„Ein Lerngerüst bauen“ – das Schreiben dialektischer Erörterungen durch den SRSD-Ansatz vermitteln
Das Schreiben einer dialektischen Erörterung stellt eine relevante Schreibaufgabe dar und ist obligatorischer Bestandteil der Prüfungen zum mittleren Schulabschluss (MSA-Prüfungen) in Klasse 10 im Fach Deutsch in Berlin und Brandenburg. Allerdings liegen für deutsche nicht-gymnasiale Schulen weder wissenschaftliche Untersuchungen zur Erhebung der Schreibkompetenz im Bereich der dialektischen Erörterung noch erprobte Unterrichtskonzepte zur Vermittlung dieser Textsorte vor.
Eine Möglichkeit, die Schreibkompetenz aller Schüler:innen mithilfe des Scaffolding-Konzeptes zu fördern, stellt der Self-Regulated-Strategy-Development-Ansatz (SRSD-Ansatz) dar. Für die erstmalige Adaption des SRSD-Ansatzes für die Textsorte der dialektischen Erörterung an nicht-gymnasialen Schulen geht die vorliegende Arbeit daher folgender globaler Fragestellung nach: Wie wirken sich die Unterrichtsreihe zum dialektischen Erörtern nach dem Self-Regulated-Strategy-Development-Ansatz und die dazugehörigen Scaffolds im Rahmen eines Forschungs- und Unterrichtsprojekts auf die Schreibkompetenz von Lerngruppen der 9. Klassenstufe an zwei Brandenburger Gesamtschulen, gemessen anhand der Textlänge und der holistischen Textqualität dialektischer Erörterungen, aus?
Die genannte Unterrichtsreihe zum dialektischen Erörtern wurde innerhalb von sechs Unterrichtsstunden (jeweils 90 Minuten) im Sinne des Design-Based-Research-Ansatzes im Rahmen des Projekts Fair Debattieren und Erörtern durchgeführt. Aus dem Korpus dieses Projekts wurden die Erörterungstexte von insgesamt sieben Klassen der 9. Jahrgangsstufe an zwei Gesamtschulen in Brandenburg (N = 381) extrahiert, wozu Schüler:innentexte von vier randomisierten Interventionsgruppen mit zwei unterschiedlichen Treatmentreihenfolgen DE (erst Debattieren, dann Erörtern, n = 115) und ED (erst Erörtern, dann Debattieren, n = 118) sowie von drei Kontrollgruppen (n = 148) gehören. Die Texte wurden anhand von vier originalen Schreibaufgaben aus MSA-Prüfungen der Jahre 2013 und 2017 zu vier Messzeitpunkten im Längsschnitt erhoben und im Mixed-Methods-Design folgendermaßen analysiert:
a) Auf quantitativer Ebene werden die holistische Textqualität (Double-Blind-Rating auf einer 5-Punkt-Skala) und die Textlänge als Indikatoren für Schreibkompetenz von dialektischen Erörterungen zu allen vier Messzeitpunkten betrachtet und Mittelwertsunterschiede zu einem Messzeitpunkt durch einfaktorielle Varianzanalysen berechnet.
b) Auf qualitativer Ebene erfolgt die Beschreibung der Unterrichtsreihe zum dialektischen Erörtern für die vier Interventionsgruppen sowie deren weitere Analyse anhand von digitalen Feldnotizen der Trainer:innen. Diese digitalen Feldnotizen werden systematisch durch das Verfahren der qualitativen zusammenfassenden Inhaltsanalyse nach Mayring (2022) ausgewertet.
Die qualitativen Ergebnisse zeigen, dass in allen vier Interventionsgruppen die Schüler:innen alle sechs Phasen der Unterrichtsreihe nach dem SRSD-Ansatz durchlaufen. Die Schaffung einer motivierenden Schreibumgebung durch die Umsetzung von – für den SRSD-Ansatz charakteristischen – Designprinzipien der Unterrichtsreihe können dabei als potenziell ergiebige Faktoren für die Förderung der Schreibkompetenz aller Schüler:innen identifiziert werden.
Die quantitativen Ergebnisse zeigen, dass diejenigen Schüler:innen die höchste Textqualität und Textlänge aufweisen, welche zuvor die Unterrichtsreihe zum dialektischen Erörtern absolvierten (t2 und t3).
Des Weiteren werden in der vorliegenden Dissertationsarbeit Aspekte hinsichtlich eines Transfers der untersuchten Unterrichtsreihe in die Schreibforschung, die Schulpraxis sowie die Lehrkräfteaus- und -weiterbildung erläutert.Writing a pro-and-con-argumentation is a relevant writing task and is a compulsory part of the examinations for the intermediate school leaving certificate (MSA examinations) in year 10 in the subject German in Berlin and Brandenburg. However, for German non-grammar schools, there are neither scientific studies to assess writing skills in the area of pro-and-con-argumentation nor tried and tested teaching concepts for teaching this type of text.
The self-regulated strategy development approach (SRSD approach) is one way of promoting the writing skills of all pupils with the help of the scaffolding concept. For the first adaptation of the SRSD approach for the text type of pro-and-con-argumentation at non-grammar schools, this study therefore pursues the following global question: How do the series of lessons on pro-and-con-argumentations according to the Self-Regulated Strategy Development approach and the associated scaffolds as part of a research and teaching project effect the writing skills of 9th grade learning groups at two comprehensive schools in Brandenburg based on the text length and holistic text quality of pro-and-con-argumentations?
