    Integrative Metabolomics for Assessing the Effect of Insect (Hermetia illucens) Protein Extract on Rainbow Trout Metabolism.

    Nutrition of high trophic species in aquaculture is faced with the development of sustainable plant-based diets. Insects seem particularly promising for supplementing plant-based diets. However, the complex effect of whole insect meal on fish metabolism is not well understood, and even less is known about insect meal extracts. The purpose of this work was to decipher the metabolic utilization of a plant-based diet supplemented with the gradual addition of an insect protein extract (insect hydrolysate at 0%, 5%, 10% and 15%). 1H-NMR profiling was used to assess metabolites in experimental diets and in fish plasma, liver and muscle. A significant dose-dependent increase in growth and feed efficiency with increasing insect extract amounts was observed. The incremental incorporation of the insect extract in diet had a significant and progressive impact on the profile of dietary soluble compounds and trout metabolome. The metabolites modulated by dietary insect extracts in plasma and tissues were involved in protein and energy metabolism. This was associated with the efficient metabolic use of dietary free amino acids toward protein synthesis through the concomitant supply of balanced free amino acids and energy substrates in muscle. The findings provide new insights into how the dietary food metabolome affects fish metabolism

    Role of the interferons in CD64 and CD169 expressions in whole blood: Relevance in the balance between viral- or bacterial-oriented immune responses

    Introduction CD64 expression increases on neutrophils during bacterial infections. Recently an increase in CD169 expression has been discovered on monocytes during viral infections. Generally, interferons alpha (IFNs alpha) and IFNs gamma are key drivers of the infectious host immune response. The purpose of this study was to explore if a link exists between these IFNs and both biomarkers. Methods Whole blood samples from healthy volunteers were stimulated with either IFNs, interleukins, or infectious extracts, to mimic an infectious state. Expressions of CD64 and CD169 were assessed in these samples by multiple flow cytometry methods, over precise kinetics. Results The expression of CD64 was statistically higher in samples stimulated with IFN gamma, and CD169 in those stimulated with IFN alpha (and all other type I IFNs). Surface expressions are directly induced by their respective IFNs via Janus kinase/signal transducer and activator of transduction pathways within 6 to 8 hours of incubation. Mixing both types of IFNs seemed to indicate that they partially inhibit each other. Conclusions The induction of CD169 on monocytes and CD164 on neutrophils by type I and type II IFNs confirms the relevance of these markers for assessing between a viral- vs bacterial-oriented immune response

    The multicomponent medication Spascupreel attenuates stress-induced gut dysfunction in rats

    Background Irritable bowel syndrome (IBS) is a common disorder worldwide. It is characterized by abdominal pain/discomfort and changes in bowel habits. Due to the multifactorial pathophysiology and the heterogeneity of IBS patients, appropriate treatment of IBS is still a challenge. Spascupreel (SP-11), as a multicomponent medication, has the potential to modulate multiple pathophysiological pathways simultaneously. Therefore, the objective of the current study was to investigate the effects of oral SP-11 treatment on stress-induced changes of peripheral and central functions in a rat model mimicking human IBS. Methods Naive Wistar rats were treated with SP-11 (0.9 tab/kg) or NaCl 0.9% by oral gavage for 4 days before 2-hour partial restraint stress (PRS) procedure. Twenty minutes after PRS, central and peripheral stress-induced changes affecting IBS were assessed. These include the hypothalamic-pituitary-adrenal (HPA) axis response through plasma ACTH and corticosterone measurements, visceral pain in response to colorectal distension, gut permeability, colonic mast cell number, and sensitization as well as gut transit time. Results Treatment with SP-11 reduced the HPA axis activation in response to PRS. At the gut level, a reduction in colonic hypersensitivity to colorectal distension, a normalization of gut transit time acceleration, a reduced mast cell sensitization, and a trend toward reduced gut hyperpermeability were observed. Conclusions These data suggest that stress-induced IBS signs can be reduced using SP-11 in rats. The observed effects and the good tolerability of the drug make SP-11 an innovative candidate in the management of IBS

    Insect pollination: an ecological process involved in the assembly of the seed microbiota

    The assembly of the seed microbiota involves some early microbial seed colonizers that are transmitted from the maternal plant through the vascular system, while other microbes enter through the stigma. Thus, the seed microbiota consists of microbes not only recruited from the plant vascular tissues, but also from the flower. Flowers are known to be a hub for microbial transmission between plants and insects. This floral-insect exchange opens the possibility for insect-transmitted bacteria to colonize the ovule and, subsequently, the seed to pass then into the next plant generation. In this study, we evaluated the contribution of insect pollination to the seed microbiota through high-throughput sequencing. Oilseed rape (OSR) flowers were exposed to visits and pollination by honey bees (Apis mellifera), red mason bees (Osmia bicornis), hand pollinated or left for autonomous self-pollination (ASP). Sequence analyses revealed that honey bee visitation reduced bacterial richness and diversity in seeds, but increased the variability of seed microbial structure, and introduced bee-associated taxa. In contrast, mason bee pollination had minor effects on the seed microbiota. Our study provides the first evidence that insect pollination is an ecological process involved in the transmission of bacteria from flowers to seeds

    Supermarkets and household food acquisition patterns in Vietnam in relation to population demographics and socioeconomic strata: insights from public data

    Food environments in Southeast Asia's emerging economies are rapidly evolving, alongside fast-paced socioeconomic and demographic changes. The widespread expansion of supermarkets and parallel restructuring of traditional markets in Vietnam are likely to impact patterns of household food acquisition. Using provincial-level time series data on the abundance of supermarkets and multiyear household survey data, this paper examines the impact of the differential country-wide presence of supermarkets with indices of food quality and quantity acquired by households. We classified provinces into three clusters based on the number of supermarkets: high (HighSM), medium (MedSM), and low (LowSM). We found that a higher number of supermarkets associated with the exceedable Vietnamese recommendation composition of macronutrients at the household-level, but not food quantity.Households with higher per capita food expenditure in HighSM provinces tended to procure foods with higher protein content and lower shares of fat and carbohydrate as compared to similar households in the others provinces. Ethnic minority households in MedSM clusters obtained food with lower carbohydrate and higher fat:protein ratios in comparison to ethnic majority households. Additionally, larger-sized households in HighSM provinces typically bought foods with higher fat shares than smaller-sized households. In contrast, in MedSM and LowSM provinces, larger-sized households typically procured foods with higher protein and lower fat shares. The diversity of foods obtained by households in MedSM and LowSM provinces decreased over time. Within the Midlands and Northern Mountains Area we observed a decrease in the diversity of food acquired among households in the LowSM clusters. This study elucidates potential impacts of the supermarket expansion on household food baskets. Insights from this study can be used to provide evidences for policy recommendation and to design and target interventions aimed at strengthening food environments to address the challenge of the double burden of malnutrition in the country
