    Assessment wine aroma persistence by using an in vivo PTR-ToF-MS approach and its relationship with salivary parameters

    To better understand wine aroma persistence, the nasal cavity of nine volunteers was monitored by Proton Transfer Reaction-Time of Flight-Mass Spectrometry (PTR-ToF-MS) after they rinsed their mouths with three rose wines (one control and the same wine supplemented with two tannin extracts) during four minutes. Wines were aromatised with a mixture of five target aroma compounds. Results showed that wine aroma persistence was highly compound-dependent: while esters disappeared very fast, other compounds such as linalool remained in the oral cavity for longer times after wine expectoration. A low effect of tannins (at 50 mg/L) on nasal cavity parameters was observed, with the exception for the compound ethyl decanoate that was significantly higher released in the presence of tannins. Strong interindividual differences on aroma persistence were also found. Significant positive correlations with the salivary total protein content and negative with the salivary flow were observed for specific compounds. This work has studied for the first time in vivo wine aroma persistence in real time from an analytical perspective

    Early impairments in the retina of rats fed with high fructose/high fat diet are associated with glucose metabolism deregulation but not dyslipidaemia

    Way of life changes such as high consumption of processed foods rich in fat and sugar and sedentary lifestyle are associated with the increasing prevalence of metabolic syndrome (MetS) that affects about 35% in the American population. MetS is the main risk factor for diabetes mellitus, which is associated with vascular changes in the retina. However, the early consequences of MetS in the retina are not well described. We therefore aimed at characterizing the early effects of a high fructose and high fat diet (HFHF) on the function and structure of the rat retina, and evaluate the associations with metabolic changes. Brown Norway rats of 6 weeks of age were fed for 8 days, 5 weeks or 13 weeks with HFHF diet, or a standard chow. After only 4 weeks of this diet, rats exhibited a reduction in cone photoreceptor sensitivity to light. Moreover, we observed that MetS significantly exacerbated laser-induced choroidal neovascularization by 72% and 67% 2 weeks and 3 weeks post laser treatment, respectively. These retinal abnormalities were associated with deregulation of glucose metabolism but not lipid metabolism. These data showed retinal modifications in HFHF-induced MetS in the rat, at very early stage of the disease

    Investigation of Miscanthus and sunflower stalk fiber-reinforced composites for insulation applications

    The development of materials based on renewable and low-cost resources is today’s crucial interest for civil engineering. This work intends to investigate two new vegetable fibers as composite reinforcement for insulation application in the building industry. Miscanthus and sunflower stalk have mainly been selected for their high fiber content, low water content after harvesting, and availability. These criteria lead to good structural properties and allow a reduction in production and transportation costs. This study aims at demonstrating the feasibility to produce cohesive panels from these lignocellulosic fibers and their related interesting mechanical and thermal properties according to various natural binders

    Potential of Sentinel-2 satellite images to monitor vine fields grown at a territorial scale

    Aim: The aim of this short note is to provide first insights into the ability of Sentinel-2 images to monitor vine growth across a whole season. It focuses on verifying the practical temporal resolution that can be reached with Sentinel-2 images, the main stages of Mediterranean vineyard development as well as potential relevant agronomic information that can be seen on the temporal vegetation curves arising from Sentinel-2 images. Methods and results: The study was carried out in 2017 in a production vineyard located in southern France, 2 km from the Mediterranean seashore. Sentinel-2 images acquired during the whole vine growing cycle were considered, i.e. between the 3(rd) of March 2017 and the 10(th) of October 2017. The images were used to compute the classical normalized difference vegetation index (NDVI). Time series of NDVI values were analyzed on four blocks chosen for exhibiting different features, e.g. age, missing plants, weeding practices. The practical time lag between two usable images was closer to 16 days than to the 10 theoretical days (with only one satellite available at the date of the experiment), i.e. near 60% of the theoretical one. Results show that it might be possible to identify i) the main steps of vine development (e.g. budburst, growth, trimming, growth stop and senescence), ii) weed management and inter-row management practices, and iii) possible reasons for significant inter-block differences in vegetative expression (e.g. young vines that have recently been planted, low-productive blocks affected by many missing vines). Conclusions: Although this experiment was conducted at a time when Sentinel-2b was not fully operational, results showed that a sufficient number of usable images was available to monitor vine development. The availability of two Sentinel satellites (2a and 2b) in upcoming seasons should increase the number of usable images and the temporal resolution of the time series. This study also showed the limitations of the Sentinel-2 images' resolution to provide within-block information in the case of small blocks or blocks with complex borders or both. Significance and impact of the study: This technical note demonstrated the potential of Sentinel-2 images to characterize vineyard blocks' vigor and to monitor winegrowers' practices at a territorial (regional) scale. The impact of management operations such as weeding and trimming, along with their incidence on canopy size, were observed on the NDVI time series. Some relevant parameters (slope, maximum values) may be derived from the NDVI time series, providing new insights into the monitoring of vineyards at a large scale. These results provided areas for further investigation, especially regarding the development of new indicators to characterize block-climate relationships