The aforementioned series of lessons on pro-and-con-argumentation was conducted within six lessons (90 minutes each) using the design-based research approach as part of the Fair Debating and Written Argumentation project. From the corpus of that project, the pro-and-con-argumentation texts of a total of seven 9th grade classes at two comprehensive schools in Brandenburg (N = 381) were extracted, including student texts from four randomised intervention groups with two different treatment sequences DE (first debating, then writing, n = 115) and ED (first writing, then debating, n = 118) as well as from three control groups (n = 148). The texts were collected longitudinally at four measurement points using four original writing tasks from MSA examinations from 2013 and 2017 and analysed as follows using a mixed-method design:
a) On a quantitative level, holistic text quality (double-blind rating on a 5-point scale) and text length are considered as indicators of writing competence of pro-and-con-argumentations at all four measurement points and mean differences at one measurement point are calculated using single-factor analyses of variance.
b) On a qualitative level, the series of lessons on pro-and-con-argumentations is described for the four intervention groups and further analysed using the trainers' digital field notes. These digital field notes are systematically analysed using the qualitative summarising content analysis method according to Mayring (2022).
The qualitative results show that in all four intervention groups, the students go through all six phases of the series of lessons according to the SRSD approach. The creation of a motivating writing environment through the implementation of design principles - characteristic of the SRSD approach - in the series of lessons can be identified as potentially productive factors for promoting the writing skills of all students.
The quantitative results show that those students who had previously completed the pro-and-con-argumentation series of lessons (t2 and t3) had the highest text quality and text length.
Furthermore, this dissertation discusses aspects regarding the transfer of the investigated series of lessons into writing research, school practice and teacher training and further education
Entstehung und Zerfall von Magnetfeldern
In order to gain a comprehensive understanding of the dynamic processes occurring on the Sun, it is essential to closely study of the dynamic interplay between plasma flows and magnetic fields. The interplay between plasma flows and magnetic fields within active regions is the primary driver of the formation and evolution of these regions. Once magnetic flux bundles have pierced the photosphere and become visible as dark surface structures, they extend as a canopy structure into the chromosphere and higher atmospheric layers. The emergence of magnetic features affects the typical surface flows, which are generally characterized by global large-scale flow motions and divergence centers caused by granulation. Moreover, their appearance impacts the entire atmosphere, potentially affecting the entire heliosphere causing explosive events such as flares and especially coronal mass ejections (CMEs).
Nevertheless, the processes by which active regions emerge, evolve and decay remain poorly understood. What are the determining factors that make the leading or trailing part the dominant one? What processes, discernible at the photospheric or chromospheric level, contribute to the heating of the corona? What are the mechanisms by which emerging magnetic flux bundles and plasma motions on the solar surface interact with each other? What are the consequences for the evolution of active regions? What processes regulate the evolution of magnetic flux bundles as they rise into the chromosphere, beyond the observed extension in a canopy-like structure? What are the conditions that give rise to the formation of a penumbra? What are the determining factors that influence whether a penumbra fully develops into its characteristic fine filamentary structure or only exhibits a more rudimentary form? To what extent do photospheric flow fields exert an influence on those typically observed in the chromosphere? Furthermore, it is little understood at which moment the gradual process of sunspot decay begins. This raises the question of whether a critical value of the magnetic field marks the onset of penumbral decay, and consequently the decay of the entire sunspot. In this context, the role played by the surrounding plasma motions requires further scrutiny. Is it solely responsible for the advection of small-scale moving magnetic features (MMFs), or does a change in the surrounding flows and magnetic fields instigates penumbral decay?
This doctoral research investigates the complex processes of emergence, evolution, and decay of active regions on the ever-changing Sun. In order to achieve this objective, surface features resulting from the influence of strong magnetic fields, such as pores and sunspots, are examined at the highest spatial, spectral, and temporal resolution, as well as with high polarimetric sensitivity. The data analyzed in this work comprises high-resolution ground-based imaging and spectro-polarimetric data. However, the high-resolution observations only unveil their full potential when placed in context with synoptic data, thus, establishing statistical significance and accurately placing them in the long-term evolution of active regions. The combination of this data enabled a detailed analysis of the three-dimensional magnetic and flow fields in the photosphere and chromosphere.
Considering the complex interaction of plasma motions and magnetic fields, sunspots play a special role in this context. High-resolution observations of the evolution of sunspots, from formation to decay, provide significant insights into the dynamics of the Sun. Optimally, observations should start prior to the initial disturbance of the quiet Sun by emerging magnetic flux bundles and continue to cover the entirety of the active region life cycle. This would include, the formation of a penumbra, which transforms a simple pore into a sunspot, thereby introducing further complexity into the magnetic and flow structure. Later, the evolution into a complex active region must be observed, which consists of multiple sunspots,
pores, and related features such as light bridges. These are embedded in a complex plasma flow and magnetic structure until the magnetic flux eventually dissipates and decays. Nevertheless, research should not be limited to sunspots alone, but should also encompass smaller features in the periphery and those related to spots, which should be included in the analysis. The objective of this research project is to enhance our understanding of active region dynamics. The utilization of sophisticated methods, including Background-subtracted Solar Activity Maps (BaSAMs), Local Correlation Tracking (LCT), and the Stokes Inversions based on Response function (SIR) code, enabled comprehensive studies of several active regions observed in the photosphere and chromosphere. These studies encompassed pores, sunspots with rudimentary and fully formed penumbra, light bridges, and several small-scale features during different stages of evolution.
These research studies are the foundation of three first-author, peer-reviewed articles in scientific journals.
1. The first publication resulting from this doctoral research covered the topic High-resolution Imaging of Solar Pores and was published in 2023 in the journal Astronomy & Astrophysics (DOI:10.1051/0004-6361/202245410).
Context. Light bridges are bright, long, and narrow features that are typically connected to the formation or decay processes of sunspots and pores.
Aims. The interaction of magnetic fields and plasma flows is investigated in the trailing part of an active region, where pores and magnetic knots evolve into a complex sunspot. The goal is to identify the photospheric and chromospheric processes, which transform the mainly vertical magnetic fields of pores into a sunspot with multiple umbral cores, light bridges, and rudimentary penumbrae.
