An increase in the intake of legumes is recommended in the promotion of plant-sourced (PSP) rather than animal-sourced (ASP) protein intake to produce a more sustainable diet. This study evaluated the quality of novel PSP isolates from pea (PEA) and fava bean (FAVA) and an ASP isolate of whey (WHEY) and compared the magnitude and temporal pattern of peripheral arterial aminoacidemia following ingestion of 0.33 g·kg−1 body mass of protein isolate in healthy young adult men (n = 9). Total indispensable amino acids (IAA) comprised 58% (WHEY), 46% (PEA), and 42% (FAVA) of the total amino acid (AA) composition, with the ingested protein providing 108% (WHEY), 77% (PEA), and 67% (FAVA) of the recommended per diem requirement of IAA. Reflecting the AA composition, the area under the curve (∆AUC0-180), post-ingestion increase in total IAA for WHEY was 41% (p < 0.001) and 57% (p < 0.001) greater than PEA and FAVA, respectively, with PEA exceeding FAVA by 28% (p = 0.003). As a sole-source, single-dose meal-size serving, the lower total IAA for PEA and FAVA would likely evoke a reduced post-prandial anabolic capacity compared to WHEY. Incorporated into a food matrix, the promotion of PSP isolates contributes to a more sustainable diet.Funder: Enterprise Ireland, Innovation Partnership; FundRef: https://doi.org/10.13039/10.13039/501100001588; Grant(s): IP/2019/087

Predicted Structure and Potential Binding of the Nematode Tetraspanin 7 Protein (tsp-7) With Human Tissue Inhibitor Metalloprotease 1 (TIMP-1): a Possible Role In Parasite- Host Interactions

    Get PDF
Human parasitic nematodes evade the host immune system through cross communication by unknown mechanisms. The proteins of the parasitic nematodes are still poorly annotated. CD63 is a human tetraspanin and binds to TIMP-1 a metalloproteinase inhibitor, which has recently been found to play an active role in inflammation by acting as a cytokine when bound to CD63. Caenorhabditis elegans is a free-living nematode, a model organism for both human and parasitic nematodes. C. elegans tetraspanin protein -7 (tsp-7) is an orthologue of human CD63. The model organism used can provide information about potential tetraspanin proteins from parasitic nematodes, which are believed to play a role in the cross communication between parasite and host. The tsp-7 protein sequence from C. elegans shares a high percentage similarity, identity, and homology with a variety of human parasitic nematode proteins. Using a number of computer-based programmes including ‘Alpha-Fold,’ which is currently the most powerful Artificial Intelligence protein structure prediction programme, the structure of the C. elegans tsp-7 was determined. From the predicted structure, the tsp-7 protein has one highly likely, membrane exposed tyrosine phosphorylation site, with implications of its potential involvement in signalling pathways and immune responses. Using multiple protein docking databases and programmes, tsp-7 has been shown to theoretically bind to human TIMP-1. Two mutant strains of C. elegans were obtained from Caenorhabditis Genetics Centre, University of Minnesota, USA, with amino acid deletions within the tsp-7 protein (mutant tm5046 and mutant tm5761). These tsp-7 mutant strains were shown to have an overall longer life span than the wild type C. elegans (N2). The mutant tm5761 showed a greater stress response compared to the N2 wild-type, when exposed to various chemical stimuli, however, mutant tm5046 was more similar to wild type C. elegans. Under heat stress, mutant tm5761 and wild type N2, were less tolerant to heat compared to the tm5046 mutant. In general, mutant tm5046 was more stable and displayed a reduced stress behavioural response compared to mutant tm5761. A primary polyclonal antibody was produced against the tsp-7 large extracellular loop domain, believed to be the key active site in protein-protein interactions. The expression of tsp-7 in mutant tm5761 was detected at lower concentrations than the wild type and mutant tm5046 in an ELISA assay and it also could not be detected by western blotting. Mutant tm5046 was detected weakly by western blotting. Cos-7 cells transfected with the tsp-7 protein-GFP tagged plasmid were incubated with active human TIMP-1. The tsp-7 tagged cells showed co-localisation with TIMP-1, using immuno-fluorescence microscopy, thus adding to the evidence that tsp-7 could interact with human TIMP-1. Such an interaction may well play a role in parasite-host interactions and its effects on the immune response. As C. elegans tsp-7 shares high similarity with many parasitic nematodes that are known to infect humans, it is a good protein candidate to further explore the potential interactions of parasitic nematodes with humans, in terms of cross communication and interaction with the host immune system

An exploratory study of how emotion tone presented in a message influences Artificial Intelligence (AI) powered recommendation system

This innovative study aims to explore how emotion tone presented in a message influences the judgement of Artificial Intelligence (AI) bots. We established a scenario by using vacation rental introduction as input message to conduct experiments to explore the influences. Our findings suggest that AI bots demonstrate preferences on the input message presented in positive tones rather than input message with negative tones. Our pioneering study can serve as a crucial starting point for future studies, in particular opening up fresh avenues for future research endeavours and engenders discussions and debates concerning the development of recommendation system

