    Stiftelsen NILU har, i samarbeid med Transportanalyse AS, utarbeidet trafikk- og luftkvalitetsberegninger for Oslo og Bærum kommuner. Arbeidet omfatter en kartlegging av luftkvaliteten ved trafikkberegninger og utslipps- og spredningsberegninger for relevante forurensningskomponenter (PM10, PM2,5 og NO2) for Dagens situasjon 2022, Referansesituasjonen 2030 og for 2030 med tiltak. Det er beregnet risiko for overskridelse av dagens grenseverdier i forurensningsforskriften og for grenseverdier i revidert EU-direktiv som vil innføres fullt fra 2030.Stiftelsen NILU, in collaboration with Transportanalyse AS, has carried out traffic and air quality assessments for the municipalities of Oslo and Bærum. The work includes an evaluation of air quality based on traffic modeling and emission and dispersion calculations for relevant pollutants (PM 10 , PM2.5 , and NO2 ) for the today’s situation (2022), the reference scenario for 2030 and 2030 with implemented measures. The risk of exceeding current limit values in Norwegian regulation and the limit values in the revised EU directive, which will be fully implemented from 2030, has been calculated.publishedVersio

    Hazard assessment of nanomaterials: how to meet the requirements for (next generation) risk assessment

    Background Hazard and risk assessment of nanomaterials (NMs) face challenges due to, among others, the numerous existing nanoforms, discordant data and conflicting results found in the literature, and specific challenges in the application of strategies such as grouping and read-across, emphasizing the need for New Approach Methodologies (NAMs) to support Next Generation Risk Assessment (NGRA). Here these challenges are addressed in a study that couples physico-chemical characterization with in vitro investigations and in silico similarity analyses for nine nanoforms, having different chemical composition, sizes, aggregation states and shapes. For cytotoxicity assessment, three methods (Alamar Blue, Colony Forming Efficiency, and Electric Cell-Substrate Impedance Sensing) are applied in a cross-validation approach to support NAMs implementation into NGRA. Results The results highlight the role of physico-chemical properties in eliciting biological responses. Uptake studies reveal distinct cellular morphological changes. The cytotoxicity assessment shows varying responses among NMs, consistent among the three methods used, while only one nanoform gave a positive response in the genotoxicity assessment performed by comet assay. Conclusions The study highlights the potential of in silico models to effectively identify biologically active nanoforms based on their physico-chemical properties, reinforcing previous knowledge on the relevance of certain properties, such as aspect ratio. The potential of implementing in vitro methods into NGRA is underlined, cross-validating three cytotoxicity assessment methods, and showcasing their strength in terms of sensitivity and suitability for the testing of NMs.publishedVersio

    Air quality and transport behaviour: sensors, field, and survey data from Warsaw, Poland

    The present study describes the data sets produced in Warsaw, Poland with the aim of developing tools and methods for the implementation of human-centred and data-driven solutions to the enhancement of sustainable mobility transition. This study focuses on school commutes and alternatives to private cars for children drop off and pick up from primary schools. The dataset enables the complex analysis of interactions between determinants of transport mode choice, revealed choices, and air quality impact. We draw on four data collection methods, namely, (i) air quality and noise sensors’ measurements, (ii) in-person observations of transport behaviours, (iii) travel diaries, and (iv) social surveys. Moreover, all trip data from travel diaries are complemented with the calculated attributes of alternative travel modes. The data produced in the project can be also combined with publicly available information on air quality, public transport schedules, and traffic flows. The present data sets help to open new venues for interdisciplinary analyses of sustainable mobility transition effectiveness and efficiency.publishedVersio

    Machine Learning-Based Retrieval of Total Ozone Column Amount and Cloud Optical Depth from Irradiance Measurements

    A machine learning algorithm combined with measurements obtained by a NILU-UV irradiance meter enables the determination of total ozone column (TOC) amount and cloud optical depth (COD). In the New York City area, a NILU-UV instrument on the rooftop of a Stevens Institute of Technology building (40.74° N, −74.03° E) has been used to collect data for several years. Inspired by a previous study [Opt. Express 22, 19595 (2014)], this research presents an updated neural-network-based method for TOC and COD retrievals. This method provides reliable results under heavy cloud conditions, and a convenient algorithm for the simultaneous retrieval of TOC and COD values. The TOC values are presented for 2014–2023, and both were compared with results obtained using the look-up table (LUT) method and measurements by the Ozone Monitoring Instrument (OMI), deployed on NASA’s AURA satellite.     Extracellular Vesicles as Next-Generation Diagnostics and Advanced Therapy Medicinal Products