Methods. Conducting observations with a broad variety of telescopes and instruments provides access to different atmospheric layers and the changing morphology of features connected to strong magnetic fields. While the Helioseismic and Magnetic Imager (HMI) of the Solar Dynamics Observatory (SDO) provides full-disk continuum images and line-of-sight magnetograms, the fine structure and flows around a pore can be deduced from high-resolution observations in various wavelengths as provided by the Goode Solar Telescope (GST) at the Big Bear Solar Observatory (BBSO). Horizontal proper motions are evaluated applying local correlation tracking (LCT) to the available time series, whereas the connectivity of sunspot features can be established using the background-subtracted activity maps (BaSAMs).
Results. Photospheric flow maps indicate radial outflows, where the light bridge connects to the surrounding granulation, whereas inflows are present at the border of the pores. In contrast, the chromospheric flow maps show strong radial outflows at superpenumbral scales, even in the absence of a penumbra in the photosphere. The region in between the two polarities is characterized by expanding granules creating strong divergence centers. Variations in BaSAMs follow locations of significant and persistent changes in and around pores. The resulting maps indicate low variations along the light bridge, as well as thin hairlines connecting the light bridge to the pores and strong variations at the border of pores. Various BaSAMs demonstrate the interaction of pores with the surrounding supergranular cell. The Hα line-of-sight velocity maps provide further insights into the flow structure, with twisted motions along some of the radial filaments around the pore with the light bridge. Furthermore, flows along filaments connecting the two polarities of the active region are pronounced in the line-of-sight velocity maps.
Conclusions. The present observations reveal that even small-scale changes of plasma motions in and around pores are conducive to transform pores into sunspots. In addition, chromospheric counterparts of penumbral filaments appear much earlier than the penumbral filaments in the photosphere. Penumbra formation is aided by a stable magnetic feature that anchors the advection of magnetic flux and provides a connection to the surrounding supergranular cell, whereas continuously emerging flux and strong light bridges are counteragents that affect the appearance and complexity of sunspots and their penumbrae.

2. The second paper that covered the topic Impact of Magnetic and Flow Fields on Penumbrae and Light Bridges of Three Leading Sunspots in an Active Region was published in 2024 in the journal Solar Physics (DOI:10.1007/s11207-024-02386-9).
This study investigates penumbrae and light bridges based on photospheric and chromospheric flow fields and photospheric magnetic fields in active region NOAA 13096. The improved High-resolution Fast Imager (HiFI+) and the GREGOR Infrared Spectrograph (GRIS) acquired high-resolution imaging and spectropolarimetric data at the 1.5-meter GREGOR solar telescope at the Observatorio del Teide, Iza ˜na, Tenerife, Spain. Background-Subtracted Activity Maps (BaSAMs) have been used to locate areas of enhanced activity, Local Correlation Tracking (LCT) provides horizontal proper motions, and near-infrared full-Stokes polarimetry offers access to magnetic fields and line-of-sight velocities. The results show that the decaying active region is characterized by a triangular region between the three leading, positive-polarity sunspots with unfavorable conditions for penumbra formation. This region has a spongy appearance in narrow-band Hα images, shows signs of enhanced activity on small spatial scales, is free of divergence centers and exploding granules, lacks well-ordered horizontal flows, has low flow speeds, and is dominated by horizontal magnetic fields. Umbral cores are inactive, but the interface between pores and penumbral filaments often shows enhanced activity. Moat flows and superpenumbrae are almost always observed, when penumbral filaments are present, even in very small penumbral sectors. However, evidence of the moat flow can also be seen around pores, surviving longer than the decaying penumbral filaments. Light bridges have mainly umbral temperatures, reaching quiet-Sun temperatures in some places, show strong intensity variations, and exhibit weak photospheric horizontal flows, while narrow-band Hα flow maps show substantial inflows.

3. The third article is currently under review for publication in the journal Solar Physics and covers the topic Wide-field Image Restoration of G-band and Ca II K Images Containing Large and Complex Active Regions.
Restoration of solar images to achieve near diffraction-limited spatial resolution is commonly implemented on large-aperture solar telescopes. However, the extent of the restored field-of-view is typically only 100′′ or even smaller. This study reports on wide-field image restoration of G-band and Ca ii K images obtained with the 0.7-meter Vacuum Tower Telescope (VTT) at Observatorio del Teide, Tenerife, Spain. The Kiepenheuer Adaptive Optics System (KAOS) at the VTT provides a field-of-view with a diameter of 270′′. Time series of datasets with 100 images were acquired with a large-format (7920×6004 pixels) CMOS imager at a frame rate of 30 Hz. An experimental optical setup that enabled wide-field imaging is briefly presented. Results from speckle-masking imaging and Multi-Frame Blind Deconvolution (MFBD) demonstrate the unique potential of the VTT for wide-field imaging. Power spectral analysis quantifies the instrument performance as a function of time and with distance from the lock point of the AO system. Further science capabilities are established using Local Correlation Tracking (LCT), Background-subtracted Solar Activity Maps (BaSAMs), and image quality/seeing metrics. In summary, commercial off-the-shelf camera systems can become an asset to sub-meter-class solar telescopes and open up new science opportunities at the interface between large-aperture and synoptic solar telescopes.

This doctoral research contributes to a more comprehensive understanding of the emergence, evolution, and decay of active regions and their corresponding features, including sunspots, pores, and light bridges. The analysis focused on the photosphere and chromosphere and incorporated a variety of analytical techniques. The combination of imaging and spectral data from ground-based and space-based telescopes enabled innovative data analysis, which permitted the intricate interactions between magnetic fields and plasma flows to be elucidated. The results of this work further expand our understanding of the dynamics of active region evolution and the influence of various features on flow motions and magnetic field structure within active regions and the connection to the surrounding regions.Um ein umfassendes Verständnis der dynamischen Prozesse auf der Sonne zu erlangen, ist es unerlässlich, das dynamische Zusammenspiel zwischen Plasmabewegungen und Magnetfeldern genau zu untersuchen. Das Zusammenspiel von Plasmaströmen und Magnetfeldern innerhalb aktiver Regionen ist der Hauptfaktor für die Entstehung und Entwicklung dieser Regionen. Sobald magnetische Flussröhren die Photosphäre durchdrungen haben und als dunkle Oberflächenstrukturen sichtbar werden, breiten sie sich aus und steigen in die Chromosphäre und höhere Atmosphärenschichten auf. Das Erscheinen magnetischer Elemente an Oberfläche beeinflusst die typischen Oberflächenströmungen, die im Allgemeinen durch globale Strömungsbewegungen und durch die Granulation verursachte werden. Darüber hinaus wirkt sich ihr Auftreten auf die gesamte Atmosphäre aus und kann selbst die Heliosphäre beeinflussen, beispielsweise durch Strahlungsausbrüche und insbesondere koronale Massenauswürfe (CMEs).
