    Spatiotemporal expression of the putative MdtABC efflux pump of Phtotorhabdus luminescens occurs in a protease-dependent manner during insect infection

    Photorhabdus luminescens is an enterobacterium establishing a mutualistic symbiosis with nematodes, that also kills insects after septicaemia and connective tissue colonization. The role of the bacterial mdtABC genes encoding a putative multidrug efflux system from the resistance/nodulation/cell division family was investigated. We showed that a mdtA mutant and the wild type had similar levels of resistance to antibiotics, antimicrobial peptides, metals, detergents and bile salts. The mdtA mutant was also as pathogenic as the wild-type following intrahaemocoel injection in Locusta migratoria, but had a slightly attenuated phenotype in Spodoptera littoralis. A transcriptional fusion of the mdtA promoter (P-mdtA) and the green fluorescent protein (gfp) encoding gene was induced by copper in bacteria cultured in vitro. The P-mdtA-gfp fusion was strongly induced within bacterial aggregates in the haematopoietic organ during late stages of infection in L. migratoria, whereas it was only weakly expressed in insect plasma throughout infection. A medium supplemented with haematopoietic organ extracts induced the P-mdtA-gfp fusion ex vivo, suggesting that site-specific mdtABC expression resulted from insect signals from the haematopoietic organ. Finally, we showed that protease inhibitors abolished ex vivo activity of the P-mdtA-gfp fusion in the presence of haematopoietic organ extracts, suggesting that proteolysis by-products play a key role in upregulating the putative MdtABC efflux pump during insect infection with P. luminescens

    Modelling Nitrogen Uptake in Plants and Phytoplankton: Advantages of Integrating Flexibility into the Spatial and Temporal Dynamics of Nitrate Absorption

    Under field conditions, plants need to optimize nutrient ion and water acquisition in their fluctuating environment. One of the most important variables involved in variations of ion uptake processes is temperature. It modifies the thermodynamic processes of root uptake and ion diffusion in soil throughout day–night and ontogenetic cycles. Yet, most models of nitrogen (N) uptake in plants are built from set values of microscopic kinetic parameters, Vm and Km, derived from a Michaelis–Menten (MM) interpretation of nutrient isotherms. An isotherm is a curve depicting the response of root nitrate influx to external nitrate concentrations at a given temperature. Models using the MM formalism are based on several implicit assumptions that do not always hold, such as homothetic behavior of the kinetic parameters between the different root biological scales, i.e., the epidermis cell, root segments, root axes, and the whole root system. However, in marine phytoplankton, it has been clearly demonstrated that the macroscopic behavior in the nutrient uptake of a colony cannot be confounded with the microscopic behavior of individual cells, due to the cell diffusion boundary layer. The same is also true around plant root segments. Improved N uptake models should either take into account the flexibility of the kinetic parameters of nitrate uptake at the cellular level (porter–diffusion approach) or use the more realistic macroscopic kinetic parameters proposed by the flow–force approach. Here we present recent solutions proposed in marine phytoplankton and plant nutrient uptake models to make a more flexible description of the nutrient ion uptake process. Use of the mechanistic porter–diffusion approach developed in marine phytoplankton introduces more flexibility in response to cell characteristics and physical processes driven by temperature (diffusion and convection). The thermodynamic flow–force interpretation of plant-based nutrient uptake isotherms introduces more flexibility in response to environmental cues and root aging. These two approaches could help solve many problems that modelers encounter in these two research areas

    Microalgae and cyanobacteria modeling in water resource recovery facilities: A critical review

    Microalgal and cyanobacterial resource recovery systems could significantly advance nutrient recovery from wastewater by achieving effluent nitrogen (N) and phosphorus (P) levels below the current limit of technology. The successful implementation of phytoplankton, however, requires the formulation of process models that balance fidelity and simplicity to accurately simulate dynamic performance in response to environmental conditions. This work synthesizes the range of model structures that have been leveraged for algae and cyanobacteria modeling and core model features that are required to enable reliable process modeling in the context of water resource recovery facilities. Results from an extensive literature review of over 300 published phytoplankton models are presented, with particular attention to similarities with and differences from existing strategies to model chemotrophic wastewater treatment processes (e.g., via the Activated Sludge Models, ASMs). Building on published process models, the core requirements of a model structure for algal and cyanobacterial processes are presented, including detailed recommendations for the prediction of growth (under phototrophic, heterotrophic, and mixotrophic conditions), nutrient uptake, carbon uptake and storage, and respiration

    Contribution to the modeling and the mechanical characterization of the subsoil in the LSBB environment

