An Immunohistochemical Study of the Cytoskeletal Proteins in the Testis of the Sunda porcupine (Hystrix javanica)
application/pdfIn the testis of the Sunda porcupine (Hystrix javanica), the distribution pattern of cytoskeletal proteins was immunohistochemically examined to understand their functional roles in the testis. Immunoreactivity for α-smooth muscle actin (SMA) was found in the peritubular myoid cells of the seminiferous tubules and in the sub-tubular myoid cells of epididymal ducts. In intermediate filaments, desmin was exhibited in the sub-tubular myoid cells of the epididymal ducts, and vimentin immunoreactivity was detected in the Sertoli cells, Leydig cells, peritubular myoid cells, and sub-tubular myoid cells of the epididymal ducts. However, cytokeratin was not expressed in the testes or epididymides. Strong immunoreactivity for vimentin was demonstrated between the infranuclear part of the elevated nuclei and the basal membrane in the Sertoli cells. Furthermore, vimentin was extended from supranuclear to apical parts in some Sertoli cells. Immunoreactivity for α-tubulin was detected only in the Sertoli cells. It is concluded that, in the porcupine testes, desmin plays no role in the contractile function of peritubular myoid cells, vimentin may contribute to the anchoring and transition of the elevated nuclei in the Sertoli cells, and α-tubulin and vimentin in the Sertoli cells are likely involved in their structure and function as the main cytoskeleton. © The Mammal Society of Japan.journal articl
大規模酪農場,中小規模酪農場および肉牛飼養農場において呼吸器症状を呈する子牛の鼻スワブから分離された Pasteurella multocida、Mannheimia haemolytica および Mycoplasma 属の抗菌剤に対する薬剤感受性
application/pdf子牛の呼吸器病は酪農場や肉牛飼養農場で問題となっている.呼吸器病の治療では,抗菌剤の投与が行われるが,抗菌剤の選択は獣医師の主観的な判断に基づくことが多い.そのため,子牛の飼養目的や農場の規模により,抗菌剤の使用方針,使用頻度あるいは菌の薬剤感受性は異なることが推察される.本研究では, l診療所が管轄する地域の酪農場と肉牛飼養農場において,臨床的に呼吸器病の症状を呈した子牛から鼻腔スワブを採取し呼吸器病の主要な原因菌であるPasteurellamultocida,Mannheimia haemolyticaおよびMycoplasma属を分離して,抗菌剤13種類(ペニシリンG:PCG,アンピシリン:ABPC,セファゾリン:CEZ,セフロキシム:CXM,オキシテトラサイクリン:OTC,カナマイシン:KM,スルファメトキサザール・トリメトプリム合剤:ST,タイロシン:TS, フロルフェニコール: FF,マルボフロキサシン:MBFX, ダノフロキサシン:DNFX,オルピフロキサシン: OBFXおよびエンロフロキサシンERFX)に対する薬剤感受性をディスク拡散法により調べた.P. multocidaは全体の50.9% (114/224), M. haemolyticaは17.0% (38/224), Mycoplasma属は68.3%(153/224)の鼻腔スワブから検出された.子牛の飼養目的別に比較すると,菌によっては肉牛飼養農場に比べて酪農場で、耐性菌の検出率が有意に高い抗菌剤が認められた(P.multocidaに対するPCG,ABPC, KM. STおよびMycoplasma属に対するFF).また,酪農場の飼養規模別に比較すると,中小規模酪農場に比べて大規模酪農場で耐性菌の検出率が有意に高い抗菌剤が認められた(P. multocidaに対するOTC, M. haemolyticaに対するKM, STおよびMycoplasma属に対するOTC, KM, FF,MBFX, DNFX, OBFX. ERFX). 一方,大規模酪農場に比べて中小規模酪農場で耐性菌の検出率が高い抗菌剤も認められた(P.multocidaに対するKM, ST, FF). 一般的に,耐性菌の出現率と抗菌剤の使用頻度の聞には相関関係があり,呼吸器病に対して抗菌剤を使用する場合は, 牛群の薬剤感受性を調べ,抗菌剤の使用方針を定めた上で,定期的に再評価する必要があるjournal articl
Molecular prevalence of Theileria in sheep in Jilin province, China
application/pdfThe aim of this study was to enhance our understanding of ovine theileriosis and its timely prevention in sheep. We therefore conducted an epidemiological study in four areas in Jilin province, China. We extracted DNA from 95 blood samples from sheep and analyzed them by nested polymerase chain reaction targeting the 18S RNA gene of ovine Theileria spp. Positive samples were further analyzed by PCR using species-specific primer sets for T. luwenshuni, T. uilenbergi, and T. ovis, respectively, to detect and differentiate among these three Theileria spp. Phylogenetic analysis of positive samples based on 18S rRNA gene sequences of Theileria spp. was also conducted. Theileria spp. was prevalent in all four investigated areas, with a positivity rate of 18.9%. T. luwenshuni was universally prevalent with a positivity rate of 11.6%, whereas no T. uilenbergi or T. ovis infections were detected in these regions. Phylogenetic analysis showed that the positive samples in all four regions belonged to the T. luwenshuni cluster. This survey confirmed the epidemiology of ovine theileriosis and provided important data to support the prevention and control of this epidemic disease in northeastern China.journal articl
Milk Processing System in Rwanda