Dennoch sind die Prozesse nach wie vor kaum verstanden, durch die aktive Regionen entstehen, sich entwickeln und zerfallen. Welche Faktoren sind ausschlaggebend dafür. Welche photosphärische oder chromosphärische Prozesse tragen zur Heizung der Korona bei? Welches sind die Mechanismen, durch die Magnetfeld und Plasma interagieren? Was sind die Konsequenzen für die Entwicklung aktiver Regionen? Welche Prozesse regulieren die Entwicklung der magnetischen Flussröhren, wenn sie in die Chromosphäre aufsteigen? Welches sind die Bedingungen, die zur Bildung einer Penumbra führen? Welche Faktoren sind ausschlaggebend dafür, ob sich eine Penumbra vollständig zu ihrer charakteristischen Filamentstruktur entwickelt oder nur eine rudimentäre Form aufweist? Darüber hinaus ist wenig bekannt, wann der langsame Prozess des Sonnenfleckenzerfalls beginnt. Dies wirft die Frage auf, ob ein kritischer Wert des Magnetfelds den Beginn des Zerfalls der Penumbra und damit den Zerfall des gesamten Sonnenflecks markiert. In diesem Zusammenhang muss die Rolle der umgebenden Plasmabewegungen genauer untersucht werden. Ist sie allein für die Advektion von kleinskaligen Magnetfeldstrukturen (engl. moving magnetic features) verantwortlich, oder löst eine Veränderung der umgebenden Strömungen und Magnetfelder den Zerfall der Penumbra aus?
Diese Doktorarbeit untersucht die komplexen Prozesse der Entstehung, Entwicklung und des Zerfalls aktiver Regionen auf der sich ständig verändernden Sonne. Um dieses Ziel zu erreichen, werden Oberflächenmerkmale, die durch den Einfluss starker Magnetfelder entstehen, wie Poren und Sonnenflecken, mit höchster räumlicher, spektraler und zeitlicher Auflösung sowie mit hoher polarimetrischer Empfindlichkeit untersucht. Die in dieser Arbeit analysierten Daten umfassen hochauflösende bodengebundene Bilder und spektro-polarimetrische Daten. Diese Beobachtungen entfalten jedoch nur dann ihr volles Potenzial, wenn sie im Kontext synoptischer Daten interpretiert werden. Die Kombination dieser Daten ermöglicht eine detaillierte Analyse der dreidimensionalen Magnet- und Strömungsfelder in der Photosphäre und Chromosphäre.
In Anbetracht der komplexen Wechselwirkung von Plasmabewegungen und Magnetfeldern spielen Sonnenflecken in diesem Zusammenhang eine besondere Rolle. Hochauflösende Beobachtungen der Entwicklung von Sonnenflecken, von der Entstehung bis zum Zerfall, liefern wichtige Erkenntnisse über die Dynamik der Sonne. Optimalerweise sollten die Beobachtungen vor der ersten Störung der ruhigen Sonne durch entstehende magnetische Flussröhren beginnen und den gesamten Lebenszyklus der aktiven Region abdecken. Dies schließt die Bildung einer Penumbra ein, die eine einfache Pore in einen Sonnenfleck verwandelt und damit die Komplexität der Magnet- und Strömungsstruktur erhöht. Später wird die Entwicklung hin zu einer komplexen aktiven Region beobachtet, die aus mehreren Sonnenflecken, Poren und Lichtbrücken besteht. Diese sind in eine komplexe Strömungs- und Magnetstruktur eingebettet, bis sich der magnetische Fluss schließlich auflöst.
Das Ziel dieses Doktorarbeit ist es, unser Verständnis der Dynamik aktiver Regionen zu verbessern. Der Einsatz hochmoderner Methoden, darunter Background-subtracted Solar Activity Maps (BaSAMs), Local Correlation Tracking (LCT) und Stokes Inversions based on Response functions (SIR), ermöglichten umfassende Studien mehrerer aktiver Regionen in der Photosphäre und Chromosphäre. Impact of Cyclooxygenase 2-dependently formed Prostaglandin E2 on LPS-induced activation of different mouse macrophage populations
Charakteristisch für die Steatohepatitis (NASH), eine progressive Form der Nicht-Alkoholischen Fettlebererkrankung, ist eine chronische Entzündung, die maßgeblich durch die Makrophagen in der Leber orchestriert wird. Entzündungsvermittelt aktivierte residente Lebermakrophagen und rekrutierte, infiltrierende Monozyten-abgeleitete Makrophagen sezernieren pro-inflammatorische Cytokine wie Tumornekrosefaktor α (TNFα) sowie den Lipidmediator Prostaglandin E2 (PGE2). Über autokrine Rückkopplungsschleifen kann PGE2 wiederum die Expression der pro-inflammatorischen Cytokine modulieren. Im Rahmen dieses Projektes sollte die PGE2-abhängige Regulation der Cytokin-Expression in verschiedenen Makrophagenpopulationen aus Mäusen, in denen die Cyclooxygenase 2 (COX2), das Schlüsselenzym der Prostanoid-Synthese, zellspezifisch in Makrophagen deletiert war, untersucht werden. Zudem sollten Veränderungen in der Zusammensetzung des hepatischen Makrophagenpools in Mäusen mit Diät-induzierter NASH mittels Durchflusszytometrie charakterisiert werden.