    Extracellular vesicles (EVs) hold great promise for clinical application as new diagnostic and therapeutic modalities. This paper describes major GMP-based upstream and downstream manufacturing processes for EV large-scale production, also focusing on post-processing technologies such as surface bioengineering and uploading studies to yield novel EV-based diagnostics and advanced therapy medicinal products. This paper also focuses on the quality, safety, and efficacy issues of the bioengineered EV drug candidates before first-in-human studies. Because clinical trials involving extracellular vesicles are on the global rise, this paper encompasses different clinical studies registered on clinical-trial register platforms, with varying levels of advancement, highlighting the growing interest in EV-related clinical programs. Navigating the regulatory affairs of EVs poses real challenges, and obtaining marketing authorization for EV-based medicines remains complex due to the lack of specific regulatory guidelines for such novel products. This paper discusses the state-of-the-art regulatory knowledge to date on EV-based diagnostics and medicinal products, highlighting further research and global regulatory needs for the safe and reliable implementation of bioengineered EVs as diagnostic and therapeutic tools in clinical settings. Post-marketing pharmacovigilance for EV-based medicinal products is also presented, mainly addressing such topics as risk assessment and risk management.publishedVersio

    Can plastic related chemicals be indicators of plastic ingestion in an Arctic seabird?

    For decades, the northern fulmar (Fulmarus glacialis) has been found to ingest and accumulate high loads of plastic due to its feeding ecology and digestive tract morphology. Plastic ingestion can lead to both physical and toxicological effects as ingested plastics can be a pathway for hazardous chemicals into seabirds' tissues. Many of these contaminants are ubiquitous in the environment and the contribution of plastic ingestion to the uptake of those contaminants in seabirds’ tissues is poorly known. In this study we aimed at quantifying several plastic-related chemicals (PRCs) -PBDE209, several dechloranes and several phthalate metabolites- and assessing their relationship with plastic burdens (both mass and number) to further investigate their potential use as proxies for plastic ingestion. Blood samples from fulmar fledglings and liver samples from both fledgling and non-fledgling fulmars were collected for PRC quantification. PBDE209 and dechloranes were quantified in 39 and 33 livers, respectively while phthalates were quantified in plasma. Plastic ingestion in these birds has been investigated previously and showed a higher prevalence in fledglings. PBDE209 was detected in 28.2 % of the liver samples. Dechlorane 602 was detected in all samples while Dechloranes 601 and 604 were not detected in any sample. Dechlorane 603 was detected in 11 individuals (33%). Phthalates were detected in one third of the analysed blood samples. Overall, no significant positive correlation was found between plastic burdens and PRC concentrations. However, a significant positive relationship between PBDE209 and plastic number was found in fledglings, although likely driven by one outlier. Our study shows the complexity of PRC exposure, the timeline of plastic ingestion and subsequent uptake of PRCs into the tissues in birds, the additional exposure of these chemicals via their prey, even in a species ingesting high loads of plastic.publishedVersio

    Revisiting the strategy for marine litter monitoring within the european marine strategy framework directive (MSFD)