    Effects of a n-3 polyunsaturated fatty acid-enriched diet on embryo production in dairy cows

    Beneficial effects of n-3 polyunsaturated fatty acid (PUFA) supplementation on dairy cow reproduction have been previously reported. The objectives of the present study were to assess whether n-3 PUFA supplementation would affect in vitro embryo production (IVP) after ovarian stimulation. Holstein cows received a diet with 1% dry matter supplementation of either n-3 PUFA (n = 18, micro encapsulated fish oil) or a control, n-6 PUFA (n = 19, micro encapsulated soy oil). Both plasma and follicular fluid FA composition showed integration of total PUFA through the diet. All cows underwent an IVP protocol consisting of ovarian stimulation, ultrasound-guided transvaginal oocyte retrieval (ovum pick-up, OPU, 5 per cow) followed by in vitro maturation, fertilisation and 7 days of embryo development. A tendency toward an increase in the blastocyst rate (diet effect, p = 0.0865) was observed in n-3 cows, with 49.6 ± 5.5%, versus 42.3 ± 5.5% in control n-6 cows. A significant increase (diet effect, p = 0.0217) in the good quality blastocyst rate (freezable blastocysts) was reported in n-3 cows (42.2 ± 7.7%) compared to control n-6 cows (32.7 ± 7.7%). A significant difference in lipid composition was shown in the oocytes recovered by OPU from n-3 and n-6 treated cows, by intact single-oocyte MALDI-TOF mass spectrometry. The 42 differentially abundant identified lipids were mainly involved in cell membrane structure. In conclusion, n-3 PUFA supplementation enhanced oocyte quality and modified their lipid composition. Further studies are necessary to investigate the potential link of these lipid modifications with enhanced oocyte quality

    Chouioia cunea Yang (Hymenoptera: Eulophidae) has been widely used for biological control of the fall webworm, Hyphantria cunea (Drury) (Lepidoptera: Arctiidae), in China. The yellow mealworm, Tenebrio molitor L. (Coleoptera: Tenebrionidae), an important resource insect species distributed worldwide, is considered to be a potential alternative host for mass rearing of C. cunea to the Chinese oak silkworm, Antheraea pernyi (Guerin-Meneville) (Lepidoptera: Saturniidae), which is currently used. In this study, we investigated the effects of host age on C. cunea mass rearing by measuring parasitism, development and adult fertility of C. cunea on T. molitor pupae of different ages. The results showed no significant differences in the percentage of parasitized hosts and developmental time of C. cunea in pupae of different ages. However, the number of C. cunea adults (137.2-154.7 adults per host) that emerged from 0, 1, and 2-day-old pupae was significantly higher than that from 4-day-old pupae. The lowest percentages of unemerged adults were found in 2-day-old (1.2%) and 3-day-old (1.4%) pupae, which were significantly lower than that of 4-day-old pupae (10.3%). The emergence of adult females from 0 to 2-day-old pupae (120.2-142.3 per pupa) was significantly higher than that from 4-day-old hosts (64.6). Adult females emerging from 2-day-old pupae carried significantly more eggs (258.2 eggs/female) than those from 0 and 1-day-old pupae (178.4-178.9 eggs/female). Our findings indicated that 2-day-old pupae of T. molitor were most suitable to rear C. cunea. Overall, this research provided valuable information to optimize pupae for the mass rearing of C. cunea on host T. molitor

    A DNA vaccine encoding the Gn ectodomain of Rift Valley Fever virus protects mice via a humoral response decreased by DEC205 targeting

    The Rift Valley fever virus (RVFV) is responsible for a serious mosquito-borne viral disease in humans and ruminants. The development of a new and safer vaccine is urgently needed due to the risk of introduction of this arbovirus into RVFV-free continents. We recently showed that a DNA vaccine encoding eGn, the ectodomain of the RVFV Gn glycoprotein, conferred a substantial protection in the sheep natural host and that the anti-eGn IgG levels correlated to protection. Addressing eGn to DEC205 reduced the protective efficacy while decreasing the antibody and increasing the IFNγ T cell responses in sheep. In order to get further insight into the involved mechanisms, we evaluated our eGn-encoding DNA vaccine strategy in the reference mouse species. A DNA vaccine encoding eGn induced full clinical protection in mice and the passive transfer of immune serum was protective. This further supports that antibodies, although non-neutralizing in vitro, are instrumental in the protection against RVFV. Addressing eGn to DEC205 was also detrimental to protection in mice, and in this species, both the antibody and the IFNγ T cell responses were strongly decreased. Conversely when using a plasmid encoding a different antigen, i.e., mCherry, DEC205 targeting promoted the antibody response. Altogether our results show that the outcome of targeting antigens to DEC205 depends on the species and on the fused antigen and is not favorable in the case of eGn. In addition, we bring evidences that eGn in itself is a pertinent antigen to be included in a DNA vaccine and that next developments should aim at promoting the anti-eGn antibody response