Zunächst wurden isolierte Kupfferzellen (KC), peritoneale Makrophagen (PM) und aus dem Knochenmark abgeleitete Makrophagen (BMDM) aus Wildtyp-Mäusen (WT) und COX2-defizienten Mäusen mit Lipopolysaccharid (LPS) ± exogenem PGE2 stimuliert. Für die Aktivierung von BMDM waren 10-fach höhere LPS-Konzentrationen notwendig als in KC und PM. Dennoch wiesen KC im Vergleich zu den anderen beiden Makrophagenpopulationen eine geringere LPS-abhängige Expression pro-inflammatorischer Cytokine wie TNFα, Interleukin (IL) 1β, IL-6 und Oncostatin M (OSM) sowie des Chemokins Macrophage inflammatory protein 2 (MIP2) auf. Die LPS-induzierte Genexpression und Sekretion von TNFα wurde in allen drei Makrophagenpopulationen Dosis-abhängig durch exogenes PGE2 gehemmt, wobei die mittlere inhibitorische Konzentration an PGE2 in KC etwa 10-fach bzw. 100-fach höher war als in BMDM bzw. PM. Während die LPS-induzierte Expression von IL-1β, IL-6 und MIP2 in KC durch exogenes PGE2 reprimiert wurde, war die Expression dieser pro-inflammatorischen Mediatoren sowie des Cytokins OSM in PM induziert. In COX2-defizienten PM und BMDM wurde die LPS-induzierte endogene PGE2-Synthese vollständig gehemmt und in COX2-defizienten Kupfferzellen um etwa 60 % reduziert. Daraus resultierte infolge der aufgehobenen PGE2-abhängigen Rückkopplungshemmung eine gesteigerte LPS-induzierte TNFα-Expression in COX2-defizienten KC und PM, nicht jedoch in BMDM. In COX2-defizienten PM wurde zudem eine verminderte LPS-abhängige Expression des Cytokins OSM nachgewiesen. Demzufolge funktionierten sowohl in KC, als auch in PM die postulierten autokrinen Rückkopplungsschleifen, welche sowohl die Entzündung dämpfen (Hemmung von TNFα), als auch verstärken (Steigerung von OSM) könnten.
In vivo nahm im Verlauf der Fütterung einer NASH-induzierenden Diät in WT-Mäusen der Anteil an KC (Tim4+Clec4F+) ab, wohingegen der Anteil an Monozyten-abgeleiteten infiltrierenden Makrophagen (Tim4-Clec4F-) anstieg. Darüber hinaus wurden Monozyten-abgeleitete infiltrierende Makrophagen mit Kupfferzell-ähnlichem Phänotyp (Tim4-Clec4F+) in den Lebern NASH-Diät gefütterter Mäuse nachgewiesen, deren Anteil im Verlauf der Fütterung ebenfalls zunahm.
Zusammengenommen konnte gezeigt werden, dass sich PM und BMDM, welche als Modell für infiltrierende Makrophagen angesehen werden, im Gegensatz zu KC durch eine sehr hohe Cytokin- und Chemokin-Expression in Folge der LPS-abhängigen Aktivierung auszeichneten. Gleichzeitig reagierten PM und BMDM jedoch sehr sensitiv auf PGE2. Da der Anteil infiltrierender Makrophagen im Verlauf der NASH zunimmt, könnte eine PGE2-abhängige Hemmung der Expression pro-inflammatorischer Cytokine wie TNFα in diesen Makrophagen einen potentiell protektiven, anti-inflammatorischen Effekt darstellen. Eine Hemmung der endogenen PGE2-Synthese in Makrophagen aufgrund einer COX2-Defizienz könnte die Entzündung im Rahmen der NASH verstärken und die Progression der NASH fördern.Steatohepatitis (NASH), a progressive form of non-alcoholic fatty liver disease, is characterised by chronic inflammation that is primarily orchestrated by macrophages in the liver. Inflammation-activated resident liver macrophages and recruited, infiltrating monocyte-derived macrophages secrete pro-inflammatory cytokines such as tumor necrosis factor α (TNFα) and the lipid mediator prostaglandin E2 (PGE2). PGE2, in turn, can modulate the expression of pro-inflammatory cytokines via autocrine feedback loops. The aim of this project was to investigate the PGE2-dependent regulation of cytokine expression in different macrophage populations isolated from mice in which cyclooxygenase 2 (COX2), the key enzyme of prostanoid synthesis, was specifically deleted in macrophages. Additionally, changes in the composition of the hepatic macrophage pool in mice with diet-induced NASH were characterised using flow cytometry. Initially, Kupffer cells (KC), peritoneal macrophages (PM) and bone marrow-derived macrophages (BMDM) were isolated from wild-type (WT) and COX2-deficient mice and were subsequently stimulated with lipopolysaccharide (LPS) ± exogenous PGE2. BMDM required a 10-fold higher LPS concentration for activation compared to KC and PM. Nevertheless, KC showed lower LPS-dependent expression of pro-inflammatory cytokines such as TNFα, interleukin (IL) 1β, IL-6 and oncostatin M (OSM) as well as the chemokine macrophage inflammatory protein 2 (MIP2), compared to the other two macrophage populations. LPS-induced gene expression and the secretion of TNFα were dose-dependently inhibited by exogenous PGE2 in all three macrophage populations. The mean inhibitory concentration of PGE2 was approximately 10-fold and 100-fold higher in KC than in BMDM and PM, respectively. In KC, exogenous PGE2 repressed the LPS-induced expression of IL-1β, IL-6 and MIP2, while in PM, it induced the expression of these mediators as well as OSM. LPS-induced endogenous PGE2 synthesis was completely abolished in COX2-deficient PM and BMDM and was reduced by about 60 % in COX2-deficient Kupffer cells. As a result of the loss of PGE2-mediated feedback inhibition, LPS-induced TNFα expression was elevated in COX2-deficient KC and PM, but not in BMDM. Furthermore, LPS-induced OSM expression was reduced in COX2-deficient PM. These findings suggest that the proposed autocrine feedback loops, capable of dampening inflammation (via inhibition of TNFα) or amplifying it (via induction of OSM), are functional in both KC and PM.