    Marine litter and non-degradable plastic pollution is of global concern. Regular monitoring programs are being established to assess and understand the scale of this pollution. In Europe, the goal of the European Marine Strategy Framework Directive (MSFD) is to assess trends in Good Environmental Status and support large-scale actions at the regional level. Marine litter monitoring requires tailored sampling strategies, protocols and indicators, that align with specific objectives and are tailored for local or regional needs. In addition, the uneven spatial and temporal distributions of marine litter present a challenge when designing a statistically powerful monitoring program. In this paper, we critically review the existing marine litter monitoring programs in Europe. We discuss the main constraints, including environmental, logistical, scientific, and ethical factors. Additionally, we outline the critical gaps and shortcomings in monitoring MSFD beaches/shorelines, floating litter, seafloor litter, microplastics, and harm. Several priorities must be established to shape the future of monitoring within the MSFD. Recent developments in analytical approaches, including optimizing protocols and sampling strategies, gaining a better understanding of the spatiotemporal heterogeneity of litter and its implications for survey design and replication, and the inclusion of newly validated methodologies that have achieved sufficient technical readiness, must be considered. Although there are well-established methods for assessing beaches, floating and seafloor litter, it will be necessary to implement monitoring schemes for microplastics in sediments and invertebrates as robust analytical methods become available for targeting smaller particle size classes. Furthermore, the inclusion of indicators for entanglement and injury to marine organisms will have to be considered in the near future. Moreover, the following actions will enhance the effectiveness of monitoring efforts: (1) creating an inventory of accumulation areas and sources of specific types of litter (e.g., fishing gear), (2) monitoring riverine inputs of litter, (3) monitoring atmospheric inputs including microplastics, (4) accidental inputs during extreme weather events, and (5) studying how species at risk may be transported by litter. We provide recommendations to support long-term, effective, and well-coordinated marine litter monitoring within the MSFD to achieve a comprehensive and accurate understanding of marine litter in EU waters. This will allow the development of measures to mitigate the impacts of marine pollution and eventually to evaluate the success of the respective measures.publishedVersio

    The challenges of opportunistic sampling when comparing prevalence of plastics in diving seabirds: A multi-species example from Norway

    There is a need for baseline information about how much plastics are ingested by wildlife and potential negative consequences thereof. We analysed the frequency of occurrence (FO) of plastics >1 mm in the stomachs of five pursuit-diving seabird species collected opportunistically. Atlantic puffins (Fratercula arctica) found emaciated on beaches in SW Norway had the highest FO of plastics (58.8 %), followed by emaciated common guillemots (Uria aalge; 9.1 %) also found beached in either SW or SE Norway. No plastics were detected in razorbills (Alca torda), great cormorants (Phalacrocorax carbo), and European shags (Gulosus aristotelis) taken as bycatch in northern Norway. This is the first study to report on plastic ingestion of these five species in northern Europe, and it highlights both the usefulness and limitations of opportunistic sampling.     Holocene black carbon in New Zealand lake sediment records

    Black carbon emitted from incomplete combustion of biomass and fossil fuel burning is an important aerosol; however, available long-term black carbon data are limited to remote polar and high-alpine ice cores from few geographic regions. Black carbon records from lake sediments fill geographic gaps but such records are still scarce, particularly in the Southern Hemisphere. We applied a new incandescence-based methodology to develop Holocene refractory black carbon (rBC) records from four lake-sediment archives in New Zealand and compare these with macroscopic charcoal records. Our rBC records suggest periods with substantial rBC deposition during the Holocene before human arrival in the 13th century reflecting long-range transport and possibly local wetland fires. With Polynesian settlement, rBC deposition increased on the South Island in agreement with macroscopic charcoal records, and it is this period of burning that is proposed as the source of rBC increases evident in Antarctic ice cores. However, sites on the North Island show no contemporaneous rBC increase suggesting regional differences in biomass burning patterns between the North and South islands.     Surface-Bioengineered Extracellular Vesicles Seeking Molecular Biotargets in Lung Cancer Cells

    Personalized medicine is a new approach to modern oncology. Here, to facilitate the application of extracellular vesicles (EVs) derived from lung cancer cells as potent advanced therapy medicinal products in lung cancer, the EV membrane was functionalized with a specific ligand for targeting purposes. In this role, the most effective heptapeptide in binding to lung cancer cells (PTHTRWA) was used. The functionalization process of EV surface was performed through the C- or N-terminal end of the heptapeptide. To prove the activity of the EVs functionalized with PTHTRWA, both a model of lipid membrane mimicking normal and cancerous cell membranes as well as human adenocarcinomic alveolar basal epithelial cells (A549) and human normal bronchial epithelial cells (BEAS-2B) have been exposed to these bioconstructs. Magnetic resonance imaging (MRI) showed that the as-bioengineered PTHTRWA-EVs loaded with superparamagnetic iron oxide nanoparticle (SPIO) cargos reach the growing tumor when dosed intravenously in NUDE Balb/c mice bearing A549 cancer. Molecular dynamics (MD) in silico studies elucidated a high affinity of the synthesized peptide to the α5β1 integrin. Preclinical safety assays did not evidence any cytotoxic or genotoxic effects of the PTHTRWA-bioengineered EVs.publishedVersio
