    Genome-wide analysis, expansion and expression of the NAC family under drought and heat stresses in bread wheat (T. aestivum L.)

    The NAC family is one of the largest plant-specific transcription factor families, and some of its members are known to play major roles in plant development and response to biotic and abiotic stresses. Here, we inventoried 488 NAC members in bread wheat (Triticum aestivum). Using the recent release of the wheat genome (IWGS RefSeq v1.0), we studied duplication events focusing on genomic regions from 4B-4D-5A chromosomes as an example of the family expansion and neofunctionalization of TaNAC members. Differentially expressed TaNAC genes in organs and in response to abiotic stresses were identified using publicly available RNAseq data. Expression profiling of 23 selected candidate TaNAC genes was studied in leaf and grain from two bread wheat genotypes at two developmental stages in field drought conditions and revealed insights into their specific and/or overlapping expression patterns. This study showed that, of the 23 TaNAC genes, seven have a leaf-specific expression and five have a grain-specific expression. In addition, the grain-specific genes profiles in response to drought depend on the genotype. These genes may be considered as potential candidates for further functional validation and could present an interest for crop improvement programs in response to climate change.     When sinks become sources: adaptive colonization in asexuals

    The successful establishment of a population into a new empty habitat outside of its initial niche is a phenomenon akin to evolutionary rescue in the presence of immigration. It underlies a wide range of processes, such as biological invasions by alien organisms, host shifts in pathogens or the emergence of resistance to pesticides or antibiotics from untreated areas.[br/] In this study, we derive an analytically tractable framework to describe the coupled evolutionary and demographic dynamics of asexual populations in a source-sink system. In particular, we analyze the influence of several factors — immigration rate, mutational parameters, and harshness of the stress induced by the change of environment — on the establishment success in the sink (i.e. the formation of a self-sufficient population in the sink), and on the time until establishment. To this aim, we use a classic phenotype-fitness landscape (Fisher’s geometrical model in n dimensions) where source and sink habitats determine distinct phenotypic optima. The harshness of stress, in the sink, is determined by the distance between the fitness optimum in the sink and that of the source. The dynamics of the full distribution of fitness and of population size in the sink are analytically predicted under a strong mutation strong immigration limit where the population is always polymorphic.[br/] The resulting eco-evolutionary dynamics depend on mutation and immigration rates in a non straightforward way. Below some mutation rate threshold, establishment always occurs in the sink, following a typical four-phases trajectory of the mean fitness. The waiting time to this establishment is independent of the immigration rate and decreases with the mutation rate. Beyond the mutation rate threshold, lethal mutagenesis impedes establishment and the sink population remains so, albeit with an equilibrium state that depends on the details of the fitness landscape. We use these results to get some insight into possible effects of several management strategies

    Functional, Structural and Biochemical Features of Plant Serinyl-Glutathione Transferases.

    Glutathione transferases (GSTs) belong to a ubiquitous multigenic family of enzymes involved in diverse biological processes including xenobiotic detoxification and secondary metabolism. A canonical GST is formed by two domains, the N-terminal one adopting a thioredoxin (TRX) fold and the C-terminal one an all-helical structure. The most recent genomic and phylogenetic analysis based on this domain organization allowed the classification of the GST family into 14 classes in terrestrial plants. These GSTs are further distinguished based on the presence of the ancestral cysteine (Cys-GSTs) present in TRX family proteins or on its substitution by a serine (Ser-GSTs). Cys-GSTs catalyze the reduction of dehydroascorbate and deglutathionylation reactions whereas Ser-GSTs catalyze glutathione conjugation reactions and eventually have peroxidase activity, both activities being important for stress tolerance or herbicide detoxification. Through non-catalytic, so-called ligandin properties, numerous plant GSTs also participate in the binding and transport of small heterocyclic ligands such as flavonoids including anthocyanins, and polyphenols. So far, this function has likely been underestimated compared to the other documented roles of GSTs.     An unexpected case of Bartonella alsatica prosthetic vascular graft infection