It was determined that the feeding of a NASH-inducing diet in WT mice decreased the proportion of KC (Tim4+Clec4F+) in vivo, while it increased the proportion of monocyte-derived infiltrating macrophages (Tim4-Clec4F-). In addition, monocyte-derived infiltrating macrophages with a Kupffer cell-like phenotype (Tim4-Clec4F+) were detected in the livers of NASH diet-fed mice, with their proportion increasing during MASH development.
In summary, PM and BMDM, which are considered models for infiltrating macrophages, were characterised by significantly increased cytokine and chemokine expression after LPS-dependent activation in contrast to KC. At the same time, PM and BMDM were highly sensitive to PGE2. As the proportion of infiltrating macrophages increases during NASH progression, PGE2-mediated inhibition of the expression of pro-inflammatory cytokine such as TNFα in these cells may represent a protective, anti-inflammatory effect. Conversely, COX2 deficiency and the resulting suppression of endogenous PGE2 synthesis in macrophages may exacerbate liver inflammation and promote the progression of NASH

Divine ontology and multicultural representation in the Marvel cinematic universe
The Marvel Cinematic Universe (‘MCU’) is one of the most lucrative entertainment franchises ever, and shapes the pop-cultural imagination to no small degree. This paper examines the MCU’s receptions of divine and supernatural characters from two interrelated research foci: (1) divine ontology, and (2) multicultural representations of premodern religions and civilizations. The first topic examines how the MCU remediates godhood: what does it mean to be a god in the MCU? Where lies the distinction between gods, heroes, and superheroes? What are the socio-cultural implications of these conceptualizations? We argue that the MCU currently exhibits a variety of divine definitions and modalities. The second topic investigates how the MCU represents various premodern cultures and mythologies. It demonstrates how the MCU’s storytelling and representation of (inter)mythological communities are driven (but not entirely subsumed) by white and Western cultural concerns. In sum, this paper investigates contemporary meanings attached to premodern religion and mythology by one of the largest entertainment franchises, illuminating not only how these meanings stand in dialogue with evidence but also what they reveal about contemporary society and how present-day creators look towards world mythologies
Agility in public administration
Agilität ist eine weit verbreitete Managementmethode, die in jüngster Zeit auch in der öffentlichen Verwaltung zunehmend Anwendung findet. Daher ist das allgemeine Forschungsinteresse an den Auswirkungen von Agilität in der öffentlichen Verwaltung sehr hoch. Aus diesem Grund widmet sich die vorliegende Arbeit der Untersuchung des Einflusses von Agilität auf die organisationalen Fähigkeiten in der öffentlichen Verwaltung. Zu diesem Zweck wird eine Literaturdiskussion durchgeführt, die sich in die Ebenen Individuum, Team und Organisation gliedert. Die vorliegende Einteilung gründet auf dem theoretischen Rahmen des organisationalen Lernens, dessen Implikationen der vorliegende Beitrag zu erörtern sucht. Zum anderen fungiert das von Amit und Schoemaker (1993) entwickelte Konstrukt der Organisational Capabilities als Diskussionsgrundlage. Im Rahmen der Diskussion wurde der Dreischritt angewandt, der den Zusammenhang zwischen den Variablen erklärt. Die Ergebnisse der Literaturdiskussion zeigen, dass Agilität in der öffentlichen Verwaltung auf allen untersuchten Ebenen einen Effekt auf die organisationalen Fähigkeiten hat. Der Effekt manifestiert sich insbesondere auf individueller Ebene, während die Ergebnisse auf Organisationsebene nicht im gleichen Maße einen Zusammenhang mit der Variable Agilität aufweisen. Die Ergebnisse der Literaturarbeit sind als Ansätze zu verstehen, die einer weiteren empirischen Überprüfung bedürfen, um den Zusammenhang zwischen den Variablen eindeutig darzustellen.Agility is a widely used management method that has recently been increasingly applied in public administration. As a result, there is considerable general research interest in the effects of agility in public administration. For this reason, this paper examines the influence of agility on organisational capabilities in public administration. To this end, a literature review is conducted, which is divided into the levels of individual, team and organisation. This classification is based on the theoretical framework of organisational learning, the implications of which this paper seeks to discuss. On the other hand, the construct of organisational capabilities developed by Amit and Schoemaker (1993) serves as a basis for discussion. The discussion applies a three-step approach to explain the relationship between the variables. The results of the literature review show that agility in public administration has an effect on organisational capabilities at all levels examined. The effect is particularly evident at the individual level, while the results at the organisational level do not show the same degree of correlation with the variable of agility. The results of the literature review should be understood as approaches that require further empirical testing in order to clearly demonstrate the relationship between the variables
Untersuchung der Folgen eines DEP-1-Mangels auf die Insulin-Signalübertragung im Gehirn und den peripheren Stoffwechsel
Energy homeostasis is regulated by a complex system centered in the brain, where insulin and AMP-activated protein kinase (AMPK) play a critical role in informing the brain of the energy status of the body and coordinate adaptive changes in food intake and energy expenditure in response to altered metabolic conditions. Disruptions in these pathways can impair the central regulation of energy intake, storage, and expenditure, leading to brain insulin resistance, a condition strongly associated with obesity and its related comorbidities. Protein tyrosine phosphatases (PTPs) are often upregulated in insulin-resistant states, with a subset of PTPs interacting directly with the insulin receptor (IR). Among these, Density-Enhanced Phosphatase-1 (DEP-1), a ubiquitously expressed receptor-like tyrosine phosphatase, has been identified as a negative regulator of IR signaling in peripheral tissues, though its role in brain metabolism remains unclear.
