em que se avaliou os níveis de expressão de genes-híbridos contendo combinações de regiões genómicas dos genes referidos anteriormente, mas cuja região codificante pertencia ao gene HBB. Verificou-se que a 3’UTR tornava o transcrito mais suscetível à ação de degradação da proteína DIS3L2. Isto poderá estar relacionado com as proteínas terminais uridiltransferases, que adicionam oligonucleótidos de uracilo na extremidade 3’ dos transcritos, os quais são reconhecidos pela DIS3L2. De forma a continuar este estudo, procedeu-se à clonagem dos quatro genes híbridos referidos anteriormente. Contudo, apenas conseguimos obter três dos quatro construtos inicialmente planeados. Apesar das várias otimizações feitas durante o processo, não se conseguiu clonar o construto 5’HBB_GADD45A ORF_3’GADD45A. Em suma, os resultados dos últimos dois objetivos são inconclusivos, havendo necessidade de otimizar algumas das técnicas efetuadas e continuar com as experiências em falta para obter conclusões definitivas. Em todo o caso, observámos uma possível associação entre alguns fatores (UPF3B, RNPS1 e CASC3) de NMD e os seus níveis de mRNA em amostras de cancro do útero e do cólo do útero. Por conseguinte, o papel que o NMD tem ao nível tumorigénico e, como se relaciona com a DIS3L2 na degradação dos seus diferentes alvos, poderá revelar novos pontos de partida para o desenvolvimento de futuras estratégias terapêuticas

    Exploring the effect of clinical case definitions on influenza vaccine effectiveness estimation at primary care level: Results from the end-of-season 2022–23 VEBIS multicentre study in Europe

    Background: Within influenza vaccine effectiveness (VE) studies at primary care level with a laboratory-confirmed outcome, clinical case definitions for recruitment of patients can vary. We used the 2022-23 VEBIS primary care European multicentre study end-of-season data to evaluate whether the clinical case definition affected IVE estimates. Methods: We estimated VE using a multicentre test-negative case-control design. We measured VE against any influenza and influenza (sub)types, by age group (0-14, 15-64, ≥65 years) and by influenza vaccine target group, using logistic regression. We estimated IVE among patients meeting the European Union (EU) acute respiratory infection (ARI) case definition and among those meeting the EU influenza-like illness (ILI) case definition, including only sites providing information on specific symptoms and recruiting patients using an ARI case definition (as the EU ILI case definition is a subset of the EU ARI one). Results: We included 24 319 patients meeting the EU ARI case definition, of whom 21 804 patients (90 %) meet the EU ILI case definition, for the overall pooled VE analysis against any influenza. The overall and influenza (sub)type-specific VE varied by ≤2 % between EU ILI and EU ARI populations. Discussion: Among all analyses, we found similar VE estimates between the EU ILI and EU ARI populations, with few (10%) additional non-ILI ARI patients recruited. These results indicate that VE in the 2022-23 influenza season was not affected by use of a different clinical case definition for recruitment, although we recommend investigating whether this holds true for next seasons.ECDC/2021/019/European Centre for Disease Prevention and Controlinfo:eu-repo/semantics/publishedVersio
