Association of factor expression levels with annual bleeding rate in people with haemophilia B
Introduction: Gene therapy clinical trials measure steady‐state clotting factor expression levels (FELs) to evaluate the modulation of the bleeding phenotype, aiming to offer consistent protection against breakthrough bleeding events. The link between FELs and bleeding risk in people with haemophilia B (PwHB) is not well understood. Aim: We evaluated the association between FEL and ABR in PwHB. Methods: This cross‐sectional study extended the CHESS burden of illness studies in Europe and the United States. Recruitment of additional adult males with haemophilia B supplemented the existing CHESS sample size of PwHB and FELs. PwHB receiving prophylaxis were excluded, as fluctuating FELs may have confounded the analysis. Demographic and clinical characteristics were reported descriptively. Any recorded baseline FEL was reported by the haemophilia‐treating physicians according to the medical records. Generalised linear models with log link explored the association between changes in FEL and ABR. Results: The study included 407 PwHB and no inhibitors receiving on‐demand treatment. Mean age was 36.7 years; 56% from the EU, 44% from the United States. Mean baseline FEL was 9.95 IU/dl (SD, 10.47); mean ABR was 2.4 bleeds/year (SD, 2.64). After adjusting for covariates, the model showed that for every 1% increase in FEL the average ABR decreased by .08 (p < .001). Predicted number of bleeding events according to FEL showed a significant non‐linear relationship between FEL and ABR (p < .05). Conclusion: This analysis showed a significant relationship between FEL and ABR, where increases in FEL were associated with decreases in ABR among men with HB in Europe and the US
Finite difference method for time-fractional Klein-Gordon equation on an unbounded domain using artificial boundary conditions
A finite difference method for time-fractional Klein-Gordon equation with the fractional order on an unbounded domain is studied. The artificial boundary conditions involving the generalized Caputo derivative are derived using the Laplace transform technique. Stability and error estimates of the proposed finite difference scheme are proved in detail by using the discrete energy method. Numerical examples show that the artificial boundary method is a robust and efficient method for solving the time-fractional Klein-Gordon equation on an unbounded domain
A Novel Double-Threshold Neural Classifier for Non-Linearly Separable Applications

When real-time operation is desired, hardware solutions tend to be more amenable as compared to algorithmic/heuristic solutions. This paper presents a novel current-mode dual-threshold neuron designed and implemented at 32nm CMOS technology node. Subsequently, a current-mode double-threshold classifier is presented which is capable of classifying input patterns of non-linearly separable problems. Thereafter, application of the current-mode dual-threshold neuron in the realization of the XOR function using only a single neural unit is discussed. The proposed neuron as well as both the applications discussed are capable of operating from sub-1V power supplies. Computer simulations using HSPICE yield promising results with the values of delay and power consumption estimated to be lower than existing circuits
Screening for Mental Illness using GMHAT App of patients with Type 2 Diabetes Mellitus at a tertiary care hospital in India – A cross sectional study
People with diabetes will have some coexisting mental illness most of the time and its evaluation and management are essential for the well‑being of the person. With this background, the Global Mental Health Assessment Tool (GMHAT) app was used to screen for mental illness among type 2 diabetes mellitus patients in Mysore, India. Methods: A cross‑sectional study was conducted on 100 patients who were on treatment for type 2 diabetes for at least 6 months. Interviews were conducted using a structured GMHAT. Results: The mean age of the study participants was 48.8 ± 11.6 years. Among the 100 diabetic patients, 39% (n = 39) had mental illness and among the 39 subjects, 21% had depression, 14% had anxiety disorder and 4% suffered from organic disorder. Apart from diabetes, 29, 17, 13, 27, 23 and 20% had hypertension, chronic kidney disease, liver disease, retinopathy, neuropathy and nephropathy, respectively, as comorbid conditions. Conclusion: There is a higher prevalence of mental illness in patients with diabetes mellitus. The prevalence rate increases with an increase in the chronicity of diabetes. The use of the GMHAT app will help in rapid assessment and accurate diagnose