This thesis aimed to investigate the molecular mechanisms through which DEP-1 deficiency modulates brain insulin signaling and its subsequent effects on energy metabolism. To explore the functional consequences of DEP-1 deficiency on metabolism in vitro a DEP-1 knockout (KO) model was established in mouse immortalized undifferentiated Neuro-2a cells using CRISPR/Cas9 technology. Furthermore, considering the high expression of DEP-1 and IR in the forebrain, the metabolic effects of DEP-1 deficiency in these brain regions were examined by deleting Dep-1 using CamKIIa Cre mice, in both sexes, to generate forebrain-specific DEP-1 knockout mice (DEFO KO) which were fed a normal chow diet.
In vitro, DEP-1 KO neuronal cells demonstrated increased IR phosphorylation and downstream signaling upon acute insulin stimulation, along with unexpected activation of the AMPK cascade due to enhanced phospholipase C gamma 1 (Plcg 1) signaling. While in vivo, no significant alterations in key metabolic parameters, such as body weight, glucose tolerance, insulin sensitivity, food intake, and activity levels were observed in either sex. However, female DEFO KO mice demonstrated increased fat depots and enhanced fat oxidation. Organ specific analysis revealed male DEFO KO mice to have heightened insulin and AMPK signaling in random-fed cortical sections (a region where DEP-1 was deleted). Peripheral manifestations included elevated fatty acid oxidation in brown adipose tissue (BAT) and enhanced lipolysis in white adipose tissue (WAT), despite maintaining normal insulin sensitivity. Conversely, female DEFO KO mice exhibited enhanced insulin signaling solely in the hippocampus (another region where DEP-1 was deleted), alongside signs of energy deprivation characterized by increased lipolytic gene expression in WAT and elevated ketogenic markers in the liver, despite being fed ad libitum. Both male and female DEFO KO mice displayed higher levels of beta-adrenergic receptor expression in BAT, WAT, and the liver, accompanied by elevated serum norepinephrine levels, suggesting heightened sympathetic nervous system activity.
In conclusion, at a molecular level DEP-1 was identified as a novel neuronal suppressor of simultaneous activation of both insulin and AMPK signaling by regulating Plcg 1 activity. Additionally, DEP-1 deficiency in the forebrain triggers a stress signal due to the parallel activation of anabolic and catabolic pathways, resulting in lipolysis with increased adipose tissue mass in females and enhanced BAT activity in males.Die Energiehomöostase wird durch ein komplexes zentralnervöses System reguliert, wo Insulin und die AMP-aktivierte Proteinkinase (AMPK) eine entscheidende Rolle dabei spielen, das Gehirn über den Energiestatus des Körpers zu informieren und adaptive Veränderungen der Nahrungsaufnahme und des Energieverbrauchs als Reaktion auf veränderte Stoffwechselbedingungen zu koordinieren. Störungen in diesen Signalwegen können die zentrale Regulation der Energieaufnahme, -speicherung und -ausgabe beeinträchtigen und zu einer Insulinresistenz des Gehirns führen, einem Zustand, der stark mit Fettleibigkeit und den damit verbundenen Komorbiditäten in Verbindung gebracht wird. Protein-Tyrosin-Phosphatasen (PTPs) werden bei Insulinresistenz häufig hochreguliert, wobei eine Untergruppe von PTPs direkt mit dem Insulinrezeptor (IR) interagiert. Unter diesen wurde die Density-Enhanced Phosphatase-1 (DEP-1), eine ubiquitär exprimierte rezeptorähnliche Tyrosinphosphatase, als negativer Regulator der IR-Signalübertragung in peripheren Geweben identifiziert, wobei ihre Rolle im Gehirnstoffwechsel noch unklar ist.
Diese Arbeit zielte darauf ab, die molekularen Mechanismen und Folgen auf die Insulinsignalkaskade sowie den Stoffwechsel zu untersuchen, die durch einen DEP-1-Mangel entstehen. Um die funktionellen Folgen eines DEP-1-Mangels auf den Stoffwechsel in vitro zu untersuchen, wurde ein DEP-1-Knockout (KO)-Modell in immortalisierten undifferenzierten Neuro-2a-Zellen der Maus unter Verwendung der CRISPR/Cas9-Technologie etabliert. Angesichts der hohen Expression von DEP-1 und IR im Vorderhirn wurden die metabolischen Auswirkungen eines DEP-1-Mangels in diesen Hirnregionen untersucht. DEP-1 wurdein CamKIIa-Cre-Mäusen beiderlei Geschlechts deletiert, um vorderhirnspezifische DEP-1-Knockout-Mäuse (DEFO-KO-Mäuse) zu erzeugen, die mit normalem Haltungsdiät ernährt wurden.
In vitro zeigten neuronale DEP-1-KO-Zellen eine erhöhte Insulinresistenz-Phosphorylierung und nachgeschaltete Signalübertragung bei akuter Insulinstimulation sowie eine unerwartete Aktivierung der AMPK-Kaskade aufgrund einer verstärkten Phospholipase-C-Gamma-1-Signalübertragung (Plcg 1). In vivo wurden bei beiden Geschlechtern keine signifikanten Veränderungen der wichtigsten Stoffwechselparameter wie Körpergewicht, Glukosetoleranz, Insulinsensitivität, Nahrungsaufnahme und Aktivitätsniveau beobachtet. Weibliche DEFO-KO-Mäuse wiesen jedoch erhöhte Fettdepots und eine verstärkte Fettoxidation auf. Eine organspezifische Analyse ergab, dass ad libitum gefütterte männliche DEFO-KO-Mäuse in kortikalen Abschnitten (eine Region, in der DEP-1 deletiert wurde) eine erhöhte Insulin- und AMPK-Signalübertragung aufwiesen. Zu den peripheren Manifestationen gehörten eine erhöhte Fettsäureoxidation im braunen Fettgewebe (BAT) und eine verstärkte Lipolyse im weißen Fettgewebe (WAT), obwohl die Insulinsensitivität normal blieb. Im Gegensatz dazu zeigten weibliche DEFO-KO-Mäuse eine verstärkte Insulinsignalisierung ausschließlich im Hippocampus (eine weitere Region, in der DEP-1 deletiert wurde), zusammen mit Anzeichen von Energiemangel, der durch eine erhöhte lipolytische Genexpression im weißen Fettgewebe und erhöhte ketogene Marker in der Leber gekennzeichnet war, obwohl sie ad libitum gefüttert wurden. Sowohl männliche als auch weibliche DEFO-KO-Mäuse wiesen eine höhere Expression von beta-adrenergen Rezeptoren in BAT, WAT und der Leber auf, begleitet von erhöhten Noradrenalinspiegeln im Serum, was auf eine erhöhte Aktivität des sympathischen Nervensystems hindeutet.