    Bartonella alsatica is a wild rabbit pathogen causing bacteremia rarely reported in humans, with only three cases published so far, including one lymphadenitis and two endocarditis cases. Here, we report the case of a 66-year-old man who suffered from acute renal failure due to a membranoproliferative glomerulonephritis. Fluorodeoxyglucose (FDG) positron emission tomography/computed tomography (PET/CT) showed diffuse FDG uptake around the aortobifemoral graft with no indication of infection. A white blood cell scan showed an accumulation of labeled neutrophils on the left femoral part of the graft. The patient underwent surgery and an abscess around the left iliac part of the graft was found intraoperatively. Intraoperative samples were all negative, but 16S rRNA gene-based PCR was positive, and the sequence was positioned among the Bartonella species cluster. Specific PCRs targeting groEL/hsp60, rpoB and gltA genes were performed and led to the identification of B. alsatica. Accordingly, indirect immunofluorescence serological analyses were positive for Bartonella henselae and Bartonella quintana. The patient had a history of regularly hunting wild rabbits. He was treated with 100 mg of doxycycline twice a day for six months and his renal function significantly improved with no sign of persistent infection. This case highlights the contribution of serology assays and molecular-based methods in prosthetic vascular graft infection diagnosis

    Organising the generalisation of distributed and decentralised technological innovations: Distributed and decentralised ex novation. : The critical phase of the deinstitutionalisation of pesticide uses in France

    This communication is grounded on previous elaboration of the notion of out-novation (Levain et al., 2015) or ex-novation (Wolff et al., 2007; Martin et al., 2016) after the seminal inputs of reflection about de-institutionalization process (Maguire et Hardy, 2009) and the idea of detachment in innovation process (Goulet & Vinck, 2012). The aim is to tackle the pending issues of sustainability transition where intermediaries have a key role in “pushing and tracking” transformative policy towards the abandonment of established but weak practices within a stable sociotechnical regime. Many sustainability transition paradox and challenges are at stake in the present agri-food system, calling for more research on intermediation and learning processes in a call for generalization of sustainable practices (Elzen et al., 2012 ; Hermans et al. 2013 ; Klerkx ert al. 2010). The role and function of intermediaries in concomitant deinstitutionalisation process of certain practices and out-scaling process of novel practices has not been that much put under empirical investigation and intermediation in transition suffers from a lack of theorizing (Steyeart et al., 2016). This communication intends to frame an answer based on the case study of intermediation of pesticide use in France during the last decade (Cerf et al., 2017). It reports about a longitudinal study with participatory momentum inclined to elicit the views, practices and conditions of action of those sustainability transition intermediaries. Some key findings point the need to open the understanding of the chasm of out-novation. In many circumstances the dynamic of sociotechnical regime and de facto governance of transformative process, do not mean only de novo creation, nor substitution, but more often a subtle agency and structure of de-framing, re-agencing and including critical mind-sets, new sociotechnical agency and human practices (Turnheim et al., 2015). The chasm of innovation (Bernstein &Singh, 2008) has its counter part in what one could name the chasm of out-novation or ex novation. As a generalisation process relays on the often silent but determinant contributions and actions of innovation brokers, sustainability transitions supposes intermediaries that convey the uptake of a critical zone of out-scaling. This perspective has to be clearly put upper in the agenda of Transition Studies

    Towards a reflective approach to research project management

    This chapter describes how we supported the project leaders of TATA-BOX in their task of designing a management system for the project. We did so by fuelling their reflectivity: rather than making suggestions on how to manage the project – in a normative approach –, we analysed the on-going project management and mirrored what had been done after a year. The TATA-BOX project leaders would thus be able to decide how to adjust their management and to carry on – in a reflective approach. We report on this process in this chapter: after giving some theoretical background on the concept of reflectivity and its role in helping the project leaders to manage TATA-BOX, we describe: (1) how we worked with them over 6 months, 1 year after the project began, and (2) the different methods we used to meet the project leaders’ expectations. We then discuss the efficiency of these methods, their effects on the management of the project, and some lessons learned for the management of such research projects generally

    Blood circulation mainly aims at distributing the nutrients required for tissue metabolism and collecting safely the by-products of all tissues to be further metabolized or eliminated. The simultaneous study of arterial (A) and venous (V) specific metabolites therefore has appeared to be a more relevant approach to understand and study the metabolism of a given organ. We propose to implement this approach by applying a metabolomics (NMR) strategy on paired AV blood across the intestine and liver on high fat/high sugar (HFHS)-fed minipigs. Our objective was to unravel kinetically and sequentially the metabolic adaptations to early obesity/insulin resistance onset specifically on these two tissues. After two months of HFHS feeding our study of AV ratios of the metabolome highlighted three major features. First, the hepatic metabolism switched from carbohydrate to lipid utilization. Second, the energy demand of the intestine increased, resulting in an enhanced uptake of glutamine, glutamate, and the recruitment of novel energy substrates (choline and creatine). Third, the uptake of methionine and threonine was considered to be driven by an increased intestine turnover to cope with the new high-density diet. Finally, the unique combination of experimental data and modelling predictions suggested that HFHS feeding was associated with changes in tryptophan metabolism and fatty acid β-oxidation, which may play an important role in lipid hepatic accumulation and insulin sensitivity