Data was collected using an online survey questionnaire from a voluntary response and snowball sample of 230 participants. Thematic, descriptive statistical, and inferential statistical analyses were undertaken. We identified 878 interventions that could be undertaken by ID nurses from 7 countries. We categorised the interventions into five themes: effectuating nursing procedures, enhancing impact of ID services, enhancing impact of mainstream services, enhancing quality of life, and enhancing ID nursing practice. Findings demonstrate that ID nurses play important roles in improving the health and healthcare experiences of people with IDs.AAM uploaded to the harvested data record 21/12/202

    Feminist Theologies: A Companion

    Get PDF
According to feminist liberation theologian Marcella Althaus-Reid, if there is one thing that feminist theologies can claim in common it is that ‘the body always takes charge: our armpits come first’, more specifically our ‘bushy’ armpits. No doubt the choice of armpits here is deliberate by Althaus-Reid given the way this part of women’s bodies has been disparagingly associated with feminism and with the western feminist project of resisting restrictive beauty standards. To begin feminist theologies with women’s ‘bushy armpits’ is to begin with a confidence in women’s flesh and with a preparedness to confront and transgress social and religious norms that contain women’s bodies and mark them as disgusting. This chapter explores the ways in which Christian feminist theologies have approached the body, paying particular attention to feminist theological discussion around body image, especially concerning beauty, fatness and thinness. It first considers how feminist theologies have exposed, challenged and reclaimed aspects of Christian body theology before considering how feminist theologians have explored body image and women’s struggles for bodily integrity

Unconditionally stable and convergent difference scheme for superdiffusion with extrapolation

The version of record of this article, first published in [Journal of Scientific Computing], is available online at Publisher’s website: https://doi.org/10.1007/s10915-023-02395-zApproximating the Hadamard finite-part integral by the quadratic interpolation polynomials, we obtain a scheme for approximating the Riemann-Liouville fractional derivative of order α∈(1, 2) and the error is shown to have the asymptotic expansion (d3τ3-α+d4τ4-α+d5τ5-α+⋯)+(d2∗τ4+d3∗τ6+d4∗τ8+⋯) at any fixed time, where τ denotes the step size and dl, l=3, 4, ⋯ and dl∗, l=2, 3, ⋯ are some suitable constants. Applying the proposed scheme in temporal direction and the central difference scheme in spatial direction, a new finite difference method is developed for approximating the time fractional wave equation. The proposed method is unconditionally stable, convergent with order O(τ3-α), α∈(1, 2) and the error has the asymptotic expansion. Richardson extrapolation is applied to improve the accuracy of the numerical method. The convergence orders are O(τ4-α) and O(τ2(3-α)), α∈(1, 2), respectively, after first two extrapolations. Numerical examples are presented to show that the numerical results are consistent with the theoretical findings

© 2023 The Author(s). Published by Informa UK Limited, trading as Taylor & Francis Group. The aim of this study was to assess and compare menstrual health literacy in professional women’s football. A three-section questionnaire was completed by professional players (n = 25), development players (n = 22) and staff (n = 19). The mean total knowledge score (out of 19) was lower for development players (5.4 ± 2.9) than professional players (7.8 ± 3.2) and staff (9.1 ± 4.8) (p 50% correct answers. For each group, knowledge of the menstrual cycle (MC) was greater than knowledge of hormonal contraceptives (HC) (p < 0.001). Previous MC and HC education did not correspond to higher knowledge scores in professional players (p = 0.823) or development players (p = 0.274). In professional and development players, comfort of communication was influenced by the sex of whom they were communicating with (p < 0.001), with a preference for females. In conclusion, results from the present study suggest refined education strategies and new approaches are required for both players and staff to improve menstrual health literacy in professional women’s football.VoR uploaded and AAM archived 14/12/202

The Accepted Manuscript is of an article published by Taylor & Francis in Person-Centered & Experiential Psychotherapies on 22/12/2023, available online: https://doi.org/10.1080/14779757.2023.2297384Little or no research has been conducted to explore the experiences of counselors who have a diagnosis of Attention-deficit/hyperactivity disorder (ADHD). The purpose of this research, therefore, was to explore and better understand the challenges and opportunities that a diagnosis of ADHD can present for these practitioners in order to inform counseling practice, supervision and training. This qualitative study utilized semi-structured interviews to obtain data from three participants, who were all person-centered counselors with ADHD. This data was then analyzed using Interpretive Phenomenological Analysis, which revealed five Group Experiential Themes (GETs). Each of these themes helped to illustrate both the challenges that participants faced as a result of their ADHD, such as inattention, impulsive disclosures, and emotional dysregulation, as well as the opportunities that it presented such as heightened curiosity and greater unconditional positive regard. This research addressed a gap within the literature and serves to enhance understanding of the ways in which a counselor’s diagnosis of ADHD can impact person-centered practice. It is hoped that this understanding can encourage greater support and acceptance of counselors with ADHD within the profession