Zusammenfassend wurde DEP-1 auf molekularer Ebene als neuartiger neuronaler Suppressor der gleichzeitigen Aktivierung von Insulin- und AMPK-Signalwegen durch Regulierung der Plcg-1-Aktivität identifiziert. Darüber hinaus löst ein DEP-1-Mangel im Vorderhirn aufgrund der parallelen Aktivierung anaboler und kataboler Signalwege ein Stresssignal aus, das bei weiblichen Mäusen zu einer Lipolyse mit erhöhter Fettgewebsmasse und bei männlichen Mäusen zu einer verstärkten Aktivität der BAT führt
Synergien des Sediment- und Kohlenstoffkreislaufs im Río Bermejo, Argentinien
Rivers transport sediments and organic matter from continental sources to marine sinks. During fluvial transport and temporary storage on floodplains, these materials undergo transformations, for instance, the conversion of particulate into dissolved forms, or molecular alterations of organic matter. Such processes influence sediment fluxes and biogeochemical cycles. The carbon cycle, a central regulator of Earth’s climate, is closely linked to the sedimentary cycle. Short-term carbon exchange, organic-clastic binding over centuries, and silicate weathering that fixes atmospheric CO2 over millennia can all be activated during floodplain storage, making floodplains relevant players in the carbon balance.
This dissertation examines how sediment type, floodplain morphology, and seasonality govern carbon cycling in the foreland floodplain of the Río Bermejo in Argentina. Flowing over 1,300 km through a foreland basin without significant tributary or distributary input, the Río Bermejo is characterized by high suspended sediment loads and extensive floodplain deposition. This makes it an ideal system to investigate synergies between sediment and carbon dynamics. Using organic and inorganic proxies as well as stable isotopes, the study tracks particulate, dissolved, and gaseous carbon both in-stream and on the floodplain.
The results show: while coarse bedload is rapidly exported along the riverbed, key transformations of organic matter occur in fine, suspendable material, particularly during intermittent floodplain deposition. Labile organic matter is respired, long-stored carbon is mobilized, and silicate weathering is promoted. These processes are strongly modulated by hydrological seasonality. Integrating carbon fluxes with sediment transport provides deeper insight into carbon mobilization and release in dynamic fluvial systems.Flüsse transportieren Sedimente und organische Substanz von kontinentalen Quellen zu marinen Senken. Kurzfristige C-Austauschprozesse, organisch-klastische Bindung über Jahrhunderte sowie silikatische CO2-Fixierung über Jahrtausende können alle in Auen aktiviert werden, was diese zu relevanten Akteuren im Kohlenstoffhaushalt macht.
Diese Dissertation untersucht, wie Sedimenttyp, Auenmorphologie und Saisonalität den Kohlenstoffkreislauf in der Vorlandflussaue des Río Bermejo (Argentinien) steuern. Der Río Bermejo fließt über 1300 km durch ein Vorlandbecken ohne nennenswerte Zu- oder Abflüsse. Die Kombination aus hoher Schwebstofffracht und großräumiger Sedimentakkumulation im Vorland schafft ein ideales System, um Synergien zwischen Sediment- und Kohlenstoffdynamiken zu untersuchen. Anhand organischer und anorganischer Proxys sowie stabiler Isotopen werden partikulärer, gelöster und gasförmiger Kohlenstoff im Fluss und auf der Aue analysiert.
Die Ergebnisse zeigen: Während grobe Geschiebefracht am Flussbett rasch exportiert wird, findet die entscheidende Umwandlung organischer Substanz im feinkörnigen, schwebfähigen Material statt, insbesondere währened intermittierender Auenablagerung. Labile organische Substanz wird dort respirativ umgesetzt, langzeitig gespeicherter Kohlenstoff mobilisiert und Silikatverwitterung begünstigt. Diese Prozesse werden stark von hydrologischer Saisonalität beeinflusst. Die Betrachtung von Kohlenstoffflüssen im Kontext von Sedimenttransport ermöglicht ein vertieftes Verständnis von Kohlenstoffmobilisierung und -freisetzung in dynamischen Flusssystemen
Regelungen zur Förderung der Mediation
Das Gesetz zur Förderung der Mediation und anderer Verfahren der außergerichtlichen Konfliktbeilegung verfehlt das Ziel, das ihm den Namen gab. Zur Behebung der konzeptionellen Mängel des geltenden Rechts gestaltet der Autor ein umfassendes, kohärentes und gleichzeitig maßvolles Regelungskonzept für eine adäquate Förderung der Mediation.
Dafür wird zwischen indirekten, insbesondere kostenassoziierten Förderungsmöglichkeiten einerseits und der direkten Förderung durch Mediationskostenhilfe andererseits differenziert, wobei die bestehenden Regelungen der ZPO und des FamFG als Referenzen fruchtbar gemacht werden. Die auf dieser Grundlage entwickelten Normen werden in einen Gesetzesentwurf gegossen, der vollständig übernommen werden kann, wenn sich der Gesetzgeber auf den Weg macht, Mediation effektiv zu fördern