    Multi-resolution study of thermal unmixing techniques over Madrid Urban Area: case study of TRISHNA mission

    This work is linked to the future Indian-French high spatio-temporal TRISHNA (Thermal infraRed Imaging Satellite for High-resolution natural resource Assessment) mission, which includes shortwave and thermal infrared bands, and is devoted amongst other things to the monitoring of urban heat island events. In this article, the performance of seven empirical thermal unmixing techniques applied on simulated TRISHNA satellite images of an urban scenario is studied across spatial resolutions. For this purpose, Top Of Atmosphere (TOA) images in the shortwave and Thermal InfraRed (TIR) ranges are constructed at different resolutions (20 m, 40 m, 60 m, 80 m, and 100 m) and according to TRISHNA specifications (spectral bands and sensor properties). These images are synthesized by correcting and undersampling DESIREX 2008 Airborne Hyperspectral Scanner (AHS) images of Madrid at 4 m resolution. This allows to compare the Land Surface Temperature (LST) retrieval of several unmixing techniques applied on different resolution images, as well as to characterize the evolution of the performance of each technique across resolutions. The seven unmixing techniques are: Disaggregation of radiometric surface Temperature (DisTrad), Thermal imagery sHARPening (TsHARP), Area-To-Point Regression Kriging (ATPRK), Adaptive Area-To-Point Regression Kriging (AATPRK), Urban Thermal Sharpener (HUTS), Multiple Linear Regressions (MLR), and two combinations of ground classification (index-based classification and K-means classification) with DisTrad. Studying these unmixing techniques across resolutions also allows to validate the scale invariance hypotheses on which the techniques hinge. Each thermal unmixing technique has been tested with several shortwave indices, in order to choose the best one. It is shown that (i) ATPRK outperforms the other compared techniques when characterizing the LST of Madrid, (ii) the unmixing performance of any technique is degraded when the coarse spatial resolution increases, (iii) the used shortwave index does not strongly influence the unmixing performance, and (iv) even if the scale-invariant hypotheses behind these techniques remain empirical, this does not affect the unmixing performances within this range of resolutions

    Control of paratuberculosis: who, why and how. A review of 48 countries

    Paratuberculosis, a chronic disease affecting ruminant livestock, is caused by Mycobacterium avium subsp. paratuberculosis (MAP). It has direct and indirect economic costs, impacts animal welfare and arouses public health concerns. In a survey of 48 countries we found paratuberculosis to be very common in livestock. In about half the countries more than 20% of herds and flocks were infected with MAP. Most countries had large ruminant populations (millions), several types of farmed ruminants, multiple husbandry systems and tens of thousands of individual farms, creating challenges for disease control. In addition, numerous species of free-living wildlife were infected. Paratuberculosis was notifiable in most countries, but formal control programs were present in only 22 countries. Generally, these were the more highly developed countries with advanced veterinary services. Of the countries without a formal control program for paratuberculosis, 76% were in South and Central America, Asia and Africa while 20% were in Europe. Control programs were justified most commonly on animal health grounds, but protecting market access and public health were other factors. Prevalence reduction was the major objective in most countries, but Norway and Sweden aimed to eradicate the disease, so surveillance and response were their major objectives. Government funding was involved in about two thirds of countries, but operations tended to be funded by farmers and their organizations and not by government alone. The majority of countries (60%) had voluntary control programs. Generally, programs were supported by incentives for joining, financial compensation and/or penalties for non-participation. Performance indicators, structure, leadership, practices and tools used in control programs are also presented. Securing funding for long-term control activities was a widespread problem. Control programs were reported to be successful in 16 (73%) of the 22 countries. Recommendations are made for future control programs, including a primary goal of establishing an international code for paratuberculosis, leading to universal acknowledgment of the principles and methods of control in relation to endemic and transboundary disease.