application/pdfThe purpose of this paper is to 1)understand milk processing system in Rwanda through a ˆeld survey, and 2)identifycharacteristics of milk processing techniques in Rwanda. The milk processing system in Rwanda used the no heating/lactic acid fermentation from the fermentation processes, in which raw milk is used without pasteurizing by heating, andnaturally fermented by the naturally mixing microbial ‰ora. The characteristics of milk processing technique in Rwandais that they adopted the milk fat separation and preservation techniques to make butter from raw milk, but they did notdevelop the milk protein separation and preservation techniques to make cheese. The year-round breeding and theconstant supply of raw milk seem to be the reason why agro-pastoralists did not need to fractionate and preserve milkprotein from raw milk and buttermilk. Since they do not make cheeses in Rwanda, the diet system in which they takenaturally fermented milk and buttermilk developed inevitably. The characteristics of milk fat separation and preserva-tion techniques in Rwanda is that the ˆnal form of milk fat is butter, because Rwanda is located in the highlands where itis relatively cold condition throughout the year. It was thought that the people of Rwanda regard butter on the basis of itsstrength of ‰avor and how much it can improve the taste when added to dishes rather than its shelf life. アポクリン導管癌の猫の1症例
application/pdf5歳4カ月齢の雑種猫、避妊雌が皮膚腫瘤を主訴に近医を受診し、切除手術を受けアポクリン導管癌と診断された。しかし早期に再発を認めたため、帯広畜産大学動物医療センターを紹介受診した。CT検査にて皮下への広範囲の浸潤が確認され、前進皮弁を用いた拡大切除と術後の放射線治療を行った。術後は一時閉眼障害がみられたが大きな問題となることはなく、術後7カ月で慢性腎臓病にて斃死するまで明らかな再発、転移は認められなかった。journal articl
Acute stimulation of a smooth muscle constrictor by oestradiol-17β via GPER1 in bovine oviducts
application/pdfOviducts play roles in reproductive processes, including gametes transport, fertilization and early embryo development. Oviductal transport is controlled by various factors such as endothelins (EDNs) and nitric oxide (NO), smooth muscle contracting and relaxing factor, respectively. EDNs and NO production depend on an ovarian steroid hormone, oestradiol-17β (E2) and E2 quickly exerts their biological functions through G protein-coupled oestrogen receptor 1 (GPER1), which mediates rapid intracellular signalling. Because follicular fluid which contains a high concentration of E2 enters the oviduct, we hypothesized that E2 in the follicular fluid participates via GPER1 in producing EDNs and NO. To test this hypothesis, we investigated 1) the expression and localization of GPER1 in bovine oviductal tissues and 2) rapid effects of E2 via GPER1 on EDN1, EDN2 and inducible NO synthase (iNOS) expression in cultured bovine oviductal isthmic epithelial cells. GPER1 was observed in the oviductal epithelium, stroma and smooth muscle, and its expression was highest in the isthmus. Short-term treatments (≤1 hr) of E2 increased EDN2 mRNA expression in the isthmic epithelial cells, although E2 did not affect EDN1 and iNOS mRNA expressions. Results of GPER1-selective agonist G-1 and GPER1-selective antagonist G-15 treatments revealed acute stimulation by E2, which is mediated via GPER1. The overall findings suggested that E2 in follicular fluid rapidly stimulates EDN2 expression via GPER1 in the isthmic epithelial cells. Follicular fluid may play a role in retention of the ovulated oocyte in the end of ampulla by contracting the isthmus for successful fertilization. © 2017 Blackwell Verlag GmbHjournal articl

Genetic analysis of babesia isolates from cattle with clinical babesiosis in Sri Lanka
application/pdfBovine babesiosis is a serious threat to the cattle industry. We preparedblood DNA samples from 13 cattle with clinical babesiosis from the Badulla (n 8),Jaffna (n 3), and Kilinochchi (n 2) districts in Sri Lanka. These DNA samplestested positive in PCR assays specific for Babesia bovis (n 9), Babesia bigemina(n 9), and Babesia ovata (n 1). Twelve cattle were positive for B. bovis and/or B.bigemina. One cow was negative for the tested Babesia species but was positive forBabesia on microscopic examination; the phylogenetic positions of 18S rRNA and cytochrome oxidase subunit III gene sequences suggested that the cow was infectedwith Babesia sp. Mymensingh, which was recently reported from a healthy cow inBangladesh. We then developed a novel Babesia sp. Mymensingh-specific PCR assayand obtained positive results for one other sample. Analysis of gene sequences fromthe cow with positive B. ovata-specific PCR results demonstrated that the animalwas infected not with B. ovata but with Babesia sp. Hue-1, which was recently reported from asymptomatic cattle in Vietnam. The virulence of Babesia sp. Hue-1 isunclear, as the cow was coinfected with B. bovis and B. bigemina. However, Babesiasp. Mymensingh probably causes severe clinical babesiosis, as it was the sole Babesiaspecies detected in a clinical case. The present study revealed the presence of twobovine Babesia species not previously reported in Sri Lanka, plus the first case of severe bovine babesiosis caused by a Babesia species other than B. bovis, B. bigemina,and Babesia divergens. © 2018 American Society for Microbiology.