    The present research work aims at better characterizing the specific underground environment of the LSBB (Low Noise Inter-Disciplinary Underground Science and Technology, Rustrel, France) using mechanical wave propagation information. The LSBB experimental environment is characterized by a system of cylindrical galleries, some of them presenting a concrete layer. In the global project, three steps are considered : firstly the construction of an efficient forward mechanical wave propagation model to calculate the displacement vector and stress tensor components; secondly a sensitivity analysis to extract the pertinent parameters in the configurations and models under study (viscoelastic or poroviscoelastic media with potential anisotropy); and lastly an inversion strategy to recover some of the pertinent parameters. In this proposal, the first step, under progress, is described. The work carried out is in the continuity of the work presented by Yi et al. (2016) [1] who studied the harmonic response of a cylindrical elastic tunnel, impacted by a plane compressional wave, embedded in an infinite elastic ground. The interface between the rock mass and the linen is an imperfect contact modeled with two spring parameters, Achenbach and Zhu (1989) [2]. We choose a semi-analytical approach to calculate the two-dimensional displacement and stress fields in order to get a fast tool, from the numerical point of view. The main steps of the theoretical approach are : use of the Helmholtz decomposition, solving the wave equation based on the separation method and the expansion in Bessel function series in the harmonic domain. The harmonic results are validated by comparison with Yi et al. (2016) [1] and new ones are presented. Moreover, the transient regime case obtained with the use of a Fourier transform on the time variable, is under progress

    Adaptive Introgression: An Untapped Evolutionary Mechanism for Crop Adaptation

    Global environmental changes strongly impact wild and domesticated species biology and their associated ecosystem services. For crops, global warming has led to significant changes in terms of phenology and/or yield. To respond to the agricultural challenges of this century, there is a strong need for harnessing the genetic variability of crops and adapting them to new conditions. Gene flow, from either the same species or a different species, may be an immediate primary source to widen genetic diversity and adaptions to various environments. When the incorporation of a foreign variant leads to an increase of the fitness of the recipient pool, it is referred to as "adaptive introgression". Crop species are excellent case studies of this phenomenon since their genetic variability has been considerably reduced over space and time but most of them continue exchanging genetic material with their wild relatives. In this paper, we review studies of adaptive introgression, presenting methodological approaches and challenges to detecting it. We pay particular attention to the potential of this evolutionary mechanism for the adaptation of crops. Furthermore, we discuss the importance of farmers' knowledge and practices in shaping wild-to-crop gene flow. Finally, we argue that screening the wild introgression already existing in the cultivated gene pool may be an effective strategy for uncovering wild diversity relevant for crop adaptation to current environmental changes and for informing new breeding directions

    The transcription factor NHR-8: A new target to increase ivermectin efficacy in nematodes

    Resistance to the anthelmintic macrocyclic lactone ivermectin (IVM) has a great impact on the control of parasitic nematodes. The mechanisms by which nematodes adapt to IVM remain to be deciphered. We have identified NHR-8, a nuclear hormone receptor involved in the xenobiotic response in Caenorhabditis elegans, as a new regulator of tolerance to IVM. Loss-of-function nhr-8(ok186) C. elegans mutants subjected to larval development assays and electropharyngeogram measurements, displayed hypersensitivity to IVM, and silencing of nhr-8 in IVM-resistant worms increased IVM efficacy. In addition, compared to wild-type worms, nhr-8 mutants under IVM selection pressure failed to acquire tolerance to the drug. In addition, IVM-hypersensitive nhr-8(ok186) worms displayed low transcript levels of several genes from the xenobiotic detoxification network and a concomitant low Pgp-mediated drug efflux activity. Interestingly, some pgp and cyp genes known to impact IVM tolerance in many nematode species, were down regulated in nhr-8 mutants and inversely upregulated in IVM-resistant worms. Moreover, pgp-6 overexpression in nhr-8(ok186) C. elegans increased tolerance to IVM. Importantly, NHR-8 function was rescued in nhr-8(ok186) C. elegans with the homolog of the parasitic nematode Haemonchus contortus, and silencing of Hco-nhr-8 by RNAi on L2 H. contortus larvae increased IVM susceptibility in both susceptible and resistant H. contortus isolates. Thus, our data show that NHR-8 controls the tolerance and development of resistance to IVM in C. elegans and the molecular basis for this relates to the NHR-8-mediated upregulation of IVM detoxification genes. Since our results show that Hco-nhr-8 functions similarly to Cel-nhr-8, this study helps to better understand mechanisms underlying failure in drug efficacy and open perspectives in finding new compounds with NHR-8 antagonist activity to potentiate IVM efficacy

    En 2012, la direction de l’enseigne Lidl en France annonçait un changement majeur de la stratégie du pionnier du « low cost » alimentaire. Un vaste plan est alors mis en place en termes d’investissement, d’organisation des magasins de communication et d’image du distributeur. Les perceptions des consommateurs de l’image de Lidl ont été recueillies via deux enquêtes (2011 et 2016). Nos résultats montrent un « repositionnement » perçu de l’image de Lidl et un gain relatif en compétitivité.In 2012, the French Lidl top management announced a major strategy change of the "low cost" food specialist. A comprehensive plan is then implemented both in terms of investment and organization of stores and in terms of communication and image change of the retailer. Based on two surveys of French consumers, we asked about consumer perceptions of the image of Food hard discounter of Lidl. Our results show a perceived "repositioning" Lidl, and a relative gain in competitiveness