Core competencies in applied infectious disease epidemiology: a framework for countries in Europe
In 2009, the European Centre for Disease Prevention and Control (ECDC) developed a competency framework to support European Union countries and the European Commission in ensuring a competent public health workforce for Europe. The coronavirus disease (COVID-19) pandemic emphasised the importance of harmonised public health strategies and competencies across international boundaries, specifically for infectious diseases. This perspective presents the process to update the competency framework for applied infectious disease epidemiology, highlighting ECDC's efforts to support countries with using the framework. ECDC commissioned the Association of Schools of Public Health in the European Region (ASPHER) to update the framework through publication and dissemination of a technical report and a self-assessment tool linked to training resources. A mixed methods approach to gather input from experts in relevant specialities included qualitative interviews with 42 experts, workshops with ECDC Technical Advisory Group and an online survey of 212 public health professionals across Europe and beyond. Modifications resulted in 157 core competencies in 23 domains, each mapping to one of six subject areas of importance in applied infectious disease epidemiology. The framework serves as a basis to update the curriculum of the ECDC Fellowship programme with two alternative paths: intervention epidemiology or public health microbiology

We argue that geography as a discipline is particularly well placed to contribute to theoretical and practical framings of ‘green recovery’ as manifested at and through a range of scales, including the micro (individual), meso (household) and what we term ‘meso+’ (neighbourhood). Specifically, we signpost what might be considered ‘green shoots’ worthy of urgent empirical investigation – shifts in everyday life and practice catalysed by COVID-19 and with the potential to underpin longer-lasting transformations towards socially, economically and environmentally sustainable localities
A unique ternary Ce(III)-quercetin-phenanthroline assembly with antioxidant and anti-inflammatory properties
Quercetin is one of the most bioactive and common dietary flavonoids, with a significant repertoire of biological and pharmacological properties. The biological activity of quercetin, however, is influenced by its limited solubility and bioavailability. Driven by the need to enhance quercetin bioavailability and bioactivity through metal ion complexation, synthetic efforts led to a unique ternary Ce(III)-quercetin-(1,10-phenanthroline) (1) compound. Physicochemical characterization (elemental analysis, FT-IR, Thermogravimetric analysis (TGA), UV–Visible, NMR, Electron Spray Ionization-Mass Spectrometry (ESI-MS), Fluorescence, X-rays) revealed its solid-state and solution properties, with significant information emanating from the coordination sphere composition of Ce(III). The experimental data justified further entry of 1 in biological studies involving toxicity, (Reactive Oxygen Species, ROS)-suppressing potential, cell metabolism inhibition in Saccharomyces cerevisiae (S. cerevisiae) cultures, and plasmid DNA degradation. DFT calculations revealed its electronic structure profile, with in silico studies showing binding to DNA, DNA gyrase, and glutathione S-transferase, thus providing useful complementary insight into the elucidation of the mechanism of action of 1 at the molecular level and interpretation of its bio-activity. Previous studies have been based on a small number of samples and have not included samples from Ireland or the Netherlands. We utilized mtDNA control region sequencing of individuals stranded around Ireland, Scotland, and the Netherlands between 1995-2019 (n=180) to assess regional diversity. Following alignment of 631bp, nine haplotypes were identified, of which five were newly described (haplotype diversity (h)=0.511). Pairwise tests revealed significant differentiation between the Irish and Scottish samples. Potential confounding factors are discussed but, given the dangers of failing to recognize population structure this needs to be investigated further using nuclear markers and individual-based approaches before population isolation is assumed. The analyzed samples included six mass stranding events. One of the mass strandings reported two haplotypes, confirming mixing of matrilines within a North Atlantic stranding for the first time. This study shows that stranding sample databases are a useful tool for assessment of genetic diversity and provides new insights into genetic diversity of long-finned pilot whale haplotypes in the eastern North Atlantic and adjacent waters
