Gli1+ Periodontal Mesenchymal Stem Cells Fate Change in Periodontitis With Diabetes
Diabetes and periodontitis are common chronic diseases. Diabetic patients have 3 times more risk of getting periodontitis, and periodontitis often causes more complications in diabetic patients. Periodontium tissue is very hard to repair in these two conditions. Periodontal mesenchymal stem cells (MSCs) play a crucial role in maintaining periodontium homeostasis and tissue repair. However, little is known about how periodontal MSCs in vivo respond in diabetic patients with periodontitis, posing a challenge for periodontium tissue regeneration. In this study, we used Gli1 as a periodontal MSC marker and combined a Gli1 cre ERT2 mouse model with lineage tracing techniques to investigate periodontal MSCs in vivo by inducing periodontitis in diabetic mice. Our findings show that the number of Gli1+ MSCs that contributed to PDL homeostasis decreased in periodontitis and diabetes. In addition, the number of Gli1+ MSCs undergoing osteogenic differentiation decreased during the progression of diabetic periodontitis. In a tooth extraction model, the contribution of Gli1+ MSCs to the tooth socket repair was significantly reduced in mice with periodontitis.
The blood vessels were characterized using both immunofluorescence staining method and PEGASOS tissue clearing-based 3D imaging technique. It was found that the number of blood vessels increased in the periodontium of teeth with periodontitis and diabetic periodontitis, but the integrity of the vasculature endothelium was disrupted. Bulk-RNA-seq was used to analyze gene expression and signaling pathway changes of Gli1+cells in samples from animals with periodontitis and diabetes. Inflammation-related gene expression was found to be upregulated while the expression of Wnt signaling-related genes was decreased. Collectively, these findings indicate that the function of Gli1+ MSCs in periodontitis and diabetes were compromised, including reduced contribution to periodontium homeostasis and impaired injury repair response. The relationship between Gli1+ MSCs and vasculature was markedly weakened during the process of diabetic periodontitis. In the future study, more emphasis will be put on the therapeutic method of increasing the number and function of periodontal MSCs in diabetic periodontitis and improving the perivascular niche of MSCs in periodontium for a better tissue regeneration result

Improving Affordability for Aggies: Recommendations for OER Course Marking at Texas A&M
Texas A&M University faculty have been working to improve the affordability of textbooks and other course materials for students. These efforts are part of larger campus initiatives to support student success. In order to maximize the impact of this textbook affordability, students have to know that courses with affordable course materials are available. They must have the ability to use this information to make informed choices during registration. To achieve this, the TAMU class scheduling system needs modifications that will enable faculty to effectively communicate to students about courses using affordable course materials
